들깨잎에 함유된 Superoxide Dismutase의 특성 및 항산화 활성 검정 (Test of Superoxide Dismutase Characteristics and Antioxidant Activity in Perilla Leaves)

  • SOD(superoxide dismutase)는 유해 활성산소에 의한 생리적 장해를 방지하는 방어기작의 한 구성요소이다. 들깨잎 SOD의 특성을 알아보기 위한 NBT(nitro blue tetrazolium) 환원법 을 이용하여 들깨와 자소잎의 SOD동위효소의 종류와 활성 및 이들의 항산화 활성을 Fe$^{2+}$/ascorbate, Fe$^{3+}$ -ADP/NADPH첨가제를 처리하여 조사하였다. 들깨잎에는 품종에 따라 3~4개의 주요 SOD동위효소가 있었다. SOD는 함유하고 있는 금속조효소에 따라 3종류로 분류되는데, 들깨잎에는 두개의 Cu/ZnSOD와 두개의 FeSOD을 함유하고 있었으나, 자소잎은 단지 Cu/ZnSOD만을 함유하고 있었다. Cu/ZnSOD는 들깨 품종에 따른 차이가 없었으나, FeSOD는 분자량이 다른 두개의 FeSOD동위효소의 존재 양상이 품종에 따라 차이를 나타내었으며, SOD활성 및 항산화 활성도 품종에 따라 비교적 큰 차이를 보였다. 비효소적인 Fe$^{2+}$/ascorbate첨가 및 Fe$^{3+}$ -ADP/NADPH첨가에 의해 유도된 지질 과산화의 억제에 미치는 영향을 조사한 결과 공시재료중 밀양 2호가 가장 강한 항산화 활성을 가지고 있는 것으로 확인되었다. 자소 잎의 경우는 SOD활성과 유사하게 항산화 활성 정도 가장 낮아서 들깨잎과는 뚜렷 이 구별되었다.

베르베린을 처리한 간세포암에서 자가포식 경로와 관련된 세포자멸사 (Berberine Suppresses Hepatocellular Carcinoma Proliferation via Autophagy-mediated Apoptosis)

  • 간세포암종은 전 세계적으로 암 관련 사망의 두 번째 주요 원인으로 새로운 치료 전략을 필요로 한다. 간세포암종 환자를 치료에 사용되는 화학요법제는 독성이 있고 심각한 부작용이 있는 것으로 알려졌다. 본 연구는 건강한 간세포에서 세포독성을 일으키지 않으면서 종양세포를 표적으로 하는 부작용을 감소시키는 항암제의 효능을 조사하였다. Berberine은 이소퀴놀린 알칼로이드의 식물 유래 화합물로 다양한 약리학적 특성으로 인해 암 치료의 잠재적 후보군으로 보고되고 있다. 간암 세포 생존율에 대한 berberine의 효과는 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide 분석을 사용하여 측정하였다. HCC 증식은 콜로니 형성 분석을 통해 실시하였다. 세포이동에 대한 berberine의 효과는 상처 치유 분석으로 실시하였다. Berberine은 HepG2 세포와 같은 간암 세포의 증식과 콜로니 형성 및 세포이동을 억제하였다. Berberine 처리는 Beclin-1 및 LC3-II 등의 자가포식 관련 유전자 발현과 단백질 발현을 증가시켰으며, Caspase-3 및 Caspase-9등의 세포자멸사의 mRNA 발현 및 활성을 증가시켰다. 또한, 세포유래 이종이식 동물실험에서 berberine 처리에 따른 종양의 크기와 무게가 감소함을 확인하였다. 본 연구 결과는 간세포암에 대한 베르베린의 효능을 조명하여 표적 및 맞춤형 치료의 기회를 제시할 수 있음을 시사한다.

FT-NIR을 이용한 상추(Lactuca sativa L) 종자의 비파괴 선별 기술에 관한 연구 (Study on non-destructive sorting technique for lettuce(Lactuca sativa L) seed using fourier transform near-Infrared spectrometer)

  • Nondestructive evaluation of seed viability is one of the highly demanding technologies for seed production industry. Conventional seed sorting technologies, such as tetrazolium and standard germination test are destructive, time consuming, and labor intensive methods. Near infrared spectroscopy technique has shown good potential for nondestructive quality measurements for food and agricultural products. In this study, FT-NIR spectroscopy was used to classify normal and artificially aged lettuce seeds. The spectra with the range of 1100~2500 nm were scanned for lettuce seeds and analyzed using the principal component analysis(PCA) method. To classify viable seeds from nonviable seeds, a calibration modeling set was developed with a partial least square(PLS) method. The calibration model developed from PLS resulted in 98% classification accuracy with the Savitzky-Golay $1^{st}$ derivative preprocessing method. The prediction accuracy for the test data set was 93% with the MSC(Multiplicative Scatter Correction) preprocessing method. The results show that FT-NIR has good potential for discriminating non-viable lettuce seeds from viable ones.

