Isolation and Evaluation of an Antiviral Producing Serratia spp. Strain Gsm01 against Cucumber mosaic virus in Korea (한국에서 CMV에 항바이러스 효과를 나타내는 Serratia spp. Gsm01 균주의 분리 동정 및 효과 검정)

  • An Antiviral producing bacterial strain was isolated from ginseng root environment in Hongcheon, Kangwon province of Republic of Korea. Identification of this bacterial strain was performed by physiological and biochemical tests along with 16S rRNA analyses. The results revealed that the bacterium was closer to genus Serratia, which was named as Gsm01. The strain was grown in Mannitol-Glutamate-Yeast (MGY) broth for 48 h. The culture was centrifuged and the filtrate obtained was tested for its ability to control Cucumber mosaic virus strain Y (CMV-Y) in greenhouse and field experiments. In the green house experiments, CF was evaluated for its ability to protect local host, Chenopodium amaranticolor and systemic host of CMV, Nicotiana tabacum cv. Xanthi-nc. It was found that, CF treatment reduced viral infection by 98% in local host; C. amaranticolor. The N. tabacum cv. Xanthi-nc plants treated with CF did not show visible viral symptoms 15 days post inoculation (dpi) and remained symptomless throughout the periods of the study. To evaluate effectiveness of CF under field conditions, experiment was carried out in a polyvinyl house. It was observed that, 52% plants were protected from viral diseases compared to non-treated plants, increasing the crop yield. This is the first report showing antiviral activity of a Serratia spp. against CMV.

Effects of Light-emitting Diodes on In Vitro Growth of Virus-free Sweet Potato Plantlets (LED가 고구마 바이러스 무병묘의 기내 생장에 미치는 영향)

  • The in vitro growth of virus-free sweet potato [Ipomoea batatas (L.) Lam.] plantlets was investigated under different light sources: fluorescent lamp (control); red (660 nm), blue (460 nm), white light-emitting diodes (LED), and two mixtures of blue and red LED (R:B = 8:2, and 7:3). Single node explants (10 mm) of three cultivars ('Matnami', 'Shincheonmi', and 'Yeonhwangmi') were cultured on Murashige and Skoog medium supplemented with $0.2mg{\cdot}L^{-1}$ 6-benzyladenine for 4 weeks. Explants were exposed to $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ photosynthetic photon flux at a distance of 20 cm, constant temperature of $25^{\circ}C$, and under 16/8-h (day/night) photoperiod. Using the same method, the in vitro growth of 10 cultivars under red LED was also compared. After 3 weeks, vine length was highest in plantlets cultured under red LED, and lowest in plantlets cultured under blue LED. Fresh and dry weights were also greatest in plantlets cultured under red LED. Compared to the control, vine thickness was significantly higher in plantlets grown under white LED and the 7:3 R:B LED mixture. Significant differences were observed among the 10 cultivars grown under red LED. 'Matnami', 'Shincheonmi', and 'Shinhwangmi' all had excellent vine lengths, and fresh and dry weights. Compared to the control, vine elongation of sweet potato plantlets was most effective under red LED, and culture duration was about 1 week shorter.

A Study on Hepatitis Infection Risk of Funeral director related to wearing PPE(Personal Protect Equipment) (한국 장례종사자의 개인보호물품 착용에 따른 A형 간염, B형 간염 위험도 및 로지스틱 분석)

  • We are investigated about the Hepatitis infection risk of the funeral director related to wearing PPE(Personal Protect Equipment) within Metropolitan, Chungcheong and Gyeongsang regions in Korea. We are classified that the vaccinated group was classified as low risk group and the non-vaccinated group was classified as high risk group. And we are analyzed the risk rate of infection based on whether or not to wear PPE(personal protective equipment) among high-risk groups. The result is as follows. The mask wearing rate of high-risk group about HAV(hepatitis A virus) is three times lower than that of low-risk group. The surgical glove wearing rate of high-risk group about HBV(hepatitis B virus) is twice lower than that of low-risk group. The surgical glove and mask not wearing rate among the high-risk group observe that potential infection risk was high 4.23 times and 3.5 times, respectively. We are concluded that increasing the risk of potential infection risks from the funeral director could result in increased risk of infection to national public health, including the bereaved family. We are suggested that the funeral director must be bound to vaccinate against hepatitis and make PPE mandatory. It is deemed necessary for the government to develop policies to promote personal health and national health care.

