The Change of ATPase-positive Dendritic Cell and the Effect of Green Tea in Mouse Skin by Ultraviolet B Irradiation (자외선 B 조사에 의한 마우스 피부 ATPase 양성 가지세포의 변화 및 녹차투여의 효과)

  • In this study we assessed the influences of ultraviolet (UV) light B radiation on epidermal ATPase-positive dendritic cell (DC) and the effect of green tea treatment in ICR mouse. The extent of changes following 200 mJ/$cm^2$ (0.5 mW/sec) was studied at 0, 6, 12, 18, 24, 30 or 36 hours after exposure. SBCs were decreased by 6 hours after irradiation. There was tendency to decrease from 6 hours to 24 hours and had little further change from then to 36 hours after irradiation. The mice that received 0, 50, 100, 200, 300 or 400 mJ/$cm^2$ of UVB were examined 24 hours after irradiation. The DCs were decreased as the radiation dose increases from 100 to 400 mJ/$cm^2$. The frequency of UVB (200 mJ/$cm^2$)-induced DC decrease was reduced by treatment of green tea (i.p. and topical application, p<0.01).

  • The interest in the use of borate crystals in ultraviolet(UV) nonlinear optics(NLO) has increased because all solid-stste UV lasers obtained with NLO are in highly demand. Much effort has been spent on developing borates series, such as{{{{ { beta -BaB }_{2 } {O }_{4 }(BBO), {LiB }_{ 3}{O }_{5 }(LBO)}}}} and{{{{{ CsKiB}_{6 }{O }_{10 }(CLBO)}}}} in this decade. Recently another new borate crystals, {{{{{ YCa}_{4 }O({BO }_{3 })_{3}}}}} and{{{{{Gd }_{x }{Y }_{1-x }{Ca }_{4 }O({BO }_{ 3})({Gd }_{x }{Y }_{1-x }COB)}}}} have been developed by the present authors. Here, the growth and NLO properties of YCOB and {{{{ {Gd }_{ x} {Y }_{ 1-x} }}}}CO B crystal are reported and their properties discussed in relation to those of other nonlinear optical crystals, such as LBO.

Accumulation of Flavonols in Response to Ultraviolet-B Irradiation in Soybean Is Related to Induction of Flavanone 3-β-Hydroxylase and Flavonol Synthase

  • There are several branch points in the flavonoid synthesis pathway starting from chalcone. Among them, the hydroxylation of flavanone is a key step leading to flavonol and anthocyanin. The flavanone 3-${\beta}$-hydroxylase (GmF3H) gene was cloned from soybean (Glycine max cultivar Sinpaldal) and shown to convert eriodictyol and naringenin into taxifolin and dihydrokaempferol, respectively. The major flavonoids in this soybean cultivar were found by LC-MS/MS to be kamepferol O-triglycosides and O-diglycosides. Expression of GmF3H and flavonol synthase (GmFLS) was induced by ultraviolet-B (UV-B) irradiation and their expression stimulated accumulation of kaempferol glycones. Thus, GmF3H and GmFLS appear to be key enzymes in the biosynthesis of the UV-protectant, kaempferol.

Degradation of Natural Dyed Silk Fabrics under Ultraviolet Light(UV) -Focused on Gardenia and Sappanwood- (자외선에 의한 천연 염색 견직물의 취화 연구 -치자, 소목 염색을 중심으로-)

  • The purpose of this study was to investigate the degradation of silk fabrics dyed with gardenia and sappanwood by Ultraviolet Light(UV). To asses the effect of uv on dyed silk fabrics, physical and chemical properties of samples were explored. K/S value rapidly decreased with increasing exposure time. Mordanting improved ultraviolet-cut ability and the sappanwood dyed samples were superior to those of gardenia dyed in ultraviolet-cut ability. Color progressively faded away as uv exposure time increased, accordingly, $L^*,\;a^*,\;b^*$, H/VC, ${\Delta}E$ were changed. Morphological change observed with SEM represented degradation of silk fabrics from the outer fibril to the inner fibril. Tensile Strength abruptly decreased as uv exposure time increased and the mordanted samples showed higher tensile strength than the unmordanted. FT-IR analysis confirmed that main peaks at 3297 and $1704cm^{-1}$ band for silk fabric were due to N-H and C=O stretching, gardenia peaks at 1654 and $668cm^{-1}$ band representing C=O(ester), C=C(alken) and O-C=O(carboxylic acids) of crocin and sappanwood peaks at $1715cm^{-1}$ band representing C=O(cyclic keton) of brazilin appeared on the samples exposed for 14 days, but these peaks indicating colorants after 28 days of uv exposure faded away due to prolonged exposure of uv.

