Cyanobacterial Taxonomy: Current Problems and Prospects for the Integration of Traditional and Molecular Approaches

  • The application of modern ecological, ultrastructural and molecular methods, aided by the cultivation of numerous cyanobacterial morphotypes, has substantially changed our knowledge of these organisms. It has led to major advances in cyanobacterial taxonomy and criteria for their phylogenetic classification. Molecular data provide basic criteria for cyanobacterial taxonomy; however, a correct phylogenetic system cannot be constructed without combining genetic data with knowledge from the previous 150 years research of cyanobacterial diversity. Thus, studies of morphological variation in nature, and modern morphological, ultrastructural, ecophysiological and biochemical characters need to be combined in a “polyphasic” approach. Taxonomic concepts for generic and infrageneric ranks are re-evaluated in light of combined phenotypic and molecular criteria. Despite their usefulness in experimental studies, the limitations of using strains from culture collections for systematic and nomenclatural purposes is highlighted. The need for a continual revision of strain identification and proper nomenclatural practice associated with either the bacteriological or botanical codes is emphasized. Recent advances in taxonomy are highlighted in the context of prospects for understanding cyanobacterial diversity from natural habitats, and the evolutionary and adaptational processes that cyanobacteria undergo.

IS 아웃소싱 프로젝트 관리를 위한 통제의 실증적유형에 관한 탐색적 연구 (An Exploratory Study on the Taxonomy of Control Types in IS Outsourcing Project Management)

  • This study examines control types of IS outsourcing project management focusing on two sets of questions: What control types are implemented during IS outsourcing projects? How various control types affect the performance of IS outsourcing projects. In order to meet these objectives, four control types are developed using typology-approach: dynamic, contract-oriented, partnership-oriented, and passive. And then four control types are identified based on taxonomy-approach using cluster analysis from 66 projects: dynamic, contract-oriented, passive, and middle. The result shows that the derived taxonomy-types are similar to the typology-types except partnership-oriented type. The result also indicates that dynamic and contract-oriented types are in the highest performance, while passive type is in the least performance.

데이터기반 의사결정을 위한 정책 및 사업 속성 분류체계 개발 연구 (A Study on Development of Policy Attributes Taxonomy for Data-based Decision Making)

  • Purpose Due to the complexity of policy environment in modern society, it is accepted as common basics of policy design to mix up a variety of policy instruments aiming the multiple functions. However, under the current situation of written-down policy specification, not only the public officers but also the policy researchers cannot easily grasp such frameworks as policy portfolio. The purpose of this study is to develop "Policy Attributes Taxonomy" identifying and classifying the public programs to help making decisions for allocative efficiency with effectiveness-based information. Design/methodology/approach To figure out the main scheme and classification criteria of Policy Attributes Taxonomy which represents characteristics of public policies, previous theories and researches on policy components were explored. In addition, to test taxonomic feasibility of certain information system, a set of "Feasibility Standards" was drawn from "requirements for well-organized criteria" of eminent taxonomy literatures. Finally, current government classification system in the area of social service was tested to visualize the application of Taxonomy and Standards. Findings Program Taxonomy Schemes were set including "policy goals", "policy targets", "policy tools", "logical relation" and "delivery system". Each program and project could be condensed into these attributes, making their design more easily distinguishable. Policy portfolio could be readily made out by extracting certain characteristics according to this scheme. Moreover, this taxonomy could be used for rearrangement of present "Program Budget System" or estimation of "Basic Income".

중소기업의 전략적 지향성과 탐험과 활용관점에 의한 개방형 혁신전략의 실증적 유형과 경영성과 (Strategic Orientation of SMEs and Exploration and Exploitation Perspective of Open Innovation And Business Performance)

  • 본 연구에서는 분류학적 접근(taxonomy approach)을 통해, 기업이 전략적 지향성과 조직 기술 학습전략의 차원인 탐험과 활용의 연관성을 통해 기업의 경영성과의 관계에 대해 새로운 시각을 제시한다. 한국의 중소기업 141개 회사의 전략지향성과 조직의 탐험과 활용 및 경영성과에 대한 설문지를 통한 인지적 자료 사용하여 이들의 다양한 전략지향성과 기술 학습전략은 어떠한 귀납적 유형으로 도출되는지를 살펴보고자 하였으며, 도출된 유형 중 어떠한 전략지향성과 기술학습을 추구 할 때 기업의 성과에 어떠한 긍정적인 영향을 미치는 지를 살펴보고자 하였다. 구체적으로는 시장-탐험형을 추구하는 유형이 경영성과가 가장 높은 것으로 나타났다. 이와 같은 결과는 시장지향성을 추구하며, 탐험적 기술혁신을 추구하는 것이 다른 유형의 전략유형에 비해 유의적인 경영성과의 차이를 보인다는 것을 의미하며, 한국의 중소기업들이 전략의 지향성과 기술학습의 전략을 선택할 때 매우 유용한 정보를 제공할 수 있다. 반대로 기술-탐험형 전략 유형은 경영성과가 가장 낮은 것으로 나타나 이들의 전략을 선택할 때 역시 고려해야 할 요소로 파악된다.

