철골 모멘트 접합부에서 웨브의 모멘트 전달효율과 변형도 집중과의 관계에 관한 해석적 연구 (Analytical Study on Relationship Between Moment Transfer Efficiency of a Beam Web and Strain Concentration at Steel Moment Connections)

  • 본 논문에서는 4개의 철골 해석모델과 1개의 합성보 해석모델에 대한 단조가력 해석의 결과를 나타냈다. 철골 기둥-보 접합부에서 웨브의 모멘트 전달효율과 변형도 집중에 대해서 조사하였다. 해석결과 각형기둥을 가진 모델의 모멘트 전달효율은 각형기둥 플랜지의 면외변형 때문에 H-기둥을 가진 모델에 비해서 빈약했다. 스캘럽과 얇은 각형기둥 두께 및 슬래브도 또한 모멘트 전달효율의 저하를 가져오는 원인으로서, 이는 보-기둥 접합부의 파단을 초래할 가능성이 크다. 해석 결과는 실험결과와 비교하였다. 실험결과를 기초로 하면, 각형기둥 또는 슬래브가 있는 접합부의 변형능력은 빈약한 모멘트 전달효율과 이에 따른 플랜지의 변형도 집중에 의해 감소했다.

  • As the research about increasing the efficiency of dredging soil transport, the technology, which reduce the friction between pipe wall and fluid in the pipe and disturbed generating pipe blockage, has been developed. So for the purpose of applying this technology to real construction site, main test has been tried at the real scale test in field. As a test result, this paper will show 30% flow efficiency increasing by permitted electro magnetic force to the pipe. And test result was evaluated as a ultra sonic velocity profiler. To propose the design technique and the execution manual of the high efficiency dredging material transport technic, this research have confirmed flow status changing depending on a soil material kind under electro-magnetic field and analyze the effect of electro-magnetic field which affects to each dredged soil material transportation. For achieving this research, EMF(Electro-Magnetic Field) generator is installed on the dredger(20,000HP) and through monitored flow status, dredging soil flow rate and sampled material specification is confirmed. Also dredger operating condition is measured and dredger power for soil transportation, hydraulic gradient and flow rate are compared, as transportation efficiency is calculated by this parameter, it is possible to check transportation efficiency improvement depending on each dredged soil material under electro-magnetic field. To verify the technique of dredged soil transfer using electromagnetic field, which is the core technique of the high efficiency dredged soil transfer, and the technique of expert system for pipeline transfer and the flow state. This could lead to a verification of transfer efficiency according to the characteristics of the dredged soil (sand, clay, silt) and the transfer distance (5km, 10km, 15km), which is planned to be used for a technology development of pump power reduction and long-distance transfer applying the high efficiency dredged soil transfer technology.

  • In the last 10 years, porcine somatic cell nuclear transfer to generate transgenic pig has been performed tremendous development with introduction and knockout of many genes. However, efficiency of porcine somatic cell nuclear transfer is still low and embryo transfer (ET) is one of important step for production efficiency. In porcine ET for production of transgenic cloned pig, we can consider many of points to increase production rates. In respect of seasonality and weather, porcine ET usually is not performed in summer and winter. Cloned transgenic embryos must be transferred into reproductive tracts of recipients where embryos are located after natural fertilization with similar estrous cycle. If cloned embryos with 2~4 cell stage are transferred, they must be transferred into oviducts in periovulatory stage. Number and deposition sites of transferred cloned embryos are important. And we must compare the methods of ET between surgical and non-surgical ones in respect of production efficiency. Sow recipients after natural estrus is most preferred recipients however its cost is must be considered. Here we will review many of current studies about porcine embryo transfer to increase production efficiency of transgenic pigs and strategies for further studies.

RHS기둥-보 접합부의 모멘트전달효율 평가 (Evaluation of Moment Transfer Efficiency of a Beam Web at RHS Column-to-Beam Connections)

  • 본 논문에서는 철골 각형강관단면(RHS) 기둥-보 접합부에서 웨브의 모멘트 전달효율을 평가하였다. 먼저, 5개의 철골보접합부에 대한 비선형 유한요소해석을 수행하였다. 이들은 접합부 상세가 다르게 설계되었고, 따라서 휨저항 성능이 각기 다르다. 해석결과 RHS 기둥을 가진 모델은 기둥 플랜지의 면외변형 때문에 WF(Wide Flange) 기둥을 가진 모델에 비해 모멘트 전달효율이 저하함을 보였다. 스캘럽(WAH)과 얇은 강관기둥 두께도 모멘트 전달효율의 저하를 가져오는 원인으로서, 결과적으로 보-기둥 접합부의 파단을 초래할 가능성이 크다. 해석과 이전의 실험결과를 기초로 하면, 응력집중은 모멘트 전달효율과 반비례하고, 접합부의 변형능력은 모멘트 전달효율의 저하에 따라 감소하는 것을 알 수 있다. 더 나아가서 바닥슬래브가 있는 합성보 접합부에 대한 유한요소해석결과는 중립축이 상부플랜지 방향으로 상승함으로써 모멘트 전달효율이 저하했고, 이러한 영향은 접합부의 조기 취성파단을 초래하는 것을 보였다.

