Microbiologically influenced corrosion (MIC) of metal is common in the natural environment and sulfate reducing bacteria are representative microorganisms for MIC. We found that biofilm fomlation by SRB on the metal surface might be controlled by quorum sensing, which is a cell density dependent regulation of cell metabolism. As cell free culture fluids (spent media) of Desulfovibrio vulgaris and D. desulfuricans were tested for quontrn sensing related test strains, it was found that spent media of two SRB induced increased luminescence of Vibrio harveyi BB886 (sensor 1+, sensor 2-) and BB170 (sensor 1-, sensor 2+). Quorum activities of D. vulgaris and D. desulfuricans appeared to be parallel to growth patterns, i.e., it was low in the lag phase, highly increased in the exponential phase, and reached maximum in the stationary phase. Interestingly, however, luminescence of V. harveyi BB886 and BB170 induced by a unit cell mass of the SHB showed a maximal peak in the late lag phase. Hence, it was suspected that quorum sensing of these two SHB play unknown roles in shifting cells from dormant to growth stages.
In this study, we have developed a microbe-carrier that combined Desulfovibrio desulfuricans and zeolite for removal of Zn and As in contaminated seawater. Desulfovibrio desulfuricans, one of the sulfate-reducing bacteria (SRB) microorganism was exhibited stable growth characteristics in highly salted water and strong resistance to Zn and As contaminated seawater. Moreover, zeolites are one of the most useful carrier to remove heavy metals from wastewaters. The results showed that SRB immobilized zeolite carrier can enhance removal ratio of Zn and As. In addition, heavy metals tended to be better removed in medium at conditions of $37^{\circ}C$. In case of heavy metal concentration, they were effectively removed ranging from 50 to 100 ppm. These results show that SRB-zeolite carriers hold great potential to remove cationic heavy metal species from industrial wastewater in marine environment.
This study, collaborated Gifu University, Japan, was performed to analyze chemical pollutants and microorganism and to clarify the distribution of sulfate-reducing bacteria and their insolubilization of heavy metal ions in leachates sampled seasonally between 1994 and 1996 from Nanjido waste landfill site, sampled 4 times between 1995 and 1996 from Pusan and Daejeon waste landfill site, and sampled 1 time between 1992 and 1994 from Hokkaido, Nagoya, Osaka and Hukuoka waste landfill site in Japan. The results were as follows: 1. The temperatures of internal leachate and leachate effluent were 40$\circ$C and 30$\circ$C, respectively, and the pH values of both leachates were about 8.0 at Nanjido waste landfill site. The concentration of SO$_4^{-2}$ gradually increased with the degree of stabilization and that of NO$_3$-N was detected in a part of sampling sites at one and half years, and in all sampling sites at 3 years after completion of landfill. 2. The organic substances in leachate of Nanjido waste landfill site decreased with the degree of stabilization and they were very fluctuated with measuring point and time. The concentration of organic substance and heavy metals in internal leachate were higher than in leachate effluent and those of Cd, Hg, and Pb were lower than detection limit except a part of samples in 1996. 3. APCs in internal leachate and leachate effluent were not much different and the minimum of APCs in internal leachate and leachate effluent were $1.0\times 10^4$/ml and $4.0\times 10^1$/ml, respectively. 4. The maximums of SRBs in Nanjido, Pusan, and Daejeon waste landfill site were 9180 MPN/ml, 24000 MPN/ml, and 348 MPN/ml, respectively and the maximum of SRBs in Japan waste landfill site was 9300 MPN/ml. 5. During 2-week-SRB culture, the values of MPN were high at 50$\circ$C for initial culture period and at 30$\circ$C for last culture period. MPN started to appear at first day and rapidly increased between 7th day and 9th day. 6. Cadmium and copper were insolubilized by SRB within 6 hr and iron and zinc were done within 48 hr. The rates of insolubilization of Cd, Cu, Fe, Zn, T-Cr were 100%, 99.5%, 95.0%, 99.8%, 16.1% after 48 hr treatment with SRB, respectively.
