감귤에서 분리한 정유 성분의 항균활성 연구 (Study of Anti-microbe Activity of Essential oil(Unshiu oil) purified from Citrus Unshiu S.Marcov)

  • Volatile odor substance originating from drying and making dry peel of tangerine from the fruit skin were collected through modern equipment. The anti-microbial activity of the substance against various skin-residing bacteria including acne, dandruff, athelete's foot, and gingivitis inducing microorganisms were tested. Anti-microbial activity was observed in purified oil, where 87 to 92% was D-limonene. Against P. acnes, 103$cfu/m{\ell}$ of P. acnes were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.3%. Against P. ovale, a dandruff inducing bacteria, 104$cfu/m{\ell}$were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.1%. Against T. rubrum and T. Mentagrophytes, both of which are athelete's foot inducing microorganisms, 83% of T. Mentagrophytes and 99.9% of T. rubrum were suppressed at 0.1% Unshiu oil, and the MIC were 0.3% and 0.05% respectively. Against S. aureus, a skin infection inducing bacteria, 103$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against B. subtilis, a non-pathogenic sporulating bacteria, 104$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against C. albicans, found in mucous membranes, 104$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against Aspergillus niger, an otomycosis inducing microorganism, 99.9% were suppressed at 0.1% Unshiu Oil. The results above indicate that low concentration of purified oil extracted from tangerine had strong antimicrobial activities against bacteria and fungi residing on the skin, and that it may be developed into skin disorder treating products in the future.

감귤 오일 마이크로캡슐 가공 직물에 대한 EEG 신호와 주관적 향기감성 및 선호도 (EEG Signal, Subjective Fragrance Sensation, and Preference of Citrus Oil Microcapsule-Loaded Fabric)

  • This study investigated EEG signal, subjective fragrance sensation, and the preference of differently colored cotton knit treated with Citrus unshiu oil containing microcapsules as well as examined their relationships for providing regression models on subjective fragrance preference. Color variables combining 2-level hue (Yellow and Green) and 3-level tone (strong, pale, and grayish) were applied by dyeing prior to microcapsule treatment. We invited 28 female college students aged 20's for EEG signal experiments and subjective fragrance sensations with fragrant knit by rubbing. EEG signals at $mid-{\alpha}$, $fast-{\alpha}$, and $low-{\beta}$ showed significant differences depending on color; Green had more relative power values and grayish tone did more at $low-{\beta}$. Even though subjective sensation showed no significant differences depending on color, some of them such as Fresh, Comfort, and Natural showed significant correlations with EEG signal at $low-{\beta}$, which means that the fragrance sensations of Citrus unshiu fragrance are concerned with attention and alertness for Koreans. Fragrance preference was regressed significantly using some EEG signals and subjective sensation. The results could be utilized to value up fragrant textiles by Citrus unshiu oil.

Comparison of the Cold-Pressed Peel Oil Composition between ]Korean and Japanese Satsuma Mandarin (Citrus unshiu Marcov. forma Miyagawa-wase) by GC, GC-MS and GC-O

  • The comparison of the volatile flavor components from Korean and Japanese Satsuma mandarin (C. unshiu Marcov. forma Miyagawa-wase) peel oils, isolated by cold-pressing, was performed by gas chromatography, mass-spectrometry and gas chromatography-olfactometry (GC-O). Eighty-five volatile components were identified in each oil by GC and GC-MS. Forty-three components were detected in each oil by GC-O. The total amount of monoterpene hydrocarbons was 95.88% (Korean mandarin) and 95.29% (Japanese mandarin). Limonene, ${\gamma}$-terpinene, myrcene and $\alpha$-pinene were the main components of the cold-pressed oils from the both samples. The volatile composition of the Japanese mandarin was characterized by a higher content of sesquiterpene hydrocarbons, especially bicyclogermacrene, $\alpha$-humullene and valencene. The volatile composition of two samples can easily be distinguished by the percentages of aldehydes, ketones and esters, which were found at higher levels in the Japanese mandarin. The sweet and fruity flavor was stronger in the Korean mandarin oil while herbaceous flavor was stronger in Japanese sample. From GC-O data it is suggested that the sweet and fruity flavor of the Korean mandarin resulted from terpinolene and linalool, and the herbaceous note of the Japanese mandarin from $\alpha$-humullene, nepal, ι-carvone and perill aldehyde.