Effects of Cultural Conditions on the In Vitro Propagation of Plantlets derived from Apical Meristem in Ipomoea batatas L. (고구마 정단분열조직 유래 식물체의 기내 증식에 미치는 배양조건의 영향)

  • Sweet potato is a crop vegetatively propagated by vine cuttings, an ineffective method for maintaining pathogene-free stock plants. As an alternative method, single-node cultures of virus-free plantlets derived from apical meristem in sweet potato (cv. Yulmi) was examined. Effective pH range, sugar concentration and nodal order were investigated to establish an in vitro mass propagation system with high quality virus-free stock plantlets to farmhouse. Although the plantlets grew at wide range of pH, the most effective pH of the medium was 4.8 in single-node cultures. High sugar concentration of 60∼80 g/L resulted in increased growth response in shoot length, root length, number of node, leaf area and fresh and dry weight of shoot and root, whereas reducing sugar contents below 6% was showed reduced growth response. The first node including meristem tip was the best for the rapid growth of plantlets and the other nodes also showed a very similar growth response. Uniform plantlet can be obtained massively at the same time by culture of single node except for the first node including meristem tip. In conclusion, the most effective pH range and sugar concentration of medium for the growth of plantlets via single-node cultures was 4.8, 60∼80 g/L respectively. The first node was the best for the rapid propagation of plantlets in nodal order.

Effect of Glutathione on Lead Induced Modulation of NO Synthesis in RAW 264.7 Cell (RAW 264.7 Cell에서 납에 의한 NO 생성의 조절에 미치는 Glutathione의 효과)

  • Objectives : To evaluate the elect of glutathione(GSH) on lead induced modulation of nitric oxide(NO) synthesis, and to examine how lead modulates NO production in macrophages. Methods : This study was observed in a culture of RAW 264.7 cells, which originated from a tumor in a Balb/c mouse that was induced by the Abelson murine leukemia virus. The compounds investigated were lead chloride, N-acetyl-cystein(NAC), and Buthionine Sulfoximine( BSO). Results : ATP synthesis in RAW 264.7 cells was unchanged by each lead concentration exposure in a dose dependent manner. The NO synthesis was decreased when exposed to lead($PbCl_2$) concentration $0.5{\mu}M$. The presence of $300{\mu}M$ NAC, used as a pretreatment in the culture medium, caused the recovery of the lead induced decrease in NO synthesis, but in the presence of $300{\mu}M$ BSO as a pretreatment, there was no recoverey. Pretreatment with NAC and BSO had no affect on ATP synthesis at any of the lead concentrations used. Conclusions : These results indicated that GSH has a protective effect toward lead toxicity, and suggested that the inhibition of NO production in macrophage due to lead toxicity may be related to cofactors of iNOS (inducible nitric oxide synthase)

Prevalence of enteropathogens in the feces from diarrheic Korean native cattle in Gwangju area, Korea (광주지역 한우 분변 내 설사병 병원체 조사)

