In situ Hybridization for the Detection and Localization of the Bitter Taste Receptor Tas2r108 in the Murine Submandibular Gland

  • Mammals have 3 pairs of major salivary glands i.e., the parotid, submandibular, and sublingual glands. Saliva secretion of these glands is modulated by taste perception. Salivary glands are composed mainly of acinar and ductal cells. Primary saliva is secreted by acinar cells and modified during ductal flow. Recently, of the murine 35 bitter taste receptors, Tas2r108 was expressed at highest levels in the submandibular gland by qPCR. Further, Tas2r108-transfected cells respond to a range of bitter compounds, such as denatonium, quinine, colchicine, diphenidol, caffeine and dapson. The objective of the present study was to characterize the expression of Tas2r108 mRNA in acinar and/or ductal cells of the submandibular gland using in situ hybridization (ISH). Male 42-60 days old DBA2 mice were used in the study. Messenger RNAs were extracted from the submandibular gland for generating digoxigenin (DIG) labeled-cRNA probes. These probes were transcribed in anti-sense and sense orientation using T7 RNA polymerase. Dot blot hybridization was performed using DIG labeled-cRNA probes, in order to estimate integrity and optimal diluting concentration of these probes. Subsequently, ISH was performed on murine submandibular gland to detect Tas2r108 mRNA. Dot blot hybridization data demonstrated that Tas2r108 DIG labeled-cRNA anti-sense probes specifically detected Tas2r108 cDNA. ISH results showed that the anti-sense probes labeled acinar and ductal cells in the submandibular gland, whereas no staining was visible in sense controls. Interestingly, the Tas2r108 expression levels were higher in acinar than ductal cells. These results suggested that Tas2r108 might be more associated with primary saliva secretion than with ductal modification of saliva composition.

An Expression Levels Analysis of the Bitter Taste Receptors in the Murine Exocrine Glands

  • Recent findings indicate that Type 2 taste receptors (T2Rs) are expressed outside the gustatory system, including in the gastrointestinal tracts and the exocrine glands, such as the submandibular (SM), parotid (P), lacrimal (L) glands and pancreas (PC). Specifically, T2Rs are found in some of the gastrointestinal endocrine cells, and these cells secreted peptide hormones in response to stimulation by bitter-tasting compounds. The results show that T2Rs may have significant physiological roles besides bitter taste reception. The functions of the T2Rs in the exocrine glands remain poorly understood. An expression levels analysis of T2Rs will help to determine those functions in the exocrine glands. The expression levels of the T2Rs in the exocrine glands were discovered via the qPCR. C57BL/6J mice of 42~60-day-old were used. Messenger RNAs were extracted from S, P, L and PC. Cloned DNAs were synthesized by reverse transcription. Quantitative PCRs were performed using the SYBR Green method. The expression levels of the T2Rs were calculated as relative expression levels to that of the GAPDH. The statistical significance among the observed exocrine glands was tested using the variance analysis (ANOVA test). Tas2r108, out of murine 35 T2Rs, was the most highly expressed in every observed exocrine gland. This finding was similar to previous results from tongue papillae, but the expression levels were lower than those of the tongue papillae. Tas2r137 of SM, P, L and PC were expressed a little lower than that of tongue papillae. The T2Rs in the exocrine glands may play slightly different roles from those in the tongue. We suggest that physiological studies such as a patch clamp and functional $Ca^{2+}$ imaging of acinar cells are necessary for understanding the Tas2r108 functions.

Self-care, Self-efficacy and Social Support of Patients Receiving Radioactive Iodine Therapy (방사성요오드 치료 환자의 자가간호와 자기효능감 및 사회적지지)

  • This study presents a descriptive research on the degree of self-care related to the prevention of radiation exposure after radioactive iodine therapy and on the relations between self-efficacy and social support to propose nursing interventions required for patients after radioactive iodine therapy. The research period lasted from March to May, 2013. The subjects included 108 patients that were receiving radioactive iodine therapy after thyroidectomy at a cancer hospital in Gyeonggi Province. The subjects scored mean 52.10 on self-care out of full 56 and as for the general characteristics of the subjects, there were significant differences in their self-care according to whether they had a child or not(t=-2.312, p=.023) and interest in health(t=5.689, p<.001). There were significant positive correlations between their self-care and their self-efficacy(r=.610, p<.001), family support(r=.646, p<.001), and medical staff support(r=.276, p=.004). Interest in health(t=5.301, p<.001) was predictor on level 1 of hierarchical regression and interest in health(t=2.140, p=.035) and family support(t=3.353, p=.001) turned out to influence the self-care of the subjects, recording total 46.3% explanatory power. The most important predictor was interest in health(${\beta}$=1.309, p=.035) of self-care.

