Isolation of Isoflavones and Soyasaponins from the Germ of Soybean (콩 배아로 부터 Isoflavone과 Soyasaponin의 동시 분리)

  • The objective of present study was to simultaneously isolate of isoflavone and soyasaponin compounds from the germ of soybean seeds. Soy germ flours were defatted with hexane for 48h at room temperature, and methanolic extracts were prepared using reflux apparatus at $90^{\circ}C$ for 6h, two times. After extraction, extracts were separated with preparative RP-$C_{18}$ packing column ($125{\AA}$, $55-105{\mu}m$, $40{\times}150mm$), and collected 52 fractions were identified with TLC plate (Kieselgel 60 F-254) and HPLC, respectively. Among the identified isoflavone and soyasaponin fractions, isoflavone fractions were re-separated using a recycling HPLC with gel permeation column (Jaigel-W252, $20{\times}500mm$). Final fractions were air-dried, and the purified compounds of two isoflavones (ISF-1-1, ISF-1-2) and four soyasaponins (SAP-1, SAP-2, SAP-3, SAP-4) were obtained. Two isoflavone compounds (ISF-1-1, ISF-1-2) were acid-hydrolyzed for the identification of their aglycones, and confirmed by comparing with 12 types of isoflavone isomers. While the four kinds of soyasaponins were identified by using a micro Q-TOF mass spectrometer in the ESI positive mode with capillary voltage of 4.5kV, and dry temperature of $200^{\circ}C$. Base on the obtained results, it was conclude that ISF-1-1 is the mixture isomers of daidzin (43.4%), glycitin (47.0%), and genistin (9.6%), but ISF-1-2 is the single compound of genistin (99.8% <). On the other hand, soyasaponin SAP-1 is the mixture compounds of soyasaponin A-group (Aa, Ab, Ac, Ae, Af); SAP-2 is soyasaponin B-group (Ba, Bb, Bc) and E-group (Bd, Be); SAP-3 is soyasaponin B-group (Ba, Bb, Bc), E-group (Bd, Be), and DDMP-group (${\beta}g$); SAP-4 is soyasaponin B-group (Ba, Bb, Bc), E-group (Bd, Be), and DDMP-group (${\beta}g$, ${\beta}a$), respectively.

The Study on Anti-cancer Effects of Distilling Fresh-ginseng Herbal acupuncture against implanted Sarcoma-180 in vivo and A549 human epithelial lung cancer cells in vitro (A549 human lung cancer cell과 Sarcoma-180 복강암에 대한 수삼(水蔘) 증류약침(蒸溜藥鍼)의 항암효과(抗癌效果))

  • Objectives ; This study was to investigate the anti-cancer effects of herbal acupuncture with distilled fresh ginseng. The herbal acupuncture was injected to Chung-wan($C.V_{12}$) and Wisu($BL_{21}$) of mice that were subjected to Sarcoma-180 adbominal cancer cell and A549 human epithelial lung cancer cells in vitro. Methods : Anti-cancer effects of distilled fresh ginseng herbal acupuncture were tested by measruing Cox, Bcl-2, and Bax by using RT-PCR in A549 human epithelial lung cancer cells in vitro. And four weeks old Balb/c line male mice weighing around $20\;{\pm}\;3g$ were used to measure survival rate and anti-cancer effect to outputs of interleukin-2 and interleukin-4 using flow cytometry, possibility of mRNA menifestation using RT-PCR, and Cox mRNA. The results are as follows. Results : 1. In measuring mRNA menifestation in Cox, Bcl-2, and Bax by using RT-PCR in A549 human epithelial lung cancer cells in vitro, the result showed that fresh ginseng decreased Cox-2 which is directly involved in Inflammation process. 2. Survival rate was measured in an anti-cancer effect experiment against Sarcoma-180 abdorminal cancer. Median survival time of controlled group was 27 days, of experiment group I was 21 days, and of experiment group II was 27 days. Therefore, experiment group I showed -22.2% increase in survival rate and experiment group II showed no difference compare to controlled group. 3. There was no difference between condition group and controlled and experiment group in measuring outputs of interleukin-2 and interleukin 4 by using flow cytometry 4. In measuring outputs of interleukin-2 by using ELISA, there was no significant difference between condition group and controlled group and there was decrease in experiment group II compared to conditioned and controlled group. 5. In measuring cytokine mRNA menifestation by using RT-PCR, experiment group I showed increase of mRNA menifestation in interleukin-2,4 and $interferon-{\gamma}$ and experiment group II showed no significant difference in $interferon-{\gamma}$. Conclusion : According to the results, fresh ginseng herbal-acupuncture took a little effects in cancer. In using distilled fresh ginseng herbal acupuncture has effect on Cox-2 decrease. However, the difference in concentration of fresh ginseng showed no effect on killing cancer cell. It is assumed that inaccurate concentration of herbal acupuncture and fresh ginseng component could be the reason for this result. Therefore, future consideration will be studies on herbal acupuncture concentration.

