Anti-inflammatory Effect of Nypa fruticans Wurmb. on tumor necrosis factor (TNF)-α-induced Inflammatory response in HaCaT cells (TNF-α로 유도된 HaCaT 각질형성세포의 염증반응에서 해죽순의 항염증 효과)

  • Objectives : Nypa fruticans Wurmb. (NF) have been used as a traditional medicine to treat inflammatory diseases in East-South Asia. However, it is largely undiscovered whether NF water extract could exhibit anti-inflammatory activities against tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)-induced inflammatory responses on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the anti-inflammatory activity of NF water extract on TNF-${\alpha}$-induced inflammatory responses in HaCaT cells. Methods : To investigate the anti-inflammatory activites of NF water extract in HaCaT cells, the inflammatory model of HaCaT cells was established under a suitable concentration (10 ng/ml) of human TNF-${\alpha}$ (hTNF-${\alpha}$). HaCaT keratinocyte cells were pre-treated with NF water extract for 1 h, and then stimulated with hTNF-${\alpha}$. Then, the cells were harvested to measure the inflammatory mediators such as inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 and prostaglandin $E_2$ ($PGE_2$), and pro-inflammatory cytokine including TNF-${\alpha}$ and interleukin (IL)-6. In addition, we examined the inhibitory mechanisms of NF, mitogen activated protein kinases (MAPKs) and inhibitory kappa B alpha ($I{\kappa}-B{\alpha}$) Results : The treatment of NF inhibited the hTNF-${\alpha}$-induced elevation of iNOS, COX-2, and $PGE_2$ in HaCaT cells. In addition, NF treatment inhibited the hTNF-${\alpha}$-induced elevation of TNF-${\alpha}$ and IL-6. Furthermore, NF treatment inhibited the activation of MAPKs but not degradation of $I{\kappa}-B{\alpha}$. Conclusions : Taken together, our result suggest that treatment of NF could inhibit the hTNF-${\alpha}$-induced inflammatory responses via deactivation of MAPKs in HaCaT cells. This study could suggest that NF could be a beneficial agent to prevent skin damage or inflammation.

- A Study on Motivations for Parenthood and Parental Role Concepts in Korean College Students - (대학생의 부모됨 동기와 부모역할개념에 관한 연구)

  • The purpose of this study is to investigate Korean youth's motivations for parenthood and concepts of parental role and to examine closely the relationship with the sociodemographic variables(age, sex, religious orientation, growth region, father's occupational level, parental education degree). the subjects are 465 students of the three universities in National Capital region. The data are collected by questionnair methods and analyzed by t-test, F-test, factor analysis. The major findings are as follows: 1) The motivations for parenthood are classified into five factors. These are acquisition of social status factor, continuation of self factor, achevement and creativity factor, tradition factor, primary group ties factor. The korean youth think the most of achievement and creativity factor among five motivation factors. 2) The korean students have somewhat stereo-typed concepts of parental role and place the stress on training the behaviors of the child and encouraging the development of the child. 3) The sociodemographic variables to have relationship with the motivations for parenthood are age, sex, growth region and father and mother's educational degrees. And the variables related with the concepts of parental role are sex, father's occupational level. 4) There are relationships between traditional role concepts of a father and motivation of acquisition of social status factor and tradition factor. And traditional role concepts of a mother are related with motivations of acquisition of social status factor and primary group ties factor among the motivations for parenthood.

  • Transforming growth factor-$\beta$ (TGF-$\beta$) is a highly pleiotropic cytokine playing pivotal roles in immune regulation. TGF-$\beta$ facilitates tumor cell survival and metastasis by targeting multiple cellular components. Focusing on its immunosuppressive functions, TGF-$\beta$ antagonists have been employed for cancer treatment to enhance tumor immunity. TGF-$\beta$ antagonists exert anti-tumor effects through #1 activating effector cells such as NK cells and cytotoxic $CD8^+$ Tcells (CTLs), #2 inhibiting regulatory/suppressor cell populations, #3 making tumor cells visible to immune cells, #4 inhibiting the production of tumor growth factors. This review focuses on the effect of TGF-$\beta$ on T cells, which are differentiated into effector T cells or newly identified tumor-supporting T cells.