The Volatile Composition of Kiyomi Peel Oil (Citrus unshiu Marcov×C. sinensis Osbeck) Cultivated in Korea

  • The volatile composition of Kiyomi peel oil cultivated in Korea was studied by using gas chromatography and gas chromatography-mass spectrometry. The peel oil from the Kiyomi fruit was prepared by using a cold-pressing extraction method. Among the 65 components quantified in Kiyomi oil, 25 terpene hydrocarbons and 40 oxygenated compounds were identified, with peak weight percentages measuring 94.5% and 4.9%, respectively. Limonene was the predominant compound (87.5%), followed by myrcene (2.4%), sabinene (0.9%), $\alpha$-pinene (0.8%), $\beta$-sinensal (0.8%), (Z)-$\beta$-farnesene (0.7%), neryl acetate (0.6%), valencene (0.5%), $\alpha$-farnesene (0.5%), and $\alpha$-sinensal (0.5%). A unique characteristic of the volatile profile of the Kiyomi oil was the proportion of aldehydes (2.7%), which resulted from the relative abundance of $\alpha$- and $\beta$-sinensal. Another unique characteristic of the Korean Kiyomi oil was its relative abundance of $\beta$-sinensal, (Z)-$\beta$-farnesene, neryl acetate, valencene, $\alpha$-sinensal and nootkatone. Valencene and $\alpha$- and $\beta$-sinensal were regarded as the influential components of Korean Kiyomi peel oil.

감귤 주스 착즙박을 이용하여 재배된 버섯균사체의 용매추출에 의한 휘발성 성분 (Solvent Extracted Volatile Components of Mushroom Mycelia Cultivated with Citrus Juice Processing Wastes)

  • 감귤 부위별 및 감귤 주스 가공 후 폐기되는 착즙박을 배지로 하여 재배된 간, 표고, 새송이, 산호침, 참부채 및 영지 버섯) 균사체의 GC/MS에 의한 휘발성 물질을 분석하였다. 감귤 분말에서 휘발성 물질은 29종류이었는데, ${\delta}-elemene$, ${\beta}-elemene$, ${\beta}-caryophyllene$, caryophyllene, ${\gamma}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 감귤류의 essential oil 중의 성분들이었으나 8-hydroxy-linalool, ${\beta}-selinene$, tetradecanoic acid 및 pentadecanal은 essential oil 중의 성분들이 변화한 것이었다. 이외에도 tangeretin, nobiletin, 및 stigmasterol이 검출되었다. 버섯 균사체 분말에서 검출된 휘발성 성분들은 18종이었으며, ${\beta}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 버섯이나 균사체에서 보고되지 않은 것으로 보아 균사체 배지로 사용한 착즙박에서 유래한 것으로 생각된다. caryophyllene, hexadecanoic acid, decanoic acid 및 tetradecanoic acid는 기 보고된 버섯이나 균사체의 성분들이었으며, Limonene-1,2-epoxide, ethyllinoleate, 1,2-benzenedicarboxylic acid, dicyclohexyl ester 및 ($3{\beta}$,24S)-stigmast-5-en-3-ol (${\gamma}-sitosterol$)은 감귤이나 버섯 균사체에서 보고되지 않은 성분들이 검출되었다.

식물정유 7종의 항산화능 분석 및 Candida albicans 생장 억제 정유의 생리활성 성분 구명 (Antioxidant Properties of 7 Domestic Essential Oils and Identification of Physiologically Active Components of Essential Oils against Candida albicans)

