Photoprotective Effect of Topical EGb 761 and Korean Red Ginseng in C57BL/6 Mice

  • Exposure to UVB radiation can cause diverse biological photodamage to skin. Eeb 761 and Korean red ginseng are the major and most effective natural drug against a variety of oxidative damage. But, the protective effects against UVB radiation have not been clearly identified. In this study, we investigated the protective effect of topical EGb 761 and Korean red ginseng on pigmentation by UVB radiation. Pro-inflammatory cytokines($IL-l{\beta}$, IL-6 and $TNF-{\alpha}$) and melanogenesis proteins(tyrosinase, TRP-1 and TRP-2) mRNA were measured by reverse transcription-polymerase chain reaction(RT-PCR) analysis. The in vivo protection against pigmentation was calculated using chromameter. The mRNA level of IL-lf and TNF-a were increased by UVB irradiation in treated and non-treated group, while no significant changes were observed in IL-6 level. Topical treatment with EGb 761 and Korean red ginseng remarkably reduced expression of tyrosinase, TRP-1 and TRP-2 in the non-irradiated and irradiated skin. Application of EGb 761 and Korean red ginseng significantly protected the WB-induced skin pigmentation and Korean red ginseng was more effective. Our study suggests that topical ECb761 and Korean red ginseng can regulate melanogenic proteins and protect UVB radiation on skin pigmentation.

자외선에 의한 비타민 $D_3$ 합성과 직물(제2보) -동물 실험을 통하여 - (UVB Photosynthesis of Vit, D3 and Fabrics (Part ll) - The animal study -)

  • Because there is a great concern' today about the damaging effect of chronic exposure to sunlight the use of sunscreen providing the photoprotection effect against ultraviolet (UV) was widely increased. As a result of common use of level of photosynthetic Vit. D3 in human skin decreased these days. In our experiment the animals covered with fabrics with 50% (fabric B) and 100% (fabric A) protection rate against ultraviolet B (UVB) were used to measure serum 25(OH)D3, ALP, total clacium and phosphorus. Vitamin D deficiency diet group had no effect on concentration of serum phosphorus. But the concentrations of serum 25(OH)D9 and total calcium were more decreased in vitamin D deficiency diet rats than in normal diet rats. Alkaline phosphatase activity in sunlight irradiated groups covered with 50% (fabric B) and 100% (fabric A) WB protection fabrics was more significantly decreased than vitamin D deficiency diet group. In conclusion, sunlight irradiateted groups were compared to effective to protect born disease due to the Vit. D deficeincy group.

Phloroglucinol Attenuates Ultraviolet B-Induced 8-Oxoguanine Formation in Human HaCaT Keratinocytes through Akt and Erk-Mediated Nrf2/Ogg1 Signaling Pathways

  • Ultraviolet B (UVB) radiation causes DNA base modifications. One of these changes leads to the generation of 8-oxoguanine (8-oxoG) due to oxidative stress. In human skin, this modification may induce sunburn, inflammation, and aging and may ultimately result in cancer. We investigated whether phloroglucinol (1,3,5-trihydroxybenzene), by enhancing the expression and activity of 8-oxoG DNA glycosylase 1 (Ogg1), had an effect on the capacity of UVB-exposed human HaCaT keratinocytes to repair oxidative DNA damage. Here, the effects of phloroglucinol were investigated using a luciferase activity assay, reverse transcription-polymerase chain reactions, western blot analysis, and a chromatin immunoprecipitation assay. Phloroglucinol restored Ogg1 activity and decreased the formation of 8-oxoG in UVB-exposed cells. Moreover, phloroglucinol increased Ogg1 transcription and protein expression, counteracting the UVB-induced reduction in Ogg1 levels. Phloroglucinol also enhanced the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2) as well as Nrf2 binding to an antioxidant response element located in the Ogg1 gene promoter. UVB exposure inhibited the phosphorylation of protein kinase B (PKB or Akt) and extracellular signal-regulated kinase (Erk), two major enzymes involved in cell protection against oxidative stress, regulating the activity of Nrf2. Akt and Erk phosphorylation was restored by phloroglucinol in the UVB-exposed keratinocytes. These results indicated that phloroglucinol attenuated UVB-induced 8-oxoG formation in keratinocytes via an Akt/Erk-dependent, Nrf2/Ogg1-mediated signaling pathway.

