Bone regenerative effects of recombinant human bone morphogenetic protein-2 employed protein transduction domain (Protein transduction domain을 이용한 recombinant human bone morphogenetic protein-2의 골재생효과)

  • Bone morphogenetic proteins(BMPs) are regarded as members of the transforming growth $factor-{\beta}$ superfamily with characteristic features in their amino acid sequences. A number of studies have demonstrated the biologic activities of BMPs, which include the induction of cartilage and bone formation. Recently there was a attempt to overcome a limitation of mass production, and economical efficieny of rh-BMPs. The method producing PTD by using bacteria have advantages of acquiry a mass of proteins. Hences, a new treatment which deliver protein employed by protein transduction domain(PTD) has been tried. The purpose of this study was to evaluate the bone regenerative effect of TATBMP-2 and TAT-HA2-BMP-2 employed by PTD from HlV-1 TAT protein for protein translocation in the rat calvarial model. An 8mm calvarial, critical size osteotomy defect was created in each of 32 male Spraque-Dawley rats(weight $250{\sim}300g$). The animals were divided into 4 groups of 32 animals each (4 animals/group/healing interval). The defect was treated with TATBMP-2/ACS(Absorbable collagen sponge) (TATBMP-2 0.1mg/ml), TAT-HA2-BMP-2/ACS(TAT-HA2-BMP-2 0.1mg/ml), ACS alone or left untreated for surgical control(negative control). The rats were sacrificed at 2 or 8 weeks postsurgery, and the results were evaluated histologically. The results were as follows: New bone formation were not significantly greater in the TATBMP-2/ACS group relative to negative, and positive control groups. New bone was evident at the defect sites in TAT-HA2-BMP-2/ACS group relative to negative, positive control and TATBMP-2 groups. There were a little bone regeneration in TATBMP-2 groups. While, enhanced local bone formation were observed in TAT-HA2-BMP-2 group. But, The results was not the same in all rat defects. Therefore, further investigations are required to develop a method. which disperse homogenously, and adhere to target cells.

Effect of Temperature and Aging on the Relationship Between Dynamic and Static Elastic Modulus of Concrete (온도와 재령이 콘크리트의 동탄성계수와 정 탄성계수의 상관관계에 미치는 영향)

  • This paper investigates the relationships between dynamic elastic modulus and static elastic modulus or compressive strength according to curing temperature, aging, and cement type. Based on this investigation, the new model of the relationships we proposed. Impact echo method estimates the resonant frequency of specimens and uniaxial compression test measures the static elastic modulus and compressive strength. Type I and V cement concretes, which have the water-cement ratios of 0.40 and 0.50, are cured under the isothermal curing temperatures of 10, 23, and 50$\^{C}$ Cement type and aging have no large influence on the relationship between dynamic and static elastic modulus, but the ratio of dynamic and static elastic modulus comes close to 1 as temperature increases. Initial chord elastic modulus which is calculated at lower strain level of stress-strain curve, has the similar value to dynamic elastic modulus. The relationship between dynamic elastic modulus and compressive strength has the same tendency as the relationship between dynamic and static elastic modulus according to cement type, temperature and aging. The proposcd relationship equations between dynamic elastic modulus and static elastic modulus or compressive strength properly estimates the variation of relationships according to cement type md temperature.

Prediction of EFT/B Signal Transfer Characteristics in Mobile Charging Circuit (모바일 충전회로에서 EFT/B 신호의 전달특성 예측에 대한 연구)

  • This paper presents a methodology and a model that can analyze the high frequency transfer characteristics from socket in the AC power port to the 5 V DC output port in the mobile charging circuit. This is to predict the output signals coming from the IEC(International Electrotechnical Commission) Standard(IEC 61000-4-4), EFT/B(Electric Fast Transient and Burst) immunity test for mobile charging circuit. Since the mobile charging circuit is energized from the AC power socket from the power line, it is necessary to know the high frequency transfer characteristics with activated AC power line. A simple CDN(Coupling-Decoupling Network) is designed and manufactured for measuring S-parameters of mobile charging circuit with and without AC power line activated. The result shows that the S-parameters of the specific mobile charging circuits are almost the same, independent of AC power line activation. Consequently, the S-parameters without AC line could be used to predict the output response to the EFT/B signals, and it was shown that the proposed methodology predicts the output responses quite accurately, which proves the validness of the methodology presented in this paper.