  • 본 연구에서는 국내 자생식물 7종의 정유 중 우수한 항산화 효과가 있는 온주밀감, 육계나무 정유 2종을 선별하였고, 피부염을 유발하는 Candida albicans에 대한 항미생물 활성 평가를 통해 식물정유의 항미생물 활성성분을 구명하고자 하였다. 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거활성 측정 결과 SC50값이 온주밀감 정유 0.010 mg/mL, 육계나무 정유 0.09 mg/mL을 나타냈으며, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) 라디칼 소거활성 측정 결과 온주밀감 정유 0.09 mg/mL, 육계나무 정유 0.06 mg/mL로 높은 항산화 활성을 나타냈다. Minimum Inhibitory Concentration는 육계나무 정유 1.25 mg/mL, 온주밀감 정유 5 mg/mL 값으로 육계나무 정유에서 뛰어난 항미생물 활성이 나타났다. Thin layer chromatography (TLC) bioassay를 통해 2종 정유의 성분 분획에 따른 C. albicans에 대한 항미생물 활성을 평가하였고, preparative TLC (prep. TLC)를 통해 활성분획을 획득하고, 동일 Rf값을 가지는 성분들에 대해서 GC/MS 분석을 수행하여 항미생물 활성성분을 구명하였다. 그 결과 항산화 및 항미생물 활성을 나타내는 주성분은 각각 육계나무 정유의 경우 cinnamyl acetate, eucalyptol, linalool, citral 온주밀감 정유의 경우 linalool이었다. 2종 정유에 공통적으로 항산화 및 항미생물 활성을 나타낸 분획부 성분분석 결과 linalool은 항산화, cinnamyl acetate, eucalyptol, citral, geranyl acetate은 항산화와 항미생물 능력이 있다고 사료된다.

진피류(陳皮類) 한약재의 Hesperidin과 정유성분 비교 (Analysis of the Content of Hesperidin and Essential Oils from the Peels of Various Citrus Species)

  • Objectives: This study was carried out to evaluate the quality of the peels of various Citrus species. Method: The contents of hesperidin from fruit peels used as Citri Pericarpium such as C. natsudaidai, C. grandis, C. unshiu, and C. sunki, were analyzed by HP-TLC, HPLC, and essential oils of those were analyzed by GC/MS. Results: HPLC analysis showed that the hesperidin from the peel of C. unshiu and C. reticulata was satisfied the standard of Korean Pharmacopoeia. The essential oil was analyzed by GC/MS. As a result, limonene, furfural, 5-methyl-2-furfural, linalool oxide(cis), linalool oxide(trans), terpinen-4-ol, $(-)-{\alpha}$-terpineol, germacrene D, 4-methyl-2,6-di-tert-butylphenol was detected in all 4 kinds of Citrus species. Conclusions: As a result of chemotaxonomical similarity analysis with essential oils, the peels of C. natsudaidai and C. grandis are closely related, while C. unshiu is distantly related to the others.

Effect of Foliar Application of Gibberellic Acid and Machine Oil Emulsion Mixture on Flowering and Fruit Quality of Satsuma Mandarin (C. unshiu Marc. cv. Miyagawa) Cultivated on Open Fields

  • In satsuma mandarin (C. unshiu Marc. cv. Miyagawa), alternate bearing is frequently occurred. It is very difficult to control and maintain the optimum production because satsuma mandarin is a very sensitive citrus cultivar. We carried out the experiment to find out the effects of foliar application of gibberellic acid ($GA_3$) and machine oil emulsion mixture on flowering and fruit quality of satsuma mandarin grown on open field. The treatments were composed of control (no application), $GA_3$ (25, 50 and $100mg{\cdot}L^{-1}$), machine oil emulsion 100 times and mixture of $GA_3$ (25 and $50mg{\cdot}L^{-1}$) with machine oil emulsion 100 times on 16 year-old 'Miyagawa' satsuma mandarin on December 30, 2012. Foliar application of $GA_3$ during winter season led to a reduction of the flowering in the following year. $GA_3$ decreased flowering in dose dependant manner. However, application of $GA_3$ and machine oil emulsion (100 times-diluted mixture) inhibited effectively the flowering compared to $GA_3$ only. Also, flower-leaf ratio was significantly decreased by the application of $GA_3$ 25 (0.54) and $50(0.41)mg{\cdot}L^{-1}$ with machine oil emulsion mixture in comparison to $GA_3$ 25 (0.86) and $50(0.60)mg{\cdot}L^{-1}$ only. The number of leaves per fruit increased in application of $GA_3$ and machine oil emulsion mixture as concentration of $GA_3$ increased. Fruit weight and sugar-acid ratio also increased in comparison to the control whereas fruit size, soluble solid content and acidity remained unchanged. From the results, it was suggested that $GA_3$ application can reduce the number of flowers on the alternate bearing of satsuma mandarin. Moreover, application of $GA_3$ (25 and $50mg{\cdot}L^{-1}$) with machine oil emulsion 100 times mixture can reduce the flowering effectively without altering fruit maturity and soluble solid contents. Therefore, it can be concluded that the application of $GA_3$ 25 and $50mg{\cdot}L^{-1}$ with machine oil emulsion 100 times might be useful in controlling alternate bearing in satsuma mandarin.