Restricting Answer Candidates Based on Taxonomic Relatedness of Integrated Lexical Knowledge Base in Question Answering

  • This paper proposes an approach using taxonomic relatedness for answer-type recognition and type coercion in a question-answering system. We introduce a question analysis method for a lexical answer type (LAT) and semantic answer type (SAT) and describe the construction of a taxonomy linking them. We also analyze the effectiveness of type coercion based on the taxonomic relatedness of both ATs. Compared with the rule-based approach of IBM's Watson, our LAT detector, which combines rule-based and machine-learning approaches, achieves an 11.04% recall improvement without a sharp decline in precision. Our SAT classifier with a relatedness-based validation method achieves a precision of 73.55%. For type coercion using the taxonomic relatedness between both ATs and answer candidates, we construct an answer-type taxonomy that has a semantic relationship between the two ATs. In this paper, we introduce how to link heterogeneous lexical knowledge bases. We propose three strategies for type coercion based on the relatedness between the two ATs and answer candidates in this taxonomy. Finally, we demonstrate that this combination of individual type coercion creates a synergistic effect.

Identification of Viral Taxon-Specific Genes (VTSG): Application to Caliciviridae

  • Virus taxonomy was initially determined by clinical experiments based on phenotype. However, with the development of sequence analysis methods, genotype-based classification was also applied. With the development of genome sequence analysis technology, there is an increasing demand for virus taxonomy to be extended from in vivo and in vitro to in silico. In this study, we verified the consistency of the current International Committee on Taxonomy of Viruses taxonomy using an in silico approach, aiming to identify the specific sequence for each virus. We applied this approach to norovirus in Caliciviridae, which causes 90% of gastroenteritis cases worldwide. First, based on the dogma "protein structure determines its function," we hypothesized that the specific sequence can be identified by the specific structure. Firstly, we extracted the coding region (CDS). Secondly, the CDS protein sequences of each genus were annotated by the conserved domain database (CDD) search. Finally, the conserved domains of each genus in Caliciviridae are classified by RPS-BLAST with CDD. The analysis result is that Caliciviridae has sequences including RNA helicase in common. In case of Norovirus, Calicivirus coat protein C terminal and viral polyprotein N-terminal appears as a specific domain in Caliciviridae. It does not include in the other genera in Caliciviridae. If this method is utilized to detect specific conserved domains, it can be used as classification keywords based on protein functional structure. After determining the specific protein domains, the specific protein domain sequences would be converted to gene sequences. This sequences would be re-used one of viral bio-marks.

한국 철도시스템의 해외진출을 위한 시장유형론적 접근 (A Market Taxonomy Approach for the Export of Korean Railway Systems)

  • 한국 철도시스템의 해외진출을 위한 효과적 전략을 모색함에 있어 시장유형론적 접근이 필수적이다. 시장선택을 통하여 우선순위 시장을 선별하고 시장세분화를 통하여 각 하위시장의 특성에 적합한 마케팅 전략을 설계하여야 한다. 이를 위하여 본 연구는 글로벌 철도공급시장에 적합한 시장유망도 지표체계와 시장세분화 방안을 제시하였다. 아울러 시장유형론적 관점에서 최근 한국의 주요 철도기업이 추진하고 있는 수출유망 프로젝트를 분석하고 철도시스템 수출을 위한 전략적 시사점을 도출하였다.

First record of a marine microalgal species, Jaagichlorella roystonensis (Trebouxiophyceae) isolated from Jungmun Saekdal Beach, Jeju Island, Korea

  • A eukaryotic marine microalga was isolated from Jungmun Saekdal Beach, Jeju Island, Korea and an integrated approach, including molecular phylogeny and morphology, was used to determine its taxonomical status. Molecular phylogenetic evidence inferred from the small subunit (SSU) 18S rRNA sequence and internal transcribed spacer (ITS) secondary structure analysis clearly showed that the isolate belonged to the recently described species, Jaagichlorella roystonensis. Distinctive morphological keys of the species were also observed by light microscopy and scanning/transmission electron microscopy(S/TEM). In this study, a Korean marine J. roystonensis species was described for the first time and was subsequently added to the national culture collections in Korea.

대학도서관 서비스 가치의 분류체계 적용에 관한 연구 (An Application of the Derived Taxonomy of Service Value for University Libraries)

  • 본 연구는 Saracevic과 Kantor(1997)가 개발한 도서관 서비스 가치의 분류체계를 적용하여 국내 도서관 이용자 16명을 대상으로, 이용자가 경험하고 표현하는 서비스 가치를 평가하였다. 이용자와의 직접적인 면담을 통해 이용자가 도서관서비스를 사용하게 된 동기 사용과정에서의 상호작용, 그리고 서비스의 이용을 통해 경험한 결과를 이미 개발된 분류체계의 구체적인 요소를 적용하였다. 이를 통해 다양한 목적으로 도서관 서비스가 사용되며 이용자 본인의 말로 표현된 서비스 가치를 포착, 분류할 수 있었다. 본 연구에서 사용된 면담 기법 등을 적절히 활용하면 이용자들로부터 서비스 가치와 관련된 구체적이고, 사실적인 언급을 수집할 수 있으며, 기존에 개발된 분류체계는 높은 포괄성과 적용성을 지닌 것으로 평가되었다.

Single-cell PCR on protargol-impregnated euplotid ciliates: a combined approach of morphological and molecular taxonomy

  • Ciliates are considered one of the most diverse protozoa and play significant roles in ecology. For successful taxonomic study of these microscopic eukaryotes, a staining procedure is necessary, due mainly to intrinsic difficulties in recognizing characteristics from living cells. Although molecular taxonomy has been used to resolve the ambiguities associated with traditional morphology-based taxonomy, extraction of genomic DNA from stained ciliate cells is not available yet. In the present study, we describe a method to extract genomic DNA from a single protargol-impregnated euplotid cell. By using $HgCl_2$ as a fixative and modulating the exposure time of bleach solution in the protargol impregnation, high-quality genomic DNA can successfully be extracted from a stained single cell with minimal loss of morphological integrity. This technique will contribute to the effectiveness of combined approaches of molecular and morphological taxonomy from single ciliate cells.