Enhancement of Performance of Shell and Tube Heat Exchanger Using Pertinent Leakage Flow Between Baffle and Tube Bundles (배플과 관군간의 적정 누설유동을 이용한 쉘-관 열교환기의 성능향상)

  • In this study, the effects of the leakage flow between the baffle and tube bundles on the performance of a shell and tube heat exchanger (STHE) were examined using the commercial software ANSYS FLUENT v.14. A computational fluid dynamics model was developed for a small STHE with five different cases for the ratio of the leakage cross-sectional area to the baffle cross-sectional area, ranging from 0 to 40%, in order to determine the optimum leakage flow corresponding to the maximum outlet temperature. Using fixed tube wall and inlet temperatures for the shell side of the STHE, the flow and temperature fields were calculated by increasing the Reynolds number from 4952 to 14858. The present results showed that the outlet temperature, pressure drop, and heat transfer coefficient were strongly affected by the leakage flow, as well as the Reynolds number. In contrast with a previous researcher's finding that the leakage flow led to simultaneous decreases in the pressure drop and heat transfer rate, the present study found that the pertinent leakage flow provided momentum in the recirculation zone near the baffle plate and thus led to the maximum outlet temperature, a small pressure drop, and the highest heat transfer rate. The optimum leakage flow was shown in the case with a ratio of 20% among the five different cases.

  • Natural ventilation in a four and one-half span, double polyethylene commercial greenhouse was investigated with actual data collected at Quailcrest Farm near Wooster, Ohio. Moreover, a computational fluid dynamics (CFD) numerical technique, FLUENT V4.3, was used to predict natural ventilation rates, thermal conditions, and airflow distributions in the greenhouse. The collected climate data showed that the multi-span greenhouse was well ventilated by the natural ventilation system during the typical summer weather conditions. The maximum recorded air temperature difference between inside and outside the greenhouse was 3.5$^{\circ}C$ during the hottest (34.7$^{\circ}C$) recorded sunny day; the air temperatures in the greenhouse were very uniform with the maximum temperature difference between six widely dispersed locations being only 1.7$^{\circ}C$. The CFD models predicted that air exchange rates were as high as 0.9 volume per minute (A.C. .min$^{-1}$ ) with 2.5m.s$^{-1}$ winds from the west as designed.

Public Officials' Perceptions on Improving Quality of Community Health Care Service (공무원 인식유형에 따른 보건서비스 개선 방안)

  • The purpose of this study is to understand the perceptions of public officials in improving primary health care service for local communities by applying Q methodology that draw upon schematic model in subjectivity study. A survey was carried out among public officials at Gangneung Community Health Center, South Korea, to classify the 49 selected Q-statements into a normal distribution using a 11 point scale. The collected data was analyzed using QUANL program, and principal component factor analysis using varimax rotation was used to identify public officials' perceptions of improving quality of health care for local communities as a meaningful service provider. Type I can be categorized by a strong concern for the service acceptance, Type II can be categorized by advocating for service usability, Type III can be categorized by the pursuit of service specialization, Type IV can be categorized by roles for service accessibility, and Type V can be categorized by the suggestions for operation and management. The results provide information used to further improve quality of community health care service with the aim to promote the effectiveness of health care delivery and to increase health service utilization.

