아시아에서 분리된 viral haemorrhagic septicaemia virus (VHSV) isolates의 계통분석학적 비교 (Phylogenetic Analysis of Viral Haemorrhagic Septicaemia Virus (VHSV) Isolates from Asia)

  • 바이러스성 출혈성 패혈증을 일으키는 VHSV는 국내의 넙치양식에 심각한 피해를 주고 있다. 본 연구에서는 우리나라에서 분리한 8종의 VHSV 분리주들의 glycoprotein (G)의 염기서열을 분석하여 Korean VHSV 분리주들과 기보고된 일본 및 중국의 분리주들을 계통발생학적으로 비교하여 G-protein의 8 부위(G34, C528, A755, T834, T951, T1147, T1221, T1336)에서 국내 VHSV 분리주들의 특이적인 서열을 확인하였고, 전 세계적으로 분리된 VHSV의 분리주들과의 비교결과 8 부위 중 3 부위 (A755, T834, T1221)의 국내VHSV 분리주 특이적 염기서열들을 확인하였다. 또한 중국 유래 VHSV 분리주의 특이적 염기서열 11 부위 (C498, C545, C548, C642, G648, T678, A738, C804, C834, G851, C1139)와 일본 유래 VHSV 분리주의 특이적 염기서열 1부위 (C179)를 확인하였다. 이상의 연구결과는 국내 특이적인 염기서열을 확보하여, 구역화된 수산생물 질병의 예찰, 모니터링 및 질병 관리에 유용하게 사용될 것으로 사료된다.

  • Fetal bovine serum (FBS) is essential for cell culture and is used in the determination of infectivity titer and propagation of viruses. To clarify the effects of FBS on the propagation of viral haemorrhagic septicaemia virus (VHSV), which is a causative agent of mass mortalities of olive flounder Paralichthys olivaceus in Korea, VHSV was inoculated into an EPC (epithelioma papulosum cyprinid) cell line supplemented with MEMs (minimal essential medium) with FBS concentrations of 0%, 2%, 5%, and 10% (MEM0, MEM2, MEM5, and MEM10), respectively, and infectivity titers were compared. Cytopathic effects were observed in all experimental groups at 2 days post virus inoculation (dpi) and all cells were detached from cell culture flasks at 7 dpi. Infectivity titers increased to 3 dpi, persisted to 7 dpi, and decreased when cells were detached. The titer of VHSV in EPC cells in MEM0 was the lowest while those in the other experimental groups showed similar levels. In conclusion, 2% (v/v) of FBS was sufficient to propagate VHSV in EPC cells and the withdrawal of VHSV from cell culture flasks should be performed before cell detachment.

바이러스성출혈성패혈증바이러스에 대한 식물 추출물의 항바이러스 효능 탐색 (Antiviral effects of various plant extracts against viral haemorrhagic septicaemia virus (VHSV))

  • Since viral haemorrhagic septicaemia virus (VHSV) was first reported in European rainbow trout (Oncorhynchus mykiss) in the 1930s, it has caused high prices in freshwater and saltwater fish around the world, causing enormous economic damage to the aquaculture industry. We have been seeking required countermeasures against viruses because of economic damage to the aquaculture industry. However, commercial vaccines have the limitations of being costly to use in farms and being effective to only one pathogen. The aquaculture industry these days is taking on new alternatives to vaccines, antibiotics and chemicals. In this study, the suitability of antiviral effects against VHSV was evaluated in vitro for various plant extracts to judge their effectiveness. Atriplex gmelinii, Ixeris repens, Arctium lappa, and Sargassum coreanum were tested to know the correlation between the amount of virus and the concentration of extract investigates if these extracts have antiviral effects. Virus and extracts at various concentrations were inoculated simultaneously as 1:1 ratio into EPC cell lines. There are no antiviral effects with Atriplex gmelinii, Ixeris repens and Arctium lappa. Extract of Sargassum coreanum only has the antiviral activity in a dose-dependent manner. These results show that extract of Sargassum coreanum can be used in aquaculture industry as an antiviral materials.

