Ecology and Early Growth of Monostroma nitidum (Ulvales, Chlorophyta) in Korea (녹조류 참홑파래 (Monostromu nitidum)의 생태와 초기생장)

  • This study investigated the ecology and growth of Monostroma nitidum Wittrock in both its natural habitat and the laboratory. The maximum length, width, and weight of M. nitidum in March were 9.0$\pm$4.7 cm, 9.6$\pm$3.6 cm, and 1.52$\pm$1.13 g, respectively. Yellowish-green or yellowish-brown reproductive thalli began to appear in January, and over 80% of the thalli matured by March. The male and female spores were ca. 6 $\mu$m long, and elongate and ovoid in shape. The spores had two flagella and one-eye spot, and tended to swim toward light. Maximum number of spores released from matured thalli was 236 cells/mL after 70 minutes at a light intensity of 100 $\mu$mol/m$^2$/s. The zygote diameter ranged from 3.4-6.0 $\mu$m (mean 4.2 $$m) and increased to 69.8 $\mu$m 14 weeks after culture. The mass release of zoospores was observed from thalli in the dark (3 to 12 days), after 30 min under dry conditions in the shade, at 25$^{\circ}C$, and a light intensity of 100 $\mu$mol/m$^2$/s. The maximum number of zoospores released was 109.8 cells/mL after 60 min of induction. M. nitidum fronds on the net increased to 6.8-7.2 cm in length, and 6.6-8.9 cm in width during the winter.

  • This paper presents a comparative study of wave height estimation method that was used for signal to noise ratio and shadowing ratio based on X-band marine radar. If the signal to noise ratio, and is widely used as a method for estimating an wave height, a new method is presented for shadowing ratio. In the case of radar images used in this study it is measuring the data from the coast of Ulsan Jujeon, compared with marine meteorological information from the Meteorological Agency measured a light beacon. We compared the measured data for about 34 days, the typhoon was measured, incluidng a period in the East Sea, and verify the results for various distribution of wave height. For estimate wave height using a shadowing ratio analysis, it does not require calibration and real-time advantages of this part, coming confirmed the possibility of the measurement, the cause detection error for radar image was caused due to determine.

Effect of Oral Administration of Seaweed-derived Fucoidan on the Resistance of the Rockfish Sebastes schlegelii to High Temperature and Streptococcus iniae Infection (해조류 유래 후코이단(Fucoidan) 경구 투여가 조피볼락(Sebastes schlegelii)의 고수온 내성 및 Streptococcus iniae 감염 저항성에 미치는 영향)

  • Immunostimulating effects of fucoidan administration to rockfish Sebastes schlegelii at a concentration of 20 g/kg of diet were evaluated under high water temperature condition. The oral administration of fucoidan mixed with feed at a concentration of 20 g/kg of diet for 2 weeks increased the interleukin 1β gene in the intestine and kidney of fish by 5.7 and 6.3 times, respectively. In addition, when the water temperature was gradually increased from 24 to 31.4℃ for 2 weeks, LT50 delayed by 24 h in the fucoidan treated group compared to that in the control group, and mortality also reduced. Streptococcus iniae infection at a concentration of 1.50×100 CFU/fish at 28℃ delayed LT50 by 12 h in the fucoidan-treated group. Furthermore, the overall survival rate was 0% in the control group and 20% in the fucoidan-treated group. This study confirmed the applicability of dietary additives such as fucoidan as an immune activator of rockfish under high temperature condition.