Effects of High-temperature Annealing of CeO$_2$ Buffer Layers on the Surface Morphology of YBa$_2Cu_3O_{7-{\delta}}$ Films on CeO$_2$-buffered R-cut Sapphire Substrates (CeO$_2$ 완충층에 대한 고온 열처리가 CeO$_2$ 완충층을 지닌 R-cut 사파이어 기판 우에 성장된 YBa$_2Cu_3O_{7-{\delta}}$ 박막의 표면상태에 미치는 영향)

  • YBa$_2Cu_3O_{7-{\delta}}$ (YBCO) films grown on CeO$_2$-buffered r-cut sapphire substrates (CbS's) were prepared and their structural and electrical properties were measured. Post-annealed CeO$_2$ films were used as buffer layers for the experiments. It turned out that the YBCO films grown on post-annealed CbS's had the rms roughness of less than 20 ${\AA}$ and peak-to-peak roughness of about 30 ${\AA}$ when the YBCO film thickness was 3000 ${\AA}$. Meanwhile, YBCO films on in-situ grown CeO$_2$ buffer layers on r-cut sapphire substrates appeared to have the peak-to-peak roughness of more than 450 ${\AA}$. X-ray diffraction data revealed that the YBCO flms were epitaxially grown along the c-axis with the typical FWHM of(005) ${\theta}$ -2 ${\theta}$ peak about 0. 16 $^{\circ}$ and ${\Delta}$ ${\omega}$ of the (005) peak about 0.5 $^{\circ}$. T$_c$ > 87 K, ${\Delta}$T < 1 K and R(look)/R(100K) ${\ge}$3 were observed from the YBCO films. Applicability of the YBCO films for high-frequency applications was described.

The Effect of Magnetic Field on Enhancing the Anisotropy of Melt-spun Nd-Fe-Co(-Zr)-B Alloy (급속응고중 외부자장에 의한 Nd-Fe-Co(-Zr)-B계 합금의 자기이방성 향상)

  • Melt-spun $Nd_{14}Fe_{76}Co_4B_6$ and $Nd_{10.5}Fe_{79}Co_2Zr_{1.5}B_7$ ribbons were prepared under an externally applied magnetic field. Magnetic properties in terms of anisotropy were evaluated by discussing the effect of textured structure of the ribbon samples as well as its powders. About 32 % increase in $(B{\cdot}H)_{max}$ and 18.8 % increase in $B_r$ were observed along the perpendicular direction of the ribbon plane which is more prominent for the Nd-Fe-Co-Zr-B than for the Nd-Fe-Co-B alloy. The enhancement of magnetic anisotropy was monitored by measuring the anisotropy constant of each alloy as a function of quenching rate of the ribbon. It was found that for the melt-spun ribbon quenched at slow rate(less than 7 m/s) the magnetic field effect was overwhelmed by the heat gradient effect through the ribbon thickness while the field effect was prominent at intermediate quenching rate (more than 7~11 m/s). The reproducible maximum energy product, $(B{\cdot}H)_{max}$=16.4 MGOe can be obtained from the Nd-Fe-Co-Zr-B alloy.

