  • A detailed evaluation of relative toxicity of fluoride to die 5th instar larvae of PM and NB$_4$D$_2$ races of silkworm and the effects of lethal, sublethal and prevailing levels of fluoride in groundwater on the growth, economic characters and fecundity of the silkworm, Bombyx mori (Lepidoptera: Bombycidae) were studied. The feeding of mulberry, Morus alba leaves treated with lethal and sublethal doses of fluoride to PM and NB$_4$D$_2$ races of silkworms from the beginning of the 5th instar to the end of the feeding period resulted in significant reduction in growth, single cocoon weight, single shell weight, silk index, average filament length, and fecundity when compared to controls. These manges were more pronounced on exposure to lethal dose than sublethal dose and in general, the changes induced by fluoride were more striking in NB$_4$D$_2$ than PM, indicating the greater resistance of PM to higher fluoride levels. Groundwater quality in sericulturally important villages of Karnataka and Andhra Pradesh States has been studied with special reference to the presence of fluoride. On exposure to dose of prevailing levels in the waters of sericulturally important areas, die changes observed on growth, economic characters and fecundity were slightly lower when compared to controls and the decrease was found to be insignificant (P$\geq$0.05). It was concluded that, though minute dose (4.0 ppm) of fluoride did not have any toxic impact, it is toxic at higher concentrations to silkworms.

  • Sodium arsenite ($NaAsO_2$) was injected to the rat subcutaneously for the study of the acute toxicity of arsenite on hepatocytes, and the effects of pretreatment of arsenite and glutathione on the lethalty of the arsenite treated rats. Arsenite treated rat hepatocytes showed vacuolated cytosol and shrinked nuclear and expanded perinuclear space and cytoplasmic membrane whirl. Rats pretreated with BSO (L-Buthionine-SR-Sulfoximine), less survived than arsenite treated alone. It means that glutathione acts as a protecting agent against the arsenite. Subcutaneous sublethal dose (10mg/kg body weight) treatment was showed the protecting activity to lethality of lethal dose (15mg/kg body weight) treated rat. 10mg/kg body weight sublethal dose effects appeared in six hours intervals of between treatments.

  • To determine the dose·survival and repair characteristics of the jejunal crypt cells, experimental study was carried out using total 70 mice. Single or split irradiations of 1,100 to 2,200 rad were delivered to whole bodies of $C_{57}$ BL mice, using a cesium 137 animal irradiator and those mice were sacrificed after 90 hours. The number of regenerating crypts per jejunal circumference was counted by a jejunal crypt cell assay technique and dose·response curve was measured. The results were as follows : 1. The average number of jejunal crypts per circumference in control group was 140. In a single irradiation group, the number of regenerated jejunal crypts was, 125, 56, 2 in each subgroup of 1,100 rad, 1,400 rad and 1,800 rad respectively. In split irraiation group, it was 105,44,2 in each subgroup of 1,400rad 1,800rad and 2,200rad respectively. 2. Mean lethal dose of mouse jejunal crypt cell was 167 and 169 rad respectively in a single and split irradiation. 3. Repair dose of sublethal damage was 280 rad. 4. Sublethal damage was completely repaired within 4 hours between the split dose of irradiation.

  • Despite concerns about the significant toxicity of copper pyrithione (CuPT) at environmental concentrations, effects of CuPT on benthic organisms have received little attention. Here, we analyzed the detrimental effects of CuPT at sublethal concentrations (1/50, 1/20, and 1/10 of the 96 h-LC50 value) for 14 days in the marine polychaete Perinereis aibuhitensis. Reduced burrowing activity and significantly decreased the acetylcholinesterase activity in response to relatively high concentrations of CuPT were identified as CuPT-triggered cholinergic inhibition. The lipid peroxidation marker, malondialdehyde levels were dose-dependently increased, whereas intracellular glutathione was depleted by relatively high concentrations. In the CuPT-treated polychaete, significant fluctuations in the enzymatic activities of the antioxidant defense system (catalase, superoxide dismutase, glutathione reductase, and glutathione peroxidase) were observed with significantly modulated glutathione 𝘚-transferase activity. These results indicate that even sublethal levels of CuPT would have detrimental effects on the health status of the marine polychaete.

  • Which sublethal cadmium pretreatment may prevent from lethal cadmium's killing mice and which cadmium pretreatment-induced protection against cadmium lethality to mice may be related with their hepatic glutathione contents were investigated. When cadmium chloride was subcutaneously injected to mice (ICR strain) at various doses, all mice died, which treated with cadmium at dose of 300 $\mu$moles/kg or more, and none died, which treated with cadmium at dose of 80 $\mu$moles/kg or less. Subcutaneous pretreatment of sublethal cadmium decreased sacrifice of mice which subsequently injected with lethal cadmium, with most effectiveness at pretreatment dose of cadmium of 40 $\mu$moles/kg b.w. and at 48 hours of interval between sublethal cadmium pretreatment and lethal cadmium treatment. Even if a great part of the cadmium-pretreated mice were sacrificed while treated with lethal cadmium, they survived longer than the non-pretreated mice. Sublethal cadmium pretreatment (40 $\mu$moles/kg b.w.) 48 hours before lethal cadmium treatment to mice didn't decrease hepatic glutathione contents of the survived mice, while decreases in the glutathione in livers were observed in the mice just after died. These results indicate that sublethal cadmium pretreatment-induced protection against cadmium lethality to mice may be related to their hepatic glutathione contents. • The pine wood nematode (PWN), Bursaphelenchus xylophilus is a well-known devastating pathogen of economic importance in the Republic of Korea and other countries. In the Republic of Korea, trunk injection of nematicides is the preferred method of control. In this study, the efficacy of 16 locally produced formulations of emamectin benzoate against the PWN are compared through determining their sublethal toxicities and reproduction inhibition potentials. Nematodes were treated with varying concentrations of the tested chemicals in multi-well culture plates, and rates of paralysis and mortality were determined after 24 h. Reproduction inhibition potential was tested by inoculating pre-treated nematodes onto Botrytis cinerea, and in pine twig cuttings. Despite the uniformity in the concentration of the active ingredient, efficacy was contrastingly different among formulations. The formulations evidently conformed to three distinct groups based on similarities in sublethal activity (group 1: LC95 of 0.00768-0.01443 mg/ ml; group 2: LC95 of 0.03202-0.07236 mg/ml, and group 3: LC95 of as high as 0.30643-0.40811 mg/ml). Nematode paralysis generally occurred at the application dose of 0.0134-0.1075 ㎍/ml, and there were significant differences in nematode paralysis rates among the products. Nematode reproduction was only evident at lower doses both on B. cinerea and pine twigs, albeit the variations among formulations. Group 1 formulations significantly reduced nematode reproduction even at a lower dose of 0.001075 ㎍/ml. The variations in efficacy might be attributed to differences in inert ingredients. Therefore, there is need to analyze the potential antagonistic effects of the large number of additives used in formulations.