Chemical Constitution, Morphological Characteristics, and Biological Properties of ProRoot Mineral Trioxide Aggregate and Ortho Mineral Trioxide Aggregate

  • Purpose: This study sought to compare the elemental constitution, morphological characteristics, particle size distribution, biocompatibility, and mineralization potential of Ortho MTA (OMTA) and ProRoot MTA (PMTA). Materials and Methods: OMTA and PMTA were compared using energy-dispersive spectrometry, particle size analysis, and scanning electron microscopy. The biocompatibility and mineralization-related gene expression (osteonectin and osteopontin) of both MTAs were also compared using methylthiazol tetrazolium assay and reverse transcription-polymerization chain reaction analysis, respectively. The results were analyzed by Kruskal-Wallis test with Bonferroni correction. P-value of <0.05 was considered significant. Result: The morphology of OMTA powders was similar to that of PMTA. The constituent elements of both MTAs were calcium, silicon, and aluminum. The mean particle sizes of OMTA and PMTA were 4.60 and 3.34 mm, respectively. Both MTAs had equally favorable in vitro biocompatibility and affected the messenger RNA expression of osteonectin and osteopontin. Conclusion: Within the limitations of this study, OMTA could be a promising biomaterial in clinical endodontics.

Biological Activities of Methyl-4-[[(2E)-3,7-dimethyl-oxy]-3-hydroxybenzoate

  • Methyl-4-[[(2E)-3,7-dimethyl-2,6-octadienyl]oxy]-3-hydroxybenzoate (5) has been identified from the New Zealand liverwort Trichocolea hatcheri (T. hatcheri) on the basis of spectroscopic evidence. This compound was tested for its growth inhibitory effects against tumor cell lines, using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. It showed growth inhibition activity against Staphylococcus epidermidis (MIC, 1,000 ${\mu}$g/mL). These results suggest that compound 5 possesses antitumoral , antimicrobial and antioxidative activities.

Epigallocatechin gallate의 인체 피부흑색종세포와 인체 구강유상피암종세포에 대한 성장억제효과 (The Growth Inhibitory Effects of Epigallocatechin Gallate Against Human Skin Melanoma Cells and Human Oral Epitheloid Carcinoma Cells)

  • Epigallocatechin gallate (EGCG) was reported to exert weak cytotoxicity against normal healthy cells such as C3H10T1/2 cells, but profound inhibitory effects on the initiation or promotion stage of chemical carcinogenesis in mammary gland, blood and mouse skin. This study was carried out to develop antitumor agents with weak side effects and strong antitumor activity. Human skin melanoma cells (HBT 69) and human oral epitheloid carcinoma cells (OCL 17) were cultured in RPMI-1640 media containing 10% fetal bovine serum, antibiotic, and fungizone. After incubation for 24 hrs, the cells were treated with various amounts of (EGCG) for 48 hrs. The growth inhibitory effects of EGCG in human oral epitheloid carcinoma cells were evaluated by the 3- (4,5-djmethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR), and sulforhodamine B protein (SRB) assays of colorimetric methods. The light microscopic study was also carried out to observe morphological changes of the treated cells. These results obtained were as follows; 1. Significantly inhibitory effects of EGCG against cultured human oral epithelioid carcinoma cells. 2. Significantly inhibitory effects against cultured human skin melanoma cells treated with 50 $\mu$M EGCG, but decreased inhibitory effects in 100 $\mu$M EGCG. 3. Degenerative changes against cultured human oral epitheloid carcinoma cells. 4. Degenerative changes against human skin melanoma cells treated with 50 UM EGCG, but recovered degenerative changes in 100 $\mu$M EGCG.

한국산 생약으로부터 항암물질의 개발 (제9보). 비색분석법에 의한 포공령 추출물의 항암평가 (Development of Anticancer Agents from Korean Medicinal Plants. Part 9. Antitumor Evaluation of Taraxaci Herba Extracts by Colormetric Methods.)