Genetic Transformation of Geobacillus kaustophilus HTA426 by Conjugative Transfer of Host-Mimicking Plasmids

  • We established an efficient transformation method for thermophile Geobacillus kaustophilus HTA426 using conjugative transfer from Escherichia coli of host-mimicking plasmids that imitate DNA methylation of strain HTA426 to circumvent its DNA restriction barriers. Two conjugative plasmids, pSTE33T and pUCG18T, capable of shuttling between E. coli and Geobacillus spp., were constructed. The plasmids were first introduced into E. coli BR408, which expressed one inherent DNA methylase gene (dam) and two heterologous methylase genes from strain HTA426 (GK1380-GK1381 and GK0343-GK0344). The plasmids were then directly transferred from E. coli cells to strain HTA426 by conjugative transfer using pUB307 or pRK2013 as a helper plasmid. pUCG18T was introduced very efficiently (transfer efficiency, $10^{-5}-10^{-3}\;recipient^{-1}$). pSTE33T showed lower efficiency ($10^{-7}-10^{-6}\;recipient^{-1}$) but had a high copy number and high segregational stability. Methylase genes in the donor substantially affected the transfer efficiency, demonstrating that the host-mimicking strategy contributes to efficient transformation. The transformation method, along with the two distinguishing plasmids, increases the potential of G. kaustophilus HTA426 as a thermophilic host to be used in various applications and as a model for biological studies of this genus. Our results also demonstrate that conjugative transfer is a promising approach for introducing exogenous DNA into thermophiles.

Polscope-Assisted Enucleation for Nuclear Transfer in Mice

  • Efficiency of somatic cell nuclear transfer was investigated in mice. First, oocyte activation was induced by SrCl₂, and the rate of development was compared with embryos from normal fertilization. Although more than one half of SrCl₂-treated oocytes developed to blastocysts (146/262, 55.7%), the rate of blastocyst formation was significantly lower than normal fertilization controls (59/79, 74.6%). Second, enucleation of oocytes was performed using Polscope that enables non-invasive visualization of metaphase spindles. Such approach could not only avoid damage of oocytes during an exposure to UV light often employed in conventional enucleation procedures, but could also assure the removal of nuclei from all oocytes operated because of monitoring the location of spindles during an entire process of enucleation. Morphologically normal blastocysts were obtained from the transfer of cumulus cell nuclei into enucleated oocytes. However, the rate of development into the blastocyst stage was still low (4/93, 4.3%). This reflects that the nuclear transfer procedure used in this study was not sufficiently optimized, and other factors may also impact greatly the efficiency of nuclear transfer. Including an induction of oocyte activation and method of enucleation tested in this study, a lot more elements are remained to be optimized to improve the efficiency of somatic cell nuclear transfer in mice.

  • In almost all real situations, there will be a radiant interchange between adjacent fins with the base surface as well as with the external environment. In the problem of this study, a rectangular fin is attached to a based. Our concern is whether the convective fin efficiency can be increased by the radiation heat exchanged between the fin and the base surface and how much. If the fin temperature toward the tip increased by the effect of radiation, the convective heat transfer increase due to the temperature difference between the ambient temperature and the surface temperature of the fin. If this true, the efficiency of the fin due to the radiation will increase. Attention is directed toward several parameters which have major roles on getting values of the fin efficiencies in several different values of parameters. Many different cases are, therefore, to be examined to have maximum fin efficiency by varying the values of each parameter.

  • The purpose of the present study is to investigate the convective heat transfer characteristics and the validity of fin efficiency of the circular finned-tube heat exchanger by using commercial CFD code. The heat transfer coefficient obtained by using the laminar model was 22% overestimated to the experimental data. The fin surface temperature compared with the experimental data measured by the liquid crystal method. The fin efficiency by the present numerical experiment, defined as normalized and averaged fin surface temperature, was greater than the theoretical fin efficiency and the difference is increased at high value of the factor m.

Numerical study of direct contact membrane distillation process: Effects of operating parameters on TPC and thermal efficiency

  • Membrane distillation (MD) is one of the water treatment processes which involves the momentum, heat and mass transfer through channels and membrane. In this study, CFD modeling has been used to simulate the heat and mass transfer in the direct contact membrane distillation (DCMD). Also, the effect of operating parameters on the water flux is investigated. The result shows a good agreement with the experimental result. Results indicated that, while feed temperature is increasing in the feed side, water flux improves in the permeate side. Since higher velocity leads to the higher mixing and turbulence in the feed channel, water flux rises due to this increase in the feed velocity. Moreover, results revealed that temperature polarization coefficient is rising as flow rate (velocity) increases and it is decreasing while the feed temperature increases. Lastly, the thermal efficiency of direct contact membrane distillation is defined, and results confirm that thermal efficiency improves while feed temperature increases. Also, flow rate increment results in enhancement of thermal efficiency.
