Antioxidant Activities in Germinated and Non-germinated Seeds of Korean Weedy Rice

  • Weedy rice (Oryza sativa) is a semi-wild rice grown in paddy fields and is more adapted to the environment than cultivated rice. Therefore, it is expected that the seeds of weedy rice might contain some chemicals related to its environmental adaptation, such as antioxidants, which may be useful for nutritional and medicinal purposes. This study was carried out to investigate the antioxidant activities of weedy rice compared to a Korean-bred rice cultivar, Sindongjin (SDJ), and to screen lines that show higher antioxidant activity in 199 accessions of weedy rice germplasm collected in Korea. DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ranged from 31.0 to 91.7%, with an average of 82.5%, and 19 accessions that showed more than 91% antioxidant activity were selected. The 19 accessions were re-screened against non-germinated brown rice (BR) and germinated brown rice (GBR) using four assays, total phenol determination, DPPH radical scavenging, ABTS radical scavenging, and reducing power activities. The results showed that accession 'WD3' had the highest antioxidant capacity in both BR and GBR, suggesting that WD3 is a promising potential source of antioxidants and could be developed as a potentially functional substance material.

Effect of Extraction Solvent on the Antioxidant Activity of Lentinula edodes GNA01 Extract (추출용매에 따른 이슬송이버섯(Lentinula edodes GNA01) 추출물의 항산화 활성)

  • Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, $Fe^{2+}$ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product.

Total Flavonoid Content and Antioxidant Activities of Turmeric (Curcuma longa L.) Extracts in Jindo Korea (진도산 울금(Curcuma longa L.) 추출물의 총 플라보노이드 함량 및 항산화 활성)

  • The present study were conducted to determine physiological activities and antioxidant effects [2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, reducing power, Ferric Reducing Antioxidant Power (FRAP) and Fe2+ (ferrous ion) chelating capacity] of 70% methanol, chloroform:methanol, 2:1 volume ratio (CM) and ethyl acetate extract of turmeric (Curcuma longa L.). Bioactive compound of tannin $0.125{\pm}0.007mg$ Catechin Equivalent (CE)/g dry weight. Turmeric extracts yield were 70% methanol 16.54%, CM 5.64% and ethyl acetate 4.14%, respectively. Antioxidant activity of the samples exhibited a dose-dependent increase. Results showed that extraction solvent had significant effects on total flavonoid content and antioxidant effects of ethyl acetate. But ferrous ion-chelating capacity of 70% methanol extract was higher than CM and ethyl acetate extract. From the results of this study, turmeric can be utilized as a valuable and potential nutraceutical for the functional food industry.

Total Phenolic Content and Antioxidant Activities of Leaves and Bark Extract of Adenanthera pavonina L.

  • Adenanthera pavonina L. is an important medicinal plant for Indonesian traditional medicine ("Jamu"). The present study is to determine the phenolic content and evaluate the antioxidant activity of ethyl acetate and ethanolic extracts of leaves and bark of A. pavonina. The total phenolic content was determined using the Folin-Ciocalteu reagent methods, and the antioxidant activity was performed by phosphomolybdate assay, through EC50 values. The highest phenolic content (81.379 mg GAE/g) was found in the hydro-ethanolic extract of the leaves and (80.630 mg GAE/g) in the bark. The antioxidant activity of the two samples was 42.270 ㎍/mL (leaves) and 45.261 ㎍/mL (bark). Plants have the potential as drugs with good antioxidant potential and phenolic compound content. Further scientific studies of this plant are supported to evaluate the antioxidant activity using other methods to validate these preliminary experiments properly.

Antioxidant activities and determination of phenolic compounds isolated from oriental plums (Soldam, Oishiwase and Formosa)

  • The purposes of this study were to determine phenolic compounds and to evaluate antioxidant activities of plums (Soldam, Oishiwase and Formosa). Soldam contains the highest amount of total phenolics among cultivars (Formosa: 4.0%, Oishiwase: 3.3%, Soldam: 6.4% for total phenolic) as well as the total flavonoids of which constituents were mainly myricetin and anthocyanidin. The antioxidant activities were measured by DPPH, ABTS radical scavenging, and SOD-like activities. The DPPH radical scavenging activity of Korean plum extracts (200 ${\mu}g/mL$) showed more than 43%, and the Soldam turned out to be the highest : ID50 value: 160-177 ${\mu}g/mL$ for Formosa and Oishiwase; 58-64 ${\mu}g/mL$ for Soldam. The ABTS radical scavenging activity of Korean plum extracts (200 ${\mu}g/mL$) was found to be more than 50%. The SOD-like activity of Korean plum extracts (200 ${\mu}g/mL$) showed more than 70%. Among three kinds of cultivars, Soldam had the highest antioxidant activity. The nitrite scavenging activity of Soldam was 61.5%, which is the highest, compared with that of the other cultivars, about 50%. From these results, Korean plums turned out to be phytochemical rich fruit as well as to show high antioxidant activities.

Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity (국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

Antioxidant activity and NO production of the Alisma orientale Juzep fermented by Paenibacillus kribbensis AM49 (Paenibacillus kribbensis AM49로 발효시킨 택사의 항산화 활성 및 NO 생성 효과)

  • Objectives : The purpose of this study was to investigate the antioxidant activities and nitric oxide (NO) production in RAW 264.7 macrophages in extract of Alisma orientale Juzep (EAOJ) and fermented extract (FAOJ) by Paenibacillus kribbensis AM49 (P. kribbensis AM49). Methods : The Alisma orientale Juzep was fermented with P. kribbensis AM49 at 37℃ for 72 hours. We measured total polyphenol and total flavonoid, DPPH radical scavenging activity, FRAP activity and reducing power by spectrometric assay in EAOJ and FAOJ at concentrations at 0.5, 1, 5, 10 mg/㎖. Positive control was used ascorbic acid. Furthermore, we examined effect of EAOJ and FAOJ on the cell viability and NO production in RAW 264.7 macrophages. Results : The total polyphenol and total flavonoids content of FAOJ were increased 9.16 mg/g, 2.59 mg/g to 12.58 mg/g, 3.45 mg/g. DPPH radical scavenging activity, FRAP activity and reducing power were dose dependently increased according to the treatment concentration (0.5, 1, 5, 10 mg/㎖) of EAOJ and FAOJ. In particular, DPPH radical scavenging activity, FRAP activity of FAOJ was significantly increased at 5, 10 mg/㎖. Reducing power of FAOJ at 10 mg/㎖ was similar to ascorbic acid at 0.1 mg/㎖. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased at the concentrations of 250, 500, 1000 ㎍/㎖. Conclusions : These results suggest that FAOJ by P. kribbensis AM49 has effects to antioxidant activity. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased.

The Effects of Yegangji-tang on Hyperlipidemic SHR and Antioxidant Activity (예강지탕(銳降脂湯)이 고지혈증 SHR 및 항산화에 미치는 영향)

  • Objective : Yegangji-tang (YJT) is noted in China as effective medicine for hyperlipidemia. This experiment was performed to determine the effects of Yegangji-tang (銳降脂湯: YJT) on antioxidant activity and hyperlipidemia induced by hypercholesterolemic diet in mice. Methods : We observed three experimental groups: normal, control, and treatment. Rats were provided a normal diet in the normal group, and the other groups were provided a hypercholesterolemic diet to induce hyperlipidemia. After 2 weeks, YJT was treated to the treatment group. We observed the change of body and liver weight, change of serum total cholesterol, LDL cholesterol, HDL cholesterol and triglycerides, lipid peroxidation and antioxidant activity of liver tissue, cholesterol gene revelation control efficiency, and histologic change of liver and abdominal aorta. Results : Total cholesterol, LDL cholesterol and triglycerides decreased significantly with YJT, whereas HDL cholesterol increased significantly. Lipid peroxidation decreased and antioxidant activity of the liver increased significantly. Gene revelation of ACAT and HMG-CoA reductase in hepatic tissue decreased significantly with YJT. We also found that tissue defects of the liver and abdominal aorta were controlled by YJT on histologic study. Conclusion : These results suggest that YJT might be effective in treatment and prevention of hyperlipidemia.

Efficient Flavonoid Extraction from Apple Peel by Subcritical Water and Estimation of Antioxidant Activity (아임계수를 이용한 사과 과피 플라보노이드의 효율적 추출 및 항산화 활성 평가)

  • The effect of subcritical water for the extraction of total polyphenols and flavonoids from apple peel was investigated, and then the antioxidant activity of the extracts was estimated. Maximum yields of total polyphenolic compounds($36.4{\pm}1.9mg$ quercetin equivalent(QE)/g dried material) and flavonoids($9.9{\pm}0.8mg$ QE/g dried material) were obtained by subcritical water extraction(SWE) with operating conditions of $190^{\circ}C$, 1,300 psi, and 20 min. Furthermore, the highest antioxidant activity($76.1{\pm}1.1%$) was observed in the extract obtained from SWE using the same conditions. The flavonoids from the SWE of apple peel were compared to three conventional extraction methods in terms of their extraction efficiency and antioxidant activity. The SWE was significantly more effective than hot water ($90^{\circ}C$), methanol, and ethanol extraction for flavonoid yield by 4.7-, 2.2-, and 1.3-fold, respectively, and for antioxidant activity by 11.0-, 4.9, and 2.8-fold, respectively.

Optimization of Extraction Process to Increase the Antioxidant Activity of Sargassum serratifolium Extracts (톱니모자반(Sargassum serratifolium) 추출물의 항산화 활성 증가를 위한 추출공정 최적화)

  • Sargassum serratifolium is a rich source of antioxidant meroterpenoids. The optimal extraction temperature and time for obtaining maximal antioxidant yield, antioxidant activity, and phenolic content from Sargassum serratifolium were determined using response surface methodology (RSM). The ranges of the independent variables for extraction temperature and time were 30-70℃ and 12-36 h, respectively. With increasing temperature and time, the yield increased significantly, while DPPH (2,2-dipheny-1-picrylhydrazyl) radical-scavenging activity and total phenolic content decreased significantly. The optimal extraction temperature and time obtained by RSM were 54℃ and 7 h, respectively, providing a yield of 8.2%, DPPH radical-scavenging activity of 60%, and total phenolic content of 163 mg GAE/g. The findings of this study provide useful information for the development of Sargassum serratifolium extraction processes for the food and cosmetic industries.
