Cold-Stress Response of Probiotic Lactobacillus plantarum K25 by iTRAQ Proteomic Analysis

  • To understand the molecular mechanism involved in the survivability of cold-tolerant lactic acid bacteria was of great significance in food processing, since these bacteria play a key role in a variety of low-temperature fermented foods. In this study, the cold-stress response of probiotic Lactobacillus plantarum K25 isolated from Tibetan kefir grains was analyzed by iTRAQ proteomic method. By comparing differentially expressed (DE) protein profiles of the strain incubated at 10℃ and 37℃, 506 DE proteins were identified. The DE proteins involved in carbohydrate, amino acid and fatty acid biosynthesis and metabolism were significantly down-regulated, leading to a specific energy conservation survival mode. The DE proteins related to DNA repair, transcription and translation were up-regulated, implicating change of gene expression and more protein biosynthesis needed in response to cold stress. In addition, two-component system, quorum sensing and ABC (ATP-binding cassette) transporters also participated in cell cold-adaptation process. These findings provide novel insight into the cold-resistance mechanism in L. plantarum with potential application in low temperature fermented or preserved foods.

The Impact of Interaction between Cloud and Longwave Radiation on the Asian Monsoon Circulation (구름-장파복사 상호작용이 아시아 몬순에 미치는 영향)

  • Three-dimensional distributions of longwave radiation flux for the April-September 1998 period are generated from radiative transfer calculations using the GEWEX Asian Monsoon Experiment (GAME) reanalysis temperature and humidity profiles and International Satellite Cloud Climatology Project (ISCCP) cloudiness as inputs to understand the effect of cloud radiative forcing in the monsoon season. By subtracting the heating of the clear atmosphere from the cloudy radiative heating, cloud-induced atmospheric radiative heating has been obtained. Emphasis is placed on the impact of horizontal gradients of the cloud-generated radiative heating on the Asian monsoon. Cloud-induced heating exhibits its maximum heating areas within the Indian Ocean and minimum heating over the Tibetan Plateau, which establishes the north-south oriented differential heating gradient. Considering that the differential heating is a ultimate source generating the atmospheric circulation, the cloud-induced heating gradient established between the Indian Ocean and the Plateau can enhance the strength of the north-south Hadley-type monsoon circulation. Cooling at cloud top and warming at cloud bottom, which are the vertical distributions of cloud-induced heating, can exert on the monsoon circulation by altering the atmospheric stability.

Molecular Characterization and Expression Analysis of Insulin-like Growth Factor-1 and Insulin-like Growth Factor Binding Protein-1 Genes in Qinghai-Tibet Plateau Bos grunniens and Lowland Bos taurus

  • Insulin-like growth factor-1 (IGF-1) and insulin-like growth factor binding protein-1 (IGFBP-1) play a pivotal role in regulating cellular hypoxic response. In this study, we cloned and characterized the genes encoding IGF-1 and IGFBP-1 to improve the current knowledge on their roles in highland Bos grunniens (Yak). We also compared their expression levels in the liver and kidney tissues between yaks and lowland cattle. We obtained full-length 465 bp IGF-1 and 792 bp IGFBP-1, encoding 154 amino acids (AA) IGF-1, and 263 AA IGFBP-1 protein, respectively using reverse transcriptase-polyerase chain reaction (RT-PCR) technology. Analysis of their corresponding amino acid sequences showed a high identity between B. grunniens and lowland mammals. Moreover, the two genes were proved to be widely distributed in the examined tissues through expression pattern analysis. Real-time PCR results revealed that IGF-1 expression was higher in the liver and kidney tissues in B. grunniens than in Bos taurus (p<0.05). The IGFBP-1 gene was expressed at a higher level in the liver (p<0.05) of B. taurus than B. grunniens, but it has a similar expression level in the kidneys of the two species. These results indicated that upregulated IGF-1 and downregulated IGFBP-1 are associated with hypoxia adaptive response in B. grunniens.

Cloning and Expression of Yak Active Chymosin in Pichia pastoris

  • Rennet, a complex of enzymes found in the stomachs of ruminants, is an important component for cheese production. In our study, we described that yak chymosin gene recombinant Pichia pastoris strain could serve as a novel source for rennet production. Yaks total RNA was extracted from the abomasum of an unweaned yak. The yak preprochymosin, prochymosin, and chymosin genes from total RNA were isolated using gene specific primers based on cattle chymosin gene sequence respectively and analyzed their expression pattern byreal time-polymerase chain reaction. The result showed that the chymosin gene expression level of the sucking yaks was 11.45 times higher than one of adult yaks and yak chymosin belongs to Bovidae family in phylogenetic analysis. To express each, the preprochymosin, prochymosin, and chymosin genes were ligated into the expression vector $pPICZ{\alpha}A$, respectively, and were expressed in Pichia pastoris X33. The results showed that all the recombinant clones of P. pastoris containing the preprochymosin, prochymosin or chymosin genes could produce the active form of recombinant chymosin into the culture supernatant. Heterologous expressed prochymosin (14.55 Soxhlet unit/mL) had the highest enzyme activity of the three expressed chymosin enzymes. Therefore, we suggest that the yak chymosin gene recombinant Pichia pastoris strain could provide an alternative source of rennet production.

