단삼 등 천연물의 항암작용 (Anticancer Activity of Natural Products including Salvia miltiorrhiza)

  • The cellular growth inhibition of 20 natural products was screened using SRB (sulforhodamine B) assay against 4 human cancer cell lines(SNU-1, SNU-C$_{4}$, Hep3B, Kato III). Ethanol extracts of Salvia miltiorrhiza, Saussurea lappa and Chelidonium majus showed potent anticancer activity among them, and further, it was fractionated into methylene chloride, hexane and methanol. Methylene chloride and methanol fraction of Salvia radix showed significant inhibitory activity against 4 human cancer cell lines. The effect of Salvia miltiorrhiza on anticancer activity in vitro models was evaluated with methylene chloride fraction of Salvia miltiorrhiza. Life span of ICR mice implanted with sarcoma-180 was increased by 40-61% and BDF$^{1}$ mice implanted with L1210 was increased by 66-89% upon intraperitoneal administration with methylene chloride fraction of Salvia miltiorrhiza. Based on these result, we suggested that Salvia miltiorrhiza showed anticancer activity on the in vivo and in vitro models

누에 및 뽕나무 유래 물질의 인간(人間) 암세포주(癌細胞株)에 대한 세포독성(細胞毒性) (Cytotoxic Activity of Bombyx mori and Morus alba Derived Materials against Human Tumor Cell Lines)

  • 동결건조(凍結乾燥)한 누에 유래(4령유충(齡幼蟲), 암 수 번데기, 암 수 성충(成蟲)) 및 건조 뽕나무 유래 재료(잎, 오디, 상백피(桑白皮))의 메탄을 추출물, 백강잠(白彊蠶) 및 누에 4령유충(齡幼蟲) 잠분(蠶糞)의 메탄올 추출물의 5종 인간(人間) 암세포주(癌細胞株)(A549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS, HCT-15 colon tumor cell lines)에 대한 세포독성(細胞毒性)을 sulforhodamine B법을 이용하여 in vitro 검정하였다. 공시시료중 잠분(蠶糞)의 70% 메탄올 열탕추출물(熱湯抽出物)은 이들 암세포주(癌細胞株)에 대하여 강한 세포독성(細胞毒性)을 나타내었으나, 잠분(蠶糞)의 메탄올 추출물 및 오디와 상백피(桑白皮)의 메탄을 추출물은 중간 정도의 활성을 보였다. 기타 물질들은 이들 암세포주(癌細胞株)에 거의 독성을 보이지 않았다. 70% 메탄올 열탕추출물(熱湯抽出物)이 강한 세포독성(細胞毒性)을 나타내어, 용매 분획한 결과 클로로포름과 에틸아세테이트 획분이 암세포주(癌細胞株)에 대하여 가장 강한 세포독성(細胞毒性)을 보였다. 결론적으로, 잠분(蠶糞), 상백피(桑白皮) 및 오디의 항암활성(抗癌活性)은 이들의 약리작용의 일부를 설명할 수 있을 것으로 생각된다.

메주 메탄올 추출물 및 분획물의 항돌연변이 및 인체 암세포 성장 억제 효과 (Inhibitory Effect of Methanol Extracts and Solvent Fractions from Meju on Mutagenicity and Growth of Human Cancer Cells)

  • 메주의 헥산 및 메탄올 추출물과 그것을 용매로 분획하여 얻어진 디클로로메탄, 에틸아세테이트, 부탄올, 물 분획물들에 의한 SOS chromotest 실험계에서 항돌연변이 효과 및 SRB assay를 이용한 인체 암세포들의 성장 억제 효과에 대하여 검토하였다. SOS chromotest 실험계의 경우, 첨가농도 $100{\mu}g/assay$에서 메주의 디클로로메탄 및 에틸아세테이트 분획물은 각각 91%, 91%로 MNNG에 대하여 강한 항돌연변이 효과를 나타났다. AGS 인체 위암세포와 Hep 3B 인체 간암세포를 이용하여 항암효과를 실험한 결과, 메주 에틸아세테이트분획물은 첨가농도 $200{\mu}g/assay$에서 각각 93% 및 91%로 암세포 성장을 크게 저해시켰고 그 저해 효과는 디클롤메탄 분획물보다 더 높았다. 인체 위암세포인 AGS를 이용한 SRB assay을 이용한 억제 효과 실험에서 메주의 에틸아세테이트 분획물은 첨가농도 2 mg/assay에서 인체 위암 및 간암세포의 증식을 각각 64% 및 71%로 억제하여 분회물들 중 가장 높은 저해효과를 보였고 헥산 및 디클로로메탄 분획물은 그 다음으로 억제 효과가 높았다. 이러한 결과는 된장 메탄올 추출물과 그 분획물의 결과와 유사한 경향을 보였으므로 메주와 된장이 여러 종류의 미생물, 곰팡이류와 세균류들에 의해 발효과정을 거치는 동안 원재료인 콩에서는 없었던 혹은 함량이 적은 성분들이 생성되거나 증가되어 항암효과를 나타내는 것으로 추정되어 진다.