  • Calf diarrhea is a common disease in young claves and is still a major cause of productivity and economic loss in livestock farms. Fecal samples from Korean native cattle (n=100) with diarrhea from 64 farms in Gwangju area, Korea from september 2017 to December 2018 were examined for shedding of important protozoan parasitic, viral and bacterial pathogens using culture, rapid test kit and PCR methods. Of 57 (89.1%) of the 64 Korean native cattle farms examined had samples infected with at least one of the investigated pathogens. Among 100 fecal samples, 88 samples were positive for at least one the twelve pathogens and 51 samples were simultaneously positive for two or more pathogens by culture and PCR assay. Bovine group A rotavirus (BRV) was the most common pathogen, found in 43/100 (43.0%) samples on 32/64 (50.0%) farms. Subsequently, kobuvirus (30.0%), pathogenic E. coli (29.0%), bovine parvovirus (17.0%), Giardia spp. (13.0%), Eimeria spp. (10.0%), Clostridium perfringens type A (8.0%), bovine torovirus (8.0%), bovine viral diarrhea virus (6.0%), bovine coronavirus (5.0%), bovine norovirus (2.0%) and Cryptosporidium spp. (2.0%) were detected. Nebovirus, kırklareli virus, bovine adenovirus, Salmonella spp. and intestinal parasites were not detected. Of the 72 calves sampled in this age group, 64 (88.9%) samples were positive for at least one enteropathogen. BRV was identified in 34/72 (47.2%) samples from 27/48 (56.3%) farms. Subsequently, pathogenic E. coli (30.6%), kobuvirus (29.2%), BPaV (22.2%), Giardia spp. (15.3%), Eimeria spp. (9.7%), BVDV (6.9%), Cl. perfringens type A (6.9%), BCoV (4.6%) and Cryptosporidium spp. (2.8%) were detected in fecal samples. A total of ninety-six strains of E. coli were isolated from one hundred fecal samples collected from Korean native cattle with diarrhea. The presence of stx1, stx2, eaeA, LT, STa, STb, ehxA, saa, F4, F5(K99), F6, F17, F18 and F41 genes in the isolates was investigated by PCR. Out of ninety-six E. coli isolates screened for specific genes, 30 strains E. coli were identified to harbor shiga toxin-producing E. coli (STEC) 7 (7.3%), enterohemorrhagic E. coli (EHEC) 8 (8.3%), enteropathogenic E. coli (EPEC) 6 (6.3%), enterotoxigenic E. coli (ETEC) 2 (2.1%) and STEC/ETEC hybrid 7 (7.3%). This study provides epidemiological estimates of the prevalence of Korean native cattle's enteropathogens in Gwangju area, Korea, which would be used for cattle farmers and veterinarians to select appropriate therapeutic method.

On a Cleaning of COVID-19 Prevention Masks with Electrolytic Decomposition Water (전기분해수로 코로나방역용 마스크의 세정에 관한연구)

  • Various COVID-19 quarantine guidelines and measures are being taken by country at the WHO, but the number of confirmed cases has not decreased significantly. In order to prevent the inflow and outflow of COVID-19 through individual droplets, it is mandatory to wear a mask anytime, anywhere. However, as virus bacteria entering the mask amplify, it pollutes the mask and causes a disgusting smell. In this paper, a new method of preventing the spread of COVID-19 was proposed by sterilizing the mask with a dental gait spray introduced into the mask that has been used for a long time. Dental gargle water is usually produced by electrolysis of tap water, and the unstable ion water (HOCl) dissolved in water penetrates the cell barrier of various viruses and fails to act in its nucleus, causing water to self-purify. As a result of the experiment, when the mask used for a long time was washed with gargle water spray, the washed mask was dried after 10 minutes, and the smell of virus droplets or saliva almost disappeared. In particular, as a result of MOS testing the fit of the subjects who participated in the mask cleaning, it was excellent at 4.4 on average. Therefore, the mask was disposable, but if the spray was washed in the proposed method more than twice a day, the mask could be used in a comfortable state for more than a week.

Studies on isolation of rhinopneumonitis virus from Korean horses and its immunogenicity II. Studies on characters and immunogenicity of equine herpesvirus (국내 말로부터 비폐렴바이러스의 분리 및 면역원성에 관한 연구 II. 국내 분리 말 비폐렴바이러스의 특성 및 면역원성 조사)

  • The study was carried out to characterize the properties of Korean isolates of EHV from aborted fetuses and determine envelope protein profiles. The results obtained were summarized as follows; 1. Two strains of EHV was isolated from 2 liver samples among 10 aborted fetuses from which the virus isolation was attempted. 2. Morphological and some enzymatic properties of the Korean isolates of EHV which was designated as $LC_1$ and $LC_2$ was identical to those of a reference strain of Australia-N of EHV-1. The Korean isolates of EHV could be propagated on ED cell culture and they formed typical plaques 1 to 2 days after infection in the ED cells from which typical cuboidal particles of 150~170 nm diameter herpesvirus were observed. The virus could be detected specifically from neucleus and cytoplasm of infected cells by flourescent antibody technique using FITC labelled anti-Aust IV(EHV-1) antiserum. The Korean isolates, $LC_1$ and $LC_2$ were specifically neutralized by anti Aust IV antiserum and reacted positively to CELISA. 3. The structural polypeptides of purified enveloped virions of $LC_1$ and $LC_2$ isolates of EHV were determined by SDS-polyacrylamide gel electrophoresis to identify the envelope glycoproteins. $LC_1$ and $LC_2$ strains revealed 14 glycoproteins ranging in molecular weight from 190 kD to 31 kD while 17 structural proteins of Aust IV(EHV-1), of which 14 were identical to those of $LC_1$ and $LC_2$, were identified. Upon immunoblotting by rabbit antiserum against EHV isolates and EHV-1(Aust IV), 4 immunogenic proteins of $LC_1$ and $LC_2$ were 135 kD, 88 kD, 64 kD and 59 kD, of which 135 kD, 88 kD and 64 kD proteins were also found in Aust IV(EHV-1).

