Genetic Relationships of Internal Transcribed Spacer (ITS) Regions on Entomopathogenic Fungi by RFLP (Entomopathogenic Fungi의 ITS 영역에 대한 RFLP 분석)

  • A similarity coefficient were analyzed by RFLP of fourteen species of entomopathogenic fungi, isolated from inhabiting pupa and adult insect at forest. Each rDNA ITS I and ITS II with primers of ITS 1 and ITS 4 was amplified by PCR. The amplified products were conserved to 500 bp were not demarcated between genus and species. Four Paeciliomyces tenuipes, two Beauveria bassiana and six Cordyceps militaris were treated by seven restriction enzymes and confirmed in species except JB3 by electrophoresis band. However, the band of C. scarabaeicola showed the identity with B. bassiana. The result of this experiment indicated that the teleomorph of C. scarabaeicola was the same as that of B. bassiana. CfoI and HpaII of seven restricted enzymes were easily discriminating in the genus between Paecilomyees and Cordyceps. Especially, CfoI was more effective to classify the genera of Paecilomyees, Cordyceps and Beauveria than other restriction enzymes. The band patterns of RFLP of P. tenuipes, C. militaris, C. scarabaeicola and B. bassiana were also analyzed by UPGMA program of NTSYS-pc and showed 100% significance. Thus, the similarity coefficient tended to be lower between genera by RFLP analysis, but was higher between species.

Diversity of I-SSR Variants in the Populations of Torreya nucifera (비자나무 집단(集團)에서의 I-SSR 변이체(變異體)의 다양성(多樣性))

  • Inter-simple sequence repeat (I-SSR) markers were analyzed from diploid genomes of 95 nutmeg trees (Torreya nucifera Siev. et Zucc.) in 5 populations. A total of 62 I-SSR amplicons were observed and 7 of them were monomorphic in 95 individuals. DNA fingerprint of each tree was verified by pooling the observed I-SSR amplicons. Most of the genetic diversity was allocated within population (90.65%) and all the populations revealed similar level of I-SSR amplicon diversity within population. Degree of population differentiation (${\phi}_{ST}=9.35%$) was moderate on the basis of criteria obtained from isozyme analysis. Based on the results of the cluster analysis of UPGMA, genetic relationships among 5 populations were not coincided with the pattern of geographic distribution. Non-significant confidence interval at each node also suggests that all the nutmeg populations are genetically not much differentiated.

Community Analysis based on Functional Feeding Groups of Aquatic Insects in Wonju Stream (원주천 수서곤충의 섭식기능군을 이용한 군집분석)

  • Community analysis based on functional feeding groups of aquatic insects in Wonju stream was assessed with the composition of aquatic insects collected for four times at the area from May to November, 2004. The total 92 species of aquatic insects in 62 genera, 37 families, and 8 orders were collected during the period of the investigation. The distribution of the numbers and movements of aquatic insects proved the Woniu stream to be healthy except for St. 7 and St. 9. In the main streams, the occupancy ratio of shredders and scrapers of the functional feeding groups(FFGs) gradually decreased from St. 1 to St. 9 while collectors-gathering and collectors-filtering remarkably increased. In St. 2, the shredders and scrapers posted a higher ratio compared to those in tributaries, and it had the quality of mountain streams. With the results of analysis on the similarity matrix(UPGMA) between each sites of Wonju stream, St. 1, 2, St. 3-8 and St. 9 were classified as the upper stream, the middle stream, and the lower stream area, respectively.

Morphological Variations and Genetic Variations Inferred from AFLP (Amplified Fragment Length Polymorphism) Analysis of Cottus Populations (Scorpaeniformes: Cottidae) in Korea (한국산 둑중개속 어류 개체군들의 형태 변이 및 AFLP 분석을 통한 유전 변이)