Overview of UV-B Effects on Marine Algae (자외선이 해조류에 미치는 영향에 관한 고찰)

  • Numerous observations revealed strong evidence of increased middle ultraviolet radiation or UV-B (280 ~ 320 nm) at the earth's surface resulting from stratospheric ozone depletion. UV is the waveband of electromagnetic radiation which is strongly absorbed by nucleic acids and proteins, thus causing damage to living systems. It has been recorded in the East Sea, Korea that solar UV-B impinging on the ocean surface penetrates seawater to significant depths. Recent researches showed that exposure to UV-B for as short as 2h at the ambient level (2.0 Wm$^{-2}$) decreased macroalgal growth and photosynthesis and destroyed photosynthetic pigments. These may suggest that UV-B could be an important environmental factor to determine algal survival and distribution. Some adaptive mechanisms to protect macroalgae from UV-damage have been found, which include photoreactivation and formation of UV-absorbing pigments. Post-illumination of visible light mitigated UV-induced damage in laminarian young sporophytes with blue the most effective waveband. The existence of UV-B absorbing pigments has been recognized in the green alga, Ulva pertusa and the red alga, Pachymeniopsis sp., which is likely to exert protective function for photosynthetic pigments inside the thalli from UV-damage. Further studies are however needed to confirm that these mechanisms are of general occurrence in seaweeds. Macroalgae together with phytoplankton are the primary producers to incorporate about 100 Gt of carbons per year, and provide half of the total biomass on the earth. UV-driven reduction in macroalgal biomass, if any, would therefore cause deleterious effects on marine ecosystem. The ultimate impacts of increasing UV-B flux due to ozone destruction are still unknown, but the impression from UV studies made so far seems to highlight the importance of setting up long-term monitoring system for us to be able to predict and detect the onset of large -scale deterioration in aquatic ecosystem.

Characterization of Arthrospira platensis Mutants Generated by UV-B Irradiation (자외선 조사에 의해 유도된 미세조류 Arthrospira platensis 변이주의 특성)

  • Arthrospira platensis (A. platensis) is an economically important filamentous microalgae. The isolation of mutants by chemical or physical mutagen is a clue for the strain improvement. In this study, effects of ultraviolet-B (UV-B) radiation on A. platensis were investigated. Cells (or microalgae) were exposed to UV-B (15 Watt, 254 nm) for 1, 3, 5, and 10 min, and resulting mutants were designated UM1, UM3, UM5, and UM10, respectively. Especially, the lipid content of UM5 mutant was considerably increased by 8~11 fold compared to that of wild types. Moreover, the carotenoid content and antioxidant enzyme (peroxidase and superoxide dismutase) activity were increased. It was shown that UV-induced mutants can accumulate bioactive compounds, which will be useful for the industrial production of valuable products.

Protective effect of Thymus quinquecostatus extracts UVB-induced matrix metalloproteinase-1 via suppressing MAPKs phosphorylation in human keratinocyte (사람의 각질세포에서 UVB 유도에 따른 MMP-1의 발현 조절과 MAPKs 인산화에 타임 추출물이 미치는 효과)

  • Ultraviolet rays are electromagnetic waves with a shorter wavelength than visible light, and ultraviolet rays that pass through the ozone layer are the main cause of skin aging. Chronic exposure of skin tissue to ultraviolet light activates the Mitogen-activated Protein Kinases (MAPKs) signaling pathways in human keratinocytes, resulting in increased production of matrix metalloproteinases (MMPs). In this study, we investigated the herbal extracts from Jeju Island on the anti-aging effect in human keratinocytes (HaCaTs) by ultraviolet stimulation. We examined that herb extract from Jeju Island were decreased in anti-aging activity on measuring the level of MMP-1 gene and protein expression in ultraviolet-induced keratinocytes. As a result, it was confirmed that Thymus quinquecostatus extract (TQE) significantly reduced the expression of MMP-1 in a dose-dependent manner by UV irradiated HaCaTs. According to our data, TQE significantly attenuated UV-induced phosphorylation of the MAPKs signaling elements ERK1/2, JNK1/2 and p38 proteins. These results suggest that the MAPKs pathway may contribute to the inhibitory effect of TQE on UV-induced MMP-1 production in human keratinocytes. Our results suggest that TQE may be a protective agent against skin aging by preventing UV-induced MMP-1 production.