  • The acute toxicity and sublethal effects of nitrite on the dark-banded rockfish, Sebastes inermis (mean body weight: $83.3{\pm}7.2$ g), were studied under static conditions for a period of 96 h. The acute toxicity of nitrite was at the 50% lethal concentration ($LC_{50}$) of 700 mg/L. The sublethal effects on selected hematological parameters of the dark-banded rockfish, such as its osmolality, hematocrit, cortisol, alanine aminotransferase (ALT), and aspartate aminotransferase (AST), were measured after 0, 6, 12, 24, 48, 72, and 96 h of exposure to 0, 50, 100, 200, 400, or 700 mg/L nitrite. Sublethal nitrite caused a progressive reduction in the hematocrit of the fish, depending on the nitrite concentration and the exposure period. Exposure to 100-700 mg/L nitrite for 96 h caused a reduction in the hematocrit and an increase in cortisol, ALT, and AST compared with the control levels. Abnormal ultrastructural changes in the gills and liver tissues were observed in fish exposed to 700 mg/L nitrite for up to 96 h compared with the control tissues. Ultrastructural changes included atrophic gill mitochondria and hepatocytes that developed smooth endoplasmic reticulum and atrophic mitochondria. Although no rockfish mortality occurred at 500 mg/L nitrite, all the hematological parameters examined responded adversely to a nitrite dose of 200 mg/L for 96 h. These results show that although the acute toxic concentration of nitrite for the dark-banded rockfish is > 700 mg/L, sublethal concentrations of nitrite also negatively affect its hematological parameters.

  • Effects of sublethal dose of imidacloprid on biological characteristics, such as longevity, fecundity and excretions from brown planthopper (BPH) and its repellency to imidacloprid were examined. Topical application of imidacloprid to BPH at the dose of $LD_{10}$ and $LD_{30}$ (These values were determined at 48 hours after topical application of imidacloprid) was remarkably increased the mortality from 3 days after application and showed over 90% mortality on 4 days after application. Topical application of imidacloprid at dose of $LD_{10}$ (16 days) showed no difference compared to the control (16.9 days) in the longevity, while application of $LD_{30}$ was reduced the longevity of BPH to 13.5 days. Application of sublethal dose of imidacloprid to BPH adults reduced the number of eggs laid per day and for adult stage, and the hatchability of their eggs. By root zone application at the dose of $LC_{30}$, 50% longevity and 40% fecundity was reduced respectively, but showed no effect on number of eggs laid per day. The day on which BPH laid 50 % of eggs in number were 7.6 days at the $LC_{10}$ and 5.9 days the of $LC_{30}$ respectively. Hatchabilities of the groups treated with imidacloprid were lower than that of the control. Topical application to BPH at the dose of $LD_{10}$ did not reduce the amount of excretes, but application at the dose of $LD_{30}$ was reduced 26.5% of excretion compared to the control. Leaf dipping into the solution of concentration of $LC_{10}$ and $LC_{30}$ was reduced 96.4% and 98.0% of excretion, respectively. And root zone treatment at the concentration of $LC_{10}$ and $LC_{30}$ reduced 79.5% and 96.6% of excretion, respectively. Repellency rates of BPH to imidacloprid were 20% regardless of the dose of application.

  • Abamectin offers great protection against Bursaphelenchus xylophilus, a well-known devastating pathogen of pine tree stands. Trunk injection of nematicides is currently the most preferred method of control. This study aimed to evaluate the potency of the commonly used formulations of abamectin against B. xylophilus. Twenty-one formulations of abamectin were evaluated by comparing their sublethal toxicities and reproduction inhibition potentials against B. xylophilus. Nematodes were treated with diluted formulation concentrations in multi-well culture plates. And, populations preexposed to pre-determined concentrations of the formulations were inoculated onto Botrytis cinerea culture, and in pine twig cuttings. Potency was contrastingly different among formulations, with LC95 of 0.00285 and 0.39462 mg/ml for the most, and the least potent formulation, respectively. Paralysis generally occurred at an application dose of 0.06 ㎍/ml or higher, and formulations with high sublethal toxicities caused significant paralysis levels at the tested doses, albeit the variations. Nematode reproduction was evident at lower doses of 0.00053-0.0006 ㎍/ml both on Botrytis cinerea and pine twigs, with significant variations among formulations. Thus, the study highlighted the inconsistencies in the potency of similar product formulations with the same active ingredient concentration against the target organism, and the need to analyze the potential antagonistic effects of the additives used in formulations.
