A Study on Optimal Design of UV Contactor using an Optical Radiation Model (광학모델을 이용한 자외선 접촉조 최적 설계에 관한 연구)

  • Because of refractory property of light, the travel path of UV light becomes longer than the straight line and shorter solely in water as UV light passes sequentially through air, quartz and water. Note that water significantly absorbs UV light. Hence, UV intensity shall be estimated to be lower when refraction is neglected than it is considered. Reflection is also critical for the design of UV radiation system. While the reflection at the interface of air and quartz is low enough to ignore, it is too high to be ignored at the interface of quartz and water. Assuming constant power, smaller length to width ratio of UV reactor is beneficial and single-lamp system is preferred to multi-lamps. Under the given cross section, optimal lamp positions could be decided. For example of an elliptical reactor with dual lamps, the optimal lamp locations shall be the 1/3 and 2/3 position of the longer axis.

Persistence of Marine Vibrio vulnificus in Oysters within Environmental Parameters (환경 변수에 대한 생굴에서의 해양 Vibrio vulnificus의 인내성)

  • This project studies marine Vibrio vulnificus in oysters in the marine environment and attempts to correlate this bacteria's presence within various environmental parameters; we design this study to determine how different storage temperatures affect the survival of V. vuinficus in oysters and whether V. vulnificus is able to persist in oysters after exposure to UV light-disinfected seawater. Experimental depuration systems consist of aquaria containing temperature-controlled seawater treated with UV light and 0.2 ㎛ pore size filtration. Results showed that depuration at temperatures higher than 25℃ caused V. vuinificus counts to increase in oysters. Throughout the process, depuration water contained high concentrations of U vuinificus indicating"that the disinfection properties of UV radiation and 0.2 ㎛ pore size filtration were less than 어e release of V. vuinificus into seawater, In contrast, when depuration seawater was maintained at 10℃, the numbers of V. vuinificus were very little and multiplication in oysters was inhibited.

Ultraviolet-activated peracetic acid treatment-enhanced Arabidopsis defense against Pseudomonas syringae pv. tomato DC3000

  • Disinfecting water containing pathogenic microbes is crucial to the food safety of fresh green agricultural products. The UV-activated peracetic acid (UV/PAA) treatment process is an efficient advanced oxidation process (AOP) and a versatile approach to disinfecting waterborne pathogens. However, its effects on plant growth remain largely unknown. This study found that low-dose UV/PAA treatment induced moderate oxidative stress but enhanced the innate immunity of Arabidopsis against Pseudomonas syringae pv. (Pst) DC3000. When applied as water sources, 5- and 10-ppm UV/PAA treatments slightly reduced biomass and root elongation in Arabidopsis seedlings grown under hydroponic conditions. Meanwhile, treatments of the same doses enhanced defense against Pst DC3000 infection in leaves. Accumulation of hydrogen peroxide and callose increased in UV/PAA-treated Arabidopsis samples, and during the post-infection period, UV/PAA-treated seedlings maintained vegetative growth, whereas untreated seedlings showed severe growth retardation. Regarding molecular aspects, priming-related defense marker genes were rapidly and markedly upregulated in UV/PAA-treated Arabidopsis samples. Conclusively, UV/PAA treatment is an efficient AOP for disinfecting water and protecting plants against secondary pathogenic attacks.

Removal of NOM in a Coagulation Process Enhanced by Modified Clay (개질 Clay를 첨가한 응집공정에서의 자연유기물 제거)

  • A feasibility test was conducted to evaluate the addition of turbidity substance in a coagulation process to remove natural organic matters (NOM), the precursor of disinfection by-products (DBPs). The experimental water sources were synthetic water containing 5 mg/L of humic acid and 50 mg/L of NaHCO3 and drinking water resource of Ulsan city (S Dam water, D Dam water and Nak-Dong raw water). The examined turbidity substances were kaolin, acid clay, and modified clay (0.38 meq $NH_4{^+}-N/g$ clay). In Jar tests at different concentrations of the turbidity substances (5, 10, 15, 20, 30 mg/L) using the synthetic water, the turbidity substances improved the removal of turbidity, UV-254 absorbance and dissolved organic carbon (DOC) by 23.8-38.1%, 17.0-24.5% and 2.5-44.5%, respectively. The modified clay showed higher removal efficiencies than other substances. In Jar tests using the drinking water, 10 and 20 mg/L of modified clay enhanced the removal efficiencies of turbidity, UV-254 absorbance, DOC, trihalomethane formation potential (THMFP), and haloacetic acid formation potential (HAAFP) by 3.0~4.3%, 19.1~29.0%, 12~34.9%, 4.9~36.7%, and 1.6~30.2%, respectively.