  • Morphological and genetic variation of Cottus populations in Korea were compared to each other in the watershed. Morphological variation was analyzed by meristic and morphometric characters, and by fertilized egg sizes. Genetic variation was assessed by amplified fragment length polymorphism (AFLP) fingerprinting. Cottus koreanus populations of rivers and streams running towards the eastern coasts of the Korean Peninsula were significantly different in morphometric characters from populations in rivers and streams running towards the western and southern coasts of the Korean Peninsula, but did not differ in meristric characters and fertilized egg sizes. Cottus populations in the upper streams-Yangyangnamadae Stream, Jusu Stream and Samcheogosip Stream-were the same in meristic and genetic characters. However, the Cottus sp. population from Baebong Stream is related to C. hangiongensis in meristic characteristics and to C. koreanus in the morphometric characteristics of length of the ventral fin ray and in the fertilized egg size. Pairwise genetic distances assessed by the AFLP method among C. koreanus populations were in the range of 0.110 to 0.221. Genetic distances between C. hangiongensis and C. koreanus populations varied from 0.542 to 0.621, and those between the Cottus sp. population of Baebong Stream and C. koreanus populations from 0.222 to 0.304. The result of the UPGMA dendrogram shows the Cottus sp. population of Baebong Stream was clearly separated of other C. koreanus populations.

Analysis of Genetic Relationships of Grapevine Cultivars (Vitis ssp.) in Korea Using RAPD Markers (RAPD를 이용한 한국 포도 품종의 계통유연관계 분석)

  • In this study, we used the random amplified polymorphic DNA (RAPD) technique to evaluate the genetic relationships among 29 grapevine cultivars (Vitis spp.). Sixty selective primers detected a total of 558 polymorphic bands. By UPGMA (unweighted pair-group method arithmetic average) cluster analysis with 558 polymorphic bands, the 29 grapevine cultivars were divided into six major groups at 58.8% genetic similarity. The "Super Hamburg" was clustered in group I. Group II consisted of "Wonkyo RA-23", "Muscat Hamburg", "Tano Red", and "Tankeumchu". Group III consisted of "Alden", "Wonkyo RA -21", "Wonkyo RA-30", and "Dutchess". Group IV included 14 grapevine cultivars ("Heukgoosul", "Heukbosuk", "Suok", "Wonkyo RA-29", "Wonkyo RA-22", "Kyoho", "Pione", "Beniizu", "Golden Muscat", "Jinok", "Doonuri", "Campbell Early", "Delaware", and "Schuyler"). Group V consisted of "Hongdan", "Tamnara", "Hongisul", and "Himrod seedless". Group VI included 2 cultivars ("Cheongsoo", and "S. 9110").

Morphological Characteristics Based on Phenotypes of Flower and Fruit in a New Variety of Anthoxylum schinifolium (산초나무(Zanthoxylum schinifolium) 신품종의 꽃과 열매에 기반한 형태적 특성 분석)

  • In this study, the morphological characteristics of flower and fruit in five varieties (Hancho 915ho, 930ho, 10ho, 1020ho, 1030ho) from Zanthoxylum schinifolium were investigated as new varieties with different fruiting seasons. Qualitative and quantitative characteristics were investigated, spectively. For quantitative characteristics can be measured numerically, such as size and number, the varieties were identified using ANOVA and Scheffe's multiple range test. In addition, the validity of whether the quantitative characteristics of five varieties can be explained by factor analysis was reviewed. Using UPGMA, the relationship between the five varieties was confirmed by dendrogram. As a result of ANOVA and Scheffe's multiple range test there was significant difference in inflorescence width and female flower length. It had a high contribution to distinguishing varieties, which was similar to the PCA results using factor analysis. As a result of dendrogram using cluster analysis, Hancho 915ho, 1020ho and 1030ho were similar, but there was a distinct difference in the characteristics of different flowering and fruiting periods, confirming that they were different varieties.

Genetic Variations and Relationships of Persicaria thunbergii(Sieb. & Zucc.) H. Gross ex Nakai(Polygonaceae) by the RAPD Analysis (RAPD 분석에 의한 고마리(마디풀과)의 유전적 변이 및 유연관계)

  • RAPD analyses were performed from twenty-four populations of Persicaria thunbergii(Sieb. & Zucc.) H. Gross ex Nakai. The length of amplified DNA fragments ranged from 200 to 1,900bp. 184 scorable RAPD markers were found from PCR reactions with sixteen random oligoprimers. Based on the results, populations of Persicaria thunbergii were classified into disturbance streams of urban and rural streams as well as natural streams. And the populations from natural streams showed having higher genetic similarites than those from highly disturbed streams, Also, the heterogenetic differences between the populations from natural and disturbed areas could be represented the results of the stream environmental changes.

