Studies on Embryo Cryopreservation and Twinning by Embryo Transfer of Korean Native Cattle II. Transfer of Bisected Embryos and Production of Twin Calves (한우 수정란의 동결보존 및 쌍자생산에 관한 연구 II. 이분 수정란의 이식과 쌍자 생산)

  • This study was carried out to enhance the efficiency of Korean Native cattle embryos and establish the techniques for producing the twin calves. Bisected embryos without zona pellucida which were divided by simple method not using holding pipette or whole two embryos were transferred to recipients.The pedigrees of monozygotic twin calves produced by transfer of bisected pair embryos were identified. The results obtained were as follows ; The average successful bisection rate was 89.16%. The embryos of blastocyst stage (91.66%) were bisected successfully at significantly (P<0.05) higher rate, compared with the morula stage embryos (86.66%). The average survival rate of bisected embryos following 24 hours culture was 59.02%. The survival rate of morula stage embryos (62.50%) was significantly (P<0.05) higher than that of blastocyst stage embryos (55.5%). For the production of monozygotic twin calves, ten pairs of flesh or frozen demi-em- lymphocytes antigen, the twin calves produced by transfer of bisected pair embryos of Korean Native cattle were identified in pedigrees and confirmed as monozygotes.

Efficiency of Radial Transit Routes (간선-지선 노선체계의 효율성)

  • This study analyzed the efficiency of a trunk with branches and a trunk with feeders route system comparing the user cost and operator cost of those route systems. Basically, a trunk with branches is more efficient than a trunk with feeders in the passengers' perspective if the transit system of a trunk and feeders are the same. In the case that the trunk system is hierarchically higher than the feeder system, a trunk with feeders has the competitive edge over a trunk with branches if the saving of travel cost induced by using the trunk line is larger than the increasing waiting and transfer cost. This result is consistent with the previous research by Park et al.(2007c), which analyzed the efficiency of hierarchical structure in a grid transit network. If the travel demand of each origins and destinations is low, increasing headway according to the travel demand can increase passenger load or operating efficiency in a trunk with feeders route system. In this case, a trunk with feeders route system is more prevalent as the operating cost, route length, and number of destinations increases, while it is less prevalent as the transfer cost and passengers' time value of the waiting time increases. In cases that central business district, which is located in the middle of a trunk line, generates travel demand, a trunk with feeders is more efficient than a trunk with branches with the increasing travel demand from and to the CBD. Therefore it can be concluded that to have a dual operating system between CBD and suburban is more efficient than one type transit system. The efficiency of that system can be increased through an efficient transfer system that can reduce the transfer cost.

  • In arc spot welding process, the arc is not moving and heat input is concentrated in one spot so that the heat input efficiency of arc is higher than that of GMAW. In other words, the heat input efficiency of arc change during weld time because arc start is done in spot and weld metal is filled. Therefore, the heat input model of arc spot welding should be different from that of general GMAW. In present study, the calculating model of heat input efficiency in arc spot welding was suggested by temperature monitoring near spot in arc spot welding of copper plate. The result showed that the heat input efficiency of arc was changed three times during weld time. The accuracy of calculating method of heat input efficiency was verified by heat transfer analysis of arc spot welding process using finite element method.

Circuit Model Based Analysis of a Wireless Energy Transfer System via Coupled Magnetic Resonances (결합된 자기공명을 통한 무선에너지 전력 전송 시스템의 회로 해석)

  • A Simple equivalent circuit model is developed for a wireless energy transfer system via coupled magnetic resonances and a practical design method is also provided. Node equations for the resonance system are built with the method, expanding on the equations for a transformer, and the optimum distances of coils in the system are derived analytically for optimum coupling coefficients for high transfer efficiency. In order to calculate the frequency characteristics for a lossy system, the equivalent model is established at an electric design automation tool. The model parameters of the actual system are extracted and the modeling results are compared with measurements. Through the developed model, it is seen that the system can transfer power over a mid-range of a few meters and impedance matching is important to achieve high efficiency. This developed model can be used for a design and prediction on the similar systems such as increasing the number of receiving coils and receiving modules, etc.

  • The finite element method(FEM) is the most widely used and powerful method for structural analysis. In general, in order to analyze complex and large structures, we have used the FEM. However, it is necessary to use a large amount of computer memory and computation time for solving accurately by the FEM the dynamic problem of a system with many degree-of-freedom, because the FEM has to deal with very large matrices in this case. Therefore, it was very difficult to analyze the vibration for plate structures with a large number of degrees of freedom by the FEM on a personal computer. For overcoming this disadvantage of the FEM without the loss of the accuracy, the finite element-transfer stiffness coefficient method(FE-TSCM) was developed. The concept of the FE-TSCM is based on the combination of modeling technique in the FEM and the transfer technique in the transfer stiffness coefficient method(TSCM). The merit of the FE-TSCM is to take the advantages of both methods, that is, the convenience of the modeling in the FEM and the computation efficiency of the TSCM. In this paper, the forced vibration analysis algorithm of plate structures is formulated by the FE-TSCM. In order to illustrate the accuracy and the efficiency of the FE-TSCM, results of frequency response analysis for a rectangular plate, which was adopted as a computational model, were compared with those by the modal analysis method and the direct analysis method which are based on the FEM.