Evaluation of inactivation kinetics on pathogenic microorganisms by free chlorine/UV hybrid disinfection system (전해 염소수/자외선 결합 시스템을 이용한 병원성 미생물의 불활성화 키네틱스 평가)

  • Chlorination and UV illumination are being widely applied to inactivate a number of pathogenic microbials in the environment. Here, we evaluated the inactivation efficiency of individual and combined treatments of chlorination and UV under various aqueous conditions. UV dosage was required higher in waste water than in phosphate buffer to achieve the similar disinfecting efficiency. Free chlorine generated by electrolysis of waste water was abundant enough to inactivate microbials. Based on these, hybrid system composed of sequential treatment of electrolysis-mediated chlorination and UV treatment was developed under waste water conditions. Compared to individual treatments, hybrid system inactivated bacteria (i.e., E. coli and S. typhimurium) and viruses (i.e., MS-2 bacteriophage, rotavirus, and norovirus) more efficiently. The hybrid system also mitigated the photo re-pair of UV-driven DNA damages of target bacteria. The combined results suggested the hybrid system would achieve high inactivation efficiency and safety on various pathogenic microbials in wastewater.

Removal of Algogenic Organic Matter in Drinking Water Treatment Process (정수처리공정에서 조류유래 유기물질의 제거)

  • Algae causes not only the eutrophication of lake, but also the deterioration of drinking water process. Especially, algogenic organic matters(AOM) are assumed as disinfection by-products(DBPs) precursors like humic and fulvic acids. In this study, it was investigated the characteristics changes of algogenic organic matter(AOM) by prechlorination and coagulation treatment. Evaluation of enhanced coagulation and applicability of UV oxidation process were also evaluated as the drinking water treatment system for the eutrophicated water source. prechlorination was effective process for algae removal but caused releasing of dissolved organic matter(DOC) into water due to the destruction of algae's cell. In coagulation treatment with Fe(III) coagulant, reaction pH is an important factor for the removal of AOM and triholomathanes(THMs). At pH 5, removal efficiency of DOC and THMs were dramatically improved by 50% and 28%, respectively, in comparison with the conventional coagulation treatment at about pH 7. Photo-Fenton($UV/H_2O_2/Fe^{3+}$) process among the UV oxidations is the most effective system to remove AOM, but its removal efficiency was lower than that of enhanced coagulation treatment at pH 5.

Effectiveness Analysis on the Application of Ultraviolet and Plasma Treatment Devices for Water Sterilization (용수 살균을 위한 자외선과 플라즈마 처리장치 적용에 따른 효과분석)

  • This study aimed to compare the disinfection efficiencies of the ultraviolet and plasma systems, the two systems designed and commercialized to disinfect water in aquaculture, by putting each in a 100 ℓ water tank and concentrating 1.0 ℓ of treated water to check the changes in the number of bacteria in the samples. Each system was operated for 6 hours to culture the typical seawater bacteria in the Marine agar, Thiosulfate citrate bile salts sucrose agar and Salmonella Shigella agar media, respectively, to check the number of bacteria in the media, and the changes in the number of Edwardsiella piscicida in the treated water were checked after the artificial inoculation of E. piscicida in the disinfected seawater. As a result, the two disinfection systems showed the almost similar levels of bacterial reduction efficiency between 99.5% and 99.9%. However, the result of this study showed that, with 100 ℓ of water treated for the same length of time using the two systems, the plasma system turned out to disinfect bacteria in a shorter period of time than the UV system. However, as the changes in the number of bacteria were checked for a short length of time (6 hours) in this study, it was judged that, considering the actual aquaculture environment in which the quality of water significantly changes with feed residues, excretions and coastal contamination, etc., and a lot of biofilms and organic matter exist, the plasma system would be more efficient than the UV system as the former is capable of continuously maintaining a certain level of efficiency than the latter that is limited in terms of efficiency depending on the level of turbidity and the existence of organic matter.

