Cultivation Limit of Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis at Coastal Area and Physiological Vitality of RAW 264.7 cell and HL-60 cell (해안지대 순비기나무, 번행초, 갯방풍의 재배한계 및 RAW 264.7 세포와 HL-60세포의 생리활성)

  • The cultivation boundary areas and biological activator efficacies of Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis were investigated. Investigation of coastal areas revealed the East and West cultivation boundary areas of Vitex rotundifolia and Gelhnia littoralis to be the Taean Peninsula, Taean-gun and Gangwon-do, Donghae-City. The East and West coastal cultivation boundary areas of Tetragonia tetrragonoides are Jeollanam-do and Kyeongsangbuk-do, Ulleung-gun. The natural habitat is less than 200 m from the coastal area. Most of the Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis grow naturally in a region of mixed sand, clay and loam, in direct sunlight. No dominant vegetation was located. To analyze anti-inflammatory efficacy, RAW 264.7 cells were used. Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis inhibited inflammation by 37%, 12% and 10% in $100{\mu}g/m{\ell}$, respectively. However, the anti-inflammatory efficacy of Glehnia littoralis was invalid. Analysis of anti-cancer activity was accomplished using HL-60 cells. Glehnia littoralis revealed 60% and 72% inhibition activity in $100{\mu}g/m{\ell},\;and\;200{\mu}g/m{\ell}$ respectively. No cytotoxicity inhibition activity was investigated in Vitex rotundifolia and Tetragonia tetragonoides.

Inhibitory Effect of Tetragonia tetragonoides Water Extract on the Production of $TNF-{\alpha}$ and Tryptase in Trypsin-Stimulated Human Mast Cells

  • Tetragonia tetragonoides (Aizoaceae) has been known as an anti-cancer agent. The activation of proteinase-activated receptor-2 (PAR-2) by trypsin appears to play a role in inflammation. In the present study, we examined the inhibitory effects of Tetragonia tetragonoides water extract (TTWE) on the production of tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ and tryptase in trypsin-stimulated human leukemic mast cells (HMC-1) expressing PAR-2. HMC-1 cells were stimulated with trypsin in the presence or absence of TTWE (10, 100, and $1000\;{\mu}g/ml$). The level of $TNF-{\alpha}$ secretion from HMC-1 cells was measured by enzyme-linked immunosorbent assay (ELISA). $TNF-{\alpha}$ and tryptase mRNA expression were examined by reverse transcription-PCR. Also, extracellular signal-regulated kinese (ERK) activation was assessed by Western blot analysis. Trypsin activity was measured using the substrate Bz-DL-Arg-p-nitroanilide (BAPNA). It was observed that $TNF-{\alpha}$ secretion, tryptase mRNA and $TNF-{\alpha}$ mRNA expression in trypsin-stimulated HMC-1 cells were inhibited by pretreatment of TTWE ($1000\;{\mu}g/ml$). Furthermore, the pretreatment of TTWE ($1000\;{\mu}g/ml$) resulted in the reduction of ERK phosphorylation and trypsin activity. These results suggest hat TTWE might have the inhibitory effects on the PAR-2-dependent inflammation processes and it is likely to function as PAR-2 antagonist.

  • A protocol has been developed for differentiation of adventitious shoots directly from leaf segments of Tetragonia tetragonoides O. Kuntze. Murashige and Skoog (MS) medium supplemented with 2 mg/L $N^6-benzyladenine$ (BA) and 0.5 mg/L ${\alpha}-naphthaleneacetic$ acid (NAA) supported the induction of adventitious shoots from leaf explants. Adventitious shoots were multiplied by subculturing on the double strength MS (2MS) medium supplemented with 0.5 mg/L NAA and 2 mg/L BA. Shoots were rooted on MS basal medium without any growth regulators.

  • It was found that 14 coastal habitats in South Korea have comparetively natural vegetation. The habitats were classified into three types - intertidal flats, sand dunes, and estuaries. There were four intertidal flats, five sand dunes and five estuaries. Except for Cynodon dactylon and Tetragonia tetragonoides, all of the main halophytes and sand dune plants were found in the habitats. These two species were mainly distributed on the southern coast. This study identified coastal vegetation, such as pure stands of Suaeda japonica on intertidal flats, mixed halophyte communities around the high-water mark, pure stands of Vitex rotundifolia on stable sand dunes, mixed communities dominated by Carex kobomugi on unstable sand dunes, and pure stands of Phragmites communis in estuaries. The types of coastal vegetation may depend on sediment types, the inundation time of seawater and the stability of sediments.

