Characterization and Antioxidant Activity of Gold Nanoparticles Synthesized using Bambusae Caulis in Taeniam extract (죽여 추출물로 합성한 금 나노 입자의 특성과 항산화 활성)

  • Green synthesis of gold nanoparticles(GNPs) considered more ecofriendly and cost effective than other chemical methods use of dangerous reagents and solvents, improved material and energy efficiency and enhanced design of non-toxic products. In this study, we developed a green synthesis method for using Caulis in Taeniam (BCT). BCT were characterized by UV-vis, Zetasizer, TEM, XRD, and FTIR. The antioxidant activity of BCT was determined by DPPH and ABTS radical-scavenging assays, and heme oxygenase-1 induction in RAW 264.7 macrophages. The resulting BCT appeared spherical with an average diameter of 67.171.39 nm The aAntioxidant activity was increased in a dependent manner. To conclude, the green synthesis of BCT-GNPs was successful, and it appers to be useful in the for future applications.

Induction of Prochloraz-Resistant Isolates of Botrytis cinerea in Vitro and Their Biological Properties (Prochloraz에 저항성인 잿빛곰팡이균(Botrytis cinerea)의 유기 및 이들 균의 생물학적 특성)

  • 1994~1995년 사이에 대전, 공주, 논산, 부여와 김해 등지에서 채집한 병든 딸기, 토마토, 오이에서 총 1,504 균주의 잿빛곰팡이병균을 분리하여 10 $\mu\textrm{g}$/ml의 약제를 넣은 PDA 배지에서 배양하면서 약제반응을 조사한 결과, benomyl에 대하여는 1,147(76.3%)균주, procymidone에 대하여는 614(40.8%)균주, prochloraz에 대하여는 189(12.6%)균주가 저항성으로 나타났다. UV 처리 및 약제 배지에서의 누대배양을 통하여 prochloraz저항성 균주를 선발하였는데, 이들의 생육 억제 최소 농도(MIC)는 8 $\mu\textrm{g}$/ml 이상이었고, 감수성균의 MIC는 2 $\mu\textrm{g}$/ml 이하였다. 병원력, 포자 발아율, 균핵형성과 같은 적응력과 관련된 생물학적 특성 비교에서 저항성 균주들은 감수성 균주보다 열등하거나 비슷한 정도로 나타났다.

  • A gene can be selectively overexpressed in E. coli by utilizing the phage T7 RNA polymerase's stringent recognition and active transcription of the T7 promoter. The T7 expression system was constructed such that the T7 RNA polymerase gene is under the control of lacUV5 promoter in one plasmid, and that the target gene, the promoterless chloramphenicol acetyltransferase (CAT) gene with E. coli ribosome binding site is under the control of T7 promoter in the other plasmid. Only the E. coli cells containing both plasmids show high resistance to chloramphenicol. When the copy number of the runaway plasmid containing the polymerase gene was varied by a temperature shift, amounts of the CAT protein synthesized upon induction was correspondingly changed as shown in SDS gel electrophoresis.

  • Induced androgenesis, a form of artificial parthenogenesis is an important tool for the generation and use of genetically isogenic or clonal fish strain. An optimized protocol for the genetic inactivation of mud loach (Misgurnus mizolepis) oocytes (i.e. production of androgenetic haploid) was developed using UV-irradiation. Various dose levels of UV significantly affected the fertilizing capacity of the eggs, hatchability of embryos and incidence of haploidy. Based on the extensive examinations of treatment conditions on embryo viability and haploid incidence, the optimum dose level of UV irradiation was turned out to be $10,800ergs/mm^2$ with 56.9% of hatching success and 94.6% of haploidy. The overall yield of putative androgen under optimized treatment condition was more than 50% out of total eggs inseminated. The success of androgenetic reproduction of haploid genome was verified by flow cytometry and PCR amplification of transgene that is exclusive to either one of parental sexes. However, a small portion $(8\sim11%)$ of presumed androgenetic haploid larvae was proven to contain residual DNA fragment(s) from maternal parent.

  • These studies were conducted to induce the available mutants in Takju yeasts by the irradiation of UV light. Two original strains(5-Y-5, 6-Y-6) using for irradiation of UV selected from 24 strains which were isolated from the Takju mashes And Nuruks collected from 12 local regions of Chungnam and Chungbuk provinces in Korea, and the irradiations to the yeasts with UV light were carried out at a distance 10-40cm from the sources of irradiation for 10-220 seconds. The purpose of this experiment is to report the effects of irradiating distances and times of UV light on the survival ratio of orginal yeasts, and the identification of two orginal yeasts and three mutants induced by the irradiation of UV light. The results were summarized as follows. 1) The effects of irradiating distances and times on the survival ratio on the yeasts were represented as follows. and acid productivity to the survival strains by the irradiation of UV light. The selected mutants were the strains 30-24, 40-27 which have more powerful fermentability about 10 percent than those of original strains and a strain 30-81 which have potential acid productivity. 3) The selected yeasts (5-Y-5, 6-Y-6) were identified to Saccharomyces cerevisiae by a taxonomic study of Lodder and the mutants(30-81, 40-27, 30-81) induced from above yeasts by the irradiation of UV light have almost same properties two orginal yeasts in the identical characteristics.

