혐기성 미생물로 제조한 synbiotics 혼합배양물의 첨가가 발효 TMR의 발효특성과 소실률에 미치는 영향 (Effects of Supplementation of Synbiotic Co-cultures Manufactured with Anaerobic Microbes on In Vitro Fermentation Characteristics and In Situ Degradability of Fermented TMR)

  • 본 연구는 혐기성 박테리아, 곰팡이 및 효모로 제조한 synbiotics 혼합 배양물을 TMR 제조 시에 접종하여 발효기간에 따른 반추위 in vitro 발효특성과 in situ 분해율에 미치는 영향을 조사하고자 수행되었다. 처리구는 무처리구(Control), 곰팡이와 박테리아로 제조한 synbiotic 첨가구(T1), 곰팡이와 효모로 제조한 synbiotic첨가구(T2), 그리고 박테리아와 효모로 제조한 synbiotic 첨가구(T3)로 나누어, 6회의 발효기간(1, 3, 5, 7, 14 및 21일)에 따라 처리당 3반복으로 총 72개의 F-TMR (4처리 $\times$ 6회 $\times$ 3반복)을 제조하였다. In situ시험은 반추위 누관이 장착된 Holstein (평균체중 550 kg) 젖소 2두를 사용하여 4처리의 F-TMR을 반추위내에서 현수시간별(1, 3, 6, 9, 18, 24, 48 및 72 시간)로 각각 3반복하여 두당 96개의 nylon bag (4 처리 $\times$ 8 발효시간 $\times$ 3반복)을 제조하였다. 발효기간에 따른 TMR의 온도변화는 발효기간이 진행됨에 따라 높아지는 경향 이었으며, pH는 4.39~4.98범위로서 발효기간이 진행됨에 따라 감소되는 경향이었으며, 각 발효시간대별로는 대조구에서 높았고, synbiotics 처리구에서 낮게 나타났다(p<0.05). F-TMR의 암모니아 농도는 발효 7일째까지는 처리간 차이가 없었으나 14일째 이후에는 대조구에 비해 박테리아와 효모를 첨가한 synbiotics인 T3구에서 가장 낮았다(p<0.05). Lactic acid 함량은 발효 1일째에 T3구에서 가장 낮았으나 다른 발효시간대에서는 처리간 차이가 없었다. 발효기간별 F-TMR의 미생물 성장률(OD값)은 각 발효시간대별로 처리간에 차이가 없었으며, 발효기간 7일째에 정점을 나타낸 이후로 점차 감소하는 경향이었다. In situ 건물 소실률은 발효초기인 1~3시간대에 곰팡이와 박테리아 synbiotics 첨가구인 T1구가 대조구에 비해 건물소실율이 높았으나, 발효 48시간대에는 오히려 대조구에서 건물소실율이 가장 높았다(p<0.05). 다른 발효시간대의 건물 소실율과 유효분해도는 처리간 차이가 없었으며, NDF와 ADF소실율은 건물 소실율의 결과와 비슷하였다. 결론적으로 발효의 척도가 되는 pH와 lactic acid 함량은 synbiotics 첨가구가 대조구에 비해 좋은 결과를 나타내었으며, in situ 시험에서 발효초기에 건물과 섬유소 소실율이 곰팡이와 박테리아를 조합한 synbiotics구에서 높게 나타났으나 유효 분해도에서는 차이가 거의 나타나지 않아 처리효과가 미흡한 것으로 조사되었다.

개시체중과 Synbiotics 첨가가 이유자돈의 성장에 미치는 영향 (Effects of Initial Body Weight and Synbiotics Supplementation on Growth Performance of Weaned Pigs)

  • 본 연구의 목적은 개시체중 차이와 사료 내 신바이오틱스 첨가에 따른 이유자돈의 성장특성을 비교하는 것이다. 공시 돈은 총 80두로 4 처리 4 반복(반복당 5두)로 난괴법 배치를 하였고, 처리구는 1) 개시체중이 높고 아무것도 첨가하지 않은 사료 급여, 2) 개시체중이 낮고 아무것도 첨가하지 않은 사료 급여, 3) 개시체중이 낮고 항생제 0.2% 첨가 급여, 4) 개시체중이 낮고 신바이오틱스 0.2% 첨가 급여 등 4처리구 이다. 실험기간은 총 28일로 Phase I (d 0~14)과 Phase II (d 14~28)로 구분하여 진행하였다. 개시체중이 낮고 사료 내 항생제 및 신바이오틱스 첨가 급여한 AB 그룹과 Syn 그룹은 개시체중이 낮고 아무것도 첨가 급여하지 않은 NC 그룹에 비해 일당증체량이 유의적으로 높았으나 실험종료 시 체중은 유의적 차이가 없었다. 개시체중이 높고 아무것도 첨가 급여하지 않은 PC 그룹은 개시체중 및 종료 체중이 다른 그룹에 비해 유의적으로 높았고, 전체 실험기간 일일사료섭취량은 적고 일당증체량은 유의적으로 높아 사료요구율이 낮은 경향을 나타냈다. 따라서 항생제 또는 신바이오틱스의 사료 내 첨가는 개시체중에서 이미 유의미한 차이가 있는 경우, 이 차이를 극복할 수 있을 정도의 성장 촉진을 이루기는 어려울 것으로 판단된다. 그러나 더 정확한 결론을 도출하기 위해서는 생시체중 및 이유체중의 구간별 비교, 항생제와 신바이오틱스의 종류별, 농도별 비교, 육성 및 비육기간까지의 성장특성 비교 등 추가적인 실험이 요구된다.

