Combined TGE-SGE Expression of Novel PAI-1-Resistant t-PA in CHO DG44 Cells Using Orbitally Shaking Disposable Bioreactors

  • An important modification of thrombolytic agents is resistance to plasminogen activator inhibitor-1 (PAI-1). In previous studies, a new truncated PAI-1-resistant variant was developed based on deletion of the first three domains in t-PA and the substitution of KHRR 128-131 amino acids with AAAA in the truncated t-PA. The novel variant expressed in a static culture system of Chinese Hamster Ovary (CHO) DG44 cells exhibited a higher resistance to PAI-1 when compared with the full-length commercial drug; Actylase. In the present study, the truncated-mutant protein was expressed in CHO DG44 cells in 50 ml orbital shaking bioreactors. The final yield of the truncated-mutant in the culture was 752 IU/ml, representing a 63% increase compared with the static culture system. Therefore, these results suggest that using the combined features of a transient and stable expression system is feasible for the production of novel recombinant proteins in the quantities needed for preclinical studies.

전남동부지방의 설사자돈에 대한 역학조사 (Epidemiological Survey on Piglet diarrhea in eastern Chonnam province)

  • This study was perfomed to examine the distribution of causative agent of piglets diarrhea in eastern Chonnam province from February 1994 to March 1995. The causative agents of diarrhea were examined by bacterial culture test, parasitological test and serological test against PED, TGE and Rota. The 35 isolated E. coli were tested for antibiotic sensitivity. The results obtained were as follows. 1. The incidence of 81 piglets with diarrhea was most prevalent as 39.5% in the age of 2 to 4 weeks, compare to 34.6% in the age of 5 to 8 weeks and 16.0% under 1 weeks and 9.9 % in the age of 9 to 11 weeks after birth. 2. The incidence of 81 piglets diarrhea showed bacterial diarrhea(75.3%), viral diarrhea (35.8%) and parasitological diarrhea(18.5%). When compared the isolation frequency of each etiological agent, enteropathogenic E. coli was most prevalent as 55.5% in bacterial diarrhea, rotavlrus enteritis as 18.5% in viral diarrhea and trichuriasis as 13.6% in parasitological diarrhea. 3. The complicated infection of piglets was most prevalent as 41.7% in rotavirus enteritis with enteropathogenic E coli in 24 complicated piglets diarrhea. 4. In antibiotic sensitivity test, isolates showed moderatly resistance to Tobramycin, Amikacin, Imipenem, Trimethoprim/Sulfamethoxazole, Gentamicin, Ampicillin but sensitivty to Ticarcillin/K. The 30 E. coli isolate showed multiple drug resistances in 3 different antibiotics.

Development of monoclonal antibody capture ELISA for the detection of antibodies against transmissible gastroenteritis virus

  • Transmissible gastroenteritis (TGE) is a disease confined to pigs of all ages, and can be a significant cause of economic loss in breeding herds, primarily because of the very high piglet mortality. The causative agent is a coronavirus, an enveloped positive strand RNA virus and closely related but non-enteropathogenic porcine respiratory coronavirus (PRCV). Although the TGEV has declined with its innocent relative, PRCV, further genome changes could not be excluded. Therefore, the herd-level immunity against this virus is important for the prevention of disease and should be carefully monitored. The aim of this study is to develop monoclonal antibody capture enzyme-linked immunosorbent assay (MAC-ELISA) which can rapidly and accurately determine a large numbers of serum samples for surveillance purpose, and to compare the ELISA with a TGEV-specific serum neutralization test. The MAC-ELISA was sufficiently achieved, and the comparison with the virus-specific serum neutralization assays for 713 sera from pig farms showed a high correlation ($r^2=0.812$, P<0.001). The specificity and sensitivity of MAC-ELISA for the serum neutralization test 91.9% and 91.6%, respectively, which means that the antibody detected by the MAC-ELISA could be said to be protective antibodies. In conclusion, the developed MAC-ELISA would be very helpful in evaluating protective antibodies against TGEV.