Genetic status of Acanthamoeba spp. Korean isolates on the basis of RAPD markers (RAPD 표지자 분석 에 의한 가시아메바속 한국분리주의 유전적 지위)

  • Genetic status of Acnnthamoebc sap. were tested on the basis of random amplified polymorphic DNA (RAPD) marker analysis. Four previously established Accnthcmoebn species, 4 Korean isolates of Acnnthamoeba sp., and one American isolate of Acanthcmoebc sp. were analyzed by RAPD-PCR using an arbitrary decamer primers. Amplification products were fractionated by agarose gel electrophoresis and slainrd by ethidium bromide . Eighteen primers produced DNA amplification profiles revealing clear differences among 4 species. Nine of them also produced DNA amplification profiles which included some isolate-specific amplification products. On the basis of amplified fragments by 18 primers, the pairwise similarity indices between A. culbensoni and other species (i.e. A. hntchetti, A. trinngularis, A. polyphaga) were 0.300, 0.308, and 0.313, respectively. Similarity index between A. hctchetti and A. triansulcris was 0.833. The mean similarity index among the 3 Korean isolates (YM-2, -3, -4) was 0.959 and 0.832 among them and 2 other species (A. hatchetti and A. triongulnris). The mean similarity index among YM-5 and other Korean isolates (YM-2, -3, -4) was 0.237. However, the similarity index between YM-5 and A. culbeksoni was 0.857, which suggests that YM-5 is genetically more similar to A. culbertsoni than other Korean isolates. Phonogram reconstructed by UPGMA method revealed that there are two groups: one group consists of A. hctchetti, A. tlonsulcns, and 3 Korean isolates (YM-2, -3, -4) , and the other group consists of A. cuLbensoni. A. polwphosc, HOV, and YM-5.

Evaluation of Genetic Diversity among Persimmon Cultivars (Diospyros kaki Thunb.) Using Microsatellite Markers (초위성 마커를 이용한 감(Diospyros kaki Thunb.)의 유연관계 분석)

  • The genetic diversity among 48 persimmon (Diospyros kaki Thunb.) accessions, indigenous in Korea and introduced from Japan and China, was evaluated by using simple sequence repeat (SSR) markers. From 20 SSR primer sets, a total of 114 polymorphic markers were detected among 12 pollination-constant non-astringent (PCNA), 13 pollination-variant non-astringent (PVNA), 15 pollination-variant astringent (PVA), and 8 pollination-constant astringent (PCA) cultivars. Analysis of pair-wise genetic similarity coefficient (Nei-Li) and unweighted pair-group method with arithmetic averaging (UPGMA) clustering revealed two main clusters and four subclusters for cluster I. The subclustering pattern was in accordance with the classification of persimmon cultivars based on the nature of astringency loss. Phenetic relationships among the subclusters showed a closer relatedness of the PCNA group with the PVNA group, and the PVA with the PCA group. Genetic similarity co-efficiency was 0.499 on average and the highest (0.954) similarity was observed between 'Cheongdo-Bansi' and 'Haman-Bansi'. The similarity was lowest (0.192) between 'Damopan'and 'Atago'. Identification of each cultivar with the execption of 'Cheongdo-Bansi' and 'Gyeongsan-Bansi' was possible based on the SSR fingerprints, suggesting that these SSR markers are a useful tool for protecting intellectual property on newly developed cultivars.

Genetic Diversity of Agaricus bisporus Strains by PCR Polymorphism (PCR 다형성에 의한 양송이(Agaricus bisporus) 계통의 유전적 다양성 분석)

  • Twelve Universal fungal PCR fingerprint (UFPF) primers that were modified from Universal rice primer (URP) were used to assess genetic diversity of 64 Agaricus strains including 45 A. bisporus strains and other 19 strains of other Agaricus spp. Eight primers, UFPF1, UFPF2, UFPF3, UFPF7, UFPF9, UFPF10, UFPF11, and UFPF12 produced PCR polymorphic bands within and between the Agaricus species. Primer UFPF7 produced specific PCR polymorphic bands that are distinct Korean strain from different strains. Ninety five PCR polymorphic bands were inputted for UPGMA cluster analysis. Forty five strains of A. bisporus are genetically clustered into 8 groups, showing coefficient similarity from 0.75 to 0.9 among them. The varieties, Saea, Saedo, Saejeong and Saeyeon that have recently been developed in Korea were involved in the same group with close genetic relationship of coefficient similarity over 0.96, whereas, other Korean strains were genetically related to A. bisporus strains that were introduced from USA, Eroupe and Chinese.