Effects of Synbiotics Containing Anaerobic Microbes and Prebiotics on In vitro Fermentation Characteristics and In situ Disappearance Rate of Fermented-TMR

  • This study was carried out to estimate effects of synbiotics containing anaerobic microorganisms and prebiotics on in vitro fermentation characteristics and in situ disappearance rate of fermented total mixed ration (F-TMR). For the in vitro trial, ninety vinyl bags were prepared to analyze temperature, pH, ammonia concentration, microbial growth rate and short chain fatty acid concentration. For the in situ trial, one hundred twenty nylon bags were prepared to analyze dry matter (DM), acid detergent fiber (ADF) and neutral detergent fiber (NDF) disappearance rate. Treatments consisted of a basal diet (US) with prebiotics and probiotics from anaerobic mold (MS), bacteria (BS), yeast (YS) or compound (CS). It was found that temperatures at 14 and 21 days were significantly higher (p<0.05) in the YS and CS than in the others. The pH at 21 days was lower in the CS than in the US. The synbiotic treatments had significantly increased (p<0.05) ammonia concentration at 21 days. The DM disappearance at 72 h was significantly higher (p<0.05) in the MS and CS than in the others. ADF and NDF disappearance rate tended to increase at a rate similar to the DM disappearance rate. Therefore, this study suggests that synbiotics (probiotics with prebiotics) may partially help the quality of fermentation and digestibility of TMR (MS and CS) as fiber disappearance.

Glycine max Merr enhances the viability and adhesion ability of Lactobacillus buchneri in gastrointestinal condition in vitro.

  • Probiotics are microorganisms that have beneficial effects on the health of the host. The health promoting effect by probiotics influences suppressing harmful bacteria, prevention of constipation, blood cholesterol reduction and regulation of blood pressure. Prebiotics are used to promote the growth or activity of microorganisms. Synbiotics, which are a mixture of probiotics and prebiotics, synergize in the intestines by complementing each other. Synbiotics not only improves the viability of the probiotics while passing through the gastrointestinal tract, maintain intestinal homeostasis, but also regulate balance of harmful and useful bacterial growth. Glycine max Merr (GMM) has been widely used in Asian countries to treat cancer, obesity, oxidative stress and imbalanced immune diseases. In addition, it has been reported that dietary fiber-rich grains promote bowel movements and prevent constipation. In this study, we investigated the viability of LactobacillIus buchneri (L.buchneri) strains, known as lactic acid bacteria under conditions of gastric fluid and intestinal fluid to determine the suitability of L.buchneri as probiotics. The adhesion ability of L.buchneri to caco-2 cells was also confirmed. The present studies showed that GMM extract promoted the growth and activity of L.buchneri strains as prebiotics. Also, this results suggested that the mixture of L.buchneri and GMM extract can helps maintain intestinal health and healthy body as synbiotics and health functional food material.

Effect of Synbiotic on the Treatment of Jaundice in Full Term Neonates: A Randomized Clinical Trial

  • Purpose: Jaundice accounts for most hospital admissions in the neonatal period. Nowadays, in addition to phototherapy, other auxiliary methods are used to reduce jaundice and the length of hospitalization. This study aimed to investigate the effect of probiotics on the treatment of hyper-bilirubinemia in full-term neonates. Methods: In this randomized clinical trial, 83 full-term neonates, who were admitted to the hospital to receive phototherapy in the first 6 months of 2015, were randomly divided into two groups: synbiotic (SG, n=40) and control (CG, n=43). Both groups received phototherapy but the SG also received 5 drops/day of synbiotics. Serum bilirubin, urine, stool, feeding frequency, and weight were measured daily until hospital discharge. A p-value<0.05 was considered statistically significant. Results: The mean total serum bilirubin in the SG was lower than that in the CG ($9.38{\pm}2.37$ and $11.17{\pm}2.60mg/dL$, respectively). The urine and stool frequency in the SG was significantly higher than that in the CG (p<0.05). The duration of hospitalization in the SG was shorter than that in the CG. Conclusion: Use of synbiotics as an adjuvant therapy had a significant treatment effect on jaundice in full-term neonates. Further studies including larger samples with long follow-up periods are essential to confirm the benefits of routine use of synbiotics in neonatal patients with jaundice.

