Characteristics of Sweet and Super Sweet Corn Seeds Shelled at Different Seed Moisture and Threshing Method Conditions (단옥수수와 초당옥수수 탈곡 시 종자 수분함량과 탈곡방법에 따른 종자 특성)

  • Characteristics of sweet (sugary, su) and super sweet (shrunken-2, sh2) corn seeds shelled by different threshing methods at different moisture content status were studied. Hybrid seeds of a su (Early Sunglow ${\times}$ Golden Cross Bantam 70, GCB 70) and a sh2 (Xtrasweet 82 ${\times}$Fortune) were dried to moisture content of 12, 15, 18, and 21%. Hand shelling did not give any mechanical damages to seeds, while an electrical corn thresher gave some visible mechanical damages. The emergence rate of hand shelled seeds was higher than that of machine shelled seeds by $6{\sim}14%$ for a su and by $9{\sim}18%$ for a sh2 hybrid depending on seed moisture contents in cold test. The optimum seed moisture content to reduce mechanical threshing damages and to improve seed quality was 15% for su and 12% for sh2 hybrid seeds. At the optimum seed moisture contents, germination rate at $25^{\circ}C$, emergence rate in the cold test and ${\alpha}-amylase$ activity were highest, while the percentage of damaged seeds and leakage of total sugars and electrolytes in soaking water were minimized.

Changes in Seed Vigour of Sweet and Super Sweet Corn Hybrids as Affected by Storage Conditions (단옥수수와 초당옥수수의 저장조건에 따른 종자 활력변화)

  • An experiment was conducted to characterize the seed vigour of sweet (su) and super sweet (sh2) corn seeds stored at different temperatures and relative humidities (RH). Hybrid seeds of Early Sunglow ${\times}$ Golden Cross Bantam 70 (su) and Xtrasweet 82 ${\times}$ Fortune (sh2) were stored at different temperatures ($5\;and\;15^{\circ}C$) and RH(70 and 85%) for 10 months. Results of the experiment show that seed deterioration of super sweet corn was much faster than that of sweet corn under all storage conditions. Germination rate of sweet corn seeds at $25^{\circ}C$ and emergence rate in cold test showed similar patterns. Emergence rate of super sweet corn in cold test was significantly lower than the germination rate at $25^{\circ}C$. Germination rate of both sweet and super sweet corns was positively correlated to the emergence rate in cold test, but the correlation coefficient of super sweet corn was much lower compared to the sweet corn. This implies that the viability of super sweet corn seeds should be tested in the cold test to estimate field emergence rate. Seeds of sweet corn could be stored for 5 months under all storage conditions without significant seed deterioration, while those of super sweet corn should be stored at low temperature and RH. The emergence rate of sweet corn in cold test was not correlated to the leakage of total sugars, electrolytes or ${\alpha}-amylase$ activity, while that of super sweet com was positively correlated to the ${\alpha}-amylase$ activity, negatively correlated to the leakage of electrolytes, and was not correlated to the leakage of total sugars.

Quality Characteristics of Kanjang Prepared with Meju Cultivated on Different Soybean Cultivars with Bacillus subtilis var. globigii Seed Culture (Bacillus subtilis var. globigii 종균접종 메주로 제조한 콩 품종별 간장의 품질 특성)

  • This study was carried out to select the most suitable soybean cultivars for the quality kanjang preparation. Six cultivars of soybean, Taekwangkong, Kumjungkong #1, Sinpaldalkong #2, Hwangkeumkong, Danbaegkong and Danyeobkong, were used as the raw materials for kanjang preparations. Grain type meju was prepared by the inoculation of Bacillus subtilis var. globigii seed culture on the cooked soybean of respective cultivar and cultivation at $30^{\circ}C$ for 10 days. Two month matured respective kanjang mash, which was prepared by mixing meju and 20% salt brine in the ratio of 1 : 3, was separated and the liquid portions were used as kanjang samples for this work. The highest crude protein content of 47.5% was obtained from Danbaegkong cultivar among six soybean cultivars tested. High sugar soybean cultivars were found to be Kumjungkong l and Danbaegkong and the content 15.87% and 13.33% respectively. The highest total nitrogen(TN) content of 1.18% was shown from Danbaegkong kanjang. Although the major free organic acid in kanjang was found to be succinic acid, no significant differences in free organic acid and sugars content were observed among cultivars. The highest free total amino acids(TA) and glutamic acid(GA) content in kanjang was observed to be 3365 mg% and 734.44 mg% respectively from Danbaegkong kanjang. Danbaegkong cultivar soybean which was shown to contain the highest crude protein was found to be the most suitable one for the preparation of quality kanjang with high TN, GA content and GA/TA ratio.

