해조단백질 추출에 관한 연구 2. 식염가용성 및 알콜가용성 단백질의 추출 (STUDIES ON THE EXTRACTION OF SEA WEED PROTEINS 2. Extraction of NaCl and Alcohol Soluble Proteins)

  • 분포량이 많고 식용되고 있는 중요해조인 둥근돌김, 잎파래, 구멍갈파래, 미역, 톳, 모자반, 셀만모자반, 누운청각의 식염가용성 및 알콜가용성 단백질 추출에 영향을 주는 제 요소 즉 추출용액의 농도, 용매량, 시간, 온도, pH의 영향을 검토하고 pH에 따른 TCA 침전단백질을 정량한 결과를 요약하면 다음과 같다. 1. 식염가용성 단백질에 있어서 미역, 톳, 돌김, 잎파래, 구멍갈파래는 0.25M의 식염용액에서, 모자, 셀만모자반, 누운청각은 1.0M에서 가장 좋은 추출성적을 얻었으며, 알콜가용성 단백징 전시료(미역, 모자반, 잎파래, 둥근돌김)가 $20\%$ 에타놀에서 가장 좋은 추출성적을 보였다. 2. 시료일추출용매비(w/v)는 식염가용성의 경우 1:30(w/v)에서 알콜 가용성 단백질은 건조시료 1g에 대하여 100ml을 가할 때 추출성적이 좋았다. 3. 3. 추출시간에 있어서 식염가용성 단백질은은 구멍 갈파래, 잎파래가 1시간, 둥근돌김, 누운청각이 2시간, 미역, 톳, 모자만, 셀만모자반은 3시간이 최적 추출시간이내, 추출성분이 좋았다. 4. 추출온도는 식염가용성일 경우, 잎파래, 구명 갈파래는 $40^{\circ}C$에서, 미역은 $50^{\circ}C$, 톳, 모자반, 셀만모자반, 누운청각은 $60^{\circ}C$서 최고의 추출성적을 보였고, 알콜가능성은 잎파래가 $30^{\circ}C$ 미역, 둥근돌김, 모자반이 $40^{\circ}C$에서 최고의 추출성적을 보였다. 5. pH의 영향은 각 시료마다 대동소이하며, 식염가용성 pH $8\~9$에서 알콜가용성은 pH $8\~9$에서 성적이 좋았다.

  • 식용해조인 둥근돌김(Perphyra suborbiculata), 잎파래 (Enteromorpha linza), 구멍갈파래 (Ulva pertusa) 미역 (Undaria pinnatifida, 자연생 및 양식), 모자반 (Sargassum fulvellum), 쎌만모자반(Sargassum kjellmanianum) 누운첨각(Codium coarctatum)의 NaOH 가용성 단백질 추출에 영향을 미치는 제요소 즉, 추출용액의 농도, 용매량, 온도, 시간에 따른 조단백질과 순단백질(TCA 불용성 단백질)의 추출 결과를 요약하면 다음과 갈다. 1. 전시료가 0.05M NaOH 용액에서 가장 좋은 추출성적을 얻었으나, TCA 불용성 단백질은 $0.01\~0.025M$에서 추출성적이 좋았으므로, 0.025M에서 추출함이 좋을 것 간다. 2. 시료-용매비(w/v)는 건조시료 1g에 대하여 100ml의 용매를 첨가함이 좋은 것 같다. 3. 추출시간은 녹조류는 1시간, 양식미역 2시간, 자연생미역, 쎌만모자반, 모자반은 3시간에 최고의 추출성적을 얻었다. 4. 추출온도는 전시료가 $60^{\circ}C$에서 최고의 추출성적을 보였다.

한려해상국립공원의 해조상 (Algal Flora in Hallyeo-haesang National Park, Southern Coast of Korea)

  • This study elucidated the floral composition of marine algae and community structure at Hallyeo-haesang National Park, on the southern coast of Korea. In all, 89 species, comprising 10 green, 30 brown and 49 red algae, were identified. The dominant species in terms of importance value were Ulva pertusa, Colpomenia sinuosa, Undaria pinnatifida, Padina arborescens, Hizikia fusiformis, Sargassum sagamianum and Amphiroa dilatata. The vertical distribution of marine vegetation was characterized by Enteromorpha linza - Ulva pertusa - Gelidium divaricatum in the upper intertidal zone, Hizikia fusiformis - Sargassum thunbergii in the middle intertidal zone, and Amphiroa spp. - Hildenbrandtia rubra - Corallina pilulifera in the lower intertidal zone. Functional form group analysis showed that coarsely branched forms comprised 50.3% of the algal community, whereas thick leathery forms, sheet forms and filamentous forms comprised 11.9-13.1%. R/P, C/P and (R+C)/P values were 1.91, 0.74 and 2.64, respectively. A cluster analysis of species occurrence suggested that the number of marine algal species differed greatly among the sampling sites.