자외선 B를 조사한 마우스 표피멜라닌세포 변화에 대한 분죽(Phyllosrachys nigra var. henenis Strapf)잎 추출물의 효과 (The Effect of Bamboo (Phyllostachys nigra var. henenis Strapf) Leaf Extract on Epidermal Melanocytes in Ultraviolet B-irradiated Mice)

  • C57BL/6 마우스에서 자외선 B(UVB)조사에 의한 표피 멜라닌세포의 변화에 대한 대나무(분죽, Phyllostachys nigra var. henenis Suapf ) 잎 추출물 (BLE)의 효과를 관찰하였다. 마우스에 UVB를 매일 $80mJ/cm^2(0.5mW/sec)$씩 7일간 조사하고 BLE를 UV조사 전 또는 조사 후에 복강내주사 또는 피부에 도포하여 멜라닌세포 형성 억제효과 및 형성된 멜라닌세포에 대한 미백효과를 dihydroxyphenylalanine (DOPA) 염색으로 관찰하였다. 마우스의 귀등쪽 표피를 분리하여 관찰한바, 정상대조군에서는 $mm^2$당 11-16개의 멜라닌세포가 관찰되었으며, UV조사 일주일 후 발달된 가지를 가진 DOPA 양성 멜라닌세포는 급격히 증가하였다. 멜라닌세포 형성 억제 실험에서는 평균치를 기준으로 복강내 주사군에서 29.9%, 피부도포군에서 33.3%의 유의성 있는 억제 효과가 관찰되었으며, 형성된 멜라닌세포의 감소 효과 실험에서는 평균치를 기준으로 복강내 주사군의 경우 6주에 24.4%의 감소 효과가 관찰되었고, 피부도포군의 경우 3주에 21.0%, 6주에 10.4%의 유의성 있는 감소 효과를 보였다. 이상의 결과는 BLE가 UV에 의한 멜라닌세포 형성 억제제 및 미백제로서의 적용 가능성을 제시하였다.

인체 피부 섬유아세포에서 자외선 조사에 대한 다양한 곡류 메탄올 추출물의 보호 효과 (Protective Effect of Various Grain Methanolic Extracts against UVB-induced Photo-aging in Human Skin Fibroblasts)

  • 본 연구는 국내에서 주로 소비되는 곡류를 사용하여 UVB를 조사한 인체 피부 섬유아세포에서 항산화 효능과 MMP-1 및 수용성 콜라겐의 변화를 측정하여 광노화 억제 효능을 평가하였다. 또한 곡류 추출물의 항산화 활성 및 항산화 성분을 측정하고, 항산화력과 항산화 성분과의 상관성을 비교 분석하였다. 7종의 곡류 추출물(백미, 현미, 보리, 율무, 귀리, 수수, 흑미)은 $30mJ/cm^2$ UVB에 노출된 인체 피부 섬유아세포에서 광노화에 대해 세포 보호효과가 있는 것을 확인하였다. 또한 곡류 추출물의 처리는 UVB 조사에 따른 MMP-1의 생성을 감소시키고, 수용성 콜라겐의 분해를 저해하였으며, ROS의 생성도 억제하였다. 7종의 곡류 메탄올 추출물에 대한 총 폴리페놀 함량을 분석한 결과, 흑미와 수수가 다른 곡류에 비해 높은 항산화 성분을 함유하고 있었으며, DPPH 및 ABTS 라디칼 소거능도 높은 활성을 나타냈다. 곡류 추출물의 항산화 성분과 항산화 활성과의 상관분석 결과, 폴리페놀 함량이 높을수록 DPPH 라디칼 소거능($R^2=0.9936$), ABTS 라디칼 소거능($R^2=0.9456$)이 증가하는 것으로 나타났다. 따라서 곡류 추출물은 항산화 성분을 다량 함유하고 높은 항산화 활성을 지녔으며, UVB 조사에 의한 ROS로부터 보호하여 MMP-1의 생성을 감소시키고, 수용성 콜라겐의 분해를 억제시켜 광노화 보효호과를 나타내는 것을 확인하였다. 본 연구 결과, 곡류 추출믈의 항산화 및 광노화 보호효능 입증으로 곡류 추출물을 항주름 기능성 소재에 활용할 수 있을 것으로 판단된다.