  • The purpose of this study was to investigate to potective effects against transmissible gastyoenteritis virus (TGEV) infection in piglets by administration of the TGEV antiserum orally at 5 hrs, 24hrs and 36hrs after birth. five piglets administered the antiserum were experimentally infected with TGEV at four-day-old. Control group were four piglets infected with TGEV only. Serum antibody titers against TGEV were examined by serum neutralization(SN) test, dectection for TGEV or TGEV antigen from feces and small intestines was tested by reverse transcrption-polymerase chain reaction (RT-PCR) and indirect immunoflurescence (IFA). The results obtained were as follows; 1. The piglets administered the TGEV antiserum showed higher antibody titers than those of control group and sustained during the experimental period. 2. The detection rate of TGEV in feces and small intestines by RT- PCR were 24.5% and 20.0% in TGEV antiserum treated group and 44.0% and 75.0% in control group, respectively. 3 The detection rate of TGEV antigen in the small intestine by IFA were 26.7% in TGEV antiserum treated group and 75.0% in control group, respectively. It was concluded that oral administration of antiserum against TGEV to piglets was effective in preventing TGEV infection.

면역전자현미경(免疫電子顯微鏡)(IEM) 기법(技法) 및 immunogold conjugate 면역전자현미경(免疫電子顯微鏡)(IGC-IEM) 기법(技法)을 이용(利用)한 돼지 분변내 PED 바이러스의 검출(檢出) (Detection of PED virus by the immunoelectron microscopy and immunogold conjugate immunoelectron microscopy)

  • Both immunoelectron microscopy(IEM) and immunogold conjugate immunoelectron microscopy (IGC-IEM) techniques were developed for the detection of porcine epidemic diarrhea virus(PEDV) from the feces. Fecal samples were incubated sequentially with anti-PEDV monoclonal antibody(MoAb) and immunogold conjugated goat anti-mouse IgG+IgM. Then negatively stained, mounted on the formvar carbon-coated copper EM grids and observed by the transmission electron microscope. By the direct electron microscopy(DEM), coronavirus particles were observed from 17 cases of total 33 fecal samples of grower pigs and sows. The virons of coronavirus were moderately pleomorphic but mostly spherical, with a diameter ranged from 90 to 190nm. PED virus particles were identified from 15 cases of 17 DEM positive samples by the IEM and IGC-IEM techniques. Aggregates of PED virus coated with specific antibody were seen in fecal samples incubated with homologous anti-PED virus MoAb but not in control samples incubated with anti-TGE virus MoAb. Following incubation with immunogold-conjugated secondary antibody, the gold granules were usually distributed around and among the virus particles and soluble and viral particle-associated antigen. So, IEM and IGC-IEM techniques were proved a rapid and sensitive methods for detection and identification of PED virus from fecal and intestinal contents.

  • 고추의 성장(成長)과 주요(主要) 성분(成分)의 함량(含量)에 미치는 cation의 영향을 조사(調査)코저 배양액내(培養液內)의 $K^+,\;Ca^{++},\;Mg^+$의 조성(組成)을 달리하여 재배(栽培) 실험(實驗)을 행(行)하고, 이 결과(結果)를 systemic variation Method 중(中) binary interaction방정식(方程式)에 적용(適用)시켜 이상(理想) 조건(條件)을 추구(追求)한 결과(結果)는 다음과 같다. 1. 성장(成長) 이상(理想) 조건(條件) 중(中) 초장(草長)은 CKa group에서 $57%\;K^++43%\;Ca^{++}$, KMg group에서 $58.5%\;K^++41.5%\;Mg^+$가 최적조건(最適條件) 이었으며, 마디수에서는 $59.0%K^++41%Ca^{++},\;60%\;K^++40%\;Mg$이었다. 2. 수확량(收穫量)을 최대(最大)로 하기 위한 이상조건(理想條件)은 $60%K^++40%Ca^{++},\;62%K^++38%\;Mg^{++}$이었으며 완숙과(完熟果)에 대(對)하여서는 $57.0%\;K^++43.0%\;Ca^{++},\;63.0%K^++37.0%\;Mg^{++}$이었다. 3. 각(各) 처리별(處理別) free sugar와 capsaicin의 함량(含量)은 유의성이 없었다. 4. 각(各) 처리별(處理別) nitrogen level은 거의 일정치(一定値)를 유지하였으나 Fe/Mn의 값은 $2{\sim}3$을 유지하였다.

