Workplace learning experience for corporate workers under COVID-19 : Focusing on S Energy Company (코로나 19 상황의 기업근로자의 일터 학습경험: S에너지를 중심으로)

  • This study was conducted to do in-depth exploration of coroperate employees' learning experiences for workplace under COVID-19. We collected data through depth interviews from August 10th to November 30th, 2020 with five employees of S Energy company, and a qualitative case study was conducted using Ricci's 3-step analysis procedure. As a result, In the process of adapting from the landing of COVID-19, "The Story of a Distant Country," "No More Safe Zones," "Exploring effective responses created by a Sense of Crisis," and "Learning the changed way of work for adaptation." appeared. In addition, It has been found that the experience of learning in the work environment of the With Corona era includes "learning experience for survival from a sense of crisis", "learning experience for adapting to Untact culture", "learning experience through SNS communication", and "competitive learning experience for performance creation". In conclusion, employees have adapted to changes in the workplace environment through various learning experiences, which can enhance workers' ability to cope with crisis situations and can be used as basic data for an effective learning. In the future, we suggested follow-up researches of corporate employees in various fields.

Platelet-Derived Growth Factor Receptor-α Subunit Targeting Suppresses Metastasis in Advanced Thyroid Cancer In Vitro and In Vivo

  • Thyroid cancer is the most common endocrine malignancy. Patients with well-differentiated thyroid cancers, such as papillary and follicular cancers, have a favorable prognosis. However, poorly differentiated thyroid cancers, such as medullary, squamous and anaplastic advanced thyroid cancers, are very aggressive and insensitive to radioiodine treatment. Thus, novel therapies that attenuate metastasis are urgently needed. We found that both PDGFC and PDGFRA are predominantly expressed in thyroid cancers and that the survival rate is significantly lower in patients with high PDGFRA expression. This finding indicates the important role of PDGF/PDGFR signaling in thyroid cancer development. Next, we established a SW579 squamous thyroid cancer cell line with 95.6% PDGFRA gene insertion and deletions (indels) through CRISPR/Cas9. Protein and invasion analysis showed a dramatic loss in EMT marker expression and metastatic ability. Furthermore, xenograft tumors derived from PDGFRA geneedited SW579 cells exhibited a minor decrease in tumor growth. However, distant lung metastasis was completely abolished upon PDGFRA gene editing, implying that PDGFRA could be an effective target to inhibit distant metastasis in advanced thyroid cancers. To translate this finding to the clinic, we used the most relevant multikinase inhibitor, imatinib, to inhibit PDGFRA signaling. The results showed that imatinib significantly suppressed cell growth, induced cell cycle arrest and cell death in SW579 cells. Our developed noninvasive apoptosis detection sensor (NIADS) indicated that imatinib induced cell apoptosis through caspase-3 activation. In conclusion, we believe that developing a specific and selective targeted therapy for PDGFRA would effectively suppress PDGFRA-mediated cancer aggressiveness in advanced thyroid cancers.

Physiological Activities of wild Conyza canadensis L. Extracts (야생 망초(Conyza canadensis L) 에틸알코올 추출물의 생리활성)

  • Background: Studies on the existence and activation of antioxidants in the wild Erigeron canadensis are still incomplete. Purposes: The activity of antioxidant substances was studied by extracting E. canadensis with ethyl alcohol. Methods: After washing the wild turfgrass, extraction with ethyl alcohol was used to investigate the activity of antioxidant substances using various analytical instruments. Results: The highest yield ratio of the extract was 49.3%, and it varied according to temperature and ethyl alcohol ratio. The 50:50 of water to ethyl alcohol at 180℃ was the highest yield. 100% survival rate was in the untreated group and 98-100% in the experimental group at 50 ppm or more of the extract. There was no cytotoxicity at almost all concentrations. The extract of 25 ppm was suppressed by 42% in the test group. The extract of 50 ppm reduced the free fatty acid content by 15%. Cell viability was 20% at the extract concentration of 50 ppm and 24% at 100 ppm. At an extract of 500 ppm, the free oxygen scavenging ability using DPPH and ABTs was 90-98%. When the changes in the free radical scavenging activity of DPPH and ABTs were observed in three dimensions, the antioxidant activity tended to increase at 85℃ as the temperature increased. However, at 85~130℃, it showed a rather decreasing tendency as the temperature increased. Conclusions: There were antioxidants in the ethyl alcohol extract of the wild grass, there was almost no cytotoxicity and suppressed NO production, and the scavenging function of free oxygen was also high. These results provide primary data for the various natural healing uses of the extracts of the turfgrass.

