활액막 육종에서 새로운 종양 표지자로서 TLE1의 가치 (TLE1: A New Molecular Target of Synovial Sarcoma)

  • 목적: 활액막 육종에서 TLE1의 발현과 정상 조직에서의 발현을 비교하고 환자의 예후와의 관련성을 알아 보고자 한다. 대상 및 방법: 2001년 1월부터 2008년 9월까지 삼성서울병원에서 수술 및 치료를 시행한 36예의 활액막 육종 환자들의 조직과 정상 대조군 조직들에서 TLE1의 발현을 비교 분석하였다. TLE1의 발현 정도를 음성, 약 양성, 중등도 양성, 강 양성으로 분류하였고, 환자들의 전체 생존율을 분석하여 관련성을 알아보았다. 결과: 정상 간엽조직에서는 TLE1의 발현이 관찰되지 않았고, 분화가 빠른 정상 혈관의 내피세포와 상피세포에서 TLE1의 발현이 약하게 관찰되었다. 활액막 육종 36예 중 35예에서 TLE1의 발현을 확인할 수 있었다. 이 중 28예에서는 중등도 이상의 발현, 5예에서는 약한 발현, 2예는 매우 강한 강도의 발현을 보였으며, 활액막 육종에서 TLE1의 발현 정도와 예후와는 통계적 연관성이 없었다. 결론: TLE1은 향후 활액막 육종의 진단에 효과적으로 사용될 수 있을 것으로 생각되며, 정상 조직에서의 발현도가 낮으므로 종양의 표적 치료시의 좋은 target으로 사용될 수 있는 새로운 인자로 생각된다.

과거 TLE정보를 활용한 새로운 TLE정보 생성기법 (New TLE generation method based on the past TLEs)

  • 본 논문에서는 인공위성의 궤도예측을 위해 사용되는 미국 합동우주관제센터(JSpOC, Joint Space Operation Center)의 TLE(Two Line Element) 정보에 대한 SGP4(Simplified General Perturbations 4) 모델의 장기 궤도예측 오차를 줄이기 위해 과거의 TLE 정보들을 이용한 보상기법을 적용하여 새로운 TLE 정보를 생성하는 방법에 대해 기술하고 있다. 이를 위해 과거 특정 시점에서의 TLE 정보를 바탕으로 현재까지 궤도전파를 한 데이터와 동일기간 동안 미국 합동우주관제센터에서 공개된 모든 TLE를 이용해서 궤도전파를 수행한 데이터를 비교하여 계산한 궤도잔차를 이용하였다. 이러한 궤도잔차 성분은 SGP4 궤도전파 모델에 의한 궤도오차 증가 경향을 보여주고 있기 때문에 궤도오차 보정을 위해 해당 궤도잔차 성분들을 적절한 함수로 표현하였다. 이후, 현재 시점에서 공개된 TLE 정보를 이용한 SGP4 궤도전파 데이터에 해당 잔차함수를 적용함으로써 장기 궤도전파에 따른 SGP4 모델의 궤도오차를 줄일 수 있었으며, 이를 바탕으로 새로운 TLE 정보를 생성하였다. 본 논문에서 일주일의 궤도전파에 대한 시뮬레이션을 통해 기존의 TLE를 이용한 궤도전파 오차가 4km 정도인 반면 새로운 TLE 생성기법에 의한 궤도전파 오차가 약 2km 수준으로 줄어드는 것을 확인할 수 있었다.

접촉궤도요소로부터 변환된 NORAD TLE를 이용한 정지위성의 안테나 포인팅 (ANTENNA POINTING TO THE GEO SATELLITE USING CONVERTED NORAD TLE FROM OSCULATING ORBITAL ELEMENTS)

  • 접촉 케플러 궤도요소로부터 변환된 NORAD TLE를 정지궤도 위성의 안테나 포인팅에 직접 사용하기 위한 분석을 수행하였다. 일주일에 한번씩 동서방향과 남북방향의 위치유지를 위한 궤도조정을 수행하고 하루에 두 번씩 추력기를 이용한 모멘텀 덤핑을 통해서 궤도가 계속 변하는 통신해양기상 위성에 대해서 변환된 NORAD TLE를 이용한 안테나 포인팅 오프셋 각을 계산하여 위성신호를 수신할 수 있는지 검토하였다. 이를 통해 변환된 NORAD TLE를 사용하여 위성 관제시스템의 안테나 포인팅에 관련된 인터페이스를 간단하게 수행할 수 있음을 보였다. 또한 이심률이 큰 천이궤도에 있어서 위성의 평균 근점각에 따른 변환된 NORAD TLE 값의 차이를 분석하여 천이궤도의 원지점 근처에서의 NORAD TLE 변환 값이 더 좋은 결과를 나타내는 것을 알 수 있었다.