Genetic Variability of mtDNA Sequences in Chinese Native Chicken Breeds

  • The variability of mtDNA hypervariable segment I (HVS I) sequences was investigated in a total of 48 birds belonging to 12 Chinese native chicken breeds. Sixteen haplotypes were identified from 35 polymorphic nucleotide sites which accounted for 6.4% of a sequenced 544 bp fragment. Diversity analysis of the haplotypes showed that Tibetan, Langshan and Henan cockfight chicken had only one haplotype, while ancient haplotypes existed in Taihe silky and Chahua chicken. Phylogenetic analysis of the haplotypes suggested that Chinese native chicken breeds shared 5 maternal lineages and some breeds would share the same maternal lineage, regardless of their external features and ecological types. Both divergent and phylogenetic analysis of the haplotypes indicated the close genetic relationships between the Chinese native chicken breeds and G. g. gallus and G. g. spadiceus from different areas, which implied that G. g. gallus and G. g. spadiceus were the original ancestors of the Chinese native chicken breeds.

Molecular Characterization and Tissue Distribution of Estrogen Receptor Genes in Domestic Yak

  • Estrogen and its receptors are essential hormones for normal reproductive function in males and females during developmental stage. To better understand the effect of estrogen receptor (ER) gene in yak (Bos grunniens), reverse transcription-polymerase chain reaction (PCR) was carried out to clone $ER{\alpha}$ and $ER{\beta}$ genes. Bioinformatics methods were used to analyze the evolutionary relationship between yaks and other species, and real-time PCR was performed to identify the mRNA expression of $ER{\alpha}$ and $ER{\beta}$. Sequence analysis showed that the ER open reading frames (ORFs) encoded 596 and 527 amino acid proteins. The yak $ER{\alpha}$ and $ER{\beta}$ shared 45.3% to 99.5% and 53.9% to 99.1% protein sequence identities with other species homologs, respectively. Real-time PCR analysis revealed that $ER{\alpha}$ and $ER{\beta}$ were expressed in a variety of tissues, but the expression level of $ER{\alpha}$ was higher than that of $ER{\beta}$ in all tissues, except testis. The mRNA expression of $ER{\alpha}$ was highest in the mammary gland, followed by uterus, oviduct, and ovary, and lowest in the liver, kidney, lung, testis, spleen, and heart. The $ER{\beta}$ mRNA level was highest in the ovary; intermediary in the uterus and oviduct; and lowest in the heart, liver, spleen, lung, kidney, mammary gland, and testis. The identification and tissue distribution of ER genes in yaks provides a foundation for the further study on their biological functions.

Adjoint-Based Observation Impact of Advanced Microwave Sounding Unit-A (AMSU-A) on the Short-Range Forecast in East Asia (수반 모델에 기반한 관측영향 진단법을 이용하여 동아시아 지역의 단기예보에 AMSU-A 자료 동화가 미치는 영향 분석)

  • The effect of Advanced Microwave Sounding Unit-A (AMSU-A) observations on the short-range forecast in East Asia (EA) was investigated for the Northern Hemispheric (NH) summer and winter months, using the Forecast Sensitivity to Observations (FSO) method. For both periods, the contribution of radiosonde (TEMP) to the EA forecast was largest, followed by AIRCRAFT, AMSU-A, Infrared Atmospheric Sounding Interferometer (IASI), and the atmospheric motion vector of Communication, Ocean and Meteorological Satellite (COMS) or Multi-functional Transport Satellite (MTSAT). The contribution of AMSU-A sensor was largely originated from the NOAA 19, NOAA 18, and MetOp-A (NOAA 19 and 18) satellites in the NH summer (winter). The contribution of AMSU-A sensor on the MetOp-A (NOAA 18 and 19) satellites was large at 00 and 12 UTC (06 and 18 UTC) analysis times, which was associated with the scanning track of four satellites. The MetOp-A provided the radiance data over the Korea Peninsula in the morning (08:00~11:30 LST), which was important to the morning forecast. In the NH summer, the channel 5 observations on MetOp-A, NOAA 18, 19 along the seaside (along the ridge of the subtropical high) increased (decreased) the forecast error slightly (largely). In the NH winter, the channel 8 observations on NOAA 18 (NOAA 15 and MetOp-A) over the Eastern China (Tibetan Plateau) decreased (increased) the forecast error. The FSO provides useful information on the effect of each AMSU-A sensor on the EA forecasts, which leads guidance to better use of AMSU-A observations for EA regional numerical weather prediction.