Survey of Antibiotic Resistant Bacteria in Ulleungdo, Korea (울릉도의 항생제 내성균 조사)

  • BACKGROUND: Although antibiotics have contributed to treatment of bacterial infection, the antibiotic abuse can lead to antibiotic resistant bacteria. Impact of human activities on distribution of antibiotic resistance has been intensively issued and occurrence of antibiotic resistant bacteria in contaminated environments would not be a surprise. Nonetheless, anthropogenic contamination with the dissemination of antibiotic resistance along uncontaminated environments has been less considered. The aim of this study is to investigate antibiotic resistant bacteria across Ulleungdo, known as antibiotic resistance free and anthropogenic pollution free environment in Rep. of Korea. METHODS AND RESULTS: Antibiotic resistant bacteria in coastal seawater of Ulleungdo were investigated in July 2021. Antibiotic susceptibility test using the disk diffusion method was applied with six drugs according to the Clinical and Laboratory Standards Institute (CLSI) guideline. Total 43 bacterial isolates were tested and 20 isolates among of them showed multidrug resistance. Particularly, the number and ratio of resistant bacteria were relatively high in a densely populated area of Ulleungdo. The bacterial communities were investigated using 16S rRNA gene metabarcoding approach in the coastal seawater and soils of Ulleungdo. In the bacterial communities, Firmicutes were selectively distributed only in seawater, suggesting the possibility of anthropogenic contamination in coastal seawater of Ulleungdo. CONCLUSION(S): We found antibiotic resistant bacteria in a populated area of Ulleungdo. The occurrence of antibiotic resistant bacteria in Ulleungdo seems to result from the recent anthropogenic impact. Consistent monitoring of antibiotic resistant bacteria in the uncontaminated environment needs to considered for future risk assessment of antibiotics.

  • The twenty six specimens of leptocephali (15.8-32.6 mm TL) of the family Congridae, collected from southeastern waters offshore of Jeju Island during August 2014, and were identified by means of morphology and genetics. Those specimens were identified as belonging to the family Congridae based on various combinations of morphological characters. An analysis of the partial 12S rRNA sequences (886 base pairs) of mitochondrial DNA showed that our specimens must be Gnathophis nystromi, because their sequences were concordant with those of G. nystromi adult (genetic distance= 0.001), furthermore their total myomeres being consistent with those of G. nystromi adult. Catch rates of G. nystromi leptocephali were higher in the offshore regions than coastal regions of Jeju Island. The smallest leptocephali (< 20 mm TL) were collected offshore from Jeju Island. We hypothesize that one of the spawning grounds of G. nystromi is located offshore in the Jeju Island. In conclusion, the hatched preleptocephali of G. nystromi might have been transported from offshore near Jeju Island to the Korea Strait by the Kuroshio Current and Tsushima Warm Current.

  • The forming of fishing ground and the population ecological characteristics of yellowtail, Seriola quinqueradiata, in the coastal waters off Gim-nyeong of Jeju Island were investigated. The stock of yellowtail, Seriola quinqueradiata, between Jeju Island and coastal areas of the East Sea is probably the same. Water temperature probably is a major factor for controlling distribution of yellowtails in deeper, offshore areas off Jeju Island. However, the major factor that determines aggregation of yellowtails in coastal areas of Jeju Island, especially off Gim-nyeong is probably strong tidal currents driven by distribution of yellowtails rather than hydrological conditions such as Yellowtails collected off Jeju Island were from 1 to 4yrs old and about 50% of them were $1^{-yr}$ old, probably indicating overfishing. Jack mackerel was the major prey item for yellowtails off Gim-nyeong from October to March, suggesting concurrence of the two species.

  • For the geographical distribution of Primno macropa it was distributed over all stations investigated, except surface tow in East China Sea, through all seasons. It is believed that this species is most cosmopolitan species in the western North Pacific. Veritical distributional range of P. macropa indicates at depths from surface to more than 1,500m and most deeper recored for the vertical occurrence was obtained from depth of 1,650-2,220m in Station 229 $(34^\\circ 44.3'N, 140^\\circ 04.4'E)$, off Nojima-Zaki, Central Japan. For the vertical distributional and migrational pattern it is a typical diurnal migrant in the western North Pacific; at depths from 100m to 500m at night and 400m to 700m at day in Oyashio population, and from surface to 200m at night and from 100m to 300m at day in Kuroshio population. In Kuroshio area, the population of P. macropa was distributed in somewhat shallower layers than in Oyashio area and it is suggested that the populations is different in region and season according to their stages consisting the population.

