Improvement of indirect enzyme-linked immunosorbent assay for detection of Japanese encephalitis virus antibodies in swine sera

  • Japanese encephalitis (JE) is an important zoonosis caused by the mosquito-transmitted JE virus (JEV), which is a causative agent of reproductive failure in pregnant sows. Detection of JEV antibodies in swine is performed by hemagglutination inhibition (HI), virus neutralization (VN), and the plaque reduction neutralization test (PRNT). The most stringent PRNT is the 90% endpoint PRNT ($PRNT_{90}$). These conventional assays are difficult to carry out in diagnostic laboratories with insufficient instruments or cell culture systems. An alternative assay that is easily conducted and time efficient is required. In this study, we improved the indirect enzyme-linked immunosorbent assay (I-ELISA) with clarified antigen for the detection of JEV antibodies. The I-ELISA results obtained from 175 swine serum samples were compared with HI, VN, and $PRNT_{90}$ results. The sensitivity of I-ELISA was 91.8%, 95.0%, and 94.7% compared with HI, VN, and $PRNT_{90}$ results, respectively. The specificity of I-ELISA was 92.2%, 94.7%, and 94.7% compared with HI, VN, and $PRNT_{90}$ results, respectively. Moreover, the I-ELISA results were significantly correlated with the HI (r = 0.93), VN (r = 0.95), and $PRNT_{90}$ (r = 0.92) results. These results suggest that the improved I-ELISA is useful for serosurveillance of JEV in swine.

  • Influenza viruses are RNA viruses that belong to the Orthomyxoviridae family, and those can be divided into three types; A, B, and C, which based on the differences of the inner nucleoproteins and genomic structures. All three genera differ in their genomic structure and nucleoprotein content, they are further classified into various serotypes based on the two surface glycoproteins, hemagglutinin (HA) and neuraminidase (NA). These glycoproteins play crucial roles in viral infection and replication. Hemagglutinin mediates binding of virions to sialic acid receptors on the surfaces of target cells at the initial stage of infection. Neuraminidase cleaves the glycosidic bonds of sialic acids from the viral and cell surfaces to release the mature virions from infected cells, after viral replication. Because NA plays an important role in the viral life cycle, it is considered an attractive therapeutic target for the treatment of influenza. The methanolic extracts of Phellinus baumii and Phellinus igniarius exhibited significant activity in the neuraminidase inhibition assay. Polyphenolic compounds were isolated from the methanolic extracts. The structures of these compounds were determined to be hispidin, hypholomine B, inoscavin A, davallialactone, phelligridin D, phelligridin E, and phelligridin G by spectroscopic methods. Compounds inhibited the H1N1 neuraminidase activity in a dose-dependent manner with $IC_{50}$ values of 50.9, 22.9, 20.0, 14.2, 8.8, 8.1 and $8.0{\mu}M$, respectively. Moreover, these compounds showed anti-influenza activity in the viral cytopathic effect (CPE) reduction assay using MDCK cells. These results suggests that the polyphenols from P. baumii and P. igniarius are promising candidates for prevention and therapeutic strategies against viral infection.

Evaluation for Serological Patterns and Fecal Viral Shedding by Hemagglutination Inhibition Test and Real-time PCR in Korean CPV-2 isolates

  • We evaluated the patterns of serology and fecal viral shedding for any differences by hemagglutination inhibition (HI) and real-time PCR on Korean CPV-2 isolates (CPV-2a-I, CPV-2a-V and CPV-2b). We successfully detected fecal viral shedding from samples extracted 2-3 d.p.i., regardless of the onset of clinical signs. In addition, the pattern of viral shedding differed depending on the CPV-2 isolates used for inoculation. We also observed differences in the serological pattern that was also depended on the CPV-2 isolates inoculated. The onset and amount of fecal viral shedding were not correlated with the level of antibody titers in this study. Our study is a valuable resource for understanding the different pathobiology of the CPV-2 isolates and the correlation between the patterns of serum antibody titer and fecal viral shedding.

  • Objectives : This study was conducted to evaluate the effects that Injinsugunomija Extract affected Liver function test of Alcoholic liver disease and Viral hepatitis patients. Methods: We adminstered eighty packets of Injinsugunomija Extract to some kinds of liver disease patients for one month. And we investigated the changes of their liver function test. Results: The results from this study were summarized as followed. In the 10 patients of Alcoholic Liver Disease (ALD), 3 cases improve prominent and 4 cases improve effective and 3 cases are ineffective. Then 70% 01 ALD patients has improved by Injinsugunomija Extract In the 9 patients of Viral hepatitis, 1 case improves prominent and 4 cases improve effective and 1 case become worse and 1 case become worst and 2 cases are ineffective. Then 55.6% of Viral hepatitis patients has improved by Injinsugunomija Extract. Conclusions: From these results, Injinsugunomija Extract showed the meaningful effects on improving liver function test of ALD patients and Viral hepatitis patients.