Characteristics and Comparison of 2016 and 2018 Heat Wave in Korea (2016년과 2018년 한반도 폭염의 특징 비교와 분석)

  • This study analyzed and compared development mechanisms leading to heat waves of 2016 and 2018 in Korea. The European Centre for Medium-Range Weather Forecasts Reanalysis Interim (ERA Interim) dataset and Automated Surface Observing System data are used for synoptic scale analysis. The synoptic conditions are investigated using geopotential height, temperature, equivalent potential temperature, thickness, potential vorticity, omega, outgoing longwave radiation, and blocking index, etc. Heat waves in South Korea occur in relation to Western North Pacific Subtropical High (WNPSH) pressure system which moves northwestward to East Asia during summer season. Especially in 2018, WNPSH intensified due to strong large-scale circulation associated with convective activities in the Philippine Sea, and moved farther north to Korea when compared to 2016. In addition, the Tibetan high near the tropopause settled over Northern China on top of WNPSH creating a very strong anticyclonic structure in the upper-level over the Korean Peninsula. Unlike 2018, WNPSH was weaker and centered over the East China Sea in 2016. Analysis of blocking indices show wide blocking phenomena over the North Pacific and the Eurasian continent during heat wave event in both years. The strong upper-level ridge which was positioned zonally near 60°N, made the WNPSH over the South Korea stagnant in both years. Analysis of heat wave intensity (HWI) and duration (HWD) show that HWI and HWD in 2018 was both strong leading to extreme high temperatures. In 2016 however, HWI was relatively weak compared to HWD. The longevity of HWD is attributed to atmosphere blocking in the surrounding Eurasian continent.

  • Kefir exhibits antimicrobial activity in vitro against gram-positive and gram negative bacteria, as well as some fungi. The ability of LAB to inhibit the growth of closely related bacteria is well known. This inhibition of pathogenic and spoilage microbes may be due to the production of organic acids, hydrogen peroxide, acetaldehyde, diacetyl, carbon dioxide, or bacteriocins. Lactobacilli are the major contributors to acid production and, hence, a determining factor in the flavor development in kefir. Lactic acid, proteolytic activity, and acetaldehyde are the essential flavor compounds in kefir. Both acid and bacteriocins contribute to the antimicrobial activity of kefir and kefir grains. Kefir is rich in proteins, calcium, vitamin $B_{12}$, niacin, and folic acid. Many studies have investigated the benefits of consuming kefir and have shown that it is a natural probiotic, which when consumed regularly, can help relieve intestinal disorders, promote bowel movement, reduce flatulence, and improve the overall health of the digestive system. Tibetan kefir, which is different from traditional kefir, is produced in China. It has been reported to exhibit antimicrobial activity that is nearly identical to that of traditional kefir. Kefir production is considered a rapidly growing food industry in China.

  • Objectives: Recently a renovation of the medical-welfare system to reflect the changes of disease spectrum with the demographic changes of society, the increase in income level, and marked concerns for health promotion has been demanded. In accordance with this, attempts have been made to actively integrate traditional medicine based on symptom-differentiated treatment and Western medicine based on disease treatment so that they can complement each other. China has already tried a complementary medical treatment system integrating traditional Chinese and Western medicine. So, this article reviewed major advances in research on integrated traditional Chinese medicine and Western medicine in China. Methods: The authors analyzed data from clinical articles and experimental works in the ' Chinese Journal of Integrated Traditional and Western Medicine' Results and conclusions: Each department attempted to integrate Traditional Chinese Medicine(TCM) and Western Medicine in treatment of various diseases such as malaria, AIDS, and intoxication (rarely found in Korea clinically). Especially in the departments of surgery, dentistry, radiology, and anesthesiology we could see the frequent use of combined treatment. TCM and Western medicine complemented each other very successfully, and the effect of the combined therapy was superior to that of traditional therapy alone. There were diverse methods for therapy in integrated TCM and Western medicine; bath-Tx, physical-Tx, manipulative-Tx, drug -acupuncture, Tibetan medicine, etc. were available in therapy as well as traditional methods such as acupuncture, moxibustion, and negative- Tx. The way of producing Chinese medications were diversified and formulated; making new prescriptions, compounding various kinds of new medicine called' Zhong Cheng Yao' (中成藥) which were easily made, stored, and taken. 'Diagnosis Criteria', 'The effect of TCM Treatment Criteria' were made by committee and broadly used for objectifying diagnosis, discriminating effects of treatments and treatment development, and developing new medical products.
