A Study on the Microbiological Quality of Drinking Water and Changes During Storage

  • To assess possible risks from the consumption of drinking water from various sources, a survey of the microbiological quality of tap water, commercial bottled drinking water which is exploited from natural mineral water, and natural spring water was conducted. A total of 4 different brands of commercial bottled drinking water, and 4 types of spring water from different sources, and tap water from 4 private houses were tested for four index microorganisms, and the microbial quality changes of the water during the storage at room temperature or refrigerated temperature for 7 days. Aerobic plate counts of all of the initial water samples were still within 100 CFU/ml (drinking water standard of Korea). Total coliforms, fecal coliforms, and E. coli were not detected in all of the water samples at initial. However, aerobic plate counts of three types of spring water and three types of bottled drinking water stored at room temperature showed higher levels than the standards in 5 days. Total coliforms were detected in three types of spring water after one day's storage at room temperature, and in one type of bottled drinking water after 5 days' storage. These results indicate that some of the spring water surveyed are not safe to drink, and the spring water and bottled drinking water after opening the lid should not be stored at room temperature, if they are used for drinking.

  • The stability of ceftriaxone sodium 100 mg/ml in the water for injection was tested at two temperature conditions (refrigerator and room temperature) and two storage containers (vitro and syringe). The stability of each sample was determined by high-performance liquid chromatography while storing for 5 days at room temperature and 27 days at refrigerator. The concentration of ceftriaxone sodium was 100 mg/ml which the hospital usually use as a filling preparation for I.V. push or Y-site injection. There was no significant difference between the storage in vials and syringes. There was no significant difference in the change of concentration until storing the reconstituted ceftriaxone sodium injection for 2 days at room temperature and 14 days at refrigerator. In conclusion, the ceftriaxone sodium 100 mg/ml in the water for injection can be stored in vial or syringe for 2 days at room temperature and 14 days at refrigerator after preparation.

수확시기와 저장방법에 따른 마의 塊根重과 품질변화 (Tuber Yield and Characteristics of Chinese Yam as Affected by Harvesting Date and Storage Condition)

  • To give some information on the harvest and storage of Chinese yam (Dioscorea opposita), the experiment was done to examine the effect of its harvesting date and storage method on tuber weight, tuberrelated characters during the storageor at the end of storage. The yam tubers harvested 4 times at 20 days interval from Oct. 15 were stored with different storage methods(pit, room and cold temperature) and on Mar.15. all the characters were measured. Fresh weight of the tuber harvestedafter Nov. 5 and stored were greater than that done on Oct. 15 while dry tuber weight did not have any difference between the harvesting dates. The fresh weigth done directly from the field on Mar. 15 was higher compared to the other harvesting dates owing to its higher moisture content. The rate of sound tuber stored at room or cold temperature condition was higher than that at the field at which the rate was greater at the harvest after late nov. compared to the earlier harvests. Although the two storages were similar at sound tuber rates and marketable yield, the tuber weight was more reduced when stored at room temperature than when done at cold temperature . Brightness and value 'b' of chromaticaity realted to the tuber coloring and sugar content were higher but moisture content was lowerwhen harvested on Oct. 15 than the other havesting dates. All the characteristics realted to the marketability after storage were nearly same when stored at room and cold temperature. Value 'a' of chromaticity and sugar content were higher when harvest on Mar. 15 than when harvested before Dec. 15 but the 'b' value and mositure content were reverse results.

Storage of laboratory animal blood samples causes hemorheological alterations : Inter-species differences and the effects of duration and temperature

  • Hemorheological results may be influenced by the time between blood sampling and measurement, and storage conditions (e.g., temperature, time) during sample delivery between laboratories may further affect the resulting data. This study examined possible hemorheological alterations subsequent to storage of rat and dog blood at room temperature ($22^{\circ}C$) or with cooling ($4{\sim}10^{\circ}C$) for 2, 4, 6, 24, 48 and 72 hours. Measured hemorheological parameters included hematological indices, RBC aggregation and RBC deformability. Our results indicate that marked changes of RBC deformability and of RBC aggregation in whole blood can occur during storage, especially for samples stored at room temperature. The patterns of deformability and aggregation changes at room temperature are complex and species specific, whereas those for storage at the lower temperature range are much less complicated. For room temperature storage, it thus seems logical to suggest measuring rat and dog cell deformability within 6 hours; aggregation should be measured immediately for rat blood or within 6 hours for dog blood. Storage at lower temperatures allows measuring EI up to 72 hours after sampling, while aggregation must be measured immediately, or if willing to accept a constant decrease, over 24~72 hours.