  • The threshold fatigue crack growth and the stable crack propagation behaviors were studied in 7017 T 651, 7020 T 651 and 5083 H 115 aluminum alloys. The threshold (.DELTA. K $_{th}$) fatigue crack growth can be expressed by the equation .DELTA. $K_{th}$) = .DELTA. $K_{tho}$(1-R)$^{r}$ , where R is stress ratio, .DELTA. $K_{tho}$ is .DELTA. K at R = 0 and r is material constant. The stable crack growth rate against stress intensity factor range .DELTA. K exhibits the trilinear form with two transitions and results of investigation on crack closure phenomena showed that the crack opening stress intensity factor $K_{op}$ is approximately equal to R $K_{max}$. + .DELTA. Label/Quencher-Free Detection of Exon Deletion Mutation in Epidermal Growth Factor Receptor Gene Using G-Quadruplex-Inducing DNA Probe

  • Detection of exon 19 deletion mutation in the epidermal growth factor receptor (EGFR) gene, which results in increased and sustained phosphorylation of EGFR, is important for diagnosis and treatment guidelines in non-small-cell lung cancer. Here, we have developed a simple and convenient detection system using the interaction between G-quadruplex and fluorophore thioflavin T (ThT) for discriminating EGFR exon 19 deletion mutant DNA from wild type without a label and quencher. In the presence of exon 19 deletion mutant DNA, the probe DNAs annealed to the target sequences were transformed into G-quadruplex structure. Subsequent intercalation of ThT into the G-quadruplex resulted in a light-up fluorescence signal, which reflects the amount of mutant DNA. Due to stark differences in fluorescence intensity between mutant and wild-type DNA, we suggest that the induced G-quadruplex structure in the probe DNA can report the presence of cancer-causing deletion mutant DNAs with high sensitivity.

  • Warehouse Management System (WMS) is a key control for Material Handling System (MHS) and Inventory Control System (ICS). How to design and implement for WMS is crucial factor for achieving the key performance index for Manufacturing Industry. In particular, iron and steel making industry, where the volume and weight is large and hence FIFO (First Input First Out) is not working, how to design WMS is a key factor. In this paper, we systematically define the problem of WMS via developing StR (Stakeholders' Requirements) or ORD (Operational Requirement Documents), SyR (System Requirement), and SA (System Architecture) based on the emerging technologies. In particular, IoT (Internet of Things), CPS (Cyber Physical System) concepts and enabling technologies haves been incorporated in developing Smart WMS. The deliverables of the research can provide a conceptual framework for developing the next generation industrial WMS.

  • In this paper, the new evaluation system for water quality pollution is implemented using fuzzy integral based on the conventional evaluation criterions and their evaluation factors and this is the first phase in the whole water quality pollution evaluation system development. In the final evaluation for water quality pollution the factors like BOD, COD, SS, T-N, and T-P are taken into overall accounts. It is found that the final evaluation can be represented in a linear combination of respective factor evaluation when each factor is independent one another, With respect to the combination patterns the fuzzy measurement is defined and the fuzzy integral is taken. As a result this approach shows stable and reliable evaluation for the water quality pollution evaluation system development.

The Analysis of Current Limiting Performance in a High-$T_c$ Superconductor using Flux-Lock Concepts

  • In this paper, we analyzed the fault current performance in a $high-T_{c}$ superconductor(HTS) which was installed on flux-lock reactor with an external magnetic field coil covering the HTS. In this HTS fault current limiter using flux-lock concepts, the initial limiting current level can be controlled by adjusting the inductance of the coils. Furthermore, the current limiting characteristics of $high-T_{c}$ superconducting FCL can be improved by applying the external magnetic field into the $high-T_{c}$ superconductor. This paper discusses current limiting performance according to the inductance of the coil 1 in two cases with ac magnetic field coil or not and suggests the methods to improve the current limiting factor $P_{limit}$, which is defined as the ratio of the limited current $I_{FCL}$ at the current limiting phase to the prospective short -circuit current $I_{PSC}$.TEX> PSC/.

  • Purpose: The purpose of this study is to investigate the epithelial-mesenchymal transition (EMT) induced by Snail transcription factor and Snail-transfected in vivo tumors with histopathological features. Materials and methods: We induced in vivo xenografted tumorigenesis in the oral vestibules of nude mice by a Snail transfected HaCaT cell line and investigated morphological and immunohistochemical features in Snail expressive tumors. Results: We identified tumor masses in 14 out of 15 nude mice in the HaCaT-Snail cell inoculation group, but no tumors were present in any of the HaCaT cell inoculation group. Induced tumors showed features of poorly differentiated carcinoma with invasion to neighboring muscles and bones. The HaCaT-Snail tumors showed decreased expressions of E-cadherin and cytokeratin, but showed increased expressions of vimentin and N-cadherin. Discussion: The Snail transfected xenograft can improve productivity of malignant tumors, show various histopathological features including invasive growth, and aid in the investigation of tumor progression and the interaction with surrounding tissues.

  • Steam generator tubes experience widespread degradations such as stress corrosion cracking, wear, tube rupture, denting, fatigue and so on. The resulting damages can cause tube bursting or leak of the primary water which contains radioactivity Therefore the allowable size of the damage is required to be determined on the maintenance purpose. The burst pressure of a tube with a T-type combination crack consisting of longitudinal and circumferential cracks is obtained experimentally and analytically. Fracture parameters such as stress intensity factor and crack opening angle are investigated. Also the burst pressure for a T-type combination crack is compared with that of a single longitudinal crack to develop a length-based criteria.