Streptomyces endus YP-1이 생산하는 항암활성 물질의 분리 및 정제

  • Sulforhodamine B (SRB) and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, RNA dot blot and Northern hybridization analysis were performed to screen microorganisms for the production of anticancer agent. Among microorganisms tested, strain YP-1 was selected for its cytotoxicity and ability to reduce the level of c-myc RNA. Strain YP-1 was identified as Streptomyces endus. The anticancer material produced by Streptomyces endus YP-1 was sequentially purified by solvent extraction, silica gel column chromatograpby, preparative TLC and preparative HPLC. The cancer material identified as azalomycin B by the instrumental analyses such as $^{1}$H-NMR, $^{13}$C-NMR, Mass, IR and UV absorption. It was colorless amorphous powder and its molecular weight was 1025.278. Azalomycin B, produced by Streptomyces endus YP-1, showed anticancer activity against several human cancer cell lines and reduction of c-Myc protein level in Colo320 DM cells which was determined by Western blot analysis.

Anti-Human Rhinovirus 1B Activity of Dexamethasone via GCR-Dependent Autophagy Activation

  • Objectives: Human rhinoviruses (HRVs) are the major cause of the common cold. Currently there is no registered, clinically effective, antiviral chemotherapeutic agent to treat diseases caused by HRVs. In this study, the antiviral activity of dexamethasone (DEX) against HRV1B was examined. Methods: The anti-HRV1B activity of DEX was assessed by sulforhodamine B assay in HeLa cells, and by RT-PCR in the lungs of HRV1B-infected mice. Histological evaluation of HRV1B-infected lungs was performed and a histological score was given. Anti-HRV1B activity of DEX via the glucocorticoid receptor (GCR)-dependent autophagy activation was assessed by blocking with chloroquine diphosphate salt or bafilomycin A1 treatment. Results: In HRV1B-infected HeLa cells, treatment with DEX in a dose-dependent manner, resulted in a cell viability of > 70% indicating that HRV1B viral replication was reduced by DEX treatment. HRV1B infected mice treated with DEX, had evidence of reduced inflammation and a moderate histological score. DEX treatment showed antiviral activity against HRV1B via GCR-dependent autophagy activation. Conclusion: This study demonstrated that DEX treatment showed anti-HRV1B activity via GCR-dependent autophagy activation in HeLa cells and HRV1B infected mice. Further investigation assessing the development of topical formulations may enable the development of improved DEX effectiveness.

Antitumor Evaluation of Cannabidiol and Its Derivatives by Colorimetric Methods

  • Cannabidiol derivatives (1, 2 and 3), 5-fluorouracil (4, 5-FU) and adriamycin (5, AM) were tested for their growth inhibitory effects against human tumor cell lines using two different 3-{4,5-dimeth-ylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and sulforhodamine B protein (SRB) assay. The light microscopic study showed morphological changes of the treated cells. Disruptions in cell organelles were determined by colorimetric methods; MTT assay and STB assay. These results suggest that cannabidiol (1, CBD) retains the most growth-inhibitory activity against human tumor cell lines.

포공령의 카드뮴에 대한 세포독성 억제효과 (The Inhibitory Effects of Taraxaci Herba against Cadmium induced Cytotoxicity)

  • This study was carried out to evaluate antitoxic effects Taraxaci Herba extract against Cadium by calorimetric methods. The antitoxic activity of Taraxaci Herba ex tract in NIH 3T3 fibroblasts was evaluated by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-phenyl-2H-tetrazoliumbromide), NR (Neutral red) and SRB (Sulforhodamine B protein) assay. The light microscopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows; The concentration of $10^{-2}mg/ml$ of Taraxaci Herba extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were antitoxic and tend to regenerate. These results suggest that Taraxaci Herba extract retains a potential antitoxic activity.

  • This study was carried out to evaluate antitoxic effects Palsun Brewing Water against cadmium by colorimetric methods. The antitoxic activity of Palsun Brewing Water in NIH-3T3 fibroblasts was evaluated by MTT ({3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-2H-tetrazoliumbromide} and SRB (sulforhodamine B protein) assays. The light microcopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows: The concentration of 10-2 mg/ml of Palsun Brewing Water was shown significant antitoxic activity against E. coli endotoxin and Salmonella endotoxin. The number of NIH-3T3 fibroblasts were antitoxin and tend to regenerate. These results suggest that Palsun Brewing Water retains a potential antitoxic activity.

인체암세포주에 대한 천연자원의 세포독성 검색 (I) (Cytotoxic Activities of Herbal Drugs against Human Cancer Cell Lines (I))

  • For the search of new antineoplastic agents from natural resources, two hudred and one kinds of oriental medicinal drugs were extracted with petroleum ether/ether(1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3 (human ovary adenocarcinoma) cell lines. Among them, thirty kinds of ether extracts, forty-one kinds of ethyl acetate extracts and nine kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml$. And also, twenty-four kinds of ether extracts, thirty-one kinds of ethyl acetate extracts and six kinds of methanol extracts showed significant cytotoxic activities against SK-OV-3 cell lines at the same concentration.

Screening for Inhibitor of c-myc Expression and Identification of Isolate No.2303

  • Sulforhodamine B(SRB) assay was performed on the human lung carcinoma, A549 cell line to screen soil microorganisms for production of anti-cancer agent. Among 4, 265 microorganisms, 45 isolates were selected for their cytotoxicity and tested for their effects on the expression of c-myc by RNA slot blot and Northern blot analysis resulting in selection of No.2303 isolate. This No.2303 was identified as Streptomyces sp. by ISP classification and the chemotaxonomic analysis method. NO.2303 inhibited the expression of cmyc in Col0320 DM and A549 cell lines. The culture extract of No. 2303 also inhibited the progression of the cell cycle of Go in NIH 313 cells, implying that the extract also inhibited the expression of c-myc in NIH 313 cell.
