Effects of Silkworm(Bombyx mori L..) Powder on Lipofuscin, Acetylcholine and Its Related Enzyme Activities in Brain of Rats (뇌조직의 리포푸신, 아세틸콜린 및 그 관련효소 활성에 미치는 누에분말의 영향)

  • This study was designed to investigate the effect of silkworm (Bombyx mori L.) powder on lipofuscin, acetylcholine (ACh) and its related enzyme activities in brain of rats. Sprague-Dawley (SD) male rats ($160{\pm}10g$) were fed basic diet (control group), and experimental diets (SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. In case of liver membranes, lipofuscin (LF) levels resulted in a slight decreases (4.6% and 11.5%, respectively) in SWP-200 and SWP-400 groups compared with control group. But in case of barin as the most sensitive organ, LF levels were remarkably inhibited about 16.7% and 20.0% in SWP-200 and SWP-400 groups compared with control group. There were no significant differences in acetylcholine (ACh) syntheses as a very important neurotransmitter, and choline acetyltransferase (ChAT) activities as a synthesis enzyme of ACh, and acetylcholinesterase (AChE) activities as a hydrolysis enzyme, which were concerned in transmission of neuron through synapses in brain of SWP-200 and SWP-400 groups compared with control group. Monoamine oxidase-B (MAO-B) activities were significantly inhibited (about 10.2%) in brain of SWP-400 groups compared with control group. These results suggest that inhibiting effects of LF accumulation and MAO-B activity of silkworm powder (SWP) may play a pivotal role in attenuating a various age-related changes for improvement of brain function.

  • N-Acetylglucosamine kinase (GlcNAc kinase or NAGK; EC 2.7.1.59) catalyzes the phosphorylation of GlcNAc to GlcNAc-6-phosphate (GlcNAc-6-P). Despite detailed characterization of the enzyme itself, there have been few studies on the expression of NAGK in mammalian tissues. In the rat hippocampal neuron in culture, NAGK-immunoreactivity (IR) formed clusters in somatodendritic domains. In this study we characterized the NAGK clusters that protrude out the long axis of dendritic shafts. By double-labeling of the neurons with antibodies against NAGK and various synaptic proteins, we show that NAGK is positioned at the base of spines, while there were no NAGK protrusions into inhibitory postsynaptic sites. Immunoblot analysis showed that NAGK was included in synaptosomes but not in PSD fractions. Our results indicate that the NAGK clusters at the dendritic periphery protrude into spines.

Hormonal Control of Induction and Termination of Diapause in Pieris rapae L. (배추흰나비(Pieris rapae L.)의 휴면 유도 및 타파에 대한 호르몬의 조절)

  • Non-diapausing larvae and diapausing pupae of Pieris rapae L. were treated with 20-HE and JH, and the concentration of 20-HE and JH in body were also measured to determine mechanisms of diapause induction and termination. When early last instar larvae of non-diapausing stage were injected with 20-HE or JH-I, diapause was not induced in both cases. 20-HE concentration of pre-diapausing last instar larvae was very low, and 20-HE peak of non-diapausing two-day-old pupae was not present in diapausing pupae. There was no difference in concentration of JH-I between diapausing and non-diapausing stages. When early and late diapausing pupae were injected with 20-HE, JH-I, or JH-III. diapause was terminated by 0.1-0.5 $\mu$g of 20-HE, but not by JH-I, or JH-III. The above results indicate that diapause of Pieris rapae was induced by decrease of 20-HE concentration and also terminated by gradual accumulation of 20-HE during diapause process.

  • This study examines how poem and poetic ego of Cho Ji-Hoon form synapses. It is to clarify the synaptic structure of the healing, the contact point between the literary mechanism and the mechanism of the ego. Therefore, it aims to encode the active therapy by substituting the structure into the literary therapy program. Cho Ji-Hoon's poem "Bellflower Flower" is a mesh of poem, and a mesh of semantic arguments is set up for the 'Bellflower Flower' of functor. At this time, the longing that attracts depression to the net of the semantic argument is caught. This exists as a function of healing. If we embody a literary therapy program that utilizes the synaptic structure of this healing, it will be able to experience the function of literary therapy improved than before.

Integrated Circuit Implementation and Characteristic Analysis of a CMOS Chaotic Neuron for Chaotic Neural Networks (카오스 신경망을 위한 CMOS 혼돈 뉴런의 집적회로 구현 및 특성 해석)

  • This paper presents an analysis of the dynamical behavor in the chaotic neuron fabricated using 0.8${\mu}{\textrm}{m}$ single poly CMOS technology. An approximated empirical equation models for the sigmoid output function and chaos generative block of the chaotic neuron are extracted from the measurement data. Then the dynamical responses of the chaotic neuron such as biurcation diagram, frequency responses, Lyapunov exponent, and average firing rate are calculated with numerical analysis. In addition, we construct the chaotic neural networks which are composed of two chaotic neurons with four synapses and obtain bifurcation diagram according to synaptic weight variation. And results of experiments in the single chaotic neuron and chaotic neural networks by two neurons with the $\pm$2.5V power supply and sampling clock frequency of 10KHz are shown and compared with the simulated results.