토양 및 지하수 등의 혐기성환경에 서식하는 황산염환원박테리아의 활동으로 생성되는 맥키나와이트가 용존산소에 의해 산화 및 용해되는 특성을 관찰하였다. 오염지역에 산화지하수 유입에 의한 용존산소량 증가로 인해 안정화된 핵종들(예: 환원우라늄)이 산화 및 용해되는 상황에서 일반적으로 같이 공존하는 황화광물의 역할을 알아보고자 하였다. '디설퍼비브리오 디설프리칸스(Desulfovibrio desulfuricans)'라는 황산염환원박테리아가 만든 맥키나와이트를 '과산화수소수'와 '아질산나트륨'으로 산화시키면서 발생되는 광물 용해 현상을 약 2주 동안 관찰하였다. 산화제의 종류에 따라 시료의 광물학적 산화 및 용해 반응 특성은 달랐으나, ${\mu}m$ 크기의 황화광물 입자들에 의한 용존산소의 소모와 그에 따른 황산염 농도의 증가로 인해 산화수가 초기에 안정화되었다. 이와 같은 결과로부터 알 수 있는 사실은, 황산염환원미생물에 의해 다량 만들어지는 황화광물이 지하수의 산소를 소모시켜 환원 환경의 교란을 예방할 뿐만 아니라 버퍼물질로써 환원/침전된 핵종들의 장기 안정화에 상당한 기여를 할 것으로 예상된다.
In order to remove both nitrate and sulfate present in the concentrate of RO(reverse osmosis) process, a combined bio-regeneration and ion-exchange(IX) system was studied. For this purpose, both denitrifying bacteria(DNB) and sulfate reducing bacteria(SRB) were simultaneously cultivated in a bio-reactor under anaerobic conditions. When the IX column containing a nitrate-selective A520E resin was fully exhausted by nitrate and sulfate, the IX column was bio-regenerated by pumping the supernatant of the bio-reactor, which contains MLSS concentration of $125{\pm}25mg/L$, at the flowrate of 360 BV/hr. Even though the nitrate-selective A520E resin was used, the breakthrough curves of ionic species showed that sulfate was exhausted earlier than nitrate. The reason for this result is due to the fact that the concentration of sulfate in RO concentrate was 36 to 48 times higher than nitrate. The bio-reactor was successfully operated at a volumetric loading rate of 0.6 g $COD/l{\cdot}d$, nitrate-N loading rate of 0.13 g $NO_3{^-}-N/l{\cdot}d$, and sulfate loading rate of 0.08 g $SO_4{^{2-}}/l{\cdot}d$. The removal rate of SCOD, nitrate-N, sulfate was 90, 100, and 85%, respectively. When the virgin resin was fully exhausted and consecutively bio-regenerated for 2 days, 81% of nitrate and 93% of sulfate were reduced. When the virgin resin was repeatedly used up to 4 cycles of service and bio-regeneration, the ion-exchange capacity of bio-regenerated resin decreased to 95, 91, 88, and 81% of virgin resin.
황산염의 농도변화에 따른 연속 혐기성 수소 발효에 미치는 영향을 고찰하기 위해서 혼합균주를 사용한 완전 혼합형 반응조를 운전하였다. 기질은 글루코오스를 사용하였고, 수리학적 체류시간은 1, 0.5, 0.25 일로 각각 고정하였다. 황산염 농도는 0${\sim}$20,000 mg/L로 단계별 증가시켰고 pH 5.5로 운전하였다. 실험 결과 높은 황산염 농도에 관계없이 수소가 발생하였고, HRT 0.25일로 짧아짐에 따라 수소 발생이 높게 나타났다. HRT 1, 0.5, 0.25일 각 조건별 수소 생성량과 수소 수율은 2.9, 4.6, 9.4 L/day, 2.0, 1.8, 1.6 mol $H_2$/mol glucose로 나타났으며, 잔존 황산염 96${\sim}$98, 95${\sim}$97 94${\sim}$97%로 나타나 황산염 환원이 발생하지 않았다. FISH 결과 모든 조건에서 수소생성균의 분포는 나타났지만 황산염환원균의 분포는 나타나지 않았다.