Genetic positioning of Korean viral hemorrhagic septicemia virus (VHSV) from cultured and wild marine fishes

  • Viral haemorrhagic septicaemia virus (VHSV) is an epidemic virus in olive flounder Paralichthys olivaceus farms in Korea, since the virus have first isolated in 2001. In the present study, partial glycoprotein (G) gene nucleotide sequences of seven Korean VHSV from cultured olive flounder and wild marine fishes in coastal areas of Korea were analyzed to evaluate their genetic relatedness to worldwide isolates. Phylogenetically, all Korean VHSV formed only one minor cluster including Japanese isolates, in genotype IVa, while the North America isolates formed a different minor cluster in genotype IVa. These results suggest that Korean VHSV could be an indigenous virus in Korean and Japanese coastal areas, but have not been introduced from North America.

2014년 제주도 양식 넙치, Paralichthys olivaceus에 대한 VHS 및 RSIVD 모니터링 (Monitoring of VHS and RSIVD in cultured Paralichthys olivaceus of Jeju in 2014)

  • The outbreak of viral diseases caused by viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) have been reported in cultured olive flounder, Paralichthys olivaceus. VHSV has been a serious viral disease that infects the olive flounders in South Korea. Clinical signs of VHSV infection are skin darkening, abdominal distension and haemorrhages. Outbreaks of fish iridoviral disease was first reported from red seabream, Pagrus major farms in Japan. Recently, iridovirus infection have occurred frequently from olive flounder farms in South Korea. In this study, disease surveillance was performed to monitor the prevalence of VHSV and RSIV in olive flounder in 2014. The samples were collected from 60 different olive flounder farms in Jeju from April, May, September, November and December in 2014. RT-PCR (VHSV) or PCR (RSIV) results showed that VHSV were detected in 5 farms, but RSIV has not been detected in any farms. The migration of olive flounder was restricted for the quarantine in 5 farms of VHS outbreak. The nucleocapsid protein (N) gene and glycoprotein (G) gene sequences of the 5 Korean VHSV isolates were successfully amplified and sequenced. Phylogenetic analysis was performed using the VHSV sequences reported here together comparison with the nucleotide sequences available from the GenBank database. Phylogenetic analysis indicated that most of Korea VHSV belong to the genotype IVa and closely related to the strains from Japan and China.

  • In this study, disease surveillance was performed to monitor the prevalence of viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) in olive flounder, Paralichthys olivaceus in 2015. The fish samples were collected in March (60 farms), May (55 farms) and July (52 farms) from different farms in Jeju. Reverse transcription polymerase chain reaction (RT-PCR) (VHSV) or PCR (RSIV) results showed that VHSV detected in 2 farms, but RSIV has not been detected in any farms. The sequences of the nucleocapsid protein (N) and glycoprotein (G) gene of the 2 VHSV isolates were successfully sequenced. Phylogenetic analysis was included VHSV isolates reported here together with a representative VHSV isolates available in GenBank. Phylogenetic analysis indicated that most of Korea VHSV isolates were closely related to the Japan and China genotype IVa which is clearly distinct from the North American genotype IVb.

  • Interferon regulatory factors (IRFs) are a family of transcription factors essential to the control of antiviral immune response, cell growth, differentiation and apoptosis. IRF10 of zebrafish (Danio rerio) was negative regulation of the interferonΦ1 and 3 response in vitro. In this study, we analyze the induction of in vivo immune response activation from the IRF10 gene of zebrafish and the protective effect against VHSV. As the results, the group inoculated with IRF10 expression vectors, there was no expression of IFNΦ1, suggestion that IRF10 may function as a negative regulator of IRF3, which binds to the IFNΦ1 promoter. And other types of interferon genes (IFNΦ2-4) are thought to have been activated, inducing to the expression of pro-inflammatory cytokine and Mx genes. As the results of challenge test performed at 14 days after inoculation of the expression vectors, the maximum survival rate [50% (1㎍ DNA) and 42.5% (10㎍ DNA)] for IRF10 group were recorded. Meanwhile, the survival rates of pcDNA3.1 and PBS as the control groups were 10% and 15%, respectively. This study suggests that the possibility that activation of IRF10 molecule could be exploited as a VHS control method.
