Species composition of the catches collected by trammel net in the coastal waters off Ulleungdo of Korea (울릉도해역에서 삼중자망에 의한 어획물의 종조성)

  • Species composition in the coastal waters off Ulleungdo of Korea were examined based on catches bimonthly collected by trammel net in 2013. A total of 711 individuals and 181.9 Kg were caught and catches were composed of 4 classes 15 orders 27 families 52 species including 44 Pisces, 4 Gastropoda, 3 Cephalopoda, and 1 Echinodermata. The dominant species in biomass were File fish (Thamnaconus modestus), Atka mackerel (Pleurogrammus azonus), and Greenling (Hexagrammos otakii). Data were summarized using hierarchical cluster analysis (HCA) and detrended correspondence analysis (DCA) to examine similarity in species composition for each month, and community structure in Ulleungdo was divided into two groups. Community structures in February, April and December with low temperature and well-mixed surface water were distinguished from those in June, August and October with high temperature and strong stratification, which could be attributed to temporal changes in dominant species. Atka mackerel and Spear squid mainly caught in February and April, disappearing in June, August and October, and File fish outburst was shown in October. Because the water off Ulleungdo has been under low human pressure, it could be a good case study to elucidate effects of climate change on community structure and ecosystem in the East sea. Continuous surveys and further studies are required to demonstrate migration route and distribution of dominant species and long-term changes in community structure in the water of Ulleungdo.

Environmental Feature Causing a Bloom of the Novel Dinoflagellate Heterocapsa circularisquama (Dinophyceae) in Uranouchi Bay, Kochi Prefecture, Japan (일본 Kochi현 Uranouchi만의 와편모조류, Heterocapsa circularisquama (Dinophyceae) 적조발생에 대한 환경 고찰)

  • To study the environmental features causing a bloom of the novel dinoflagellate Heterocapsa circularisquama (Dinophyceae), hydrographic and chemical aspects were measured in the Uranouchi Bay, Kochi Prefecture, Japan, from January to December, 1997. The cell density of H. circularisquama increased rapidly in early October, and dropped sharply in mid-October. Growth rate of H, circularisquama during bloom period appeared 1.50 division day$^{-1}$ under high water temperature (25$^{\circ}C$) and salinity (32 psu) conditions. Althought the result from hydrographic aspect indicated good condition for their growth, dissolved inorganic phosphorus (DIP) concentration in surface layer before bloom formation was less than 0.70uM, which is lower than their half saturation constant(Ks). Dissolved inorganic nitrogen(DIN): DIP ratio was > 30, indicating potential P-limitation. However, before bloom formation period of H. circularisquama, DIP concentrations were high in bottom layer (> 4.0 uM). Some studies reported that H. circularisquama had the ability to migrate vertically and to utilize dissolved organic phosphorus (DOP). Thus, DIP in bottom layer might have been utilized by H. circularisquama for their growth.DOP might have weakly affected their growth because of low reactive DOP concentrations owing to low DOP concentration (ca. 0.39 uM). Thus, if nutrient condition of bottom layer in Uranuchi Bay is not improved, the outbreaks of H. circularisquama red tides may became an annual feature.

Effects of Hormones on the Expression of Matrix Metalloproteinases and Their Inhibitors in Bovine Spermatozoa

  • Proteases and protease inhibitors play key roles in most physiological processes, including cell migration, cell signaling, and cell surface and tissue remodeling. Among these, the matrix metalloproteinase (MMPs) pathway is one of the most efficient biosynthetic pathways for controlling the activation of enzymes responsible for protein degradation. This also indicates the association of MMPs with the maturation of spermatozoa. In an attempt to investigate the effect of MMP activation and inhibitors in cultures with various hormones during sperm capacitation, we examined and monitored the localization and expression of MMPs (MMP-2 and MMP-9), tissue inhibitors of metalloproteinases (TIMP-2 and TIMP-3), as well as their expression profiles. Matured spermatozoa were collected from cultures with follicle-stimulating hormone (FSH), luteinizing hormone (LH), and Lutalyse at 1 h, 6 h, 18 h, and 24 h. ELISA detected the expression of MMP-2, MMP-9, TIMP-2, and TIMP-3 in all culture media, regardless of medium type (FSH-supplemented fertilization Brackett-Oliphant medium (FFBO), LH-supplemented FBO (LFBO), or Lutalyse-supplemented FBO (LuFBO)). TIMP-2 and TIMP-3 expression patterns decreased in LFBO and LuFBO. MMP-2 and MMP-9 activity in FBO and FFBO progressively increased from 1 h to 24 h but was not detected in LFBO and LuFBO. The localization and expression of TIMP-2 and TIMP-3 in sperm heads was also measured by immunofluorescence analysis. However, MMPs were not detected in the sperm heads. MMP and TIMP expression patterns differed according to the effect of various hormones. These findings suggest that MMPs have a role in sperm viability during capacitation. In conjunction with hormones, MMPs play a role in maintaining capacitation and fertilization by controlling extracellular matrix inhibitors of sperm.

