Photosensitized Lysis of Egg Lecithin Liposomes by L-Tryptophan and N-Acetylphenylalanyl-L-Tryptophan

  • The photosensitized lysis of egg lecithin lipid membranes (liposomes) have been performed to UV-B light (270-320 nm) by L-tryptophan(L-Trp) and its peptide such as N-acetylphenylalanyl-L-tryptophan(NAPT) incorporated in the liposomes(ca. 0.1% by weight) or in the external buffer (0.1-0.3 mM). Requirement of oxygenation suggests that the lysis of liposomes is caused by the photosensitized oxidation of lipids. There was significant protection against lysis photosensitized by Trp in the external buffer by low concentration of ferricyanide (0.8 mM), but there was no effect on the lytic efficiency by $N_3^-$ which is singlet oxygen($^1O_2$) quencher, indicative of an electron transfer mechanism involved in the photosensitization. The small change of the lytic efficiency with increasing pH from 4 to 9 was interpreted by large target theory and subsequently indicates that superoxide($O_2^-$) may be an active intermediate for the oxidation. The efficiency of photosensitization of Trp was higher than that of NAPT under the same experimental condition. The weak lytic efficiency of liposomes photosensitized by NAPT was enhanced by incorporating NAPT in liposomes, but it was again quenched by ${\beta}$-carotene incorporated in the bilayer of liposomes. These results indicate that a portion of liposome lysis may be due to $^1O_2$ formation from the excited NAPT.

An investigation of the nuclear shielding effectiveness of some transparent glasses manufactured from natural quartz doped lead cations

  • The influence of lead cations on natural quartz (QZ) from Egypt as a glass shielding material for the composition with nominal formula (10Na2O - (90 - x) QZ - xPbO (where x = 30, 35, 40, 45 and 50 mol %)) was examined. The studied samples are synthesized via the melt quenching method at 1050 ℃. The X-ray diffraction XRD patterns were confirmed the glass nature for studied samples. Moreover, the optical properties, and the transparency for all compositions were examined by UV-Vis spectroscopy. Also, the major elemental composition of the natural quartz were estimated via the X-ray fluorescence (XRF) technique. Further, the density and molar volume were determined. Furthermore, the nuclear shielding parameters such as, mass attenuation coefficient, effective atomic number, electronic density, the total atomic, and electronic cross sections as well as the mean free path, and the half value layer with different gamma ray energies (81 keV-1407 keV) were calculated. Besides, the results showed that the shielding behavior towards the gamma ray radiation for all glass samples was increased as the increment in PbO concentration in the glass system.

Dyeability and Functionality of Wool Fabrics Dyed with Zizania latifolia Turcz. extract (줄풀염색에 의한 모직물의 염색성과 기능성)

  • This study investigated the proper dyeing conditions, color fastness and functionality of wool fabrics dyed with Zizania latifolia Turcz. We also tried to improve light fastness through treatment with benzophenone ultraviolet absorber. The dyeing of wool fabrics using Zizania latifolia Turcz was good even without pretreatment or mordanting treatment. Optimal wool fabric dyeing conditions were colorant concentration of 200% (o.w.f.), dyeing temperature of $100^{\circ}C$, dyeing time of 80 minutes and a dye bath pH of 3. Color fastness of dyed wool fabrics to washing, rubbing, perspiration and light was 4-5, 5, 4-4-5 (acidic), 4-5 (alkaline) and 2 respectively. The results after treatment with ultraviolet absorber for improving the fastness of daylight were improved to 3-4 grade. The UV protection rate were increased after dyeing and the deodorization of ammonia gas improved to 98%. Bacterial reduction rate (Staphylococcus aureus) of wool fabrics was excellent at 99.9%. All dye fastness (except for light fastness) was excellent; in addition, the functionality of wool fabrics dyed with Zizania latifolia Turcz also improved. The results are expected to be applied to various fields because they indicate excellent results after treatment with ultraviolet absorber for improving the fastness of daylight.

Fabrication and Processing Method of Ophthalmic Hydrogel Tinted Lens Containing Indium Tin Oxide-Composited Materials

  • In this study, a multifunctional ophthalmic lens material with an electromagnetic shielding effect, high oxygen permeability, and high water content is tested, and its applicability is evaluated. Metal oxide nanoparticles are applied to the ophthalmic lens material for vision correction to shield harmful electromagnetic waves; the pyridine group is used to improve the antibacterial effect; and silicone substituted with urethane and acrylate is employed to increase the oxygen permeability and water content. In addition, multifunctional tinted ophthalmic lens materials are studied using lens materials with an excellent antibacterial effect (2,6-difluoropyridine, 2-fluoro-4-pyridinecarboxylic acid) and functional (UV protection, high wettability) lens materials (2,4-dihydroxy benzophenone, 2-hydroxy-4-(methacryloyloxy)benzophenone). To solve problems such as air bubbles generated during the polymerization process for the manufacturing and turbidity of the lens surface, polymerization conditions in which the defect rate is minimized are determined. The results show that the polymerization temperature and time are most appropriate when they are $110^{\circ}C$ and 40 minutes, respectively. The optimum injection amount of the polymerization solution is 350 ms. The turbid phenomenon that appears in lens processing is improved by 10 to 95 % according to the test time and conditions.

