Objective: The adhesion capabilities of different types of self-ligating brackets were measured with respect to Streptococcus mutans and Streptococcus sobrinus. Methods: Five types of self-ligating brackets (Clippy-C; Mini Clippy; Clarity-SL; Speed; Damon 3) were used for the experiment group and composite resin brackets (Spirit-MB), metal brackets (Victory) and polycrystalline alumina brackets (Clarity) were used for the control group. In order to assess adhesion of bacteria to the brackets, the brackets were cultured for 3, 6 and 24 hours in media containing bacteria and 20% sucrose. Results: There was no statistic difference in adhesion amount of Streptococcus mutans and Streptococcus sobrinus according to the types of brackets. A total adhesion amount according to bracket type was different. An extended incubation time increased adhesion amount. Observation under scanning electron microscope showed that Streptococcus sobrinus adhered more to Clippy-C and Victory rather than to Clarity-SL. Conclusions: Clarity-SL, a self-ligating esthetic bracket was confirmed to show lower bacterial adhesion to cariogenic bacteria, Streptococcus mutans and Streptococcus sobrinus group than other self-ligating brackets or conventional brackets, which suggests that proper use of self-ligating esthetic brackets might even be better in preventing tooth surface decalcification.

Natural extracts were isolated from 20 kinds of medicinal plants that have been traditionally widely used in Korea to develop treatments for dental caries. Medicinal plants showing antibacterial activity against a cariogenic bacterium, Streptococcus sobrinus KCOM 1157 were investigated. The natural extracts of Glycyrrhiza uralensis, Sophora flavescens, Coptis Chinensis, Mentha arvensis, Pinus densiflora, Spirodela polyrhiza showed antimicrobial activities against S. sobrinus KCOM 1157. The extracts of Pinus densiflora, Sophora flavescens, Coptis Chinensis among these medical herbs showed significant antimicrobial activity. These results suggested that the natural products of Pinus densiflora, Sophora flavescens, Coptis Chinensis could be the potential source of anticariogenic agent against dental pathogens including S. sobrinus.
The purpose of this study was to develop Streptococcus sobrinus-specific qPCR primers based on the nucleotide sequence of the RNA polymerase ${\beta}-subunit$ gene (rpoB). The specificity of the primers was determined by conventional polymerase chain reaction (PCR) with 12 strains of S. sobrinus and 50 strains (50 species) of non-S. sobrinus bacteria. The sensitivity of the primers was determined by quantitative real-time PCR (qPCR) with serial dilutions of the purified genomic DNAs (40 ng to 4 fg) of S. sobrinus ATCC $33478^T$. The specificity data showed that the S. sobrinus-specific qPCR primers (RTSsob-F4/RTSsob-R4) detected only the genomic DNAs of S. sobrinus strains with a detection limit of up to 4 fg of S. sobrinus genomic DNA. Our results suggest that the RTSsob-F4/RTSsob-R4 primers are useful in detecting S. sobrinus with high sensitivity and specificity for epidemiological studies of dental caries..

As part of our screening of anticariogenic agents from medicinal plants, the ethanol extracts of 10 herbs widely used in the fork medicine were tested for the antimicrobial activity against a cariogenic bacterium Streptococcus sobrinus. The ethanol extracts of Dryopteris crassirhizoma showed antimicrobial activities against S. sobrinus. These results suggested that the extracts of Dryopteris crassirhizoma could be the potential source of antimicrobial agent against S. sobrinus.