  • It is a recent observation that about 80 per cent of the body's immune system is localized in the gastrointestinal tract. This explains to a large extent why eating right is important for the modulation the immune response and prevention of disease. In addition it is increasingly recognized that the body has an important digestive system also in the lower gastrointestinal tract where numerous important substances are released by microbial enzymes and absorbed. Among these substances are short chain fatty acids, amino acids, various carbohydrates, poly-amines, growth factors, coagulation factors, and many thousands of antioxidants, not only traditional vitamins but numerous flavonoids, carotenoids and similar plant- and vegetable produced antioxidants. Also consumption of health-promoting bacteria (probiotics) and vegetable fibres (prebiotics) from numerous sources are known to have strong health-promoting influence. It has been calculated that the intestine harbours about 300,000 genes, which is much more than the calculated about 60,000 for the rest of the human body, indicating a till today totally unexpected metabolic activity in this part of the GI tract. There are seemingly several times more active enzymes in the intestine than in the rest of the body, ready to release hundred thousand or more of substances important for our health and well-being. In addition do the microbial cells produce signal molecules similar to cytokines but called bacteriokines and nitric oxide, with provide modulatory effects both on the mucosal cells, the mucosa- associated lymphoid system (MALT) and the rest of the immune system. Identification of various fermentation products, and often referred to as synbiotics, studies of their role in maintaining health and well-being should be a priority issue during the years to come.

The effects of synbiotics-glyconutrients on growth performance, nutrient digestibility, gas emission, meat quality, and fatty acid profile of finishing pigs

  • Glyconutrients help in the body's cell communication. Glyconutrients and synbiotics are promising options for improving immune function. Therefore, we hypothesized that combining synbiotics and glyconutrients will enhance pig nutrient utilization. 150 pigs (Landrace × Yorkshire × Duroc), initially weighing 58.85 ± 3.30 kg of live body weight (BW) were utilized to determine the effects of synbiotics-glyconutrients (SGN) on the pigs' performance, feed efficiency, gas emission, pork traits, and composition of fatty acids. The pigs were matched by BW and sex and chosen at random to 1 of 3 diet treatments: control = Basal diet; TRT1 = Basal diet + SGN 0.15%; TRT2 = Basal diet + SGN 0.30%%. The trials were conducted in two phases (weeks 1-5 and weeks 5-10). The average daily gain was increased in pigs fed a basal diet with SGN (p = 0.036) in weeks 5-10. However, the apparent total tract digestibility of dry matter, nitrogen, and gross energy did not differ among the treatments (p > 0.05). Dietary treatments had no effect on NH3, H2S, methyl mercaptans, acetic acids, and CO2 emissions (p > 0.05). Improvement in drip loss on day 7 (p = 0.053) and tendency in the cooking loss were observed (p = 0.070) in a group fed basal diets and SGN at 0.30% inclusion level. The group supplemented with 0.30% of SGN had higher levels of palmitoleic acid (C16:1), margaric acid (C17:0), omega-3 fatty acid, omega-6 fatty acid, and ω-6: ω-3 ratio (p = 0.034, 0.020, 0.025, 0.007, and 0.003, respectively) in the fat of finishing pigs. Furthermore, group supplemented with 0.30% of SGN improved margaric acid (C17:0), linoleic acid (C18:2n6c), arachidic acid (C20:0), omega 6 fatty acid, omega-6 to omega-3 ratio, unsaturated fatty acid, and monounsaturated fatty acid (p = 0.037, 0.05, 0.0142, 0.036, 0.033, 0.020, and 0.045, respectively) in the lean tissues of finishing pigs compared to pigs fed with the control diets. In conclusion, the combination of probiotics, prebiotics, and glyconutrients led to higher average daily gain, improved the quality of pork, and more favorable fatty acid composition. Therefore, these results contributed to a better understanding of the potential of SGN combinations as a feed additive for pigs.