  • In the present study, we have evaluated cytotoxic effects of Taraxaci Herba extract in human oral epitheloid carcinoma cells. An antitumor activity was measured by colorimetric assays using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) and sulforhodamine protein B (SRB). The light microscopic study showed morphological changes, Ag-NOR (argyrophylic nucleolar organizer region) number and PAS positive reaction or the treated cells. These results obtained are as follows : MTT and SRB quantities were significantly decreased in cultured KB cells treated with 10$^{-2}$ /mg/ml and 10$^{-3}$ /mg/ml concentrations. The number of Ag-NORs were significantly decreased in cultured KB cells treated with 10$^{-2}$ /mg/ml and 10$^{-3}$ /mg/ml concentrations and the rate of Ag-NORs was shifted to left side (one Ag-Nounucleus was increased and five Ag-NORs/nucleus were decreased) by the high concentration. PAS reaction of cultured KB cells treated with 10$^{-2}$ /mg/ml and 10$^{-3}$ /mg/ml concentrations was negative. These results suggest that Taraxaci Herba retains a potential antitumor activity.

정상사람림프구와 HL-60 cell에서 목향의 세포독성과 백혈병세포 분화효과에 관한 연구 (Cytotoxicity in HL-60 cells and human lymphocytes and effect of leukemia cell differentiation induced by Saussureae Radix extract)

  • Objectives : This study was focused to investigate the toxicity of Saussurea lappa (SL) extracts in HL-60 cells and human lymphocytes. We also examined the differentiation effect of SL against leukemia cells. Methods : For examining the toxicity of SL, cytokinesis-block micronucleus (CBMN) assay and single cell gel eletrophoresis (SCGE) assay were used in present study. The cell differentiation effect of SL was evaluated by nitroblue tetrazolium (NBT) reduction assay. Results : The inhibition of cell growth in HL-60 cells was observed in a dose-dependant manner after SL treatment for 24 h. According to SCGE assay, HL-60 cells treated with SL increased DNA damage at $10{\mu}g/m{\ell}$, while DNA damage was induced by 0.1, 1, $10{\mu}g/m{\ell}$ concentration of SL in human lymphocytes. Our results indicated that SL have no genotoxic effect in HL-60 cells and human lymphocytes. Additionally, the differentiation effect was induced in $1{\mu}g/m{\ell}$ SL-treated HL-60 cells. Conclusions : From above results it is suggested that SL could be beneficial for the preparation of the useful agent for treating leukemia.

Anti-Helicobacter pylori, Cytotoxic, and Anti-inflammatory Activities of White Ginseng Extract

  • The anti-Helicobacter pylori activity, cytotoxicity, and anti-inflammatory activity of white ginseng extract (WGE) were investigated in vitro in this study. The antimicrobial effects of WGE toward H. pylori strains 52 J99, SSI, and 51 were tested using the disk diffusion method. Among these H. pylori strains, H. pylori 52 was the most sensitive, having the largest inhibition zone (19 mm), followed by J99, SSI, and 51. The zone of inhibition due to WGE increased significantly with increasing dosage. The cytotoxicity of WGE toward the human cancer cell lines A-549 (human lung carcinoma), HEC-1-B (human endometrial adenocarcinoma), HeLa (human uterin adenocarcinoma), and SW-156 (human kidney carcinoma) was measured using the 3-(4,5-dimethylthizol-2-yl)-2,5-diphenylate-tetrazolium bromide (MTT) assay. WGE exhibited an inhibitory effect on cell growth at 2.0 mg/mL for all tumor cell lines. An analysis of anti-inflammatory activity using the RAW 264.7 cell line showed that the inhibition of nitric oxide (NO) production increased as the WGE content increased. These results demonstrate the potential of WGE to be used as a health-promoting substance.

빨간유글레나, Euglena rubra 건조분말의 사료첨가에 따른 나일틸라피아, Oreochromis niloticus에서의 영향 (Effects of feeding of dried Euglena rubra on Nile tilapia Oreochromis niloticus)

  • To examine the effects of Euglena rubra (E. rubra) feeding on Nile tiapia, Oreochromis niloticus were fed diets containing dried powder of E. rubra at 0.5 or 2% for 4 weeks. Growth, selected hematological and non-specific immune parameters were assessed at 2 and 4 weeks. There were no significant changes in body weigh gain and erythrocyte levels. However a significant and diet E. rubra level-related decrease in leukocyte level was noted. Significant increases were observed in respiratory burst activity (nitroblue tetrazolium reduction) and lysozyme activities following E. rubra feeding. These results indicate that E. rubra could be beneficial to fish, but excess feeding could toxic.