Prognostic and Clinicopathological Significance of Transducer-Like Enhancer of Split 1 Expression in Gastric Cancer

  • Purpose: Transducer-like enhancer of split 1 (TLE1) is a member of the Groucho/TLE family of transcriptional co-repressors that regulate the transcriptional activity of numerous genes. TLE1 is involved in the tumorigenesis of various tumors. We investigated the prognostic significance of TLE1 expression and its association with clinicopathological parameters in gastric cancer (GC) patients. Materials and Methods: Immunohistochemical analysis of six tissue microarrays was performed to examine TLE1 expression using 291 surgically resected GC specimens from the Soonchunhyang University Cheonan Hospital between July 2006 and December 2009. Results: In the non-neoplastic gastric mucosa, TLE1 expression was negative. In GC, 121 patients (41.6%) were positive for TLE1. The expression of TLE1 was significantly associated with male gender (P=0.021), less frequent lymphatic (P=0.017) or perineural invasion (P=0.029), intestinal type according to the Lauren classification (P=0.024), good histologic grade (P<0.001), early pathologic T-stage (P=0.012), and early American Joint Committee on Cancer stage (P=0.022). In the Kaplan-Meier analysis, the TLE1 expression was significantly associated with longer disease-free (P=0.022) and overall (P=0.001) survival rates. Conclusions: We suggested that TLE1 expression is a good prognostic indicator in GCs.

돼지 난자의 체내 및 체외 성숙시 Transducin-like Enhancer Protein 1(TLE-1) mRNA의 발현 (TLE-1 mRNA Expression during In Vivo and In Vitro Maturation in Porcine Oocytes)

  • Transducin-like enhancer protein 1(TLE-1) is protein associated with cell proliferation. This study analyzed change of TLE-1 mRNA expression during in vivo and in vitro maturation in porcine oocytes. Oocytes and granulose cells were collected from follicles of <2 mm, 2~6 mm and >6 mm in diameter in slaughtered pig's ovaries. Oocytes collected from follicles of 2~6 mm in diameter were used after in vitro maturation for 0, 10, 20 and 44 h. Cumulus cells and granulose cells were collected after treatment with hyaluronidase. In results, TLE-1 mRNA expression in oocytes collected from follicle >6 mm in diameter is increased, TLE-1 RNA expression in cumulus cells and granulosa cells from follicles <2 mm, 2 mm 6 mm and >6 mm in diameter. However, there is no significant difference. On the other hand, TLE-1 mRNA expression from oocytes cultured for 10 hand 44 h is increased, TLE-1 mRNA in cumulus cells cultured for 10 h is significant increased(p<0.05) than other culture periods. In conclusion, these results show that TLE-1 is expressed in all cell types of oocytes, cumulus cells and granulose cells, and associated with oocyte maturation.

Language Lateralization in Patients with Temporal Lobe Epilepsy : A Comparison between Volumetric Analysis and the Wada Test

  • Objective : Determining language lateralization is important for the presurgical evaluation of patients with medically intractable epilepsy. The Wada test has been the gold standard for lateralization of language dominance before epilepsy surgery. However, it is an invasive test with risk, and have some limitations. Methods : We compared the volumetric analysis with Wada test, and studied the clinical potential of volumetric analysis to assess language laterality in large surgical candidates with temporal lobe epilepsy (TLE). To examine the efficacy of volumetric analysis to determine language lateralization during presurgical evaluation, we compared the volumetric analysis of the bilateral planum temporale with the results of Wada test in 59 patients with chronic intractable TLE (rTLE, n=32; lTLE, n=27) who underwent epilepsy surgery. We measured the gray matter volumes of planum temporale (PT) of each patients using the VoxelPlus2 program (Mevisys, Daejeon, Korea). Results : Overall congruence of the volumetric analysis with the Wada test was 97.75% in rTLE patients and 81.5% in lTLE patients. There were more significant leftward asymmetry of the PT in rTLE patients than lTLE patients. In lTLE patients, relatively high proportion (37%) of the patients showed bilateral or right hemispheric language dominance. Conclusion : These results provide evidence that the volumetric analysis of the PT could be used as an alternatives in language lateralization. Also, the results of the Wada test suggested that there was considerable plasticity of language representation in the brains of patients with intractable TLE and it was associated with an earlier age of brain injury.