Antioxidative and Protective Activity of Polysaccharide Extract from Artemisia iwayomogi Kitamura Stems on UVB-Damaged Mouse Epidermis

  • Polysaccharide (PS) was fractionated from hot-water extract of Artemisia iwayomogi Kitamura stems. PS showed considerably higher hydroxyl radical scavenging activity than caffeic acid and glutathione. PS showed lower superoxide anion radical scavenging activity than hydroquinone and ascorbic acid. The scavenging activity of PS on the reactive oxygen species (ROS) induced by human neutrophils with zymosan was determined by the lucigenin-enhanced chemiluminescence assay. The scavenging effect of the PS on ROS as determined by the chemiluminescence assay was about 2-fold stronger than that of ascorbic acid at the same concentration. PS significantly decreased protein carbonyl and malonaldehyde contents in UVB irradiated skin homogenates, which was comparable to glutathione at the same concentration. This result suggested that PS derived from A. iwayomogi Kitamura stems may be a potent candidate as functional compound for the protection on UVB induced skin damage in cosmetics.

Effects of EGb 761 and Korean Red Ginseng on Melanogenesis in B16F10 Melanoma Cells and Protection Against UVB Irradiation in Murine Skin

  • These days there is a constant possibility of exposure to UV radiation which can cause abnormal production of melanin and result in skin disease such as hyperpigmentation and melanoma. Many materials were investigated for skin whitening and protection against UV radiation. In this study, we assessed the melanogenesis inhibitory activities of Korean Red Ginseng (KRG, Ginseng Radix Rubra) and Ginkgo (EGb 761 Ginkgo Biloba) in an attempt to develop a new skin whitening agent derived from natural products. B16F10 melanoma cells were treated for 48 hr with KRG and EGb 761. The inhibitory effect on melanogenesis was measured and related cytokines and proteins expression were also investigated by RT-PCR and Western blotting. In addition, we also assessed the effects of these substances on the skin of C57BL/6 mice. Cell growth, melanin content and tyrosinase activity were inhibited effectively in B16F10 melanoma cells treated with KRG and EGb 761. Moreover, tyrosinase mRNA expression was inhibited clearly and melanogenesis related proteins (MRPs) containing tyrosinase, TRP1 and TRP2 were also reduced by KRG and EGb761, while cytokines such as IL-$1{\beta}$ and IL-6 were induced. In the case of UV irradiated mice, we observed induction of cytokine mRNA levels and reduction of MRPs mRNA expression. In addition, a decrease in pigmentation from treatment with KRG and EGb 761 on the skin of mice was observed. These results indicate that KRG and EGb 761 inhibit melanogenesis in B16F10 cells and have display protective activities against UVB. Therefore, we suggest that KRG and EGb 761 are good candidates to be used as whitening agents and UVB protectors for the skin.