Effect of Compressive Loading on the Chloride Penetration of Concrete Mixed with Granulated Blast Furnace Slag (고로슬래그미분말을 혼입한 콘크리트의 염분침투성에 미치는 압축하중의 영향)

  • The analysis of the effect of loading on chloride penetration into concrete is very important. In this study, we confirmed that the chloride penetration rates for plain and BFS concrete were increased by 47% and 89% under compressive stress, respectively. The diffusion coefficient of BFS concrete was lower than for conventional concrete with no BFS, no loads, and under stressed states. Therefore, BFS substitution plays an important role in the repression of chloride penetration even under compressive stress. Under compressive stress,the diffusion coefficient for BFS concrete was higher with increasing stress, and this was also the case for plain concrete. However, BFS concrete was strongly influenced by compressive stress in comparison to plain concrete. We investigated the effect of the difference of specific surfaces on the diffusion coefficient. As a result, the larger specific surface of BFS exhibited a lower diffusion coefficient. This tendency was most pronounced under the high stress conditions.

Comparative Analysis of Envelope Proteomes in Escherichia coli B and K-12 Strains

  • Recent genome comparisons of E. coli B and K-12 strains have indicated that the makeup of the cell envelopes in these two strains is quite different. Therefore, we analyzed and compared the envelope proteomes of E. coli BL21(DE3) and MG1655. A total of 165 protein spots, including 62 nonredundant proteins, were unambiguously identified by two-dimensional gel electrophoresis and mass spectrometry. Of these, 43 proteins were conserved between the two strains, whereas 4 and 16 strain-specific proteins were identified only in E. coli BL21(DE3) and MG1655, respectively. Additionally, 24 proteins showed more than 2-fold differences in intensities between the B and K-12 strains. The reference envelope proteome maps showed that E. coli envelope mainly contained channel proteins and lipoproteins. Interesting proteomic observations between the two strains were as follows: (i) B produced more OmpF porin with a larger pore size than K-12, indicating an increase in the membrane permeability; (ii) B produced higher amounts of lipoproteins, which facilitates the assembly of outer membrane ${\beta}$-barrel proteins; and (iii) motility- (FliC) and chemotaxis-related proteins (CheA and CheW) were detected only in K-12, which showed that E. coli B is restricted with regard to migration under unfavorable conditions. These differences may influence the permeability and integrity of the cell envelope, showing that E. coli B may be more susceptible than K-12 to certain stress conditions. Thus, these findings suggest that E. coli K-12 and its derivatives will be more favorable strains in certain biotechnological applications, such as cell surface display or membrane engineering studies.

Contamination of Heavy Metals from Dongmyeong Au-Ag Mine Area (동명 금-은 광산 주변의 중금속 오염)

  • Researches were carried out to investigate the characteristics and concentration of heavy metal elements of stream water through Dongmyoung abandoned metal mine and soil adjacent to the mine. The pH range of water was 5.9∼7.1 that implies the water environment was acidic to neutral. The contents and distribution aspects of heavy metals in water samples varied with geochemical characteristics of element, but the concentration of heavy metals has the tendency of increase closer to the mine in general. The results of soil analysis show that total heavy metal concentration of agricultural soil near mine was far lower than those of ore tailing and dumping site. Therefore, the effects of the abandoned mine on stream water and agricultural products were supposed to be insignificant, particularly because the portion of absorbed carbonates and reducible fractions among total heavy metal concentration was relatively lower than the other. Since, however total heavy metal concentrations of mining site were relatively higher than those of adjacent region, there is a possibility of heavy metal difussion when the chemical environment of the site changes due to migration of surface and underground water. It is suggested that the preventive measures for water and soil pollution by the heavy metals would be considered around the region.

  • Specimens of eels, collected in the lagoons of Arcachon Bay from April 1977 to October 1978, were analysed for age and growty. The structure of the otolith was described based on examination of the decalccified surface using an scanning electron microscope, and was compared with obcervations made under an optical microscope. The otolith of Arcachon eels show a narrow summer growth stop in addition to a growth stop formed in winter. The narrow summer growtu in length of sedentsry eels was well represented removal of the large memgers by the migration to the sea decreases the growth ratio of sedentary eels as the fish grows older. This size selective migrationin part contributes to satisfy Von Bertalanffy's assumption. Length distribution of the exceptionally large eels in relation to age was analysed ty comparing them with "normal" sedentary eesl and with silver migsating, and it is suggested that thew exceptionally large eesl consist of the anmals which probably the [apacity for growth wen alredy young.