  • Parkinson's disease is one of the most frequent neurodegenerative diseases, which mainly affects the elderly. Parkinson's disease is often difficult to differentiate from atypical parkinson diorder such as progressive supranuclear palsy, multiple system atrophy, dementia with Lewy body, and corticobasal ganglionic degeneration, based on the clinical findings because of the similarity of phenotypes and lack of diagnostic markers. The accurate diagnosis of Parkinson's disease and atypical Parkinson disorders is not only important for deciding on treatment regimens and providing prognosis, but also it is critical for studies designed to investigate etiology and pathogenesis of parkinsonism and to develop new therapeutic strategies. Although degeneration of the nigrostriatal dopamine system results in marked loss of striatal dopamine content in most of the diseases causing parkinsonism, pathologic studies revealed different topographies of the neuronal cell loss in Parkisonism. Since the regional cerebral glucose metabolism is a marker of integrated local synaptic activity and as such is sensitive to both direct neuronal/synaptic damage and secondary functional disruption at synapses distant from the primary site of pathology, an assessment of the regional cerebral glucose metabolism with F-18 FDG PET is useful in the differential diagnosis of parkinsonism and evaluating the pathophysiology of parkisonism.

A Neural Network Design using Pulsewidth-Modulation (PWM) Technique (펄스폭변조 기법을 이용한 신경망회로 설계)

  • In this paper, a design of the pulsewidth-modulation(PWM) neural network with both retrieving and learning function is proposed. In the designed PWM neural system, the input and output signals of the neural network are represented by PWM signals. In neural network, the multiplication is one of the most commonly used operations. The multiplication and summation functions are realized by using the PWM technique and simple mixed-mode circuits. Thus, the designed neural network only occupies the small chip area. By applying some circuit design techniques to reduce the nonideal effects, the designed circuits have good linearity and large dynamic range. Moreover, the delta learning rule can easily be realized. To demonstrate the learning capability of the realized PWM neural network, the delta learning nile is realized. The circuit with one neuron, three synapses, and the associated learning circuits has been designed. The HSPICE simulation results on the two learning examples on AND function and OR function have successfully verified the function correctness and performance of the designed neural network.

  • The adaptive learning circuit is designed on the basis of modeling of MFSFET (Metal-Ferroelectric-Semiconductor FET) and the numerical results is analyzed. The output frequency of the adaptive learning circuit is inversely proportional to the source-drain resistance of MFSFET and the capacitance of the circuit. The saturated drain current with input pulse number is analogous to the ferroelectric polarization reversal. It indicates that the ferroelectric polarization plays an important role in the drain current control of MFSFET. The output frequency modulation of the adaptive learning circuit is investigated by analyzing the source-drain resistance of MFSFET as functions of input pulse numbers in the adaptive learning circuit and the dimensionality factor of the ferroelectric thin film. From the results, adaptive learning characteristics which means a gradual frequency change of output pulse with the progress of input pulse, are confirmed. Consequently it is shown that our circuit can be used effectively in the neuron synapses of neural networks.

Effects of δ-Catenin on APP by Its Interaction with Presenilin-1

  • Alzheimer's disease (AD) is the most frequent age-related human neurological disorder. The characteristics of AD include senile plaques, neurofibrillary tangles, and loss of synapses and neurons in the brain. ${\beta}-Amyloid$ ($A{\beta}$) peptide is the predominant proteinaceous component of senile plaques. The amyloid hypothesis states that $A{\beta}$ initiates the cascade of events that result in AD. Amyloid precursor protein (APP) processing plays an important role in $A{\beta}$ production, which initiates synaptic and neuronal damage. ${\delta}-Catenin$ is known to be bound to presenilin-1 (PS-1), which is the main component of the ${\gamma}-secretase$ complex that regulates APP cleavage. Because PS-1 interacts with both APP and ${\delta}-catenin$, it is worth studying their interactive mechanism and/or effects on each other. Our immunoprecipitation data showed that there was no physical association between ${\delta}-catenin$ and APP. However, we observed that ${\delta}-catenin$ could reduce the binding between PS-1 and APP, thus decreasing the PS-1 mediated APP processing activity. Furthermore, ${\delta}-catenin$ reduced PS-1-mediated stabilization of APP. The results suggest that ${\delta}-catenin$ can influence the APP processing and its level by interacting with PS-1, which may eventually play a protective role in the degeneration of an Alzheimer's disease patient.

A Study on Purification Process of Sialic Acid from Edible Bird's Nest Using Affinity Bead Technology (식용 제비집으로부터 비극성 비드기술을 활용한 시알산의 분리정제방법에 관한 연구)

  • Sialic acid, which is contained in about 60-160 mg/kg in the edible bird's nest (EBN), is known to facilitate in the proper formation of synapses and improve memory function. The objective of this study is to extract effectively the sialic acid from edible bird's nest using affinity bead technology (ABT). After preparing the non-polar polymeric bead "KJM-278-28A" having a porous network structure, and then desorbed sialic acid was concentrated and dried. The analysis of the physicochemical properties of bead "KJM-278-28A" showed that the particle size was 400-700 ㎛, the moisture holding capacity was 67-70%, the surface area (BET) was 705-900 ㎡/g, and the average pore diameter 70-87 Å. The adsorption capacity of the bead "KJM-278-28A" for sialic acid was shown a strong physical force to bind sialic acid to the bead surface of 400 mg/L. In addition, as a result of analyzing the adsorption and desorption effects of sialic acid on water, ethanol, and 10% ethanol on the bead, it was confirmed that desorption effectively occurs from the beads when only ethanol is used. As a result of HPLC measurement of the separated sialic acid solution, a total of four sialic acid peaks of N-acetyl-neuraminic acid (Neu5Ac), α,β-anomer of Neu5Ac and N-glycoly-neuraminic acid were identified. Through these results, it was confirmed that it is possible to separate sialic acid from EBN extract with efficient and high yield when using ABT.