  • It is essential to reduce the amount of fossil fuel because facing with the natural problem such as a global warming. To achieve this goal, many of interests in the use of renewable energy is growing. Especially, as one of these renewable energy systems, a solar air heater invention has been conducted for enhancing the efficiency of solar air heater. According to this trend, scale-down sized experiment apparatus was constructed and performed for searching a proper fin and confirming the heat transfer performance by fin shape on constant heat condition to enhance efficiency of solar air heater. In this experiment, heat gain, convection heat transfer coefficient, number of transfer units, Nusselt number, Reynold's number, friction factor, performance factor were investigated in order to evaluate the thermal characteristics based on the real data obtained. By comparison with the each fin performance, a zigzag shape keeping a right angle to the plate had the highest value among them.

Comparisons of Development Potential in Bovine SCNT Embryos using Donor Cells treated with Different Demethylating Inhibitors

  • To improve the developmental potential of bovine somatic cell nuclear transfer (SCNT) embryos, this study compared the developmental rates to blastocyst stage in the SCNT embryos using donor fibroblasts treated with 5-azacytidine (5AC) and S-adenosylhomocysteine (SAH) at different concentrations. Their reprogramming efficiency level was investigated with level of telomerase activity. Donor fibroblasts isolated from adult ear skin of a cow were exposed to 5AC and SAH at different concentrations during 2 passages. After nuclear transfer into enucleated recipient oocytes, the cleavage and developmental rates were significantly (p<0.05) decreased in the SCNT embryos using 5AC-treated fibroblasts (5AC-SCNT embryos), compared with those of non-treated control (control-SCNT embryos) and SAH-treated fibroblasts (SAH-SCNT embryos). The developmental rates to blastocyst stage tended to be slightly increased in the SAH-SCNT embryos at each of the concentrations, and especially, the developmental rates in the SCNT embryos using 1.0 mM SAH-treated fibroblasts were significantly (p<0.05) higher than that of control SCNT embryos. The mean numbers of total and ICM cell in blastocysts were also significantly (p<0.05) decreased in the 5AC-SCNT embryos, compared with those of other SCNT blastocysts. Further, the level of telomerase activity was also significantly (p<0.05) decreased in the 5AC-SCNT embryos than those of control and SAH-SCNT embryos. Whereas, a significantly (p<0.05) up-regulated telomerase activity was observed in SAH-SCNT embryos, compare with that of control-SCNT embryos. In conclusion, SCNT embryos using hypomethylated donor cells with SAH, not 5AC, may improve the developmental potential and reprogramming efficiency.

Frequency-Domain Circuit Model and Analysis of Coupled Magnetic Resonance Systems

  • An explicit frequency-domain circuit model for the conventional coupled magnetic resonance system (CMRS) is newly proposed in this paper. Detail circuit parameters such as the leakage inductances, magnetizing inductances, turn-ratios, internal coil resistances, and source/load resistances are explicitly included in the model. Accurate overall system efficiency, DC gain, and key design parameters are deduced from the model in closed form equations, which were not available in previous works. It has been found that the CMRS can be simply described by an equivalent voltage source, resistances, and ideal transformers when it is resonated to a specified frequency in the steady state. It has been identified that the voltage gain of the CMRS was saturated to a specific value although the source side or the load side coils were strongly coupled. The phase differences between adjacent coils were ${\pi}/2$, which should be considered for the EMF cancellations. The analysis results were verified by simulations and experiments. A detailed circuit-parameter-based model was verified by experiments for 500 kHz by using a new experimental kit with a class-E inverter. The experiments showed a transfer of 1.38 W and a 40 % coil to coil efficiency.

  • School is a basic and the most fundamental facility of city planning just like other basic public facilities including the village office. Every plan is established on the basis of school. However, the problems such as the population reduction resulting from the nuclear family-zation and low birth rate, employment and welfare of the aged people resulting from "the old aged society", and the infant nursing and education resulting from a rapid increase of the working couples become notable and accordingly more requirements are being made. Reflecting this trend, the concept and operation system should be changed. Up to now, the BTL projects of the educational facilities are gaining a reputation of being efficient in terms of national budget running, but at the same time receiving negative reputation in terms of budget saving under the civil creativity and efficiency. Through upgrading the private sector investment projects into the BTO+BTL system and further into the BTO(Build Transfer Operate), we can accomplish the original goals of the private sector investment projects, and can make the education budget operation more efficient, and can greatly improve the education environments. However, we should not underestimate in this process that these facilities should not negatively affect the education environments. In any situation, the owners of schools are students.

Protein Analysis Using a Combination of an Online Monolithic Trypsin Immobilized Enzyme Reactor and Collisionally-Activated Dissociation/Electron Transfer Dissociation Dual Tandem Mass Spectrometry

  • We demonstrated the combined applications of online protein digestion using trypsin immobilized enzyme reactor (IMER) and dual tandem mass spectrometry with collisionally activated dissociation (CAD) and electron transfer dissociation (ETD) for tryptic peptides eluted through the trypsin-IMER. For the trypsin-IMER, the organic and inorganic hybrid monolithic material was used. By employing the trypsin-IMER, the long digestion time could be saved with little or no sacrifice of the digestion efficiency, which was demonstrated for standard protein samples. For three model proteins (cytochrome c, carbonic anhydrase, and bovine serum albumin), the tryptic peptides digested by the IMER were analyzed using LC-MS/MS with the dual application of CAD and ETD. As previously shown by others, the dual application of CAD and ETD increased the sequence coverage in comparison with CAD application only. In particular, ETD was very useful for the analysis of highly-protontated peptide cations, e.g., ${\geq}3+$. The combination approach provided the advantages of both trypsin-IMER and CAD/ETD dual tandem mass spectrometry applications, which are rapid digestion (i.e., 10 min), good digestion efficiency, online coupling of trypsin-IMER and liquid chromatography, and high sequence coverage.