혈청학적 진단을 위한 돼지 편충의 체항원, 배설/분비항원의 분리 및 비교 (Isolation and comparison of somatic and excretory-secretory antigens for serological diagnosis in Trichuris suis)

  • Swine whipworm(Trichuris suis) is cosmopolitan nematode which can cause serious pathology in immature stage(larva2~larva5) of infected pigs, such as anorexia, diarrhea, anemia, and death in heavy infections. In this larval stages, it is very difficult to diagnose the infection of whipworm and to differentiate from other common swine gastrointestinal disorders such as 21 day scours which are associated with TGE virus, rota virus, coccidium, and the stress of weaning. In this experiment, the isolated antigens of Trichuris spp. were carried out to examine the structure and specificity of antigens and to select the reasonable antigens which would be used in serological diagnosis by electrophoresis, Western blotting, ELISA. The results of this experiment were as follows; 1. The common fractions of each Trichuris suis antigen were identified 28,32,45, 80kDa by SDS-PAGE with silver stain and four major fractions could be detected in positive swine sera by Western blot analysis. 2. The OD(optical density) values of somatic and excretory-secretory antigens which were reacted against positive(negative) sera from pigs infected with Trichuris suis by ELISA reader were; 1) OD values($mean{\pm}SD$) of adult somatic antigen against positive(negative) sera were $0.30{\pm}0.12(0.09{\pm}0.006)$ and third-stage larva of somatic antigen were $0.28{\pm}0.038(0.10{\pm}0.005)$. And OD values of excretory-secretory antigens of adult and third-stage larva were $0.24{\pm}0.031(0.11{\pm}0.005)$ and $0.08{\pm}0.013(0.10{\pm}0.003)$, respectively. 2) OD values of adult somatic, larval somatic antigen and adult excretory-secretory antigen response to positive sera were significantly (p<0.01) associated with negative swine sera. And the Cut-off OD values(minimum positive value) were determined to be mean negative value plus 3 SD that would minimized the risk of false positives. 3. The OD values of somatic antigens of T suis and T vulpis against swine positive(negative) sera were $0.30{\pm}0.120(0.09{\pm}0.006)$ and $0.25{\pm}0.141(0.09{\pm}0.003)$. These data mean that the somatic antigens of T suis and T vulpis were able to diagnose T vulpis infection in dogs as well as T suis infection in pigs. These results suggest that somatic antigen of third-stage larva and excretory-secretory antigen of adult T suis could be used the diagnostic antigen by serological test(ELISA) in immature Trichuris spp. infection.

이미지와 텍스트 메시지의 통합 사용자 뷰를 제공하는 전자 우편 시스템 (E-mail System Providing Integrated User's View for the Message containing Image and Text)

  • 전자 우편은 인터넷 정보 전달 서비스로서 널리 사용되고 있다. 멀티미디어 기술의 급속한 발전으로 최근 인터넷 정보 서비스는 대부분 멀티미디어를 지원하고 있다 아. 따라서 멀티미디어 메시지를 지원하는 전자 우편 시스템이 요구된다. 그러나RFC 821/822 의SMTP (Simple Mail Transfer protocol)를 사용하는 인터넷 메일 서비스를 전달할 수 있는 정보가 7bit ASCII텍스트만으로 제한되어 있다. 또한 한 라인의 길이에도 제한이 있어서 사용자 요구를 충족하기에 부족하다. MIME(Multipurpose Internet Mail EXtensions)은 RFC 822를 수정, 보완한 것으로서, 메시지를 구성하는 데이타의 종류와 크기의 제한을 해결하여 멀티미디어 데이타를 전송하기 위해 제안되었다. 본 연구에서는 MIME의 표준을 따라 정보 전송 상의 제약을 해결한 전자 우편 시스템을 설계 및 구현하였다. HTML( Hyper Text Markup Language)구문을 적용하여 서로 다른 미디어로 구성된 메시지를 통합 적으로 보여줌으로써 정보 전달의 명확성을 향상시켰다. 별도의 응용 프로그램이 필요치 않고, 사용자 편의를 더욱 증가시킨 시스템을 구성하였다. 앞으로도 동화상 및 음성 정보 등을 지원할 수 있도록 보완하여, 통합 사용자 뷰를 제공하는 완전한 멀티미디어 전자 우편 시서템을 개발하고자 한다.