  • When a low-temperature treatment was given to a small sweet pepper variety Zairaisisi, the seed browning effect appeared soon. This change attracted the studies to determine and discuss the browning metabolites, polyphenolic compounds, and changes in their between-step components. (1) Chlorogenic acids were found as a polyphenolic compound in seed, whereas no flavanol-type polyphenol was observed. (2) There was sharp increase in total polyphenol content and chlorogenic acid with a low-temperature treatment. The contents of these substrates dropped below that of room-temperature treatment after the browning effect took place. (3) A marked increase in between-step metabolites phenylalanine, tyrosine, shikimic acid contents, and thus assumed activated shikimate pathway in this process. (4) It was suggested by determining the effect of specific metabolic inhibition and respiratory inhibitor administrations on enzymes that active biosynthesis of polyphenolic compounds takes place in shikimate pathway with combination of phosphoenolpyruvate and erythrose-4-phosphate connected to TCA cycle jaming after an active EMP pathway was gone through with sugars in pepper seeds at a low-temperature. (5) It was also suggested from the observation of increased K ion flow-out in pepper seeds with a low-temperature treatment that there is an abnormality in the plasma membrance.

Nutritional Composition of White-flowered and Pink-flowered Lotus in Different Parts (백련(白蓮)과 홍련(紅蓮)의 부위별 영양성분)

  • The nutritional compositions of different parts (roots, leaves, and seeds) of two lotus species (Nelumbo nucifera Gaertner), Muan's white-flowered and Naju's pink-flowered, are as follows; crude protein content in the seed was four times higher and the carbohydrate content was three times higher than those in the root and leaf. Comparing between the species, the crude protein content of the white lotus was the higher than that of the pink lotus, but the carbohydrate content was comparatively lower. The potassium content of the minerals in all samples was much higher than those in others. Additionally, the iron content of the root was much higher than those in the leaf and seed. In both species, the major free sugar in the root was sucrose; the main sugars in the leaf were fructose and alucose, and those in the seed were stachyose, raffinose, and sucrose. Among amino acids, glutamic acid showed the highest level in the leaf and seed, while aspartic acid was the highest in the root.

Effects of Gibbrellic Acid and Benzyl Adenine on Seed Germination and the Chemical Change of Maize(Zea mays) Endosperm (GA와 BA 처리농도(處理濃度)가 옥수수(Zea mays)의 발아(發芽)와 배유(胚乳)의 양분소장(養分消長)에 미치는 영향(影響))

  • The experiment was conducted to study on the germination of seeds, and chemical change in the endorsperm and embryo of maize seeds treated with gibbrellic acid(GA) 25, G A 50, bengyl adenine(BA) 100 and B A 200 ppm. Obtained results can be summerized as follows: 1. The germination rate in the initial stage of maize seed increased most with G A 25 ppm treated but decreased most with B A 200 ppm treatment amount other treated blocks. There is no significant difference between treatments in the middle and later stage of the seed germination. 2. The quantity of Q $CO_2$ was noticeably increased together in parallel with germination at each blocks, but such fact was found that the block of treatment has low Q $CO_2$ in comparison with control and in particular, has the tendency to take very low in B A 200 ppm. 3. The amount of reducing sugar and total sugars among embryo and endosperm are promptly increased at each blocks, but B A treatment shows low rate of increase in comparison with the non treatment and G A treatment. 4. The quantity of crude protein of seed during germination are increased at the part of embryo and decreased at the part of endosperm. Such degree is most prominent in the treatment with G A 25 ppm and changing degree of treatment with 200 ppm is lowest.

Optimum Water Potential, Temperature, and Duration for Priming of Rice Seeds

  • Experiments were carried out to find out the optimum water potential, temperature, and duration for the priming of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) for better germination at sub-optimal temperatures. Seeds were primed in 0, -0.2, -0.4, -0.6, -0.8, and -1.0 MPa PEG (polyethylene glycol) solutions at $25^{\circ}C$. The optimum water potential for seed priming, the highest water potential at which rice seeds did not germinate, was -0.6 MPa. To find out optimum priming temperature and duration rice seeds were primed in -0.6 MPa PEG solution and 0 MPa (water as a control) for various durations at 15 and $25^{\circ}C$ and the seeds were germinated at 17, 20, and $25^{\circ}C$. Considering germination rate and speed, the optimum priming time in water (0 MPa) was 4 days at 15$^{\circ}C$ and 1 day at $25^{\circ}C$, while 4 days was the optimum priming time in a -0.6 MPa PEG solution, regardless of the priming temperature. Priming reduced the actual time of germination, especially at sub-optimal temperatures. Priming did not affect germination rate in -0.6 MPa PEG solution at 15$^{\circ}C$, but overpriming reduced the final germination rate in water at 15$^{\circ}C$ and in -0.6 PEG solution at $25^{\circ}C$. Total sugars and $\alpha$-amylase activity induced during the seed priming were negatively correlated with the final germination rate and there was no noted relationship with the speed or uniformity of germination.

