Review of regulatory management on standards and specifications for veterinary medical devices in Korea (동물용 의료기기 기준규격에 대한 고찰)

  • Well-established standards and specifications for medical devices not only provide clarity and consistency in licensing processes but also secure device safety and effectiveness. This study reviewed regulatory standards and specifications for veterinary medical devices in Korea based on data obtained through analyses of those medical devices. General standards for electromechanical, electromagnetic, and biological safety and individual standards for 76 electric and 70 general medical device products have been established by the Korean Ministry of Food and Drug Safety. However, the first standards and specifications for veterinary medical devices were established in March 2000 for two products (disposable needle and syringe). Similar to the large number of standards for medical devices, there have been general standards for safety and individual standards for 72 veterinary medical devices (27 electric medical devices, 12 non-electric medical devices, 10 medical supplies, and 23 other types) established since 2014. These new standards considered, among other aspects, the devices' applications, usage characteristics, and industrial environment. To date, the establishment of standards and specifications for veterinary medical devices remains limited. This study suggests that improvements should be made to the regulation of general and individual standards associated with various veterinary medical instruments, supplies, and in vitro diagnostic medical reagents.

Comparison of CXCL10 Secretion in Colorectal Cancer Cell Lines

  • Established cancer cell lines are widely used for developing biomarkers for the patient-specific treatment of colorectal cancer and predicting prognoses. However, cancer cell lines may exhibit different drug responses depending upon the characteristics of the cell line. Therefore, it is necessary to select a tumor cell line suitable for the purpose of the study by considering the cell characteristics. This study investigated the levels of CXCL10, which were recently been reported to play an important role in the outcome of tumor treatment, secreted by colon cancer cells. 2 × 105 cells/mL of each colorectal cancer cell was seeded into a 35 mm cell culture dish. After 24 h incubation, culture supernatant was used to determine the secreted CXCL10 levels. Among six colorectal cancer cell lines (HT-29, HCT116, CaCo-2, SW620, SW480, and CT26), Caco-2 cells showed the highest level of CXCL10 secretion. HT-29 cells showed the second-highest level of CXCL10 secretion. No significantly measurable level of CXCL10 secretion was detected in HCT116 cells. These results will be helpful in investigating the molecular basis of colorectal cancer.

Effects of Extract Mixture (Yg-1) of Anti-Inflammatory Herbs on LPS-Induced Acute Inflammation in Macrophages and Rats (급성염증성 발열 모델에서의 항염증성 약재 혼합 추출물(YG-1)의 효과)

  • Traditional herbs, such as Lonicera japonica, Arctii Fructus, and Scutellariae Radix have been used as traditional drug due to their anti-inflammatory and anti-oxidant activities. The aim of this study was to investigate the anti-inflammatory effects of extract mixture (YG-1) in a model of lipopolysaccharide (LPS)-induced acute inflammation in both macrophage (RAW 264.7) cells and Sprague-Dawley rats. YG-1 did not show specific cellular toxicity in RAW 264.7 cells until a concentration of $100{\mu}g/mL$. YG-1 reduced various markers related to inflammation such as IL-$1{\beta}$, COX-2, and iNOS caused by LPS in RAW 264.7 cells. Consistent with these results, YG-1 exerted significant anti-inflammatory effects in an acute inflammation rat model. Acute fever and high concentration of IL-$1{\beta}$ in serum induced by LPS were significantly reduced by YG-1. These results were similar to flubiprofen, a commercial anti-inflammatory and anti-febrile drug. Therefore, these results indicate that YG-1 has beneficial effects on LPS-induced acute inflammation and suggest that YG-1 can serve as an effective anti-inflammatory and anti-febrile drug.

Evaluation of Conventional Culture Methods and Validation of Immunoassays for Rapid Detection of Listeria monocytogenes in Dairy and Processed Foods (유제품 및 가공식품에서 Listeria monocytogenes 검출을 위한 배지법과 신속 검사키트의 유효성 검증)

  • Listeria monocytogenes is a foodborne pathogen inducing listeriosis in human. We compared two different culture methods for detection of L. monocytogenes and validated two commercial kits, $VIDAS^{(R)}$ and $REVEAL^{(R)}$ for Listeria. L. monocytogenes was inoculated into various food samples to generate partial positive samples. The inoculated samples were enriched in half-Fraser broth for 48 hr at $30^{\circ}C$. The enriched samples were streaked onto Oxford agar at 24 and 48 hr postincubation followed by biochemical confirmation and concurrently analyzed by using the two commercial kits for comparison. When the enrichment period was extended from 24 to 48 hr, the numbers of positive samples were dramatically increased from 6 to 52 out of 80 samples tested using the culture method. With the commercial kits, the numbers of positive samples were also significantly increased from 10 to 18 and 1 to 18, respectively, when the enrichment period was extended from 48 to 72 hr. There was no statistical difference between the 24 hr culture method and $VIDAS^{(R)}$ or $Reveal^{(R)}$ with 48 hr enrichment. In conclusion, the 24 hr for the culture method was insufficient to detect L. monocytogenes in various foods. The commercial kits could be adequate means for presumptive screening of L. monocytogenes in food.