  • Thin-film-transistors (TFTs) that can be prepared at low temperatures have attracted much attention because of the great potential for transparent and flexible electronics. One of the mainstreams in this field is the use of organic semiconductors such as pentacene. But device performance of the organic TFTs is still limited due to low field-effect mobility and rapid degradation after exposing to air. Alternative approach is the use of amorphous oxide semiconductors as a channel. Amorphous oxide semiconductors (AOSs) based TFTs showed the fast technological development, because AOS films can be fabricated at room temperature and exhibit the possibility in application like flexible display, electronic paper, and larges solar cells. Among the various AOSs, a-IGZO has lots of advantages because it has high channel mobility, uniform surface roughness and good transparency. [1] The high mobility is attributed to the overlap of spherical s-orbital of the heavy post-transition metal cations. This study demonstrated the effect of the variation in channel thickness from 30nm to 200nm on the TFT device performance. When the thickness was increased, turn-on voltage and subthreshold swing was decreased. The a-IGZO channels and source/drain metals were deposited with shadow mask. The a-IGZO channel layer was deposited on $SiO_2$/p-Si substrates by RF magnetron sputtering, where RF power is 150W. And working pressure is 3m Torr, at $O_2/Ar$ (2/28 sccm) atmosphere. The electrodes were formed with electron-beam evaporated Ti (30 nm) and Au (70 nm) bilayer. Finally, Al (150nm) as a gate metal was thermal-evaporated. TFT devices were heat-treated in a furnace at 250 $^{\circ}C$ and nitrogen atmosphere for 1hour. The electrical properties of the TFTs were measured using a probe-station. The TFT with channel thickness of 150nm exhibits a good subthreshold swing (SS) of 0.72 V/decade and on-off ratio of $1{\times}10^8$. The field effect mobility and threshold voltage were evaluated as 7.2 and 8 V, respectively.

A Study on Poisoning of the Reforming Catalysts on the Position of Anode in the Direct Internal Reforming Molten Carbonate Fuel Cell (직접 내부개질형 용융탄산염 연료전지의 음극판 위치에 따른 개질 촉매 피독에 관한 연구)

  • The trend of poisoning of reforming catalyst along with the position of anodic catalyst bed was studied. Keeping the conditions that steam to carbon ratio was 2.5, operating voltage was 0.75 V, current density was $140mA/cm^2$, the unit cell was operated during 24 hrs at a steady state. And then the cell was stopped, the catalysts packed in the position of inlet, middle and outlet were sampled individually and then the amount of carbon, Li and K poisoned were analysed. After 100 hrs operated, the catalysts at the same positions were analysed at the same manner. The result of this experiment was as followings. After 24 hrs operated, the poisoning amounts of Li and K in the catalyst were 0.27 wt% at inlet, 0.23 wt% at middle and the highest value 1.59 wt% at outlet. After 100 hrs, the amount of poisoning is the highest in the catalyst packed at the inlet of unit cell. The performance simulation of unit cell explained these trends of poisoning catalysts. The simulation told that the catalyst in the region of the inlet of unit cell treated the 90% of initial methane flow rate and the highest electrochemical reaction happened in this region. So the catalysts of this region were the most poisoned with carbon, Li and K and also the rate of poisoning is faster than that of the catalyst at other regions. The temperature at the region of outlet of unit cell was $30^{\circ}C$ higher than that of other regions, so more Li, and K vaporized than at other regions and little reforming reaction at this region made the catalysts poisoning rate low.

Phenomenological Study on Crystal Phase Separation in InGaN/GaN Multiple Quantum Well Structures (InGaN/GaN 다중 양자우물 구조에서의 결정상 분리 현상 연구)

  • We have investigated photoluminescence(PL) spectra of four $In_xGa_{1-x}N(x=0.15)/GaN$ multiple quantum well(MQW) structures with different well widths in order to study a phenomenon on crystal phase separation. The asymmetic behavior of PL spectra becomes stronger with increase of the well width from 1.5 nm to 6.0 nm, which indicates dual-peak nature. Analyzing the dual-peak fit PL spectra, we have observed that the intensity of low-energy shoulder peak rapidly becomes stronger, compared to that of high-energy peak corresponding to a transition in InGaN QW. It suggests that InGaN QW has two phases with tiny different In compositions, and that In-rich(InN-like) phase forms more and more relatively than stoichiometric InGaN(x=0.15) phase by the InN phase separation mechanism as the QW width increases. PL spectrum of 6.0-nm sample shows an additional peak at low-energy lesion(${\sim}2.0\;eV$) whose energy position is almost the same as a defect band of yellow luminescence frequently observed in GaN epilayers. It may be due to a defect resulted from In deficiency formed with development of the phase separation.