Improvement of Somatic Cell Nuclear Transfer Technology for the Production of Disease Model Mouse: I. Optimization of Oocyte Enucleation and Reconstruction (질환모델마우스 생산을 위한 체세포핵이식방법 개선; I. 탈핵 및 재조합난자 생산기법 최적화)

  • This study was undertaken to optimize enucleation and reconstitution methods for the production of cloned mice by somatic cell nuclear transfer Outbred ICR mouse oocytes at the metapahse- II stage were retrieved from female mice superovulated by PMSG and hCG. In Experiment 1, oocytes were enucleated in medium supplemented with cytochalasin B (CCB) of 3 levels (0, 7.5 or 15 $\mu\textrm{g}$/mL), and higher rate of encleation was obtained at 7.5 and 15 $\mu\textrm{g}$/mL than at $\mu\textrm{g}$/mL. In Experiment 2, oocytes enucleated in 7.5 $\mu\textrm{g}$/mL CCB-containing medium were reconstituted with different types of somatic cell by following methods; 1) cumulus cells by direct cell injection, 2) cumulus cells by electric fusion (1.25 kV/cm, 2 pulses for each 70 $mutextrm{s}$) or 3) STO cells by the electrofusion. Electrofusion of STO cells with enucleated oocytes yielded the greatest (P<0.05) rate of reconstitution without lysis (76%) than any other combinations. Although significant decrease in the rate of somatic cell introduction was found, the electrofusion of cumulus cells yielded better rate of reconstitution than direct injection (0 vs. 18%). In Experiment 3, the duration of electric stimulation for the fusion was changed to either 50 $mutextrm{s}$ or 90 $mutextrm{s}$, but no significant improvement of reconstitution efficacy was obtained. In conclusion, this study showed that ICR mouse oocytes could be used for the production of reconstituted oocytes and a fusion method of 1.25 KV/cm with 2 pulses using 570 cell was the optimal.

A Study on the Accelerated Life Test of BLDC Motor in Ceiling Mounted Digital Signage Rotating System (천정 거치형 디지털 사이니지 회전 시스템의 BLDC모터 가속수명시험에 관한 연구)

  • In a time when product development cycles are getting shorter and shorter, many companies are making efforts to develop products with high reliability in a short period of time, accelerated life test is widely used as a method to quickly evaluate reliability. Accelerated life test reduces the test life or the life of the product from the observed data by shortening the lifetime of the product or abruptly lowering the performance under the worse condition than the actual condition in order to shorten the test cost or the test time. In this paper, BL3640A-06P+RB35, DC12V model, which is used in the support device of an automatic rotation type digital signage, which display various information such as textures and images on a display screen in a public place or a commercial space, BLDC motors were subjected to a constant stress test and at the rotational speed of 1rpm, $180^{\circ}$ rotation and reverse rotation under actual use conditions, the stress was imposed on the rotating speed of 2rpm and the weight of the actual installed product from 22.2kgf to 10kgf were installed. The lifetime of the actual use environment condition is 23,545 hours and the rotation speed is accelerated. The life time of the acceleration condition with the additional weight is 1,380 hours. The acceleration factor is calculated as 17.06, the one year guarantee test day is 235 days to 14 days, of the period from 470 days to 28 days, and the third year from 704 days to 42 days. The test date of the BLDC motor was tested on the shortened test date, and the rotational speed and the current value were measured. It is found that there is no defect even if it operates as the test date corresponding to the specified one year warranty period and the 3 year accelerated life test which is experimented. Using the statistical technique of the regression analysis the expected time for the motor to defect to #4 samples was 20 years.

A Simple Dosing Scheme for Intravenous Busulfan Based on Retrospective Population Pharmacokinetic Analysis in Korean Patients

