  • Objectives: This study aimed to evaluate the effect of pomegranate solution (Punica granatum) on eroded dentin through antioxidant action, shear bond strength (SBS) and interface morphology. Materials and Methods: The 10% pomegranate peel extract was prepared by the lyophilization method. Punicalagin polyphenol was confirmed by high-performance liquid chromatography. Antioxidant activity was evaluated by capturing the 2,2-diphenyl1-picrylhydrazyl (DPPH) radical. For the SBS, 48 dentin fragments were divided into sound or eroded, and subdivided according to the pretreatment (n = 12): water or P. granatum. The surfaces were restored with self-etch adhesive and a bulk-fill resin (Ecosite; DMG). The SBS was done immediately (24 hours) and after thermal cycling + water storage (12 months). For scanning electron microscopy, 48 dentin fragments (24 sound and 24 eroded) received the same treatments as for SBS (n = 6), and they were analyzed after 24 hours and 12 months. Results: The P. granatum had antioxidant action similar (p = 0.246) to the phenolic standard antioxidants. After 24 hours, eroded dentin had lower SBS than sound dentin (p < 0.001), regardless of the pretreatment. After 12 months, P. granatum maintained the SBS of sound dentin (13.46 ± 3.42 MPa) and eroded dentin (10.96 ± 1.90 MPa) statistically similar. The lowest values were found on eroded dentin treated with water (5.75 ± 1.65 MPa) (p < 0.001). P. granatum on eroded dentin caused peritubular demineralization and hybrid layer with resin tags. Conclusions: The pomegranate extract had antioxidant action and preserved the adhesive interface of the eroded dentin.

  • With the advancement and proliferation of the Internet of Things (IoT), a wide range of location-based services are being offered, and various ranging methods are being researched to meet the objectives of the required services. Conventional ranging methods involve the direct exchange of signals between tags and anchors to estimate distance, presenting a limitation in efficiently utilizing communication resources in large-scale IoT environments. To overcome these limitations, active-passive ranging methods have been proposed. However, there is a lack of theoretical convergence guarantees against clock drift errors and a detailed analysis of the characteristics of ranging estimation techniques, making it challenging to derive precise positioning results. In this paper, an improved active-passive ranging method that accounts for clock drift errors is proposed for precise positioning in large-scale IoT environments. The simulation results confirmed that the proposed active-passive ranging method can enhance distance estimation performance by up to 94.4% and 14.4%, respectively, compared to the existing active-passive ranging methods.

  • Recombinant immunotoxins (RITs) are fusion proteins consisting of a targeting domain linked to a toxin, offering a highly specific therapeutic strategy for cancer treatment. In this study, we engineered and characterized RITs aimed at mesothelin, a cell surface glycoprotein overexpressed in various malignancies. Through an extensive screening of a large nanobody library, four mesothelin-specific nanobodies were selected and genetically fused to a truncated Pseudomonas exotoxin (PE24B). Various optimizations, including the incorporation of furin cleavage sites, maltose-binding protein tags, and tobacco etch virus protease cleavage sites, were implemented to improve protein expression, solubility, and purification. The RITs were successfully overexpressed in Escherichia coli, achieving high solubility and purity post-purification. In vitro cytotoxicity assays on gastric carcinoma cell lines NCI-N87 and AGS revealed that Meso(Nb2)-PE24B demonstrated the highest cytotoxic efficacy, warranting further characterization. This RIT also displayed selective binding to human and monkey mesothelins but not to mouse mesothelin. The competitive binding assays between different RIT constructs revealed significant alterations in IC50 values, emphasizing the importance of nanobody specificity. Finally, a modification in the endoplasmic reticulum retention signal at the C-terminus further augmented its cytotoxic activity. Our findings offer valuable insights into the design and optimization of RITs, showcasing the potential of Meso(Nb2)-PE24B as a promising therapeutic candidate for targeted cancer treatment.