  • "Eating cosmetic" market comes to prominence worldwide. Inner Beauty has established itself as the top trend nowadays and is leading the health functional food market. Another noticeable trend other than beauty-from-within is that products using "plant-based" natural botanical raw material are increasing. Based on these trends above, we have developed a plant extract named Agatri® for skin health using Agastache rugosa. By ingesting Agatri®, it is expected that the formation of skin wrinkles is suppressed, and skin elasticity is increased by suppressing the decrease in collagen content caused by ultraviolet rays. At the same time, it is possible to prevent skin aging by improving skin hydration and trans-epidermal water loss (TEWL). The studies show that it significantly increased production of hyaluronic acid and collagen, and decreased production of matrix metalloproteinases (MMPs). And Agatri® has been approved as an "Individually recognized functional ingredient for skin health by the Ministry of Food and Drug Safety, Korea in 2020. As a result, Agatri® is a good solution for those who are in need of skin protection from exposure to UV and aging.

Photoprotection and Anti-inflammatory Effects of Chinese Medical Plants (약용식물추출물의 광보호 효과와 항염증 효과 연구)

  • Chronic exposure to solar radiation, particularly ultraviolet (UV) light, causes a variety of adverse reactions on human skin, such as sunburn, photoaging and photocarcinogenesis. Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental facts play critical roles in cellular damage. And, repeated-UV irradiation activated the expression of the matrix metalloproteinase (MMP) and induced skin irritation. Therefore, the development of effective and safe photoprotectants that can reduce and improve the skin damage has been required. The purpose of this study was to investigate the photo-protective effect of several chinese medical plants (Juniperus chinensis) on the UV -induced skin cell damages. We tested free radical and superoxide scavenging effect in vitro. Fluorometric assays of the proteolytic activities of MMP-1 (collagenase) were performed using fluorescent collagen substrates. UVA induced MMP-1 synthesis and activity were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in skin fibroblasts. We also examined anti-inflammatory effects by the determination test of proinflammatory cytokine, interleukin 6 in HaCaT keratinocytes. Expression of prostaglandin E$_2$ (PGE$_2$) after UVB irradiation was measured by competitive enzyme immunoassay(EIA) using PGE$_2$ monoclonal antibody. In the human skin we tested anti-irritation effect on the SLS-induced damage skin after appling the extract containing emulsion. We found that Juniperus chinensis extract had potent radical scavenging effect by 98% at 100$\mu\textrm{g}$/mL. The extract of Juniperus chinensis showed strong inhibitory effect on MMP-1 activities by 97% at 100 $\mu\textrm{g}$/mL and suppressed the UVA induced expression of MMP-1 by 79% at 25$\mu\textrm{g}$/mL. This extract also showed strong inhibition on MMP-2 activity in UVA irradiated fibroblast by zymography. In the test of proinflammatory cytokines of human keratinocytes Juniperus chinensis extract decreased expression of interleukin 6 about 30%. The amount of PGE$_2$ by HaCaT keratinocytes was significantly increased at the doses of above 10 mJ/$\textrm{cm}^2$ of UVB (p < 0.05). At the concentrations of 3.2-25$\mu\textrm{g}$/mL of this extract, the production of PGE$_2$ by HaCaT keratinocytes (24 h after 10mJ/$\textrm{cm}^2$ UVB irradiation) was significantly inhibited in culture supernatants (p < 0.05). In SLS-induced skin irritation model in vivo, we found to reduce skin erythema and improve barrier recovery after appling Juniperus chinensis extract containing emulsion when compared to irritated non-treated and placebo-treated skin. Our results suggest that Juniperus chinensis extract can be effectively used for the prevention of UV and SLS-induced adverse skin reactions and applied as anti-aging and anti-irritation cosmetics.

Antioxidation and Anti-photoaging Effects of White Taraxacum Coreanum Extract by Lactobacillus plantarum (Lactobacillus plantarum에 의한 흰 민들레 발효물의 항산화 및 광노화 억제 효과)

  • Lactobacillus plantarum SM4, a strain producing β-glucosidase, was isolated from kimchi and fermented with white Taraxacum coreanum to enhance the production of bioactive compounds. The total polyphenol content(TPC), total flavonoid content(TFC), radical scavenging activity(RSA), tyrosinase inhibitory activities(TIA), and collagenase inhibitory activities(CIA) were measured to evaluate the skin whitening and anti-wrinkle effects of the fermented product. The TPC of fermented white T. coreanum was 41.8±0.26 mg GAE/g DW, which was approximately two times higher than the hot-water extraction of 21.4±0.67 mg GAE/g DW. RSA, an indicator of antioxidant activity, was 65.6±4.7% in fermentation, which is four times higher than that of the hot-water extract. TAI and CAI, which are indicators of the whitening and anti-wrinkle effects, were 87.9±4.73% and 66.7±3.48%, respectively, which were 2.4 and 1.5 times higher than those of hot-water extraction. When comparing the UVA(320 nm) protection effects of fermented and hot-water extraction, the fermented white T. coreanum showed higher protection with an absorption rate of 64.7% and 15.2%, respectively. The white T. coreanum fermented product showed higher bioactive properties and improved skin whitening, anti-wrinkle, and UV protection effects through the production of β-glucosidase from L. plantarum SM4.