황색종 잎담배 공동건조장의 환경개선

  • The Joint curing barns for flue-cured tobacco is a favorite with the tobacco farmers in Korea. However, most of farmer utilizing the joint curing barns indicated many problems such as high temperature and noise in the working room and a dry of cured leaves in the storage room. A structure of Joint curing barns has been modified to meet the needs of tobacco farmers. Compared with the unimproved joint curing barns, the improved one showed that the noise of workshop decreased about 7.7~10.8db, the amount of $CO_2$ decreased 40~50ppm in a working room and 80~100ppm in a machine room. Ammonia gas decreased 0.29mg/㎥ and the temperature of a working room dropped about 2.1~3.5$^{\circ}C$. The amount of air flow in a working room increased 23.2% at a site being 2m away from the entrance and 30.8% at a center. The inner temperature of the improved storage room showed that maximum temperature dropped about 3$^{\circ}C$, minimum temperature was high about 2$^{\circ}C$. The highest relative humidity was low 6%, the lowest one increased high about 10% when compared with the unimproved joint curing barns.

양파의 저장 온도 및 습도에 따른 품질변화 (Quality Changes Based on Storage Temperature and Humidify of Onion)

  • 양파의 저장조건을 저온(2-4$^{\circ}C$, 80% RH), 움식(3-15$^{\circ}C$, 75-85% RH), 실온방열(10-23$^{\circ}C$, 75-98% RH) 및 상온(2-$25^{\circ}C$, 62-72% RH)으로 구분하여 저장 중 물리적 및 화학적 변화를 조사하였다. 저장 중 양파의 발아율은 저온 저장된 양파를 제외하고는 모두 높게 나타났으며, 저온 저장된 양파에서는 저장 11개월까지 거의 발아되지 않았다. 양파의 부패율은 실온방열 저장된 양파를 제외하고는 7개월까지 낮았으나 7개월 이후에는 모두 급격히 부패되었다. 중량감소는 기간이 경과함에 따라 계속되었으며, 저온 저장된 양파에서 가장 적게 일어났다 수분함량은 저온 및 움식 저장된 양파에서 증가하였으나 실온방열 및 상온에서 저장된 양파에서는 감소하였다. 전당과 환원당의 함량은 시간의 경과와 함께 완만하게 감소하였다. Vitamin C는 저장 중 큰 변화가 없었으나 저온 저장된 양파의 경우에는 다소 증가하였다.

시금치 포장 및 보관온도가 품질보존에 미치는 영향 (Effect of Polypropylene Film Package and Storage Temperature on the Shelf-life Extension of Spinach (Spinacia oleracea L.))

  • 신선 엽채류인 시금치의 수확 후 포장, 저장유통온도별 품질보존 및 상품성에 미치는 영향을 조사하였다. 저장 중 중량의 변화에서 포장 후 저장 시는 중량감소가 거의 없었으나, 무포장은 저장기간이 길어질수록 그리고 저장온도가 높을수록 감소율이 현저하였다. 비타민 C함량은 저장 초기에 감소가 컸으며, 저장온도가 낮을수록 감소가 적었고, 비닐포장 간에는 큰 차이가 없었다. 시금치에서 Hunter's value는 저장온도가 높을수록 b값이 높아지는 경향이었다. 시금치 저장 중 외관 신선도에서 polypropylene 비닐포장 후 저온$(1{\sim}3^{\circ}C)$ 저장은 최고 34일, 상온$(10{\sim}15^{\circ}C)$ 저장은 8일까지 상품성이 유지되었으나, 무포장 상온보관은 3일 밖에 유지되지 않았다.