Study on the by-catch prevention device of spring frame net trap for conger eel, Conger myriaster (붕장어 그물통발의 혼획 방지기구 개발에 관한 기초적 연구)

  • Present study was conducted to improve the spring frame net trap for conger eel, Conger myriaster which prevents by-catch and protects immature fish. A series of comparative fishing experiment was carried out in water off the south-east coast of Korea from Nov. 2004 to Jul. 2005. And analyzed the amount of catch, by-catch rate, and CPUE, etc. of experimental traps which were the spring frame net traps, sorted four types of flappers. And the compared gear was a plastic conger eel pot. The conclusions were as follows. By-catch rates of spring frame net trap with the compressed flapper bound by nylon mono-filament was about 0-3%. CPUE of spring frame net trap with the compressed flapper was about 50-60% lower than that of traditional and plastic pot. And in case of conger eel that the total length is over 350mm, CPUE was little different on each type of flapper of every mesh size.

Steroid Metabolism in the Blackfin Flounder Glyptocephalus stelleri during Oocyte Maturation (기름가자미(Glyptocephalus stelleri) 성숙기 난모세포에서의 성스테로이드 호르몬 대사물질 분석)

  • We studied oocyte steroidogenesis in the blackfin flounder Glyptocephalus stelleri as a region-specific species, in the East Sea of Korea during the spawning season. Maturing oocytes (0.76, 0.82, 0.88, and 0.91 mm in oocyte diameter) were incubated in vitro in the presence of [$^3H$] $17{\alpha}$-hydroxyprogesterone ($[^3H]17{\alpha}$-OHP) as a precursor. Steroid metabolites were extracted from the incubated medium and oocytes, and the extracts were separated and identified by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and gas chromatographymass spectrometry (GC/MS). The major metabolites produced from $[^3H]17{\alpha}$-OHP were androgens [androstenedione (A4) and testosterone (T)] and estrogens [$17{\beta}$-estradiol (E2) and estrone (E1)] and progestins [$17{\alpha},20{\alpha}$-dihydroxy-4-pregen-3-one ($17{\alpha}20{\alpha}P$) and $17{\alpha}20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$)] in maturing oocytes. The metabolic rate of $17{\alpha}20{\beta}$ was elevated (29.04%) in oocytes measuring 0.88 mm (nucleus migration stage following the induction of germinal vesicle breakdown), but was very low in oocytes measuring 0.76, 0.82, and 0.91 mm (0.42, 0.67, and 2.62%, respectively). From these results, we suggest that $17{\alpha}20{\beta}P$ acts as a maturation-inducing steroid in the blackfin flounder.

Isolation and Identification of FSHβ and LHβ Subunits in the Blacktip Grouper Epinephelus fasciatus (홍바리(Epinephelus fasciatus) FSHβ와 LHβ subunit의 분리 및 동정)

  • We cloned and sequenced the cDNA encoding the $FSH{\beta}$ and $LH{\beta}$ subunits from the pituitary of the blacktip grouper Epinephelus fasciatus, which regulate vitellogenesis and maturation in vertebrates, to achieve stable and healthy gametes. The full-length cDNAs of $FSH{\beta}$ and $LH{\beta}$ were 571 bp and 617 bp, encoding 120 amino acid (aa) and 147 aa proteins, respectively. The deduced amino acid sequences of $FSH{\beta}$ and $LH{\beta}$ were highly homologous (68-97%) to those of other Perciformes; E. bruneus, Dicentrarchus labrax, Thunnus thynnus, and Pseudolabrus sieboldi. Phylogenetic analysis showed that the deduced $FSH{\beta}$ and $LH{\beta}$ amino acid sequences were categorized as a distinct subunit in the $GTH{\beta}$ family, and are closely related to the teleostei $FSH{\beta}$ and $LH{\beta}$, respectively. $FSH{\beta}$ and $LH{\beta}$ mRNA exhibited high abundance in the pituitary gland and low in other brain areas, but were not present in peripheral tissues, as determined by RT-PCR.