The Evaluation on the Effectiveness as a Cosmetic Material of Ascidian shell Extract Using Zebrafish model

  • The extracts of AS contain in alloxanthin, halocynthiaxanthin, astaxanthin and 13 kinds of carotenoids. The aim of the study was to assess the anti-oxidant activity and cell viability of AS. The anti-oxidant activity was determined by using DPPH radical inhibition activity and superoxide dismutase (SOD)-like activity. The results of cell viability assay showed that the extracts from AS were cytotoxic at concentrations above $5.0mg/m{\ell}$. This study was designed to examine inflammation induced by LPS, protection effect by UVB and the toxicity of Ascidian shell extract(ASE) as a functional cosmetic ingredient. Evaluation of embryo toxicity resulted in embryo coagulation and mortality when treated at 5.0, 10.0, $20.0mg/m{\ell}$. At the lowest concentration of $1.0mg/m{\ell}$, hatchability resulted in 100.0 % rate. The results of arrhythmia measurement in larvae showed similarity to the evaluation of embryo toxicity. This result demonstrated that toxicity is present at concentrations greater than $5.0mg/m{\ell}$. The protective effect of ASE on LPS and UVB-induced in the zebrafish was investigated. Intracellular reactive oxygen species(ROS) generated by the exposure of zebrafish to LPS, UVB-radiation were significantly decreased after treatment with ASE at $0.1mg/m{\ell}$. As a result, ASE similarly reduced UVB-induced ROS generation and cell death in live zebrafsih. Therefore, it is suggested that ASE has anti-Inflammatory effects and can possibly be used as a functional substance for skin protection in the future.

UVB 로부터 ROS를 유도한 제브라피쉬에 스피룰리나 에탄올 추출물의 항산화 및 보호효과에 관한 연구 (Anti-oxidative and protective effects of Arthrospira platensis ethanol extracts on zebrafish ROS Induced by UVB Induction)

  • 스피룰리나는 지구상의 가장 오래된 조류중 하나이며 피코시아닌, 토코페롤, 베타카로틴을 함유하고 있어 노화를 방지한다고 보고된 바 있다. 본 연구에서는 재배방법이 다른 스피룰리나 에탄올추출물의 UVB로 유도된 활성산소종(reactive oxygen species, ROS)의 저해와 항산화효과를 확인하였다. 실내배양 스피룰리나 에탄올추출물(ICAE)과 야외배양 스피룰리나 에탄올추출물(OCAE) 0.1, 0.5, $1mg/m{\ell}$의 DPPH 라디칼 소거능과 SOD 유사 활성을 측정하여 항산화효과를 확인하였다. UVB로 유도된 ROS의 저해 효과는 제브라피쉬 배아와 HaCaT 세포를 이용하여 확인하였다. ICAE와 OCAE 0, 0.01, 0.05, 0.1, 0.5, $1mg/m{\ell}$을 제브라피쉬 배아와 HaCaT 세포에 처리하고 UVB로 ROS를 유도하였고 DCFH-DA로 염색하여 세포내 ROS의 양을 검출하였다. 항산화효과를 측정한 결과 양성대조군인 ascorbic acid의 DPPH 라디칼 소거능은 73%, SOD 유사 활성은 86%로 나타났다. ICAE와 OCAE $1mg/m{\ell}$의 농도에서 각각 43, 57%의 DPPH 라디칼 소거능을 나타내었고, 20, 19%의 SOD 유사 활성을 나타내어 양성대조군인 ascorbic acid보다 낮았지만 유의적인 항산화효과가 있었다. UVB로 유도된 제브라피쉬 배아와 HaCaT 세포의 ROS가 음성대조군보다 증가하였고, ICAE와 OCAE 처리된 군은 농도 의존적으로 UVB만 조사된 양성대조군 보다 ROS가 감소하였다. 본 연구의 결과는 스피룰리나 에탄올추출물이 피부보호용 화장품 소재로 사용가치가 있음을 시사한다.

잘피 추출물의 UVB로 손상을 유도한 각질형성세포에 대한 항염 효능 (Anti-inflammation effect of extract from Zostera marina using UVB-induced damage on keratinocytes)

  • Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin.Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages.Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited.Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.
