Optimal Production of Thermostable Alkaline Phosphatase from Thermus caldophilus GK24 (Thermus caldophilus GK24로부터 내열성 alkaline phosphatase의 최적생산)

  • Thermus caldophilus GK24 was selected as sources of thermostable alkaline phosphatase from a survey of extreme thermophile. T. caldophilus GK24 was tested for production of alkaline phosphatase by addition of various concentration of sodium glutamate, bactotryptone, glucose and yeast extract to basal salts. Sodium glutamate was found to be effective for the alkaline phosphatase induction. The optimal induction medium for production of alkaline phosphatase involves the addition of 0.3% sodium glutamate, 0.2% bactotryptone and 0.5% glucose to basal salts. The activity of the enzyme in optimal induction medium increased nearly 6-fold/ml than basal medium and 27.5-fold/ml than standard medium. T. caldophilus GK24 alkaline phosphatase was found to be inducible. When starved of inorganic phosphate, T. caldophilus GK24 produces the enzyme alkaline phosphatase. The addition of inorganic phosphate to growth medium had a repressive effect on enzyme synthesis.

  • Recombinant $\beta$-glucosidase from Thermus caldophilus GK24 was easily purified partially by a heat treatment procedure, resulting in 8-fold and recovery yield of 80% from crude enzyme. When the $\beta$-glucosidase was incubated with a 80% glucose solution (w/w), gentiobiose ($\beta$1,6-glucobiose) was the major product in the reaction mixture. The optimal conditions for producing gentiobiose (11% yields of total sugar) were pH 8-9 and 7$0^{\circ}C$ for 72 h.

  • High-level expression of Thermus caldophilus GK24 alkaline phosphatase (Tca APase) was achieved in Escherichia coli using the pET-based expression plasmids, pEAP1 and pEAP2. In the case of plasmid pEAP2, the signal peptide region of Tca APase was replaced by the PelB leader peptide of expression vector pET-22b(+). Furthermore, the expression level was somewhat higher than that of plasmid pEAPl. A rapid purification procedure of Tca APase overproduced in E. coli was developed which involved heating to denature E. coli proteins followed by HiTrap Heparin HP column chromatography. Optimal temperature and pH and $Mg^{2+}$ dependence of the recombinant Tca APase were similar to those of native enzyme isolated from T. caldophilus GK24.

  • Thermus caldophilus GK24 was selected as sources of thermostable $\beta$-galactosidase from a survey of genus Thermus. T. caldophilus GK24 (Tca) $\beta$-galactosidase was found to be inducible. The enzyme was optimally active at 75$\circ$C. Enzyme induction was achieved by addition of lactose, galactose and cellobiose to basal media. The addition of glucose to culture media had a repressive effect on further enzyme synthesis. T caldophilus GK24 was tested for production of $\beta$-galactosidase by addition of various concentration of lactose, galactose and cellobiose to standard media. Cellobiose was found to be effective for the $\beta$-galactosidase induction. The optimal induction medium for production of $\beta$-galactosidase was composed of 0.2% cellobiose, 0.3% bactotryptone, 0.3% yeast extract, basal salts and Tris/HCI(pH 7.8). The activity of the enzyme in the optimal induction medium increased nearly 16.5-fold compared to the standard medium. Tca $\beta$-galactosidase was detected when cell extracts was subjected to electrophoresis in a nondenaturing polyacryamide gel and stained for activity with 6-bromo-2-naphtyl-$\beta$-D-galactopyranoside(BNG).

  • A gene (GenBank AF096282) coding for a $\alpha$-glucosidase (TcaAG, EC 3.2.1.20) from Thermus caldophilus GK24 was expressed in Saccharomyces cerevisiae, a generally recognized as safe (GRAS) host. The thermostable $\alpha$-glucosidase was produced inside of the GRAS host at 0.04 unit/mg-dry cell by the constitutively expressing ADH1 promoter and at 1.2 unit/mg-dry cell by the inductively expressing GALl0 promoter, respectively. No $\alpha$-glucosidase activities were found in the medium when the MF-alpha signal sequence from S. cerevisiae or $\alpha$-amylase signal sequence from Aspergillus oryzae were fused before the $\alpha$-glucosidase gene for the secretion.

  • The thermophilic and thermostable alkaline phosphatase was purified to near homogeneity from the osmotic lysis of Thermus caldophilus GK24, The purified enzyme had an apparent molecular mass of 108, 000 Da and consisted of two subunits of 54,000 Da. lsoelectric-focusing analysis of the purified enzyme showed a pi of 7.3. The enzyme contained two Cys residues, and its amino acids composition was quite different from that of Thermus aquaticus YT-1 alkaline phosphatase and Escherichia coli alkaline phosphatase, The optimum pH and temperature of the enzyme were 11.0-11.5 and $80^{\circ}C$ respectively. The enzyme was stable in the pH range of 9.0-12.0 at $25^{\circ}C$ for 36 h. and the half-life at $80^{\circ}C$ (pH 11.0) was 6 h. The enzyme was activated by $MgCl_2$ and inhibited by EDTA. With ${\rho}-nitrophenyl\;phosphate\;({\rho}NPP)$ as the substrate, the enzyme had a Michaelis constant $(K_m) $of $3.6{\times}10^{-5}M$, The enzyme preferentially hydrolyzed the phosphomonoester bond of AMP in ribonucleotides and glycerophosphate.

  • A thermostable adenylate kinase isolated from the sonic extracts of Thermus caldophilus cells revealed higher substrate-specificity to the nucleoside monophosphate than to the nucleoside triphosphate. A $P', P^5$-di(adenosine-5') pentaphosphate was acted as a competitive inhibitor to the various substrates. Various divalent cations were activated the enzyme activity following orders: $Mg^{2+}, Ca^{2+}, Mn^{2+}, Ba^[2+}, $ and $Fe^{2+}$-. The enzyme activity was not affected by the sulfurhydryl reagent, p-chloromeric uribenzoic acid and activated by addition of the sodium chloride or phosphoenol pyruvate to the reaction mixture.

  • The gene encoding Thermus caldophilus GK24 polyphosphate kinase (Tca PPK) was cloned and sequenced. The gene contains an open reading frame encoding 608 amino acids with a calculated molecular mass of 69,850 Da. The deduced amino acid sequence of Tca PPK showed a 40% homology to Escherichia coli PPK, and $39\%$ to Klebsiella aerogenes PPK. The Tca ppk gene was expressed under the control of the T7lac promoter on pET-22b(+) in E. coli and its enzyme was purified about 70-fold with $36\%$ yield, following heating and HiTrap chelating HP column chromatography. The native enzyme was found to have an approximate molecular mass of 580,000 Da and consisted of eight subunits. The optimum pH and temperature of the enzyme were 5.5 and $70^{\circ}C$, respectively. A divalent cation was required for the enzyme activity, with $Mg^2+$ being the most effective.