폐광산 지역에서 발생하는 광산배수의 오염원소중 하나인 구리(Cu)는 낮은 농도에서도 독성을 지니고 있어 수계환경에 노출되기 전에 처리되어야 한다. 본 연구에서는 경남 고성군 S광산의 갱내수 정화시설의 Cu 제거효율을 개선하기 위해 석회석과 폐상퇴비를 혼합한 자연정화 기반의 반응조를 이용하여 약 9개월 동안 현장 파일럿실험을 수행하였다. 반응조별 유입수 대비 pH 증가량과 Cu 제거효율은 Successive Alkalinity Producing System (SAPS) > Reducing and Alkalinity Producing System (RAPS) > 석회석반응조 순으로 나타났다. SAPS조와 RAPS조에서는 석회석의 영향과 동시에 유기물의 분해로 인한 알칼리도의 부과로 석회석반응조보다 높은 pH 환경을 조성하였다. pH가 증가할수록 Cu 제거효율이 높아지는 결과를 통해 pH 상승이 Cu를 처리하는 주된 기작임을 확인할 수 있었다. 또한 황산염환원박테리아(Sulfate Reduction Bacteria, SRB)가 SAPS조에서 가장 많이 활성화 된 것을 확인 할 수 있어, 황산염환원반응도 Cu를 제거시키는 기작에 관여함을 판단할 수 있었다. 본 연구는 S광산에서 발생하는 광산배수의 특성에 알맞은 맞춤형 정화공정을 도출하기 위해 현장 그대로의 조건에서 실험을 수행한 것에 의의가 있으며, 향후 정화시설의 개선에 있어 공법 선정에 도움을 줄 수 있을 것이다.
골재채취가 수층의 세균군집구조에 미치는 영향을 알아보기 위해 낙동강 중류, 골재채취가 이루어 지지 않은 수역인 구미대교와 장기간에 걸쳐 연속적으로 골재채취가 행해지는 지역인 성주군 소학리 (성주대교 인근)에서 $\alpha\;\beta\;\gamma-subclass$ proteobacteria, Cytophaga-Flavobacterium (CF) group 세균군과 황산염화원세균을 FISH법으로 정량분석하였다. 아울러 수온, pH, EC, 엽록소-$\alpha$ 양 등의 이화학적인 환경요인도 측정하였다. DAPI로 염색된 세균수로 측정한 총세균수의 경우, 두 정점간의 괄목할 만한 양적인 차이는 없었으나 그 시간차에 따른 변동이 골재채취 수역에서 더욱 심했다. 또한 높은 엽록소-$\alpha$ 양을 나타낸 골재채취 수역에서 검출한 CF group 세균군과 황산염환원세균이 총세균수에서 차지하는 비율이 비골재채취 수역에 비해 높았다. 이같은 결과는 저니질이 수층으로 재유입되었기 때문으로 판단된다. 그러나 본 연구에서 검출한 세균군이 총세균수에서 차지하는 비율을 합하면 그 비율은 골재채취 수역보다 비골재채취 수역에서 더 높았다. FISH법에 의한 세균의 검출가능성이 해당세균의 물질대사능과 연관이 있으므로 골재채취 수역에서의 낮은 세균 검출율은 골재채취가 세균의 물질순환에서의 역기능을 초래하고 이는 또한 연차적으로 그들의 먹이연쇄에 부정적인 영향을 미칠 가능성을 시사하는 것이다.
In this study, estimation model for iron compound originated from upflow, anaerobic fixed bed reactor, which treats sewage domestic wastewater, was developed. The estimation model was formulated by a mathematical expression which was based on the mass balance. Below the HRT of 60 minute, sulfide concentration combining with iron $FeS_2$ is the highest because the maximum sulfate consumption rate $V_{maxS}$ and half-saturation constant of sulfate $K_{mS}$ exert an important effect on the estimation model as temperature was increased. But increment of $FeS_2$ concentration is weakened above the HRT of 60 minutes and represent the lowest value at the HRT of 108 minutes. It implies that liquid phase distribution ratio of sulfide ${\alpha}r$ becomes lower as temperature was increased. While phosphorus concentration combining with iron $Fe_3(PO_4)_3$ is increased as HRT and temperature are increased, which is affected by phosphorus removal rate constant $k_p$. As the result of estimating the iron concentrations of corrosion by the model, the concentration of iron corrosion is higher than any other at the HRT of 108 minute and $20^{\circ}C$. The predicted values were compared with measured ones at different HRT(13.5, 27, 54, 108 min) and temperature(20, 25, $30^{\circ}C$). The experimental data could be fitted with the simulated curves. Therefore, the mathematical expression could be applicable to design full-scale wastewater treatment plants.