주요 식용버섯 가공원료의 상온 및 저온 저장에 따른 항산화 활성 변화 (Changes in antioxidant activity of processed edible mushrooms stored at room temperature and low temperature)

  • 본 연구에서는 국내에서 소비도 및 인지도가 높은 식용버섯인 큰느타리(Pleurotus eryngii), 느타리(Pleurotus ostreatus), 표고(Lentinula edodes), 팽이(Flammulina velutipes)의 열풍건조 및 로스팅 시료를 대상으로 상온(20-25℃) 및 저온저장(4℃) 3개월 후의 시료와 저장 전의 시료와의 항산화능, 아질산염 소거능 및 베타글루칸 성분변화를 알아보기 위하여 실험을 수행하였다. DPPH 라디칼 소거능은 큰느타리와 표고의 열풍건조 시료를 제외하고 다른 열풍건조 및 로스팅 시료의 상온저장 처리가 가장 낮은 소거능을 나타냈다. 총 폴리페놀 함량은 큰느타리와 표고 열풍건조 시료를 제외하고 상온저장 시 함량치가 가장 낮았으며, 모든 식용 버섯류의 로스팅 시료에서 저장 전에 비하여 상온 및 저온저장 시에 함량이 감소하였다. 철환원 항산화능은 표고의 로스팅 시료를 제외하고 다른 버섯류의 열풍건조 및 로스팅 시료에서 저장 전에 비하여 상온 및 저온저장 시료에서 높아지는 경향이었으며, 환원력은 열풍건조 및 로스팅 시료의 상온 및 저온저장 시에 저장 전에 비하여 증가하는 경향을 나타냈다. 베타글루칸 함량의 경우, 열풍건조 및 로스팅 시료의 상온 저장된 시료에서 가장 낮은 함량치를 보였다. 본 연구의 결과는 식용버섯 가공원료의 상온 또는 저온저장 시에 버섯에 함유되어 있는 항산화, 베타글루칸 등과 같은 유용한 생리활성 성분의 변화 정도를 예측할 수 있는 기초적 자료로서 그 활용도가 높으리라 기대된다.

약용작물 고수 종자의 형태학적 특성 및 발아조건에 관한 연구 (Seeds Characteristics and Germination of Coriandrum sativum L. on Several Storage Conditions)

  • This study was conducted to obtain the basic data related to seed characteristics and germination conditions of the Coriandrum sativum L. at different storage condition and temperature. The shape of fruit was oval with light brown color. Fruit was mericarp, biloculate, with one ovule in each locus. The length and width of seed were $1.37{\pm}0.067mm$ and $0.52{\pm}0.039mm$, respectively. Weight of 1,000 seeds was $6.55{\pm}0.15g$. Seeds which were stored at room ($15^{\circ}C$) and cold temperature ($4^{\circ}C$) in vinyl container showed the highest germination rate (93.3%) under the room temperature germination condition. Percentage of germination in cold and freeze temperature was 20% and 0%, respectively. Germination rate of room storage seeds with paper container increased to about 91.3%, however, decreased in cold ($4^{\circ}C$) and freeze storage ($-20^{\circ}C$) with paper container (76.7% and 78.0%, respectively). Germination rate of seed in vinyl stock container was better than that of paper stock container. Germination rate of seeds stored at room temperature for 4 years (2010 - 2013) ranged from 80.0% to 91.3%. Therefore, coriander seeds are expected to be stored at room temperature for increasing the germination rate and keeping a long term.

상온보관에 따른 권연벌레의 번식이 한약재 천궁, 당귀의 품질에 미치는 영향 (Effect of the Establishment of Cigarette Beetle Population on the Quality of Stored Cnidium Rhizome and Angelica Radix at Room Temperature)

  • Background: The current regulations regarding the storage of medicinal herbs do not define the storage temperature; instead, but the only requirement is that the plastic bags used for storing the medicinal herbs should be tightly sealed during storage. The practice of storing medicinal herbs at room temperature ($28^{\circ}C$), causes many problems, including growth of insect pests on the stored medicinal herbs. Thus, it is necessary to understand the effect of insect pests on the quality of medicinal herbs stored at room temperature ($28^{\circ}C$) for the improving the relevant regulations. Methods and Results: Cnidium rhizome and Angelica radix were infested with Lasioderma serricorne F. adults and incubated at $28^{\circ}C$ for 2.5 and 5 months. The population of L. serricorne was established rapidly, and left many holes, cadavers, and feces on the stored medicinal herbs, thereby greatly damaging the product appearance and hygiene. In addition, active ingredients of the medicinal herbs were significantly decreased, probably reducing their medicinal quality as well. Conclusions: These results indicate that the current regulations are not sufficient to prevent the establishment of storage insect pests and guarantee the quality and hygiene of stored medicinal herbs. Therefore, it is necessary to devise proper storage protocols and upgrade the current relevant regulations to maintain the quality of medicinal herbs during storage.