  • Busulfan is an antineoplastic agent with a narrow therapeutic window. A post-hoc population pharmacokinetic analysis of a prospective randomized trial for comparison of four-times daily versus once-daily intravenous busulfan was carried out to search for predictive factors of intravenous busulfan (iBu) pharmacokinetics (PK). In this study the population PK of iBu was characterized to provide suitable dosing recommendations. Patients were randomized to receive iBu, either as 0.8 mg/kg every 6 h or 3.2 mg/kg daily over 4 days prior to hematopoietic stem cell transplantation. In total, 295 busulfan concentrations were analyzed with NONMEM. Actual body weight and sex were significant covariates affecting the PK of iBu. Sixty patients were included in the study (all Korean; 23 women, 37 men; mean [SD] age, 36.5 [10.9] years; weight, 66.5 [11.3] kg). Population estimates for a typical patient weighing 65 kg were: clearance (CL) 7.6 l/h and volume of distribution (Vd) 32.2 l for men and 29.1 L for women. Inter-individual random variabilities of CL and $V_d$ were 16% and 9%. Based on a CL estimate from the final PK model, a simple dosage scheme to achieve the target $AUC_{0-inf}$ (defined as median AUC0-inf with a once-daily dosage) of 26.18 $mg/l{\cdot}hr$, was proposed: $24.79{\cdot}ABW^{0.5}mg$ q24h, where ABW represents the actual body weight in kilograms. The dosing scheme reduced the unexplained interindividual variabilities of CL and Vd of iBu with ABW being a significant covariate affecting clearance of iBU. We propose a new simple dosing scheme for iBu based only on ABW.

Sodium Dependent Taurine Transport into the Choroid Plexus, the Blood-Cerebrospinal Fluid Barrier

  • Taurine, a ${\beta}-amino$ acid, plays an important role as a neuromodulator and is necessary for the normal development of the brain. Since de novo synthesis of taurine in the brain is minimal and in vivo studies suggest that taurine dose not cross the blood-brain barrier, we examined whether the choroid plexus, the blood-cerebrospinal fluid (CSF) barrier, plays a role in taurine transport in the central nervous system. The uptake of $[^3H]-taurine$ into ATP depleted choroid plexus from rabbit was substantially greater in the presence of an inwardly directed $Na^+$ gradient taurine accumulation was negligible. A transient in side-negative potential gradient enhanced the $Na^+-driven$ uptake of taurine into the tissue slices, suggesting that the transport process is electrogenic, $Na^+-driven$ taurine uptake was saturable with an estimated $V_{max}$ of $111\;{\pm}\;20.2\;nmole/g/15\;min$ and a $K_M\;of\;99.8{\pm}29.9\;{\mu}M$. The estimated coupling ratio of $Na^+$ and taurine was $1.80\;{\pm}\;0.122.$ $Na^+-dependent$ taurine uptake was significantly inhibited by ${\beta}-amino$ acids, but not by ${\alpha}-amino$ acids, indicating that the transporter is selective for ${\beta}-amino$ acids. Since it is known that the physiological concentration of taurine in the CSF is lower than that in the plasma, the active transport system we characterized may face the brush border (i.e., CSF facing) side of the choroid plexus and actively transport taurine out of the CSF. Therefore, we examined in vivo elimination of taurine from the CSF in the rat to determine whether elimination kinetics of taurine from the CSF is consistent with the in vitro study. Using a stereotaxic device, cannulaes were placed into the lateral ventricle and the cisterna magna of the rat. Radio-labelled taurine and inulin (a marker of CSF flow) were injected into the lateral ventricle, and the concentrations of the labelled compounds in the CSF were monitored for upto 3 hrs in the cisterna magna. The apparent clearance of taurine from CSF was greater than the estimated CSF flow (p<0.005) indicating that there is a clearance process in addition to the CSF flow. Taurine distribution into the choroid plexus was at least 10 fold higher than that found in other brain areas (e. g., cerebellum, olfactory bulb and cortex). When unlabelled taurine was co-administered with radio-labelled taurine, the apparent clearance of taurine was reduced (p<0.0l), suggesting a saturable disposition of taurine from CSF. Distribution of taurine into the choroid plexus, cerebellum, olfactory bulb and cortex was similarly diminished, indicating that the saturable uptake of taurine into these tissues is responsible for the non-linear disposition. A pharmacokinetic model involving first order elimination and saturable distribution described these data adequately. The Michaelis-Menten rate constant estimated from in vivo elimination study is similar to that obtained in the in vitro uptake experiment. Collectively, our results demonstrate that taurine is transported in the choroid plexus via a $Na^+-dependent,saturable$ and apparently ${\beta}-amino$ acid selective mechanism. This process may be functionally relevant to taurine homeostasis in the brain.