Experimental Infection for the Neutralization of White Spot Syndrome Virus (WSSV) in Wild Captured Sand Shrimp, Crangon affinis (자연산 자주새우(Crangon affinis)에서 흰반점바이러스(WSSV)의 neutralization을 위한 인위감염(experimental infection))

  • White spot syndrome virus (WSSV) is one of the most virulent viral agents threatening the penaeid shrimp culture industry. This study was carried out to evaluate the susceptibility of the sand shrimp, Crangon affinis, to WSSV as an alternative experimental model. WSSV caused 100% mortality in C. affinis within 7 days after experimental infection by immersion. Based on challenge studies, it was confirmed that C. affinis could be a potential host in WSSV transmission. Also, the neutralization of WSSV was carried out using an antiserum raised against recombinant envelop protein rVP466 to evaluate the WSSV infection mechanism. A constant amount of WSSV (at $1{\times}10^4$ diluted stocks) was incubated with various amounts of antiserum and then mixed to 20 l reservoir for the immersion challenge of C. affinis for neutralization. At 5 days post challenge, the shrimp in the positive control immersed in the immersion reservoir containing WSSV stock showed 100% mortality. The shrimps challenged with the 3 different mixtures of WSSV and rVP466 antiserum (1:0.1, 1:0.5 and 1:1) showed 100%, 68.8% and 68.8% mortality at 14 days post challenge, respectively. These results indicated that the antiserum raised against rVP466 could block WSSV infection in C. affinis. Therefore, this study confirmed that C. affinis can be naturally infected by WSSV as another potential host and that C. affinis can be used as an alternative experimental animal instead of penaeid shrimps.

Sequence Analysis of Rubella Viruses Isolated in Korea and Expression of Rubella Specific Gene (우리나라에서 분리된 풍진바이러스의 염기서열 분석 및 유전자 발현)

  • During the recent epidemic period ($1995{\sim}1996$), seven strains of rubella virus were isolated in Korea. To analyze phylogenetic relationship between seven Korean strains and rubella virus strains from other different geographical areas, structural genes (E1, E2 and C) of Korean strains were enzymatically amplified and automatically sequenced. The sequence similarities of the E1, E2 and C genes of the cosmopolitan types were $95.8{\sim}98.1%$, $92.6{\sim}99.2%$ and $96.4{\sim}99.3%$ based on 1,441, 122 and 139 nucleotides and $96.9{\sim}98.5%$, $90{\sim}100%$ and $97.8{\sim}100%$ based on 480, 40 and 46 amino acids compared to the sequences of strain RA27/3, respectively. In contrast, the sequence similarities of the E1, E2 and C genes of the Asian types were $91.5{\sim}92.1%$, $83.6{\sim}88.5%$ and 91.4% based on nucleotides and $96.9{\sim}97.7%$, 85.5% and 97.8% based on amino acids compared to the sequences of strain RA27/3, respectively. However, immunodominent epitopes of the E1 gene of the cosmopolitan and Asian types were well conserved, and the growth patterns in cell culture and immunofluorescent antibody titers in cross-reaction test showed no differences between two different types. In phylogenetic analysis based on nucleotide sequences of each gene regions, the cosmopolitan and Asian types formed two distinct phylogenetic lineages. These data showed two distinct genotypes of rubella viruses cocirculated in Korea, but no significant differences in the antigenicity of two different rubella virus strains were found.