Screening of Peroxynitrite and DPPH Raoical Scavenging Activities from Salt Marsh Plants (염생식물로부터 Peroxynitrite와 DPPH 라디칼 소거 활성 검색)

  • A peroxynitrite is formed when superoxide and nitric oxide exist at near eqimolar ratio in biological systems. Although not a free radical by chemical nature, peroxynitrite is a powerful oxidant having a wide array of tissue damaging effects ranging from lipid oxidation and inactivation of enzymes and ion channels through protein oxidation and nitration to inhibition of mitochondrial respiration. During our search for new antioxidizing components from natural resources, twenty salt marsh plants were screened for their ONOO and DPPH radical scavenging activities. Among them, methanol extract of Rosa rugosa, lxeris tamagawaensis, Erigeron annus, Tetragonia tetragonoides, Imperata cylindrica, and Suaeda japonica inhibited more than 85% of peroxynitrite produced by 3-morpholinsydnonimine (SIN-1) at a concentration of 5 $\mu\textrm{g}$/$m\ell$. In addition, Rosa rugosa, Artemisia capillaris, Erigeron annus and Ixeris tamagawaensis showed significant scavenging effect against DPPH (1,1-diphenyl-2-picrylhydrazyl radical).

Antioxidant and Anti-inflammatory Activities of Tetragonia tetragonoides Water Extract (번행초 추출물의 항산화 및 항염증 효과)

  • Objectives : Tetragonia tetragonioides is one of the traditional herbal medicines that can be used to protect the stomach and treat cancer. However, its mechanism of overcoming gastrointestinal disorders is unclear. In this study, we investigated antioxidant and anti-inflammatory effects of Tetragonia tetragonioides Water Extract (TTWE) on RAW264.7 cells. Methods : The cell viability by TTWE was measured by using MTT assay. The free radical scavenging ability and cytokine production were analyzed by using ELISA Kit. SPSS version 25 was used for statistical analysis. Results : According to the results of this study, the cell viability measurement of TTWE significantly affected the cell viability. The radical scavenging ability of TTWE showed the highest effect compared to the positive control group when the concentration was 3.1-12.5 ㎍/ml, and significantly inhibited NO production induced by LPS. In addition, the inhibitory effect of TTWE on the production of IL-6 and TNF-α induced by LPS was significant at both TTWE concentrations of 12.5 ㎍/ml [p <0.01 (IL-6), p <0.05 (TNF-α)]. Conclusion : In conclusion, it is suggested that the antioxidant function of Tetragonia tetragonioides Water Extract could be used to prevent and treat inflammatory diseases.

Antidiabetic Activity of Polysaccharide Extract from Tetragonia tetragonoides in Streptozotocin-induced Diabetic Mice (스트렙토조토신으로 유도한 당뇨 마우스에서 번행초 다당 추출물의 항당뇨 효과)

  • Tetragonia tetragonioides seems to be a promising antiulcer medicinal plant due to the presence of water-soluble polysaccharide and cerebroside as its major constituents. There have been no previous studies using T. tetragonioides polysaccharide extract (TPE) to assess its antidiabetic effect in streptozotocin (STZ)-induced diabetes in mice. This study was designed to evaluate the antidiabetic effect of TPE in diabetic mice, which was established by one-week intraperitoneal injection of 65 mg/kg STZ. After three weeks of TPE treatment at a dose of 100 mg/kg, a maintenance of body weight, a decrement in plasma glucose, and low levels of triglyceride, lactate dehydrogenase, alkaline phosphatase, and glutamic pyruvic transaminase were observed in diabetic mice. Furthermore, the ingestion of TPE lowered the blood glucose levels during the oral glucose tolerance test (OGTT) and restored most of the tested parameters to their normal levels. Therefore, the antidiabetic potential of T. tetragonioides has been demonstrated for the first time by our research.

Anti-inflammatory and Anti-tumor Effects of Tetragonia tetragonoides Extracts (번행초 추출물의 항염증 및 종양억제 효과)

  • We examined the anti-inflammatory effect and anti-tumor activity of Tetragonia tetragonioides crude extracts and fractions. The anti-inflammatory activity of T. tetragonioides was exuded through the inhibition of lipopolysaccharide (LPS, $1{\mu}g/ml$), induced nitric oxide (NO) and interleukin (IL)-$1{\beta}$ production. The production of IL-6 and tumor necrosis factor $(TNF)-{\alpha}$ also decreased in LPS induced RAW264.7 cells after treatment with polysaccharide (PS) fraction. Furthermore, the hexane (HX) fraction strongly inhibited the granulocytes macrophage-colony stimulating factor (GM-CSF) production. In ICR mice previously inoculated with Sarcoma 180, the life prolongation effects were 16.67% with an intraperitoneal injection of methanol (MeOH) extract and polysaccharide fraction at a dose of 100 mg/kg/day. The results are an important preliminary step toward the development of effective anti-inflammatory and anti-tumor agents using T. tetragonioides.