Peroxidase Activity of Peroxidasin Affects Endothelial Cell Growth (내피 세포 성장에 영향을 미치는 PXDN의 peroxidase 활성)

  • Peroxidasin (PXDN), a multidomain heme peroxidase containing extracellular matrix (ECM) motifs, as well as a catalytic domain, catalyzes the sulfilimine crosslink of collagen IV (Col IV) to reinforce Col IV scaffolds. We previously reported that PXDN is required for endothelial cell (EC) survival and growth signaling through sulfilimine crosslink-dependent matrix assembly. In this study, we examined whether peroxidase activity is required for PXDN function in ECs. First, we constructed a mutant PXDN by point mutation of two highly conserved amino acids, Q823 and D826, which are present in the active site of the peroxidase domain. After isolation of HEK293 clones highly expressing the mutant protein, conditioned medium (CM) was obtained after incubating the cells in serum-free medium for 24 hours and then analyzed by Western blot analysis under nonreducing conditions. The results revealed that the mutant PXDN formed a trimer and that it was cleaved by proprotein convertase-like wild-type (WT) PXDN. However, peroxidase activity was not detected in the CM containing the mutant PXDN, in contrast to that of WT PXDN. In addition, the sulfilimine crosslink ability of the mutant PXDN was lost. Moreover, the CM containing the mutant PXDN failed to promote the growth of PXDN-depleted ECs, unlike the CM containing WT PXDN. These results suggest that the peroxidase activity of PXDN affects EC growth by forming a sulfilimine crosslink.

  • The purpose of this study is to present the direction of development of our navy's response strategy through analysis at the ends, ways, and means level of the Chinese navy's offensive strategy for the West Sea. As a result of the analysis, at the ends level, the Chinese Navy's offensive strategy for the West Sea strategy is being linked to a grand strategy to protect maritime rights and achieve maritime power between the U.S. and China competition, at the ways level, the Chinese Navy is expected to create a foundation for the international community to recognize the West Sea as China's inland sea through "routine entry" and "exercise authority", and in case of emergency, it will try to secure sea control in the West Sea in a short period of time by blocking Korea's maritime transportation route based on the overwhelming preemptive attack capability of aircraft carriers. At the means level, it is accelerating the construction of aircraft carrier warfare units and improving its ability to engage long-range missiles. As a direction of development of the Korean Navy's response strategy in response to this, first, Establishment and Development of National Maritime Security Strategy in conjunction with the Korean Indo-Pacific Strategy. Second, it proposes the development of the concept of effective security operations for the east sea area of the West Sea intermediate line, and third, the development of the concept of combat performance and capacity building to strengthen survival and lethality.

Effects on Lethal Concentration 50%, Hematological Parameters and Plasma Components of Mirror carp, Cyprinus carpio nudus Exposed to Waterborne Nitrate (수인성 질산 급성 노출에 의한 향어, Cyprinus carpio nudus의 반수치사농도, 혈액학적 성상 및 혈장성분에 미치는 독성 영향)

  • To determine the toxic effects of the mirror carp, Cyprinus carpio nudus on waterborne nitric acid exposure, they were exposed to waterborne nitric acid at 0, 4, 20, 100, 500, and 2,500 mg NO3-/L for 96 h. The semi-lethal concentration (LC50) of the scented fish by 96 hours of acute exposure to nitric acid was 1,433.54 mg NO3-/L. The physiological changes of waterborne nitric acid exposure on mirror carp were evaluated through hematologic properties, and RBC count showed a significantly decreased (p < 0.05). The change in the ion control ability of the mirror carp according to exposure to waterborne nitric acid was evaluated through the plasma inorganic component, and calcium and magnesium were significantly decreased (p < 0.05) in the plasma inorganic component. The health and stress status of the mirror carp due to exposure to waterborne nitric acid were evaluated through plasma organic ingredients, and the plasma organic component glucose showed a significantly increase (p < 0.05). Changes in liver damage and enzyme activity due to waterborne nitric acid exposure were evaluated through plasma enzyme components, the enzymatic activities of plasma AST, ALT, and ALP were significantly decreased (p < 0.05) in high levels of waterborne nitric acid exposure. The results of the present study suggest that waterborne nitric acid exposure to C. carpio nudus can toxicly affect survival, hematologic properties, and plasma components.

  • The elderly in rural areas are faced with the dilemma of poor community environment, weak social communication ability and insufficient pension knowledge reserve. In addition, due to the serious shortage of social security facilities in rural areas and medical resources, the elderly are struggling, and the suicide rate is far higher than that in urban areas. In order to make the elderly have a comfortable pension environment and face the increasingly serious aging problem with a positive attitude, this paper takes the community environment of the Baima Tibetan elderly in Tielou Township, Gansu Province as the research object of aging transformation. First of all, literature data were used to carry out research on the aging transformation in rural areas. On the basis of sorting out previous research topics, ERG theory was determined as the guide. Secondly, the research methods of on-site investigation, interview and other research methods are adopted to investigate the number of left-behind elderly people in this area, and classify them according to the national standards. At the same time, the image of the current situation of the community environment of the elderly. Finally, combined with the ERG theory, the transformation design of the elderly living environment is implemented, mainly from the three aspects of survival, mutual relationship and growth.