수박 과실썩음병 병원균(Acidovorax avenae subsp. citrulli)에 대한 식물유래 항균 활성물질 탐색 (Search for Plant-originated Antibacterial Compounds Against Pathogen (Acidovorax avenae subsp. citrulli) of Watermelon Bacterial Fruit Blotch)

  • 본 연구는 수박 과실썩음병의 원인균인 Acidovorax avenae subsp. citrulli 균에 대해서 항균활성을 갖는 친환경 유기농자재를 개발할 목적으로 약용식물 133종을 대상으로 진행되었다. 이들 133종 약용식물의 MeOH 추출물에 대한 bioassay를 통해 항균 활성을 검정한 결과 청피(Citrus unshiu Markovich) 추출물에서 강한 항균활성을 보였다. 청피(Citrus unshiu Markovich)로부터 항균활성물질을 구명하고자 용매분획을 하였고, 용매분획 중에서 hexane fraction이 가장 강한 활성을 나타내었다. Hexane fraction을 GC-MS로 분석하여 chromatogram상의 각각의 peak에 해당하는 mass spectrum과 Wiley library를 비교하여 profiling 한 결과, essential oil인 d-limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol과 지방산인 palmitic acid, 9,12-octadecadienoic acid, linolenic acid 그리고 steroid 화합물인 stigmasterol이 검출되었다. 이들 검출화합물 중에서 항균 활성물질로 추정되는 d-limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol의 항균 활성을 검정하기 위해 표준품을 사용하여 bioassay한 결과 두 화합물 d-limonene, ${\gamma}$-terpinene에서 높은 항균활성을 보였다. 따라서 본 연구를 통해 청피로부터 분리한 d-limonene과 ${\gamma}$-terpinene 화합물이 항균 활성물질인 것을 구명하였다. 항균력이 강한 청피 추출물 또는 d-limonene과 ${\gamma}$-terpinene을 주성분으로 하는 추출물을 수박 과실썩음병에 대한 친환경 방제용 자제로 개발이 가능할 것으로 판단되었다.

Symptomatology of Citrus mosaic sadwavirus (CiMV) in Some Citrus Cultivars and Effect of CiMV Infection on Citrus Fruit Quality

  • Citrus mosaic sadwavirus (CiMV) is a closely related virus with the Satsuma dwarf virus (SDV) along with Navel orange infectious mottling virus (NIMV), Natsudaidai dwarf virus (NDV), and Hyugagatsu virus (HV). The present study found that the typical symptoms of CiMV-infected citrus fruits include the appearance of dark blue speckles or ringspots on fruit rinds and the browning of oil glands in the spots as rind coloring began. As rind coloring progressed, the spots gradually faded, whereas the browning of the oil glands worsened to the point that the tissues surrounding the oil glands became necrotic. In very early satsuma mandarins (Citrus unshiu 'Miyamoto Wase') and 'Setoka' cultivar (C. hybrid 'Setoka') of late-maturity citrus, the symptomatic fruits were eventually dropped. And in early satsuma mandarin (C. unshiu 'Miyakawa Wase'), the peel hardness of the virus-infected fruit (1,618.3 ± 305.5, g-force) was more than twice as hard as that of the healthy fruit (636.5 ± 39.1, g-force). The ratio of flesh weight to total fruit weight was higher for the healthy fruit (77.3 ± 1.7%) than for the infected fruit (70.7 ± 0.6) and peel puffing was more severe in the infected fruit (2.9 ± 0.4 mm) than in the healthy fruit (0.9 ± 0.2 mm). The soluble solids content in infected citrus fruits was less values than the healthy fruit by 0.5-1.5 °Brix. These findings reveal that CiMV infection on citrus trees reduces the fruit quality of citrus.