국내산 조제분유의 최근 연구 개발 동향 (Current Trends of Research and Developments of Infant Formula in Korea)

  • Human milk contains the nutrients for the growth and the development of the newborn. It contains specific proteins, lipids, and other components designed to be easily digestible and which play important roles in infant nutrition. However, when the lactation is failure or milk secretion is not enough, bovine milk based dried formulations, which is called infant formula, have been widely used for infant feeding. In this study, we investigated protein composition and amino acids profile related to easy digestion; fatty acids composition and LCPUFA(DHA and ARA) related to brain and eye development; synbiotics which take advantage of both probiotics and prebiotics; and colostrum-originated functional micronutrient as major issues in the recent infant formula. Domestic infant formulas, which are based on the research of human and bovine milk rich in functional components for immunity, growth, anti-allergenicity, digestion and brain development, have been developed to be similar to human milk.

Effects of glycozyme addition on fatty acid and meat quality characteristics of growing pigs

  • Synbiotics help to improve gut health by promoting the growth of beneficial bacteria while glyconutrients provide a source of energy for the gut bacteria and may also have immunemodulating effects. The aim of the present study was to assess the effect of this combination on fatty acid and meat quality characteristics of growing pigs. In a five-week experiment, 804 growing three-way crossbred ([Landrace × Yorkshire] × Duroc) pigs with an initial body weight of 31.90 ± 2.6 kg on average were assigned to two treatments: 1) CON (basal diet) and 2) TRT1 (basal diet + 0.3% glycozyme [synbiotics and glyconutrient]), each consisting of 402 pigs. The TRT1 groups showed significantly higher values of palmitoleic acid (C16:1), capric acid (C10:0), myristic acid (C14:0), lauric acid (C12:0), elaidic acid (C18:1, t), pentadecylic acid (C15:0), gondoic acid (C20:1), lignoceric acid (C24:0), and omega-6 : omega-3 in fat than the CON groups. Moreover, in the lean tissues of the pig, the levels of C12:0, C14:0, C17:0, and C20:1 were significantly higher in TRT1 than in CON. However, significant differences were not observed after glycozyme addition in pH, water holding capacity, cooking loss, longissimus muscle area, drip loss, meat color, and sensory evaluation parameters. To conclude, the positive results of the fatty acid composition indicate that glycozyme may be an effective pig feed additive.

한국전통발효식품에서 분리한 Probiotics의 특징 및 Synbiotics 항균활성 효과 (Characteristics of Probiotics Isolated from Korean Traditional Foods and Antibacterial Activity of Synbiotics)

  • 세계 각국의 민족은 자신의 국토의 삶과 기후 풍토, 지역 특산 산물과 식습관 및 식생활 등에 따라 색다른 전통식품이 만들어 진다. 한국은 오랫동안 농경을 중심으로 먹거리를 해오며 쌀과 함께 곁들일 채소류를 중심으로 조미료가 발달되었고, 염절임 기법을 통해 발효식품을 만들었다. 이러한 발효식품에서 주로 찾아볼 수 있는 종은 Lactobacillus sp.과 Pediococcus sp. 및 Bacillus sp. 등과 같은 유산균을 분리할 수 있었고 다양한 방법으로 동정 되어왔다. 본 연구에서는 시장에서 시판되고 있는 한국전통발효식품을 통해 유산균을 분리 및 동정하였고, 유해세균에 대한 항균능력이 우수한 균주를 선별하였다. 그리고 우수 후보 균주의 인공위액 및 담즙액에 대한 내성과 용혈능 등을 검토하여 최종 균주를 선별하였다. 최종 선별된 유산균은 3종의 prebiotics와 혼합한 synbiotics로서의 항균활성 능력을 평가하였다. 1차 항균 활성에서 C13은 인체 및 어류질병세균에서 가장 넓은 항균 스펙트럼을 보였고, β-haemolysis를 생산하지 않고 인공위액과 담즙액의 내성을 지닌 M1, K1 및 C13을 synbiotics의 2차 항균 활성을 수행하였다. Prebiotics 3종(FOS, GOS, Inulin)과 선별된 균주가 혼합된 synbiotics에서는 이전 보다 항균 활성이 증진 또는 저해됨을 알 수 있었다. 16 rDNA 염기서열 결과, K1과 M1은 Bacillus tequiensis 99.72%, Bacillus subtilis 99.65%, Bacillus inaquosorum 99.72%, Bacillus cabrialesii 99.72%, Bacillus stercoris 99.58%, Bacillus spizizenii 99.58%, Bacillus halotolerans 99.58%, Bacillus mojavensis 99.51%로 분석되었다. 그리고 C13은 Bacillus velezensis 99.71%, Bacillus nematocida 99.36%, Bacillus amyloliquefaciens 99.44%, Bacillus atrophaeus 99.22%, Bacillus nakamurai 99.44%로 분석되었다.