Characteristics of The Wastewater Treatment Processes for The Removal of Dyes in Aqueous Solution(1) - Chemical Precipitation or Biological Treatment - (수용액 중의 염료 제거를 위한 폐수처리공정의 특성(1) -화학적 응집 및 생물학적 처리-)

  • This study was conducted to remove the dyes in dye wastewater by the chemical precipitation or biological treatment which are one of the main pollutants in dye wastewater. In order to remove the disperse dyes effectively in aqueous solution by chemical precipitation process, coagulation and flocculation tests were carried out using several coagulants on various reaction conditions. It was found that the Ferrous sulfate was the most effective coagulant for the removal of disperse dye(DB79), and we could get the best result for the removal of disperse dye(DB56) in the aspects of TOC removal efficiency and sludge yield. When the Ferrous sulfate dosage was 800mg/l, the sludge settling velocity was very fast$(SV_{30}=4\%)$, and the color was effectively removed in the disperse dye(DB79) solution. Although the color removal was ineffective when the Alum was used as a coagulant, the sludge yield decreased in comparison with the Ferrous sulfate or the Ferric sulfate being used in the disperse dye(DB56) solution. In order to decolorize disperse dye(DR17) by using biological treatment process, a strain which has potential ability to degrade disperse dyes was isolated from natural system. The optimal culture conditions of temperature and pH were found to be $40^{\circ}C\;and\;8.5\~9$, respectively. When yeast extract was mixed with polypeptone at the mixing ratio of 1:1 as a nitrogen source, decolorization efficiency was highest$(93\%)$ among the nitrogen sources. The strain screened was excellent to adjust to pH, and it seems to have ability to control pH needed to growth. The optimal culture conditions in concentration of $MgSO_{4.}\cdot7H_2O\;and\;KH_2PO_4$ were $0.1\%(w/v)\;and\;0.2\%(w/v)$, respectively. Strains degrading and decolorizing reactive dyes, RB198 and RR141 which were isolated from water system, are named RBK1 and RRK. And the cell growth characteristics of RBK1 and RRK were investigated. The optimal culture conditions of temperature and pH were found to be 30t' and 7.0, respectively. Optimum nitrogen source was peptone, and it was found that decolorization efficiencies by strains RBK1 and RRK, were $85\%\;and\;62\%$, respectively, with introduction of 4,000mg/l of peptone. In the case of RBK1, color removal efficiencies were very high below 400mg/l. Decolorization efficiency was over $90\%$ at 20hours of culture time. The Color degradation ability of RRK was lower than that of RBK1.

Isolation and Identification Antifungal Compounds from Vitex trifolia L. (만형자(Vitex trifolia L.)로부터 항균활성물질의 분리 및 구조결정)

  • Vitex trifolia L. is a full grown fruit of Vitex rotundifolia L. (Verbenaceae). It has been used for treating headache, dizziness, toothache and removal of fever as a traditional medicine in Korea. V. trifolia (500 g) were extracted three times with 80% aqueous MeOH at room temperature. The MeOH extract (38 g) was successively partitioned with n-hexane, EtOAc, n-BuOH and $H_2O$. Both n-hexane and EtOAc fractions showed more than 80% antifungal activity in vivo against several plant pathogens at 2000 ppm. Successive repeated silica gel, Sephadex LH-20, preparative TLC, and preparative HPLC of these fractions led to isolation of three compounds. Using mainly mass spectroscopy and nuclear magnatic resonance (NMR) spectroscopy, their chemical structures were determined as agnuside(1), chrysosplenol B(2), and artemetin(3). Compound 2 was isolated for the first time from V. trifolia. Study on in vitro and in vivo antifungal activities of the isolated compounds is in progress.

High Performance Liquid Chromatographic Determination of Free Sugars in Ginseng and Its Products (고속액체(高速液體) 크로마토그래피에 의(依)한 인삼(人蔘) 및 인삼제품중(人蔘製品中)의 유리당(遊離糖)의 정량(定量))

  • Free sugars were isolated from ginseng root and its products and analyzed by using high performance liquid chromatogrphy. To isolate free sugars from aqueous sample solution fat-soluble components, crude saponin and protein were removed from the solution by extracting with benzene, water-saturated butanol and 80% ethanol, respectively. Free sugars found from both ginseng root and its products were fructose, glucose, sucrose and maltose, and the only sugar detected from red ginseng root and its products was rhamnose. Major sugar detected from fresh ginseng and white ginseng roots was sucrose, while sucrose and maltose were major sugars of red ginseng root.