Comparison of the Chemical Compositions of Korean and Chinese Safflower (Carthamus tinctorious L.) Seed (한국산과 중국산 홍화종실의 화학적 성분 비교)

  • The chemical compositions of korean and Chinese safflower (Carthamus tinctorious L.) seed were compared in this study. The proximate compositions were 0.73 and 0.05% of moisture, 19.74 and 18.82% of crude protein, 15.47 and 14.61% of crude fat, 3.78 and 3.87% of crude ash, 14.53 and 10.46% of crude fiber, 46.49 and 52.23% of N-free extracts in the non-roasted safflower seed (NRS) and roasted safflower seed (RS), respectively. Crude fat contents in non-roasted chinese safflower seed (NRCS) and roasted chinese safflower seed (RCS) were 33.30 and 31.22%, which were higher than those of NRS and RS. Unsaturated fatty acid in NRS was 83.2% and 90.9% in NRCS. Linoleic acid was the most predominant fatty acid in NRS (74.0%) and NRCS (74.2%). Sucrose (216.5 mg/100 g) and raffinose (117.5 mg/100 g) were major free sugars in NRS, but sucrose, glucose, fructose and raffinose were in NRCS. Glutamic acid, aspatic acid and arginine were major in total amino acids. 24 kinds of free amino acid were detected in NRS and 11 kinds in RS. Total essential amino acid in NRS ($28.0\;{\mu}g/100\;g$) was higher than that in NRCS ($9.2\;{\mu}g/100\;g$). The organic acids in safflower seed were composed of formic acid, succinic acid, malic acid, oxalic acid and fumaric acid. The content of vitamin E $({\alpha}-tocopherol)$ in NRS and NRCS were 10.5 mg/100 g, 6.2 mg/100 g, NRCS and RCS were 12.8 mg/100 g, 9.4 mg/100 g, respectively. Total carotenoid content in NRCS was $452.0\;{\mu}g/100\;g$ and it was higher than in NRS. The major minerals of safflower seed were K, P, Ca, Mg.

Effect of Defatted Torreya nucifera Seed Extract on the Cross-linking of Advanced Glycation End Products to Collagen (비자 유박 추출물이 최종당화산물과 콜라겐 교차결합에 미치는 효과)

  • Advanced glycation end products (AGEs) are final products formed by glycation reaction between reducing sugars and proteins, lipids or nucleic acids. These AGEs are related to progress of skin aging. In this study, we evaluate anti-aging activity of Defatted Torreya nucifera seed extract (DTSE) through antioxidant, anti-glycation, anti-elastase and inhibitory and breaking activity on the cross-linking of AGEs to collagen assay. Results showed that DTSE contained polyphenols and flavonoids. The $IC_{50}$ values of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity were $16.4{\mu}g$ (Dried materials, DM)/mL and $16.7{\mu}g\;DM/mL$, respectively. DTSE also inhibited the formation of AGEs, elastase activity and cross-linking of AGEs to collagen as well as broke existing cross-linking of AGEs to collagen in a dose-dependent manner. Consequently, our findings suggest that DTSE could be useful as a cosmetic material with anti-aging activity.

Pharmacognostical Evaluation of Trachyspermum roxburghianum (DC) Craib Fruits

  • Sophisticated modern research tools for evaluation of medicinal plants are available but microscopic methods are one of the simplest and cheapest methods to establish the identity of the source materials. Pharmacognostical investigation of the dried, powdered and anatomical sections of the fruits of Trachyspermum roxburghianum (DC) Craib was carried out to determine its macro and microscopical characteristics along with its physical constants. Externally, the fruits, yellowish or greenish brown in colour are elongated, elliptical, slightly curved, prominently ridged and longitudinal. As seen in transectional views of the fruits from Trachyspermum roxburghianum, the mericarp has concave sides called commissural surfaces and a convex outer side called the dorsal surface. The mericarp has three primary ridges alternating with two secondary ridges on the dorsal side. On the commissural side, there are two primary ridges which are lateral in position and two secondary ridges in the commissural side. The seed is attached to the pericarp by a short stalk called a raphe. Circular, four-lobed calcium oxalate crystals are fairly abundant in the endosperm. Phytochemical studies revealed the presence of phenolic compounds, triterpenoids, proteins and sugars. The pharmacognostical profile of the fruits will assist in standardization for quality, purity and sample identification.