Anti-Pigmentation Effects of Eight Phellinus linteus-Fermented Traditional Crude Herbal Extracts on Brown Guinea Pigs of Ultraviolet B-Induced Hyperpigmentation

  • We have previously found that mycelia culture broth of eight kinds of traditional herbal extracts fermented with Phellinus linteus (previously named as 8-HsPLCB) not only inhibited melanin and tyrosinase activity, but also reduced the contents of melanogenesis-related proteins, including tyrosinase and microphthalmia-associated transcription factor, in 3-isobutyl-1-methylxanthine-stimulated B16F0 melanoma cells. For a further study, the effect of 8-HsPLCB against skin pigmentation in brown guinea pigs with ultraviolet B (UVB)-induced hyperpigmentation was investigated. 8-HsPLCB (3%) and arbutin (2%) as positive controls were applied topically twice daily for 4 weeks to the hyperpigmented areas. 8-HsPLCB showed skin-lightening effect as effective as arbutin, one of the most widely used in whitening cosmetics. Melanin index values as the degree of pigmentation showed a significant reduction week by week post 8-HsPLCB treatment and then substantially reduced by 4 weeks. The degree of depigmentation after 4 weeks of topical application with 8-HsPLCB was 32.2% as compared with before treatment (0 week). Moreover, using Fontana-Masson staining and hematoxylin-eosin staining, 8-HsPLCB reduced melanin pigmentation in the basal layer of the epidermis and epidermal thickness changes exposed to the UV-B irradiation as compared with non-treatment and vehicle treatment. The intensity of the skin-lightening effect of 8-HsPLCB was similar to arbutin. These results suggest that the skin-lightening effect of 8-HsPLCB might be resulted from inhibition of melanin synthesis by tyrosinase in melanocytes. To conclude, 8-HsPLCB treatment showed reduction of the melanin pigment and histological changes induced by UV irradiation in brown guinea pigs.

A Study on the Changes in the Property of Nylon Fabrics Dyed with Safflower Under Ultraviolet-Light(UV) - Focused on the Changes in Color, Strength, and Surface - (홍화 염색 나일론 직물의 자외선에 의한 특성 변화 - 색상, 강도 및 표면 특성 변화를 중심으로 -)

  • The purpose of this study was to investigate the changes in the property of nylon fabrics dyed with safflower red and yellow colorants under Ultraviolet(UV)-light. For this purpose, nylon fabrics dyed with safflower red and yellow colorants were compared with each other after uv-light exposure in terms of K/S value, color changes(${\Delta}E$), morphology, and strength retention. K/S value rapidly decreased with increasing exposure time, but K/S value of the samples dyed with safflower red colorants decreased less than that of samples dyed with safflower yellow colorants. In color changes, as increasing exposure time, $L^*$ increased, $a^*$ decreased, $b^*$ increased, and so ${\Delta}E$ increased in samples dyed with safflower red colorants. In color changes, as increasing exposure time, $L^*$ increased, $a^*$ and $b^*$ decreased, and thereby ${\Delta}E$ increased in the samples dyed with safflower yellow colorants, indicating fading away by uv-light and changes of hue, value and chroma value. But the color change of samples dyed with safflower yellow colorants was less than that of samples dyed with safflower red colorants. SEM pictures showed a severe degradation by uv exposure, regardless of colorants type. Tensile strength slowly decreased until 14 days, and rapidly decreased until 28 days. Strength retention of the samples dyed with safflower yellow colorants was higher than that of the samples dyed with safflower red colorants.

raf Proto-oncogene is Involved in Ultraviolet Response in Drosophila

  • Raf-1, a cytoplasmic serine/threonine protein kinase, serves as a central intermediate in many signaling pathways in cell proliferation, differentiation, and development. In this study, we investigated that B-raf, Drosophila homolog of the human c-raf-1, is involved in ultraviolet (UV) responsive events by using hypomorphic mutant $D-raf^{c110}$ and Draf-lacZ transgenic fly. At first, effect of UV damage on the survival of wild-type and $D-raf^{C110}$ strains was examined. In terms of $1/LD_{50}$ value, the relative ratio of UV sensitivities of wild-type versus $D-raf^{C110}$ strain was 1 : 2.2. By using quantitative $\beta$-galactosidase activity analysis, transcriptional activity of the D-raf gene promoter was also examined in UV-irradiated Draf-lacZ transgenic larvae. UV irradiation increased the expression of lacZ reporter gene in Draf-lacZ transgenic fly. However, in $D-raf^{C110}$ strain the transcriptional activity of D-raf gene promoter by UV irradiation was extensively reduced. Results obtained in this study suggest that D-raf plays a role in UV response, leading to better survival of Drosophila to UV damage.