Isolation and identification of canine adenovirus type 2 from a naturally infected dog in Korea

  • Canine adenovirus type 2 (CAV-2) infection results in significant respiratory illness in dogs. Isolating and culturing CAV-2 allows for investigations into its pathogenesis and the development of vaccines and diagnostic assays. In this study, we successfully isolated a virus from a naturally infected dog in Gyeonggi-do, Korea. The virus was propagated in Madin-Darby canine kidney (MDCK) and Vero cells and showed a specific cytopathic morphology that appeared similar to a bunch of grapes. The virus was first confirmed as CAV-2 based on these cytopathic effects, an immunofluorescence assay, hemagglutination assay, and electron microscopy. The viral titer of the isolate designated APQA1601 reached $10^{6.5}$ 50% tissue culture infections dose per mL in MDCK cells and exhibited no hemagglutination units with erythrocytes from guinea pig. The virus was also confirmed by polymerase chain reaction and next-generation sequencing. The APQA1601 strain had the highest similarity (~99.9%) with the Toronto A26/61 strain, which was isolated in Canada in 1976 when the nucleotide sequences of the full genome of the APQA1601 strain were compared with those of other CAV strains. Isolating CAV-2 will help elucidate the biological properties of CAV-2 circulating in Korean dogs.

개 REM134 유선종양세포의 sphere 형성을 통한 암 줄기세포 특성 분석 (Tumorsphere formation and cancer stem cell characterization of REM134 canine mammary carcinoma cells)

  • Canine mammary tumors are among the most frequently observed cutaneous tumors in female dogs. Cancer stem cells (CSCs), referred to as tumor-initiating cells, are thought to have properties similar to normal stem cells such as the ability to self-renewal and to differentiate into various cell types. Biological understanding of CSCs and the critical pathways involved in their maintenance are important in research and therapy for mammary tumors. We conducted the present study on sphere formation from REM134 cells by using methylcellulose to produce tumorspheres on a large scale and compared the specific markers of the spheres-formed and plating-cultured REM134 cells. The results revealed that the tumorspheres cultured in methylcellulose had higher seeding density and improved morphology compared to those produced in normal sphere formation medium. Expression levels of stemness markers and CSC-related markers were higher in tumorsphere-forming cells than in plating-cultured cells. Subsequently, we transplanted the tumorsphere-forming and plating-cultured cells into female nude mice to examine their tumorigenic potential. Tumor volume increased rapidly in mice transplanted with tumorsphere-derived cells compared to plating-cultured cells. We observed a novel sphere-forming condition for REM134 cells and showed that REM134 cell tumorspheres can exhibit improved CSC properties.

소비자의 페이스북 바이럴 광고 구전의도에 영향을 미치는 요인에 관한 연구 (Factors Influencing Consumer's Sharing Intent of Facebook Viral Advertising)

  • 온라인 동영상 광고 시장의 성장으로, 바이럴 광고가 주목받고 있다. 소비자, 기업, 콘텐츠, 네트워크는 바이럴 광고를 구성하는 네 가지 차원이다. 본 연구는 각 차원에 속한 세부요인들이 바이럴 광고 구전의도에 미치는 영향을 검증하였다. 수집한 설문자료에 대한 다중회귀분석을 실시한 결과, 바이럴 광고 구전의도에 유의미한 영향을 미치는 요인은 콘텐츠 차원의 설득의도, 기업 차원의 기업-광고이미지 관련성, 소비자 차원의 인지된 자기표현, 그리고 네트워크차원의 교량적 사회 자본으로 나타났다. 콘텐츠 차원의 사회적가치와 네트워크 차원의 결속적사회자본은 유의미한 영향력을 보이지 않았다. 결과적으로 바이럴 광고가 설득의도를 적게 나타낼수록, 광고 이미지와 기업의 이미지 관련성이 높을수록, 소비자가 광고를 공유함으로써 자신의이미지를 원하는대로 표현할 수 있다고 인지할수록, 그리고 소비자가 교량적 사회자본을 많이가지고 있을수록 바이럴 광고에 대한 공유의도를 높게 보이는 것으로 나타났다. 본 연구는 바이럴 광고 구전의도를 주제로 진행한 첫 번째 실증적인 연구이며, 바이럴 광고의 정의와 특징을 체계적으로 정리하여 다양한 차원의 요인들이 바이럴 광고 구전의도에 미치는 영향력을 통합적으로 검증하였다는 점에서 의의가 있다. 또한 기존의 온라인 구전 관련 연구에서는 드물게 할당표집을 사용하였고, 설문 영상으로 현재 바이럴 광고 소재로 가장 많이 사용되고 있는 감동 소재와유머 소재를 사용한 각각의 영상 두 개를 선정하여 결과의 일반화 가능성을 높였다.