Streptococcus mutans and Streptococcus sobrinus play important roles in dental caries. Coptis chinensis is a natural product with antimicrobial activity against enterobacteria; however, its effects on oral streptococci are still unknown. Therefore, the effects of C. chinensis on the growth and biofilm formation of the representative cariogenic bacteria S. mutans and S. sobrinus were investigated for the possible use of C. chinensis as an anticaries agent. The C. chinensis extract was diluted with sterile distilled water, and 0.1-2.5% of the extract was used in the experiment. The effects of the C. chinensis extract on the growth and glucan formation of S. mutans and S. sobrinus were measured by viable cell counting and spectrophotometry at 650 nm absorbance, respectively. Crystal violet staining was also carried out to confirm the C. chinensis extract's inhibitory effect on biofilm formation. The C. chinensis extract significantly inhibited the growth of S. mutans and S. sobrinus at concentrations of ≥ 0.3% as compared with the control group. The viable cell count of colonies decreased by 1.7-fold and 1.2-fold at 2.5% and 1.25%, respectively, compared with the control group. The biofilm formation of S. mutans and S. sobrinus was inhibited by > 20-fold at C. chinensis extract concentrations of ≥ 1.25% as compared with the control group. In summary, the C. chinensis extract inhibited the growth and biofilm and glucan formation of S. mutans and S. sobrinus. Therefore, C. chinensis might be a potential candidate for controlling dental caries.
This study was undertaken to develop species-specific forward and universal reverse PCR primers for the detection of Streptococcus sobrinus. These primers target the variable regions of the 16S ribosomal RNA coding gene (rDNA) and their specificity was tested against 10 strains of S. sobrinus strains and 20 different species of oral bacteria using serial dilutions of the purified genomic DNA of S. sobrinus ATCC $33478^T$. Our data show that species-specific amplicons were obtained from all the S. sobrinus strains tested but not from other species. Both direct and nested PCR could detect as little as 400 pg and 4 fg of genomic DNA from S. sobrinus ATCC $33478^T$, respectively. This result suggests that these PCR primers are highly specific and sensitive and applicable to the detection of S. sobrinus.
Plant extract has attracted considerable interest in oral disease therapy. The present study was performed to observe the antibacterial effect on cariogenic Streptococcus mutans GS5 and Streptococcus sobrinus 6715, and periodontopathic Actinobacillus actinomycetemcomitans Y4 of phytoncide from Chamaecyparis obtusa Sieb. et Zucc employing the measurement of optical density, viable cell counts, and antibiotic sensitivity. The results were as follows: 1. Minimum inhibitory concentration of the phytoncide for S. mutans, S. sobrinus, and A. actinomycetemcomitans was observed to be 0.5%, 1%, and 0.2%, respectively. 2. Minimum bactericidal concentration of the phytoncide for S. mutans, S. sobrinus, and A. actinomycetemcomitans was determined to be 0.5%, 2%, and 0.2%, respectively. 3. The bacteria exposed to the phytoncide become more sensitive to antibiotics. The phytoncide enhanced significantly antibacterial activity of ampicillin against S. mutans and S. sobrinus. It also increased significantly the activity of penicillin and amoxicillin against S. sobrinus. In contrast, the phytoncide augmented the activity of amoxicillin and cefotaxime against A. actinomycetemcomitans but the increase was not statistically significant. The overall results indicate that phytoncide from Chamaecyparis obtusa Sieb. et Zucc used for this study has a strong antibacterial activity against cariogenic and periodontopathic bacteria and that it also has permeabilizing effect on certain antibiotics against these bacteria. Therefore, the phytoncide may be used as a candidate for prevention and therapeutic agent against oral infectious disease including dental caries and periodontal disease.
Acceplor reaction of glucosyltr~ansferase of Streptococcus ,SO~~-~IZLIS with f ~ ~ t o ~ y l o l i g o ~ a ~ ~ h a r i d e ~ was studied for the biosynthesis of novel olgisaccharides. First, bacilracin resistant mutants were selected by mutagenesis of Streptococcus sobrimis ATCC27351. Among these mutants 4 strains were selected by resistance to bacitracin and increase of glucosyltransferase. Acceptor reaction of maltose was analyzed by TLC and image analysis. There were differences in the specificity of the acceptor reaclion by Ule glucosylumsferase between mother strain (Streptococcus sobrinus ATCC2735) and bacitracin resistant mutants (Streptococcus sobrinus BR24C, Strepfococcus sobrinus CH-5). Molher strain did ilot show an acceptor reaction with fructosyloligosaccharides such as 1-keqtose and nystose. Acceptor reaction products of turailose and 1-kestose with glucosyltransferase (GW-S) of Streptococcus sobrini~s BR24C were TEX>\6^{3}$-$\alpha$-D-glucopyranosyl \3^{2}$-O-$\alpha$-D-fructose (glucose-fructose-glucose) and \6^{4}$-$\alpha$-D-glucopyranosyl \1^{3}$-$\beta$-D-~-h~ctofuranos~~ sucrose (glucose-glucosefructose- fructose). respectively These are novel oligosaccharides which can be produced only by enzymatic reaction.

Mutans streptococci have been reported to be implicated in dental caries. Of these streptococcal species, Streptcoccus mutans and Streptococcus sobrinus are most commonly found in human dental caries. Prevalence of these bacterial species in dental caries is found to be varied in different races and countries. Therefore, importance of these bacteria in dental caries remains to be determined The present study was performed to find out correlation S. mutans and S. sobrinus with dental caries in 125 Korean children with mixed dentition between 6 to 11 years of age. They were classified as group A(6-8 years) and group B(9-11 years) by age. For the study, stimulated saliva samples were collected from each subject. The vials containing saliva specimens were serially diluted (1:10) in saline and plated in duplicate on tryptone-yeast extract-cysteine with sucrose and bacitracin (TYCSB) for S. mutans and S. sobrinus. After genomic DNA was extracted from the samples, polymerase chain reaction (PCR) amplification was performed for identification using universal primers and specific primers to S. mutans and S. sobrinus. Data of microbial variables were compared to caries status of the subjects. According to this study, the result were as follows : 1. S. mutans versus S. sobrinus were moderate positive linear correlated in both group A(r=0.70) and group B(r=0.50). 2. Between S. mutans and dental caries there were weak positive linear correlation in both group A(r= 0.25) and group B(r=0.34). 3. S. sobrinus versus dental caries were not correlated in group A but slightly correlated in group B(r=0.21). 4. Between S. mutans and age, there were not correlation in both group. 5. S. sobrinus versus age were weak correlated in group A(r=0.32) but not correlated in group B.

The aim of this study was to determine the efficacy of a 3 tone plaque disclosing gel in assessing the risk of caries related to the population of Streptococcus mutans, Streptococcus sobrinus, and Lactobacillus spp. quantified using a quantitative real-time polymerase chain reaction (qRT-PCR). 15 healthy children of ages 9 - 12 years were randomly examined. The 3 tone plaque disclosing gel was applied on teeth surfaces, which changed the color to pink or red, blue or purple and light blue. Plaque was divided into 3 groups based on staining. Genomic DNA from each sample was subjected to a qRT-PCR assay for quantitative detection of target bacteria. The Kruskal-Wallis test was conducted for correlation between the color of plaque and the number of bacterial species. The levels of S. mutans, S. sobrinus, and Lactobacillus spp. were significantly different in the plaque samples of the 3 groups (p < 0.05). The proportion of S. sobrinus to S. mutans showed correlation to the color of plaque. The different color-dyed plaque was related to the number of acidogenic bacteria. The 3 tone plaque disclosing gel could be used as one of the indicators to assess the clinical risk of caries associated with the population of S. mutans, S. sobrinus, and Lactobacillus spp.