Loci of Orebodies, the Bupyeong Silver Deposits (부평은광상(富平銀鑛床)의 광체배태장소(鑛體胚胎場所))

  • The geology of the Bupyeong mine area is consisted of Precambrian Gyeonggi gneiss complex and Mesozoic igneous rocks; i.e., pyroclastic rocks, intrusive breccia, granite and felsic porphyries which were formed during a Jurassic to early Cretaceous resurgent caldera evolution. Granites are not observed on the surface and in the underground of the mine. Bupyeong silver deposits occur as stockworks of base metal sulfides- minor silver minerals-quartz - carbonate veinlets, hosted by pyroclastic rocks and intrusive breccia at the southwestern margin of the caldera. Silver occurs mainly as native silver, and other silver minerals, minor in quantity, are argentite, tetrahedrite-freibergite, pyrargyrite, polybasite, canfieldite and dyscrasite. The average grade of silver ore is about 180g/t Ag. Discrimination of silver ore from the country rocks depends largely on the chemical analyses of rock samples taken every two meters from tunnels, diamond-drilling cores and mining stopes, because silver minerals are hardly observed in the ore by crude eye, and silver orebodies do not properly coincide with the concentrated zone of base metal sulfides which were precipitated at the earlier stage than the stage of precipitation of native silver. General characteristics of the loci of the silver orebodies are as follows; (1) The host rocks of orebodies are pyroclastic rocks and intrusive breccia. (2) Many of the orebodies are distributed around Gyeonggi gneiss complex. Especially where the paleotopography of gneiss complex shows a gradual slope, the basal stratigraphic horizon of the pyroclastic rocks unconformably overlying the gneiss complex offered a favorable loci of high grade ore. (3) $N5^{\circ}W$ to $N15^{\circ}$ E-striking faults played an important role in the localization of the orebodies. (4) Conduits of intrusive breccia within the gneiss complex, through which the intrusive breccia intruded into the upper pyroclastic rocks, exist beneath most of the main orebodies. This suggests that the conduits of intrusive breccia served as channelways for the migration of ore fluids.

An Electron Microscopic Radioautographic Study of the Synthesis and Migration of the Glycoproteins in the Osteoclast of the Mice Maxillary Alveolar Bone (생쥐 상악치조부에서의 파골세포의 당단백 합성 및 이동에 관한 전자현미경 자기방사법적 연구)

  • The pathway and time course of fucose-containing glycoprotein synthesis and intracellular translocation in osteoclasts of the mice maxillary alveolar bone were investigated by electron microscopic radioautography. Male Balb-C mice weighing 17gm were anesthetized with Nembutal and injected via the external jugular vein with 2.5 mCi of $L-[6-^{3}H]-fucose$ (specific activity 16.8 mCi/mmol) in 0.1 ml of sterile saline solution. At 5, 10, 20, 35 minutes and 8 hours after administration of the $^{3}H-fucose$, animals were killed by intracardiac perfusion of 30ml of 2% glutaraldehyde in a modified Tyroid solution, pH 7.4. The maxillae were then removed and further fixed in Karnovsky fixative for an additional 3-4 hours. After rinsing in 0.1M cacodylate buffer for 10 minutes, the maxillae were demineralized for 2 weeks at $4^{\circ}C$ in ethylene diamine tetra acetate containing 2% glutaraldehyde. The first interdental areas were mesiodistally sectioned into slices of 1mm thickness and postfixed in osmium tetroxide. Tissues were then dehydrated and embedded in Poly Bed. To prepare electron microscopic radioautography, the dipping method of Kopriwa (1973) was employed. Thin sections were coated with a crystalline monolayer of ILford $L_4$ photographic emulsion. After exposure for 4 months at $4^{\circ}C$, the sections were developed Kodak Microdol-X and Phenidon (for compact grains), fixed in 30% sodium thiosulfate, stained with uranyl acetate and lead citrate and examined in the electron microscope (JEOL 1200 EX). At 5, 10 and 20 minutes after injection, $^{3}H-fucose$ was concentrated in Golgi cisternae of the osteoblasts. By 35 minutes the labels were observed over small vesicles in the suprannclear area of osteoclasts. At 8 hours, numerous silver grains were located on the ruffled border and cell membrane of osteoclasts. These results indicate that fucose molecules are added in the Golgi apparatus and small vesicles appear to be responsible for translocation of the glycoproteins to the marginal portion of osteoblasts. The glycoproteins are distributed on the osteoclast cell surface and especially over the ruffled border.

A study on vertical distribution observation of giant jellyfish (Nemopilema nomurai) using acoustical and optical methods (음향 및 광학기법을 이용한 노무라입깃해파리 (Nemopilema nomurai)의 수층별 분포 관찰에 관한 연구)

  • A giant jellyfish (Nemopilema nomurai), which is presumed developing in the East China Sea, is recently one of major issues in the Northeast Asia region due to its fatal damage to the fishery. The biomass estimates have generally been conducted by trawl sampling and sighting survey methods. The biological research is also needed to clarify such environmental origin or diurnal migration patterns. While trawl sampling or sighting survey methods are effective to investigate its density estimates in its distributed community of near bottom or surface, they have a problem in investigation on the vertical distribution of jellyfishes. In this case, an echo sounding detection would have an advantage to survey it more extensively and effectively. This trial was conducted to observe the vertical distribution of giant jellyfish, where thermocline strongly formed, during mooring at each station of the East China Sea and southern coastal area using acoustical and optical methods. By the results, they were observed to exit and move at the water column under the thermocline using the optical camera and echo sounder system, and the information was analyzed to find out the acoustical sound scattering characteristics relatives to 120kHz frequency. These results can be utilized effectively to estimate the vertical distribution and biomass of Giant jellyfish with comparing results from trawl sampling and sighting survey methods, hereafter.