  • 4,4'-diaminodiphenyl sulfone (dapsone) is a sulfone drug that has antibacterial effects on a variety of bacteria, especially Mycobacterium leprae; thus, it has been used to treat leprosy. Previous studies demonstrated that dapsone inhibits integrin-mediated adherence of neutrophils and production of prostaglandin $E_2$ by polymorphonuclear leukocytes. Hence, dapsone may act in immune cells and regulate cell-mediated inflammation processes. However, its anti-inflammatory effects remain unclear. The present study demonstrated that dapsone modulates the production of inflammation-related cytokines in immune cells. We employed the spleen cells of mice, which are major immune cells, and lipopolysaccharide (LPS) as a causative agent of inflammation for experiments. Dapsone induced a proportional change in splenocyte subsets and the apoptosis of spleen cells. Interestingly, dapsone decreased the production of tumor necrosis factor-alpha and interleukin (IL)-10, but not IL-6, in LPS-treated spleen cells. In other assays, we measured the dapsone-induced production of nitric oxide (NO) and the expression of activation markers of spleen cells. Dapsone decreased NO production in LPS-treated spleen cells. Taken together, our results demonstrate that dapsone has anti-inflammatory effects in immune cells and provide new insight into the potential uses of this agent.

Characteristics and antimicrobial susceptibility of Escherichia coli isolated from horse (말에서 분리한 Escherichia coli의 특성 및 항생제 감수성)

  • This study was conducted to investigate characteristics and antimicrobial susceptibility of Escherichia (E.) coli isolates isolated from vaginal mucosa and clitorial fossa of 105 Thoroughbred mares suspicious of the genital disease in Korea during the period from March 2006 to July 2007. Ninety six E. coli isolates were identified as standard biochemical properties and using BIOLOG system. Fifty three isolates (55.2%) could be classified into a total of 21 O serotypes and forty three isolates (44.8%) were non-typeable with 51 O antisera used in this study. The verotoxin 1 (VT 1) and verotoxin 2 genes were analyzed by multiplex PCR. Among them, one isolate was detected VT 1 gene (130 bp). Most of isolates showed a high susceptibility in ciprofloxacin (100%), enrofloxacin (100%), norfloxacin (100%), cefoxitin (96.9%), gentamicin (96.9%), sulphamethoxazole (96.9%), nitrofurantoin (94.8%), amikacin (93.8%), nalidixic acid (92.7%) and tetracycline (90.6%). These results may provide the basic information to establish strategies for the treatment and prevention of reproductive disease in Thoroughbred mares in Korea.

  • The possibility of the allergic encephalomyelitis caused by the combined vaccine (KGCC95VI) for the prophylaxis against Japanese encephalitis and Hantaan virus infection, recently developed by Korea Green Cross Corporation, was investigated in the Hartley guinea pigs. The KGCC-95VI was administered to the guinea pigs subcutaneously to sensitize the animals three times at one month intervals. There were no clinical signs or gross pathological findings. There were no abnormal histopathological findings at cerebrums, cerebellums, brain stems and the spinal cords. The concentration of myelin basic protein was 1.10 ng/dose quantified by ELISA, which met the guide4ine of below 2 ng/ml/dose recommended by American Society of Health -System Pharmacists(AHPS) Drug Information. Accordingly, the KGCC-95VI is considered not to induce any allergic immune responses which may lead to the experimental allergic encephalomyelitis.

Determination of tylosin in edible meats by high-performance liquid chromatography (HPLC를 이용한 식육내 타이로신의 잔류분석법)

  • A simple and rapid analytical method for the determination of tylosin in chicken, pork and muscle was established by High-Performance Liquid Chromatography(HPLC). Chicken, pork and beef muscle(5 g) were fortified by adding the $0.2{\mu}g/ml$ of standard tylosin and the drug was extracted from meats with 70% acetonitrile(ACN) and followed by liquid-liquid partition for clean-up procedure. Then $20{\mu}l$ portion of ACN elution was directly analyzed by HPLC with spectra 100 variable wavelength detector, and unfortified blank control were treated similarly. The average recovery rate of tylosin added to chicken, pork and beef muscle were $83{\pm}2.3$, $96{\pm}3.3$ and $92{\pm}1.6$(%) at the level 0.2 ppm, respectively. No tylosin residues in marketing meats. These results suggested that HPLC methodology could be acceptable for the extraction, determination and screening of tylosin residues in edible meats.

  • Biochemical and antimicrobial susceptibility tests were conducted on 40 strains of Pseudomonas aeruginosa originated from diseased chicken submitted for diagnosis to this Institute during 1978-80. An extensive study of the biochemical properties revealed that the tested strains can be identified with Pseudomonas aeruginosa. Antibiogram showed that all the strains were susceptible to gentamicin, colistin and amikacin but resist to nitrofurantoin, trimethoprim+sulfamethoxazole, ampicillin, methicillin and kanamycin, and had varing degreed of resistance to other antimicrobials including carbenicillin, sulfomamides, neomycin, streptomycin, tetracycline and chloramphenicol. Three of the most frequent resitance patterns observed were FM SXT AM ME KM CM TC SM NE SSS Pattern, FM SXT AM ME KM CM TC SM NE SSS CB Pattern and FM SXT AM EKM CM TC SM Pattern, and these resistance patterns contained 72.5% of the tested strains.

  • A study on quinolone antibacterial (pefloxacin methanesulfonate) was performed to use for the drug of fisheries. Petloxacin was proved excellent in antibacterial activity and resistance against fish pathogens when compared with the existing antibacterials. And any side effect was not observed during the period of indicated use. An outline of MIC(Minimal Inhibitory Concentration) was $1.6{\sim}6.4{\mu}g/ml$, $TLm_{48h}$ value were 380~420 ppm in Israeli carp(Cyprinus caprio) and 2100~2300 ppm in flounder(Paralichthys olivaceus). The residual time of fish body was less than 15 days. So we can treat some bacterial disease of fish by the dosage of 100 g/day/ton of fish body weight for 3 days and pefloxacin is thought to be used effectively and widely against most bacterial fish pathogens.