  • The appearence of organochlorine pesticide residues in some marinealgae, e.g. Sargassum thunbergii, Ulva pertusa, Codium fragile, and Enteromorpha linza, collected from various sites of Kwang Yang Bay, southern coast of Korea, was surveyed through the year from May 1974 to March 1975. The residues were found to appear in the order to July>September>May>November≒March≒January; $\alpha$-BHC>heptachlor epoxide>dieldrin>aldrin>endrin; and in the range of 0-93 $\mu\textrm{g}$/kg dry matter.

오동도 해조군락에 관한 연구 (A Study on the Algal Communities of Odongdo, Southern Coast of Korea)

  • 본 연구는 1982년 6월부터 1983년 5월까지 전남 여수반도 남단에 위치한 오동도 해조군락의 특성을 밟히기 위하여 5개의 line transect를 설치하고 연간 계절별로 조사된 자료를 분석 하였다. 해조류의 수직분포는 상부군이 Gelidium divaricatum, Enteromorpha linza, Porphyra yezoensis, Scytosiphon lomentaria, Blidingia nana, Ectocarpus confervoides, 중부군이 Ulva pertusa, Chondria crassicaulis, 그리고 하부군은 Sargossum sagamianum, Undaria pinnatifida, Sargassum thunbergii 및 Gelidium amansii 등의 홍조류로 대표되었다. 출현해조의 종수와 피도는 4월에 최대이고 8월에 최소로되며 종별 우점도의 변화는 연중 출현종은 10월에, 일계절 출현종은 1월 또는 4월에 각각 최대치를 나타내었다. 유사도지수에 따른 5개 transect의 cluster analysis 결과 내만성군과 외양성의 2군으로 분류되었으며 본 지역은 남해안 중${\cdot}$동부 군락에 해당되는 Myelophycus simplex alliance의 Colpomenia sinuosa-Ecklonia cava association에 속한다고 본다.

서해 경기만 해조군집 복원을 위한 기초생태조사 (Primary Survey on Algal Community of Gyounggi Bay for Restoration)

  • For getting primary cue of restoration, the algal flora and intertidal community structure of Deokjeokdo, Yeonpyeongdo and Ganghwado were investigated for from February to November 2006. The total of 28 species, including five Chlorophytes, five Phaeophytes, 17 Rhodophytes and one sea grass were identified. The occurrence of species according to sampling site was 22 species in Deokjeokdo, seven species in Yeonpyeongdo and four species in Ganghwado. Among them Gloiopeltis furcata, Ulva pertusa and Sargassum horneri in Deokjeokdo, Scytosiphon lomentaria in Yeonpyeongdo, Enteromorpha prolifera in Ganghwado were ominants. The algal zonation of intertidal zone was figured out by Gloiopeltis furcata, Caulacanthus okamurae – Corallina pilulifera – Ulva pertusa, Ahnfeltiopsis flabelliformis, Sargassum horneri, Undaria pinnatifida in Deokjeokdo, Bangia atropurpurea – Scytosiphon lomentaria – Ahnfeltiopsis flablleiformis in Yeonpyeongdo from upper to lower zone and Enteromorpha prolifera, Enteromorpha linza – Sargassum thunbergii, Myelophycus simplex in Ganghwado from middle to lower zone. The average of biomass was measured as 80.6 g dry wt. m–2 in Deokjeokdo, 32.2 g dry wt. m–2 in Yeonpyeongdo and less than 1 g dry wt. m–2 in Ganghwado.

한국 남해안 중부에 위치한 광양만 조간대의 해조상과 군집 (Composition of Marine Algal Community at the Intertidal Zone in Gwangyang Bay, South Sea, Korea)

  • The seasonal variation and vertical distribution of the marine algal community were investigated using the quadrat method from February 2006 to January 2007 at 10 sites in Gwangyang Bay, Korea. In total, 48 red, 17 brown, and 13 green algal taxa were identified. The algal vertical distribution in the intertidal zone was characterized by Ulva pertusa, while the upper tidal zone was dominated by Gelidium divaricatum. The middle zone was primarily composed of Enteromorpha linza, Sargassum thunbergii, and Chondracanthus intermedia, whereas Hizikia fusiformis, Sargassum thunbergii, Gelidium amansii, and Corallina pilulifera characterized the low tidal zone. Results of cluster analysis indicated that algal composition fell into two groups, which were composed of species in the inland and open sea. In conclusion, the number of species, diversity, and abundance of vegetation in this area were remarkably reduced compared to previous studies in Gwangyang Bay.