마우스세포주 Balb/c 3T3 A31-1-1에서 Epigallocatechin gallate(EGCG)의 세포암화 억제효과에 대한 유전자발현 해석 (Genome-based Gene Expression Analysis of EGCG-mediated Cell Transformation Suppression Effect in Mouse Cell line Balb/c 3T3 A31-1-1)

  • Previous studies showed that epigallocatechin gallate(EGCG) have substantial effects of suppressing the N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)-initiated cell transformation process on the bases of foci formation frequency and loss of anchorage dependency. In this study we tried to clarify the molecular mechanism of suppressing the cell transformation process. Mouse cell line balb/c 3T3 A31-1-1 was exposed 2 days to MNNG followed by 15 days 12-O-tetradecanoylphorbol-13-acetate(TPA) treatment for our transformation process. EGCG was added after the time point of 24 hours exposure to TPA and incubated for 19 days. 2029 genes were selected in our transformation process that showed fold change value of 1.5 or more in the microarray gene expression analysis covering the mouse full genome. These genes were found to be involved mainly in the cell cycle pathway, focal adhesion, adherens junction, TGE-$\beta$ signaling, apoptosis, lysine degradation, insulin signaling, ECM-receptor interaction. Among the genes, we focused on the 631 genes(FC>0.5) reciprocally affected by EGCG treatment. Our study suggest that EGCG down-regulate the gene expressions of up stream signaling factors such as nemo like kinase with MAPK activity and PI3-Kinase, Ras GTPase and down stream factors such as cyclin D1, D2, H, T2, cdk6.

선형가속기를 이용한 뇌종양 46예의 뇌정위다방향방사선치료 성적 (Clinical Report of 46 Intracranial Tumors with LINAC Based Stereotactic Radiosurgery)

  • 가톨릭 의과대학 강남성모병원 치료방사선과에서는 1988년 7월부터 1992년 12월 사이에 뇌종양 환자 45명의 뇌내 46개 병소에 대한 뇌정위다방향방사선치료를 실시하였으며, 이들에 대한 영상학적 및 임상적 추적조사 결과를 후향적으로 분석하였다. 뇌종양에 대한 뇌정위다방향방사선치료는 병소가 생명중추에 인접되어 있거나 다른 전신질환과 함께 있어 종양의 수술적 제거가 어려운 경우, 수술후 잔류 병소가 남아 있거나 재발된 경우, 또는 종래 부터 해오던 방사선치료에도 불구하고 잔류병소가 남아있거나 환자가 수술을 거부할 경우 등에서 실시하게 된다. 이는 수술이나 유사한 다른 치료와 비교하여 동일한 치료효과를 얻으면서도 간편하고 비침습적 이며 또한 상대적으로 저렴한 치료 경비가 소요됨으로, 최근 방사선치료장비 및 치료계획 프로그램의 개발과 함께 그 치료성적이 관심의 대상이 되고 있다. 환자 분포는 남녀 비가 22:33 이며, 연령 분포는 5-74세 (중앙값 : 43세)이었고, 추적조사 기간은 2-55개월 (중앙값 : 35개월)이었다. 6 MV 선형 가속기를 사용하여 조사야 용적은 $0.13-42.88\;cm^3$(중앙값 : $7.26\;cm^3)$이었으며, 최대치료선량은 5-35.5 Gy(중앙값 : 20.9 Gy)이었다. 종양 별로는 뇌하수체종양15예, 청각신경종 8예, 수막종 7예, 뇌교종 6예, 두개인두종 4예, 송과선종 3예, 혈관아세포종 2예, 및 뇌전이 암 1예 씩이었다. 총 46예 중 18예$(39.1\%)$에서 추적 영상검사상 종양의 소실 및 위축을 관찰하였으며, 10예$(34.8\%)$에서는 종양크기의 정지상태를 나타냈다. 임상적으로는 34예$(73.9\%)$에서 방사선치료 전보다 양호한 전신상태 및 수행능력을 유지하고 있었으며, 이중 4예 $(8.7\%)$에서는 추적 검사상 종양 크기가 약간 커졌음에도 불구하고 임상적으로는 안정된 상태이었다. 영상학적으로나 임상적으로 악화된 경우는 2예$(4.4\%)$에서 있었다. 치료에 따르는 부작용으로는 치료 직후, 일시적 두통을 호소하였으나 스테로이드 및 진통제 투여로 개선되었으며 경미한 일시적 탈모를 3예$(6.6\%)$에서 경험하였으며 향후 좀더 정밀 추적검사가 필요할 것으로 생각된다.