Effect of ME and Crude Protein Content of Diet on the Performance and Production Cost of White Semibroiler Chickens (사료의 ME가와 단백질 수준이 백세미의 생산성과 경제성에 미치는 영향)

  • An experiment was conducted to investigate the effect of ME and crude protein (CP) content of diet on the performance and production cost of white semibroiler chickens. Three hundred sixty hatched white semibroiler chickens (Hy-Line $female\;{\times}\;Ross^{(R)}$ male) were assigned to low dietary treatments of different metabolizable energy (ME) value and crude protein (CP) content: T1; ME 3,040 and 3,070 kcal/kg, CP 21.40 and 20.86%, T2; ME 2,950 and 3,000 kcal/kg, CP 20.62 and 19.70%, T3; ME 2,950 and 3,000 kcal/kg, CP 19.90 and 18.70% and T4; ME 2,900 and 2,920 kcal/kg, CP 18.50 and 18.00% for starter diet ($0{\sim}1st$ wk) and power diet ($2{\sim}5th$ wk), respectively There were significant (P<0.01) differences among treatments in weight gain, feed intake and feed conversion ratio. T1 was significantly higher in weight gain and feed intake and love. in feed conversion ratio than other treatments. Production index of T1 (153.42) was far greater than T2 (112.13), T3 (108.40) and T4 (100.95). It was concluded that semibroilers required similar ME and CP to those of regular commercial broilers (highbro).

On Convergence Impact Influencing of Satisfaction Clinical Practice of Dental Hygienist students (치위생(학)과 학생들의 임상실습만족도에 영향을 미치는 융합 요인)

  • This study was to identify factors influencing satisfaction on clinical practice of dental hygienist students located in G, J region. The data was analyzed by t-test, Anova, Pearson correlation coefficient, and multiple regression using SPSS 21.0 program. There were significant in satisfaction of score(F=2.925, p<.05), perceive heath status(F=8.108, p<.001), satisfaction on department(F=8.198, p<.001). The relationship on peers with score(r=.277, p=.01) and perceive health status correlations with relationships on peers(r=.327, p=.01), The satisfaction on clinical practice had positive correlations with score(r=.127, p=.05), perceive health status(r=.226, p=.01), relationship on peers(r=.240, p=.01). The factors influencing satisfaction on clinical practice were satisfaction of relationship on peers, satisfaction of department, perceive health status. The results suggest that development of program to improve satisfaction on clinical practice of dental hygiene students.

Genetic Diversify and Population Structure of Two Korean Pond Frog Species, Rana nigromaculata and R. plancyi (Anura, Ranidae), with a Survey of Temporal Genetic Variation in R. nigromaculata

  • Korean R. plancyi occupies a restricted area in western South Korea and shows a relatively low level of genic variability (%P=15.2, Ho=0.052, He=0.048). In contrast, R. nigromaculata is broadly distributed in South Korea. The observed low level of variability of R. nigromaculata (%P=14.3, Ho=0.042, He=0.043) is probably due to its recent colonization. Populations of R. nigromaculata exhibited considerable genetic differentiation (F$_{sT}$=0.149) and low level of gene flow (Nm=1.427) among populations, compared to those of R. Plancyi (F$_{sTF$_{sT}$}$=0.096, Nm=2.354), which occupies a restricted area. The observed levels of gene flow among populations of R. nigromaculata (Nm=1.427) over a broad geographic range is relatively higher than other amphibian species. The high level of gene flow is probably the result of the high dispersal abilities of R. nigromaculata. A survey of temporal genic variation of R. nigromaculata showed that there was no significant change on the overall average genetic diversity from 1978 (average He=0.044) to 1997 (average He=0.040). Wright's F-statistics also indicated no significant genetic differentiation from 1978 (F$_{sT}$=0.118) to 1997 (F$_{sT}$=0.108). This suggests that the environmental change appears to have had little influence on the genetic composition of R. nigromaculata in the study areas during the past 20 years. The low level of temporal variation might be due to the result of high dispersal abilities and wide migration range of this species.