Antioxidative and Protective Effects of Corn Silk (Zea mays L.) Extract on Human HaCaT Keratinocyte (옥수수수염 추출물의 항산화효과 및 피부각질세포 보호효과)

  • We investigated the antioxidative and protective effects of corn silk (Zea mays L.) ethanol extracts on human HaCaT cells and erythrocytes. The NICS-2 fraction, extracted from corn silk, exhibited favorable 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activities with $IC_{50}$valuesof$13.3{\pm}0.3{\mu}g/mL$ and $14.2{\pm}0.1{\mu}g/mL$ when compared with those of ${\alpha}$-tocopherol, a positive control, with $IC_{50}=10.4{\pm}02.2$ and $22.2{\pm}3.6{\mu}g/mL$, respectively. In addition, we investigated skin protection effects of NICS extracts of corn silk in HaCaT keratinocytes. To investigate the pharmacological potential of NICS-1 and NICS-2 extracts of corn silk on UV-B-induced damage in HaCaT cells, we measured the activity of interleukin (IL) 1a. Our results showed that all the corn silk extracts inhibited the UV-B-induced activity of IL-1a. In particular, NICS-1 extracts of corn silk significantly suppressed IL-1a activity in a dose-dependent manner without inducing cytotoxicity. These results indicate that the ethanol extracts of corn silk (Zea mays L.) could function as natural cytoprotective agents and antioxidants in biological systems, particularly the skin exposed to UV radiation, by protecting cellular membrane against reactive oxygen species (ROS).

Development of Glucoamylase & Simultaneous Saccharification and Fermentation Process for High-yield Bioethanol (고효율 바이오 에탄올 생산을 위한 당화효소 개발 및 동시당화발효 공정 연구)

  • The bioethanol for use as a liquid fuel by fermentation of renewable biomass as an alternative to petroleum is important from the viewpoint of global environmental protection. Recently, many scientists have attempted to increase the productivity of bioethanol process by developing specific microorganism as well as optimizing the process conditions. In the present study, which is based on our previous investigation on the pretreatment process, theproductivity of bioethanol obtained from simultaneous saccharification and fermentation (SSF) process was compared between various domestic materials including barley, brown rice, corn and sweet potato. Additionally, Solid glucoamylase (SGA; developed in Changhae Co.), from modified strain with UV, was used. The result was compared to commercial glucoamylase (GA). It was observed that the fermentation rate was increased together with the yield which can be derived from the final ethanol concentration. Especially, in the case of brown rice, compared to the experimental results using GA, the final ethanol concentration was 1.25 times higher and 18.4 g/L of the yield was increased. Also, the time required for reaching 95% of the maximum ethanol concentration is significantly reduced, which is approximately 36 hours, compared to 88 hours using GA. It means that SGA has excellent saccharogenic power.

Skin Care Effects of Green Tea (녹차의 피부보호효과)

  • Tea (Camellia sinenis) is a popular beverage consumed worldwide. Since green tea, mainly consumed in Asia, has various biological activities, green tea components became one of the most favorite candidates as a functional materials for cosmetics and functional foods. The biological activities of green tea for skin cue have been ranged from protection of epidermal cells to the stimulation of extracellular matrix (ECM) biosynthesis. Green tea polyphenols (GTPs), which are active ingredients of green tea, possess anti-inflammatory, anti-carcinogenic and immune potentiation properties as well as antioxidant. They also modulate intracellular signal transduction pathways. GTPs decrease ultraviolet (UV)-induced oxidative stress, thus suppress mitogen-activated protein kinase (MAPK) pathway and apoptosis in keratinocytes. In addition, GTPs prevent the Induction of inflammatory mediators, such as cyclooxygenase-2 (COX-2), interleukin-8 (IL-8), and vascular endothelial growth factor (VEGF) by tumor necrosis factor alpha $(TNF{\alpha})$ or chemical treatment in keratinocytes. GTPs treatment protects from chemical-or UV-induced skin tumor incidence in animal experiment. Besides, GTPs stimulate keratinocyte differentiation and proliferation of normal and aged epidermal cells, resectively, and suppress matrix metalloproteinases (MMPs) release. According to the progress of formulation study, green tea components will be guaranteed materials for the more effective skin cue products.