Hepatitis B Virus DNA Polymerase Displays an Anti-Apoptotic Effect by Interacting with Elongation Factor-1 Alpha-2 in Hepatoma Cells

  • Hepatitis B virus (HBV) genome P-encoded protein HBV DNA polymerase (Pol) has long been known as a reverse transcriptase during HBV replication. In this study, we investigated the impact of HBV Pol on host cellular processes, mainly apoptosis, and the underlying mechanisms. We showed a marked reduction in apoptotic rates in the HBV Pol-expressed HepG2 cells compared to controls. Moreover, a series of assays, i.e., yeast two-hybrid, GST pull-down, co-immunoprecipitation, and confocal laser scanning microscopy, identified the host factor eEF1A2 to be associated with HBV Pol. Furthermore, knockdown of eEF1A2 gene by siRNA abrogated the HBV Pol-mediated anti-apoptotic effect with apoptosis induced by endoplasmatic reticulum (ER) stress-inducer thapsigargin (TG), thus suggesting that the host factor eEF1A2 is essential for HBV Pol's anti-apoptosis properties. Our findings have revealed a novel role for HBV Pol in its modulation of apoptosis through integrating with eEF1A2.

Development of indirect ELISA for the detection of canine adenovirus type 2 antibodies in dog sera

  • Background: Canine adenovirus type 2 (CAV-2) induces infectious laryngotracheitis in members of the family Canidae, including dogs. To date, no ELISA kits specific for CAV-2 antibody have been commercialized for dogs in Korea. Objectives: We aimed to develop new indirect enzyme-linked immunosorbent assay (I-ELISA) to perform rapid, accurate serological surveys of CAV-2 in dog serum samples. Methods: In total, 165 serum samples were collected from dogs residing in Chungbuk and Gyeongbuk provinces between 2016 and 2018. The Korean CAV-2, named the APQA1701-40P strain, was propagated in Madin-Darby canine kidney cells and purified in an anion-exchange chromatography column for use as an antigen for I-ELISA. The virus-neutralizing antibody titers of CAV-2 in the dog sera were measured by virus neutralization (VN) test. Results: We compared the results obtained between the VN and new I-ELISA tests. The sensitivity, specificity, and accuracy of new I-ELISA were 98.6%, 86.4% and 97.0% compared with VN test, respectively. New I-ELISA was significantly correlated with VN (r = 0.91). Conclusions: These results indicate that new I-ELISA is useful for sero-surveillance of CAV-2 in dog serum.

Immunogenicity of a new inactivated vaccine against feline panleukopenia virus, calicivirus, and herpesvirus-1 for cats

  • Feline panleukopenia virus (FPV), feline calicivirus (FCV), and feline herpesvirus type-1 (FHV-1) are major infectious pathogens in cats. We evaluated the immunogenicity of a new vaccine containing inactivated FPV, two FCVs, and FHV-1 in animals. An FPV, two FCVs, and an FHV-1 isolate were continuously passaged 70, 50, 80, and 100 times in CRFK cells. FP70, FC50, FC80, and FH100 were propagated and used as vaccine antigens. Two inactivated feline virus vaccines, feline rehydragel-adjuvanted vaccine (FRAV) and feline cabopol-adjuvanted vaccine (FCAV) were prepared and inoculated into mice and guinea pigs. Humoral immune responses were measured using hemagglutination inhibition (HI) for FPV and virus-neutralizing antibody (VNA) for two FCVs and FHV-1 tests. Serial passages in CRFK cells resulted in increase in titers of FPV and two FCVs but not FHV-1 The FCAV induced higher mean HI and VNA titers than the FRAV in guinea pigs; therefore, the FCAV was selected. Cats inoculated with FCAV developed a mean HI titer of 259.9 against FPV, and VNA titers of 64, 256, and 3.2 against FCV17D03, FCV17D283, and FHV191071, respectively. Therefore, cats inoculated with the FCAV showed a considerable immune response after receiving a booster vaccination.