독도의 해조상 및 군집구조 (Marine Algal Flora and Community Structure in Dokdo, East Sea, Korea)

  • We investigated species composition and variation of benthic marine algae between 2003 and 2006 at Dokdo on the East Sea. The total number of benthic algae species identified was 96, which included 15 Chlorophyta, 30 Phaeophyta, 50 Rhodophyta and 1 Spermatophyta species. The representative species in this study were Ulva pertusa, Caulerpa okamurae, Codium adhaerens, C. frgile, Undaria pinnatifida, Ecklonia cava, Eisenia bicyclis, Dictyopteris undulata, Padina arborescens, Sargassum horneri, Hildenbrandtia rubra, Lithophyllum okamurae, Amphiroa dilatata, Corallina officinalis, C. pilulifera, Prionitis cornea, Grateloupia elliptica, Plocamium telgairiae, Chondrus ocellatus, Chondracanthus intermedia, Chondria crassicaulis, Polysiphonia morrowii, Melobesioidean algae and Phyllospadix sp. Wet weight biomass according to various depths ranged between 1,094.8 to $2,595.3\;g\;m^{-2}$ during the study period. Mean biomass at the investigated sites was greater in the 15m depth range than in the 5 and 10m depths. Vertical distribution was characterized by Ulva intestinalis, U. linza, Endarachne binghamiae, Bangia atropurpurea, Gloiopeltis furcata and Chondria crassicaulis at intertidal zone, Amphiroa dilatata, Corallina pilulifera, Gelidium amansii, Eisenia bicyclis and Ecklonia cava at 2m depth, Ecklonia cava, Eisenia bicyclis, Padina arborescens, Undaria pinnatifida and Sargasum horneri at 5m depth, Ecklonia cava, Sargassum horneri and Padina arborescens at 10m depth, Ecklonia cava, Myagropsis myagroides, Padina arborescens and Sargassum horneri at 15m depth. The R/P, C/P and (R+C)/P value were 1.67, 0.50 and 2.17, respectively.

Oxidative Stress and Antioxidant Activities of Intertidal Macroalgae in Korea

  • The oxidative stress level and antioxidant activities in two green algae (Ulva pertusa and Ulva linza), two brown algae (Agarum cribrosum and Dictyota dichotoma), and three red algae (Grateloupia lanceolata, Carpopeltis affinis, and Gracilaria verrucosa) collected from intertidal regions of Korea were assessed. In the two green algae, although the total glutathione content was not as high as that of the brown algae, the glutathione pool was extremely reduced, and the glutathione reductase (GRd)/glutathione peroxidase (GPx) activity ratio was high, which apparently plays an important role for protection against oxidative damage, as manifested by low lipid peroxidation. In the brown algae, which exhibited a low lipid peroxidation level that was comparable to the green algal species, the highest glutathione content, together with high GPx activity, appears to be the most important factor in their antioxidant protection. The red algal species exhibited extremely high lipid peroxidation levels. They also contained the lowest and most oxidized glutathione among the species, as well as the lowest GRd activity. In spite of the marked difference in the glutathione content, the significant difference in the activity of ${\gamma}$-glutamylcysteine ligase, the rate limiting enzyme for glutathione synthesis, among the species was not exhibited. Our results suggest that there is a significant difference in the levels of oxidative stress and antioxidant capacity among the algal species, and that the glutathione system, especially the efficiency of glutathione recycling, plays a vital role in antioxidative protection in algal species.

한국 일광만 저서 해조류의 해조상과 군집구조 (Flora and Community Structure of Benthic Marine Algae in Ilkwang Bay, Korea)

  • Marine algal flora and community structure were investigated seasonally at three sites in Ilkwang Bay on the southern east coast of Korea from May 2007 to February 2008. Total 103 species including 10 of green algae, 17 of brown algae, 76 of red algae were collected and identified. Among these species, 21 species were found throughout the year. Ulva pertusa, Enteromorpha linza, Grateloupia lanceolata, Chondracanthus intermedia and Caulacanthus ustulatus were distributed dominantly in upper intertidal zone. By contrast, crustose coralline algae, Grateloupia spp., Chondracanthus tenellus, Prionitis cornea and Sargassum spp. occurred predominantly in middle intertidal zone. Grateloupia spp., Sargassum spp., Ecklonia cava and Ulva pertusa were dominant in lower intertidal zone. Annual mean biomass in wet weight was 478.3 g m$^{-2}$. Maximum biomass was recorded in site 1 (731.8 g m$^{-2}$), and minimum was recorded in site 3 (78.5 g m$^{-2}$). The R/P, C/P and (R + C)/P value reflecting flora characteristics were 4.47, 0.59 and 5.06, respectively. Two groups produced by cluster analysis, one including sites 1, 2 and the other including site 3, showed meaningful difference in similarity, each other. Site 3 showed the limited species composition due to inflow of fresh water and absence of solid substratum. However, there was no significant difference between site 1 and site 2. In conclusion, the number of marine algae species and biomass in Ilkwang Bay were markedly reduced comparing with the previous studies. These suggest that a solution for reconstruction of the poor marine algal vegetation is considerably demanded.