Treatment of Ballast Water By Filtration -Ultraviolet radiation-Electrolytic Process (FUE 공정에 의한 Ballast Water처리)

  • Reballasting at sea, as recommended by the IMO guidelines, currently provides the best-available measure to reduce the risk of transfer of harmful aquatic organisms, but is subject to serious ship-safety limits. It is therefore extremely important that alternative, effective ballast water management and treatment methods are developed as soon as possible, to replace reballasting at sea. Filtration-Ultraviolet radiation-Electrolytic process (FUE) was evaluated for disinfection of seawater used In ballast water Optimal current density and UV light intensity were 2.0A/dm$^2$ and, 220㎼/$\textrm{cm}^2$/m with which 100% reduction time was 2sec in a Ultraviolet radiation-Electrolytic process. This study showed that FUE process was effective for the disinfection of commonly isolated bacteria and bacillus from ballast water.

Evaluation of Disinfection Characteristics of Ozone, UV Processes for Bacillus Subtilis Spores Inactivation (Bacillus Subtilis Spores 불활성화 실험을 통한 오존, UV 공정의 소독 특성 평가)

  • Ozone/UV combined process is an effective technique to enhance generation of OH radical which is non-selective and powerful oxidant. The objective of this study is to evaluate the inactivation rates of B. subtilis spores by three candidate processes (ozone alone, UV alone, ozone/UV combined processes) at 4 and $20^{\circ}$ and to investigate the effects of OH radical on inactivation of B. subtilis spores. On the UV alone process, required UV dosages for lag phase and 3-log inactivation of B. subtilis spores were determined as $8.9mJ/cm^2$ and $47mJ/cm^2$. However, the inactivation of B. subtilis spores didn't occured beyond 4.5-log inactivation despite increasing UV dose. The inactivation of B. subtilis spores by ozone alone and ozone/UV combined process was investigated with ozone CT (Concentration of disinfectant ${\times}$ Contact time) concept. As a result, inactivation of B. subtilis spores by ozone/UV combined process was faster than by ozone alone, and especially $CT_{lag}$ value B. subtilis spores in the presence and absence of t-BuOH, OH radical scavenger, was investigated to evaluate effects of OH radical formed during ozone/UV combined process. We found that OH radical plays important roles on inactivation of B. subtilis spores.

Bacteriocidal Effects of Ultraviolet Irradiation for Reducing Bovine Mastitis Derived from Environmental Contamination (우분뇨 유래 젖소 유방염 저감을 위한 자외선 조사 살균의 효과 규명)

  • Bovine mastitis is an important disease causing serious economic loss in dairy production and food poison in public health. The major causative agents of bovine mastitis include Escherichia coli (E. coli), Streptococcus agalactiae (S. agalactiae), Staphylococcus aureus (S. aureus). These bacteria were found in milk and environmental condition such as feces, water, soil and so on. Recently, many cases of mastitis are derived from environmental contamination of micro-organisms, which important factors for the spread of this disease in farm. Ultraviolet irradiation (UV) has been used as disinfection for waste and water in clinical and industrial facilities. Moreover the UV irradiation has been used as useful bactericidal agents to remove bacterial biofilms in environmental condition. In this study, we determined the bacterial replication in different percentage of water content (PWC) in sterilized saw dust and feces complexes from farm, and results showed that slightly decreased growth pattern of E. coli and S. agalactiae but increased growth pattern of S. aureus in various PWC (200, 400 and 600%) until 144 h incubation. In the bacteriocidal effect of UV irradiation to bacteria in saw dust and feces complex, the results showed that bacteriocidal effect was depended on the UV irradiation time, irradiation distance and PWC. Especially the antibacterial activity of UV irratiation is stronger in low PWC (50%), long time irradiation (50 sec), and short distance (5 cm) than other condition of this study. Furthermore UV irradiation with stirring showed increased the bactericidal effect compared without stirring. These results suggested that bovine mastitis causing agents may survive long time in environmental condition especially saw dust and feces complexes in farm and can cause a various disease including mastitis. Moreover, these data can be used as basis for application and development of UV disinfection to control of bovine mastitis from environmental contaminated bacteria in dairy farm.