APPLICABLE TRACKING DATA ARCS FOR NORAD TLE ORBIT DETERMINATION OF THE KOMPSAT-1 SATELLITE USING GPS NAVIGATION SOLUTIONS

  • NORAD Two Line Element (TLE) is very useful to simplify the ground station antenna pointing and mission operations. When a satellite operations facility has the capability to determine NORAD type TLE which is independent of NORAD, it is important to analyze the applicable tracking data arcs for obtaining the best possible orbit. The applicable tracking data arcs for NORAD independent TLE orbit determination of the KOMPSAT-1 using GPS navigation solutions was analyzed for the best possible orbit determination and propagation results. Data spans of the GPS navigation solutions from 1 day to 5 days were used for TLE orbit determination and the results were used as Initial orbit for SGP4 orbit propagation. The operational orbit determination results using KOMPSAT-1 Mission Analysis and Planning System(MAPS) were used as references for the comparisons. The best-matched orbit determination was obtained when 3 days of GPS navigation solutions were used. The resulting 4 days of orbit propagation results were within 2 km of the KOMPSAI-1 MAPS results.

EphrinB1 interacts with the transcriptional co-repressor Groucho/xTLE4

  • Ephrin signaling is involved in various morphogenetic events, such as axon guidance, hindbrain segmentation, and angiogenesis. We conducted a yeast two-hybrid screen using the intracellular domain (ICD) of EphrinB1 to gain biochemical insight into the function of the EphrinB1 ICD. We identified the transcriptional co-repressor xTLE1/Groucho as an EphrinB1 interacting protein. Whole-mount in situ hybridization of Xenopus embryos confirmed the co-localization of EphrinB1 and a Xenopus counterpart to TLE1, xTLE4, during various stages of development. The EphrinB1/xTLE4 interaction was confirmed by co-immunoprecipitation experiments. Further characterization of the interaction revealed that the carboxy-terminal PDZ binding motif of EphrinB1 and the SP domain of xTLE4 are required for binding. Additionally, phosphorylation of EphrinB1 by a constitutively activated fibroblast growth factor receptor resulted in loss of the interaction, suggesting that the interaction is modulated by tyrosine phosphorylation of the EphrinB1 ICD.

NORAD TLE TYPE ORBIT DETERMINATION OF LEO SATELLITES USING GPS NAVIGATION SOLUTIONS

  • NORAD Two Line Elements (TLE) are widely used for the increasing number of small satellite mission operations and analysis. However, due to the irregular periodicity of generation of the NORAD TLE, a new TLE that is independent of NORAD is required. A TLE type Orbit Determination (TLEOD) has been developed for the generation of a new TLE. Thus, the TLEOD system can provide an Antenna Control Unit (ACU) with the orbit determination result in the type of a TLE, which provides a simple interface for the commercialized ACU system. For the TLEOD system, NORAD SGP4 was used to make a new orbit determination system. In addition, a least squares method was implemented for the TLEOD system with the GPS navigation solutions of the KOMPSAT-1. Considering both the Orbit Propagation (OP) difference and the tendency of $B^{*}$ value, the preferable span of the day in the observation data was selected to be 3 days. Through the OD with 3 days observation data, the OP difference was derived and compared with that of Mission Analysis and Planning (MAPS) for the KOMPSAT-1. It has the extent from 2 km after sit days to 4 km after seven days. This is qualified enough for the efficiency of an ACU in image reception and processing center of the KOMPSAT-2.

NORAD TLE CONVERSION FROM OSCULATING ORBITAL ELEMENT

  • The NORAD type Two Line Element (TLE) was obtained from the osculating orbital elements by an iterative approximation procedure. The mathematical model was presented and computer program was developed for the conversion. The osculating orbital elements of the KOMPSAT-1 were converted into the NORAD TLE. Then the effect of the SGP4 atmospheric drag coefficient ($B^*$) was analyzed by comparison of the orbit propagation results with different $B^*$ values.