Highly efficient genome editing via CRISPR-Cas9 ribonucleoprotein (RNP) delivery in mesenchymal stem cells

  • The CRISPR-Cas9 system has significantly advanced regenerative medicine research by enabling genome editing in stem cells. Due to their desirable properties, mesenchymal stem cells (MSCs) have recently emerged as highly promising therapeutic agents, which properties include differentiation ability and cytokine production. While CRISPR-Cas9 technology is applied to develop MSC-based therapeutics, MSCs exhibit inefficient genome editing, and susceptibility to plasmid DNA. In this study, we compared and optimized plasmid DNA and RNP approaches for efficient genome engineering in MSCs. The RNP-mediated approach enabled genome editing with high indel frequency and low cytotoxicity in MSCs. By utilizing Cas9 RNPs, we successfully generated B2M-knockout MSCs, which reduced T-cell differentiation, and improved MSC survival. Furthermore, this approach enhanced the immunomodulatory effect of IFN-r priming. These findings indicate that the RNP-mediated engineering of MSC genomes can achieve high efficiency, and engineered MSCs offer potential as a promising therapeutic strategy.

FUN14 Domain-Containing Protein 1 Is Involved in Amyloid Beta Peptide-Induced Mitochondrial Dysfunction and Cell Injury in HT-22 Neuronal Cells (HT-22 신경세포에서 아밀로이드 베타 펩티드에 의한 미토콘드리아와 세포 손상 기전에서 FUN14 도메인 함유 단백 1의 역할)

  • FUN14 domain-containing protein 1 (FUNDC1), an outer mitochondrial membrane protein, contributes to removal of damaged mitochondria through mitophagy. In this study, to elucidate the role of the FUNDC1 in the amyloid beta peptide (Aβ)-induced neuropathy, changes in the degree of mitochondrial dysfunction and cell injury caused by Aβ treatment were examined in the HT-22 neuronal cells in which the FUNDC1 expression was transiently silenced or overexpressed. We found that Aβ treatment causes a time-dependent decrease of the FUNDC1 expression. In the Aβ-treated cells, there were a drop in MTT reduction ability, depletion of cellular ATP, disruption of mitochondrial membrane potential, stimulation of cellular ROS production, and increased mitochondrial Ca2+ load. Activation of caspase-3 and induction of apoptotic cell death were also observed. Transient silencing of the FUNDC1 expression by transfection with the FUNDC1 small interfering RNA per se caused mitochondrial dysfunction and apoptotic cell death like the effect of Aβ treatment. Conversely, in cells in which the FUNDC1 was transiently overexpressed by FUNDC1-Myc transfection, overexpression itself had no effect on the mitochondrial functional integrity and cell survival but showed a significant prevention effect against mitochondrial and cell injury caused by Aβ treatment. Overall, these results suggest that the FUNDC1 is importantly involved in the Aβ-induced mitochondrial dysfunction and cell injury in the HT-22 neuronal cells.

Regrowth Ability and Species Composition of Phytoplankton in International Commercial Ship's Ballast Water Berthed at Pusan and Daesan Ports (부산과 대산항에서 선박평형수에 유입된 식물플랑크톤의 종조성과 재성장능력)

  • The aim of this study is to assess the importance of ballast water discharge as a vector for the introduction of exotic species into Pusan and Daesan Ports, Korea. We also examined to understand the impacts of environmental factors on the survival success of introduced species by ship's ballast water in laboratory experiments. Seven ship's ballast water originated from the coastal water of China (Taicang, Ningbo and Jinshan), Japan (Tokuyama, Moji and Akita), and Singapore. According to PCA (principal components analysis) analysis, environmental factor in the each ballast and shipside waters were different by bioregion. Based on cluster analysis, the phytoplankton community structures were distinguished for ballast water origin. Most of the major taxonomic groups were diatoms and, the others were dinoflagellate, silcoflagellate and several fresh-waters species. In particular, species number and standing crops of phytoplankton in the ballast tanks decreased with the increasing age ofballast water(r = -0.35 for standing crop; r = -0.63 for species number). In the laboratory study, although phytoplankton in ballast water treatment did not survive even in optimal temperature, the in vivo fluorescence of phytoplankton viability increased under the nutrient typical of shipside water and F/2 medium at $15^{\circ}C$ and $20^{\circ}C$. The diatoms species such as Skeletonema costatum and Thalassiosira pseudonana in ballast water were successfully regrown. On the salinity gradient experiments for Shui Shan (2) vessel, several freshwater species, brackish and marine species were successfully adapted. Of these, S.costatum was able to tolerate a wide range of salinities (10 to 30 psu) and its species-specific viability was suitable for colonization.