  • The existing indoor positioning methods using beacons or tags suffer from issues such as occasional undetection or increased errors due to noise. In this paper, we propose a method for determining the indoor position of a robot using the distance and, the angle between the direction of a target object whose position is known and the direction in which the robot views the target object from the front. The method proposed in this paper utilizes a depth camera to measure distance and calculate angles. Distance is measured using depth information captured by the camera, while angles are determined using images captured by the camera to determine the orientation of the target object. The proposed method calculates coordinate displacements using distance and angle. And then the method determines the position of the mobile robot using these displacements and the coordinates of the target object. To show the applicability of the proposed method for indoor positioning, we conducted experimental implementation and compared measured displacements. The results showed errors within 50mm, but considering the size of the mobile robot, it is judged that the method proposed in this paper can be sufficiently used for indoor positioning.

  • The passive integrated transponder (PIT) telemetry is a useful method for investigating fish population dynamics, community structure and migration. It can be applied for small fishes (TL<100 mm) because of its tiny size and light weight. The survival rate of PIT tag was investigated on 4 small size cyprindae fish species, Carassius gibelio langsdorfi (n=34, standard length; $91.9{\pm}0.9mm$, body weight; $21.2{\pm}0.9g$), Hypophthalmichthys molitrix (n=16, SL; $75.1{\pm}0.9mm$, BW; $6.0{\pm}0.2g$), Pseudorasbora parva (n=30, SL; $51.4{\pm}1.1mm$, BW; $2.7{\pm}0.2g$) and Phoxinus phoxinus (n=37, SL; $70.6{\pm}1.4mm$, BW; $8.2{\pm}0.5g$) under age 1 for applicability and effectiveness. We used three type tags including a small (length 11.0 mm, diameter 2.1 mm, weight 0.088 g), middle (20 mm, 3.5 mm, 0.102 g), large (30 mm, 3.5 mm, 0.298 g) size. After 30 days of tag insertion, survival rate of 117 individuals were 58.1% (large tag, 50.0%; middle tag, 57.5%; small tag, 61.4%). Survival rates varied between three types of tags because the abdominal cavity of each individual was different size. The death was due to surgical damage. If we apply tagging systems on field research of the Korean freshwater fish, the PIT tag will be effective method for analyzing fish ecology.

  • This study was performed to compare the shear bond strength of AQ Bond $Plus^{TM}$ with AQ $Bond^{TM}$ and Single $Bond^{TM}$. Also by observing the fractured interface under scanning electro-microscope, the fracture pattern and the quality of hybrid layer were analyzed. The possibility of clinical application of all-in-one system which has an advantage to reduce chair time for children with difficult behavior pattern was evaluated, The results obtained are as follows ; 1. There was no significant difference between AQ $Bond^{TM}$ and AQ Bond $Plus^{TM}$ in shear bond strength and Single Bond showed the highest bond strength with statistical significant difference (p<0.05). 2. Adhesive fracture pattern was mainly observed in both enamel/dentin in AQ $Bond^{TM}$ and AQ Bond $Plus^{TM}$ group while Single Bond group showed equal numbers for cohesive and adhesive pattern. 3. Under scanning electro-microscope, resin tags observed in AQ $Bond^{TM}$ and AQ Bond $Plus^{TM}$ were very weak and tangled while strong and thick tags were shown with many lateral branches in Single Bond. Careful case selection and accurate clinical application is recommended when using AQ $Bond^{TM}$ and AQ Bond $Plus^{TM}$considering the result showing its weaker strength than Single $Bond^{TM}$.

  • Recently. the reintroduction of air-abrasion technology in dentistry has added a new potential method of pretreating teeth prior to placing sealants. The purpose of this in vitro study was to investigate microleakages of several pit and fissure sealants following various surface pre-treatment methods to the enamel, because there was a question concerning the validity of claim that this method was better than the conventional acid etching. Permanent molars were divided randomly into nine groups and treated accordingly: acid etching(group 1, 4, 7), air-abrasion(group 2, 5, 8), acid etching after air-abrasion(group 3, 6, 9). Then the authors placed and cured the three kinds of sealants(group $1\sim3$: Teethmate-F, group $4\sim6$: Ultraseal XT plus, group $7\sim9$: Denseal) according to the manufacturer's order. They were observed to determine the degree of microleakage. And these specimens were separated from the corresponding enamels and examined by Scanning Electron Microscope. The following results were obtained: 1. In comparing microleakage among tooth surface treatment methods, air-abrasion group(group 2, 5, 8) showed the greatest microleakage, while combination(air-abrasion + acid etching) group showed the least (p<0.05). However, no significant differences were found between group 7 and 9. 2. The mean microleakages were ranked as follows(p<0.05): In acid etching groups, group 7> group 4> group 1. In air-abrasive groups, group 8>groups 5> group 2. In combination groups, group 9>group 3> group 6. However, no significant differences were found between group 4 and 7 between group 2 and 5 and between group 3 and 6. 3. SEM showed that comparably longer resin tags were distributed regularly in acid etching groups and that shorter ones irregularly in air-abrasion groups. It also showed that these two kinds of tags were distributed simultaneously in combination groups.