Spawning Induction and Egg Development of Surf Clam, Spisula sachalinensis (북방대합, Spisula sachalinensis의 산란유발 및 난발생)

  • In order to obtain the basic information for seedling production of surf clam, Spisula sachalinensis, spawning induction and egg development were investigated. $NH_4OH$ addition and serotonin injection could induce the spawning in surf clam. Water temperature rising, sperm suspension immersion, UV-ray irradiated seawater and $H_2O_2$ addition less affected on induction of spawning than $NH_4OH$ and serotonin did. On the other hand, males were more sensitive to the treatments than females. The response time to initial spawning in the case of $NH_4OH$ addition was $3\~4$ hours. However in the case of serotonin injection, it was within 5 minutes. The number of eggs released by $NH_4OH$ addition were significantly more than those released by serotonin injection. The serotonin injection induced higher rates of germinal vesicle breakdown than the $NH_4OH$ addition. Fertilizing and hatching rates of the eggs also were the similar results. Eggs of surf clam were demersal isolated eggs and averaging $65.2{\pm}±1.8\;{\mu}m$ in diameter after spawning. Optimum range of water temperature for the development of egg was $15\~20^{\circ}C$, The required time for development of D-shaped larvae was 42 hours at $15^{\circ}C$ and 27 hours at $20^{\circ}C$, respectively.

Comparison of different ploidy detection methods in Oncorhynchus mykiss, the rainbow trout

  • The objective of this study was to determine a simple and reliable ploidy identification protocol for the rainbow trout (RT), Oncorhynchus mykiss, in the field condition. To evaluate the ploidy level and compare different detection protocols, triploid RT and gynogenesis were induced by UV irradiation and/or heat shock. The hatching rate at day 30 was 85.2% and the survival rate at day 90 was 69.4% (fingerling). The sex ratio of female RT was 93.75% in the gynogenesis group, illustrating that the UV irradiation inactivated the sperm DNA. The hatching rate and survival rate were 82.0 and 74.7%, respectively, in the triploid-induced group. The triploid induction rate by heat shock procedure was 73.9%. Cytogenetic protocols for ploidy identification such as chromosome counting, erythrocyte nuclear size comparison, and analysis of nucleolar organizing regions (NORs) by silver staining were compared. Silver nitrate staining showed the greatest success rate (22/23 and 32/32 for the triploid-induced group and gynogenesis group, respectively), followed by erythrocyte nuclear size comparison (16/23 and 19/32 for the triploid-induced group and gynogenesis group, respectively) and, lastly, chromosome preparation (2/23 and 6/32 for the triploid-induced group and gynogenesis group, respectively) with the lowest success rate. Based on our findings, silver staining for RT ploidy identification is speculated to be highly applicable in a wide range of research conditions, due to its cost-effectiveness and simplicity compared to other numerous ploidy detection protocols.

  • During the screening of inhibitors of caspase-3 induction in U937 human monocytic leukemia cells from natural sources, Petasites japonicum, which showed a high level of inhibition was selected. The inhibiting compound was purified from the methanol extract by Sephadex LH-20 column chromatography and HPLC. The inhibitor was identified as [3-(3,4-dihydroxyphenyl)-2-oxopropyl]ester, petasiphenol, by spectroscopic methods of UV, EIMS, $^1H-NMR$, $^{13}C-NMR$ and HMBC. It showed a caspase-3 inducing inhibitory activity at $8\;{\mu}g/ml$ of $IC_{50}$ value with DNA fragmentation inhibition in U937 human leukemia cells. It also showed a free radical scavenging ability of 1,1-diphenyl-2-picrylhydrazyl.

  • Auxotrophic mutants were obtained by UV-irradiation to mycelium of Ganoderma lucidum. Induction rate of auxotrophs was 5.78%. Interspecific fusion products of protoplasts were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Ganoderma lucidum and Ganoderma applanatum. Fusion products were selected by means of the comparison with the mycelial growth rate and colony morphology. Fusion products were confirmed by mycelial morphology and esterase isozyme pattern. Some segregants were observed and fusion product produced fruit bodies.

Synthesis and Luminescence Properties of Sr/SmSi5N8:Eu2+ Phosphor for White Light-Emitting-Diode

  • Red-emitting nitride phosphors recently attracted considerable attention because of their high thermal stability and high color rendering index properties. For excellent phosphor of white light-emitting-diode, ternary nitride phosphor of $Sr/SmSi_5N_8:Eu^{2+}$ with different $Eu^{2+}$ ion concentration were synthesized by solid state reaction method. In this work, red-emitting nitride $Sr/SmSi_5N_8:Eu^{2+}$ phosphor was successfully synthesized by using multi-step high frequency induction heat treatment. The effects of molar ratio of component and experimental conditions on luminescence property of prepared phosphors have been investigated. The structure and luminescence properties of prepared $Sr/SmSi_5N_8:Eu^{2+}$ phosphors were investigated by XRD and photoluminescence spectroscopy. The excitation spectra of $Sr/SmSi_5N_8:Eu^{2+}$ phosphors indicated broad excitation wavelength range of 300 - 550 nm, namely from UV to visible area with distinct enhanced emission peaks. With an increase of $Eu^{2+}$ ion concentration, the peak position of emission in spectra was red-shifted from 613 to 671 nm. After via multi-step heat treatment, prepared phosphor showed excellent luminescence properties, such as high emission intensity and low thermal quenching, better than commercial phosphor of $Y_3Al_5O_{12}:Ce^{3+}$. Using $Eu_2O_3$ as a raw material for $Eu^{2+}$ dopant with nitrogen gas flowing instead of using commercial EuN chemical for $Sr/SmSi_5N_8:Eu^{2+}$ synthesis is one of characteristic of this work.