Antifungal Activity of Paenibacillus kribbensis Strain T-9 Isolated from Soils against Several Plant Pathogenic Fungi

  • The bacterial strain T-9, which shows strong antifungal activity, is isolated from the soils of Samcheok, Gangwondo and identified as Paenibacillus kribbensis according to morphological and taxonomic characteristics and 16S rRNA gene sequence analysis. The P. kribbensis strain T-9 strongly inhibits the growth of various phytopathogenic fungi including Botrytis cinerea, Colletotricum acutatum, Fusarium oxysporum f. sp. radicis-lycopersici, Magnaporthe oryzae, Phytophthora capsici, Rhizoctonia solani, and Sclerotium cepivorum in vitro. Also, the P. kribbensis strain T-9 exhibited similar or better control effects to plant diseases than in fungicide treatment through in vivo assays. In the 2-year greenhouse experiments, P. kribbensis strain T-9 was highly effective against clubroot. In the 2-year field trials, the P. kribbensis strain T-9 was less effective than the fungicide, but reduced clubroot on Chinese cabbage when compared to the control. The above-described results indicate that the strain T-9 may have the potential as an antagonist to control various phytopathogenic fungi.

Effects of SDN® as a Microbial Culture on Milk Production, Milk Composition and Somatic Cell Counts of Lactating Cows (미생물 배양액 SDN®의 급여가 착유우의 유생산, 유성분 및 체세포수에 미치는 영향)

  • Holstein cows(n=12) were assigned to one of three diets containing control diet and diets containing a microbial culture, 50ml(T1) and 100ml(T2) SDN(R)(amicrobial culture), per day. The basal diet containing concentrate mixture, corn silage and timothy hay were fed midlactating cows for 12 weeks. Milk production tended to be higher for cows fed T2 diet (20.8kg/day) than fed T1(19.7kg/d) and control diet (19.2kg/day).  There was a tendency of an increase in 4% FCM for cows fed T2 diet(19.6kg/day) than T1(18.8kg/d) and control diet(18.4kg/day). Milk components were not found to be different between cows fed control diet and SDN(R) diets. There was a tendency an increase in milk protein for cows fed control diet(3.43%) compared with microbial diets, T1 and T2(3.08% and 3.20%). However, milk protein production was not significantly different between control diet(0.65kg/d) and T1(0.61kg/d) or T2(0.67kg/d). Somatic cell counts for cows fed T1(72,000) and T2(60,000/ml) were lower than cows fed control diet (108,000/ml) (P<0.05). In conclusion, the cows that were fed diets containing SDN(R) as a microbial culture resulted a tendency of an increase in milk production and a reduction of somatic cell counts which indicates improved milk quality and hygiene.

Analysis of Knowledge About, Attitude Toward, and Educational Needs for Dementia in Certified Caregivers (요양보호사의 치매에 대한 지식, 태도 및 교육요구도 분석)

  • Purpose: The purpose of this study was first to identify knowledge about, attitude toward, and educational needs for dementia in certified caregivers; secondly, to understand the relationships among those variables; and thirdly, to provide base-line data for education program development. Methods: The participants in this study were 670 certified caregivers who were working in nursing homes or care hospitals throughout the whole of the country and engaged in education programs for improvement of their professional abilities. Collected data were analyzed using t-tests, ANOVA, Duncan, and Pearson's correlation coefficients with the SPSS 18.0 for Windows Program. Results: The scores for knowledge about, attitude toward, and educational needs for dementia were $14.12{\pm}2.83$ out of 20, $42.27{\pm}5.54$ out of 50 (item mean 4.22), and $121.51{\pm}22.13$ out of 160 (item mean 3.79) respectively. There were statistically significant positive correlations between knowledge and attitude (r=.238, p<.001), educational needs (r=.083, p=.032), and attitude and educational needs (r=.244, p<.001). Conclusion: Tailored and continuous educational programs for certified caregivers that can improve knowledge about the causes and symptoms of dementia should be developed. Nurses and professors who are experts in dementia should be on the front lines of education for caregivers.