  • The thermotolerant methylotrophic yeast Hansenula polymorpha is an attractive model organism for various fundamental studies, such as the genetic control of enzymes involved in methanol metabolism, peroxisome biogenesis, nitrate assimilation, and resistance to heavy metals and oxidative stresses. In addition, H. polymorpha has been highlighted as a promising recombinant protein expression host, especially due to the availability of strong and tightly regulatable promoters. In this study, we investigated the possibility of employing human serum albumin (HSA) as the fusion tag for the secretory expression of heterologous proteins in H. polymorpha. A set of four expression cassettes, which contained the methanol oxidase (MOX) promoter, translational HSA fusion tag, and the terminator of MOX, were constructed. The expression cassettes were also designed to contain sequences for accessory elements including His8-tag, $2{\times}(Gly_4Ser_1)$ linkers, tobacco etch virus protease recognition sites (Tev), multi-cloning sites, and strep-tags. To determine the effects of the size of the HSA fusion tag on the secretory expression of the target protein, each cassette contained the HSA gene fragment truncated at a specific position based on its domain structure. By using the Green fluorescence protein gene as the reporter, the properties of each expression cassette were compared in various conditions. Our results suggest that the translational HSA fusion tag is an efficient tool for the secretory expression of recombinant proteins in H. polymorpha.

  • Asymmetric 1,2-distearoyl-3-oleoyl-rac-glycerol (SSO) triacylglycerol (TAG) is used as a cocoa butter replacer (CBR). In this study, it was produced by lipase-catalyzed interesterification of fully hydrogenated soybean oil (FHSBO) and oleic ethyl ester (OEE) in a batch type reactor at $75^{\circ}C$, 250 rpm. Different molar ratios (FHSBO : OEE=1:1, 1:2 and 1:3, w/w) and various reaction times (1, 2, 3, 4, and 5 hr) were also tested. The optimized condition for SSO was a FHSBO : OEE molar ratio of =1:1 at reaction times of 2, 3, 4, and 5 hr. Enzymatic synthesis generated SSO/SOS, as well as the other TAGs (e.g., PSO/POS, SOO/OSO, SSS), ethyl esters, monoacylglycerol (MAG), and diacylglycerol (DAG). After scale-up, fractionation by solvent (methanol and acetone) fractionation and column chromatography was applied. To reduce ethyl esters, high-melting TAGs (e.g., SSS), and SOO/OSO in reactants, solvent fractionation was applied. Using a silica gel column (sample : silica gel=2:1, wt%), MAG and DAG were removed at $25^{\circ}C$. The major fatty acid composition of the final products (with a high SSO/SOS content) was palmitic acid (C16:0, 10.9~12.9 area%), stearic acid (C18:0, 52.2~54.9 area%), and oleic acid (C18:1, 34.2~35.5 area%). In reversed-phase HPLC analysis, the major TAG species of the final product (FHSBO : OEE=1:1, 2 hr) were SSO/SOS (82.31 area%) and PSO/POS (14.51 area%). Based on the $[SS]^+$ : $[SO]^+$ ratio obtained by RP-HPLC/APCI-MS, the final product had a higher SSO (AAB type TAG) content than cocoa butter (CB). The solid fat index (SFI) of CB and the final product obtained were similar with a narrow melting point range around ~32 to $35^{\circ}C$.
