Porphyromonas gingivalis에 대한 오리나무 줄기 추출물의 항균활성 및 생물막 형성 억제 효과 (Antibacterial and Antibiofilm Activities of Alnus japonica Stem Extract against Porphyromonas gingivalis)

  • 본 연구에서는 천연물유래 구강건강소재로써 염료식물의 이용 가능성을 알아보기 위해 오리나무, 옻나무, 치자나무, 황칠나무, 황벽나무, 회화나무 줄기 추출물의 구강미생물에 대한 항균활성을 조사하였다. 6 종의 국내 자생 염료식물의 줄기에서 추출한 에탄올 추출물 중 오리나무 줄기 추출물(1 mg/disc)이 치주질환 원인균인 P. gingivalis KCTC5352에 대해 가장 우수한 항균활성을 나타내었다. 오리나무 줄기 추출물은 P. gingivalis KCTC5352에 대해 양성대조구로 사용한 chlorhexidine과 유사한 항균활성을 나타내었으며 P. gingivalis KCTC5352의 MIC는 0.4 mg/ml였고 MBC는 0.6 mg/ml였다. 오리나무 줄기 추출물이 0.2-2.0 mg/ml 농도로 처리된 배양액에서 P. gingivalis KCTC5352의 바이오필름 생성율과 세균 생육은 추출물의 농도가 증가할수록 저해되는 경향을 보였다. 또한 P. gingivalis KCTC5352의 superoxide dismutase (SOD)와 섬모에 대한 mRNA 발현도 추출물의 농도가 높아질수록 감소하는 것을 확인할 수 있었다. 이상의 결과를 종합하면 오리나무 줄기 추출물은 치주질환 원인균인 P. gingivalis KCTC5352에 대한 항균활성과 생물막 생성 억제능이 우수하기 때문에 천연물유래 구강건강소재로써 이용 가능성이 높을 것으로 판단된다.

담쟁이덩굴 추출물과 분획물의 항산화, 항당뇨 및 항염증 효과 (Antioxidant, Antidiabetic, and Anti-Inflammatory Effects of Extracts and Fractions from Parthenocissus tricuspidata Stems)

  • 본 연구에서는 담쟁이덩굴 추출물이 항산화, 항당뇨 및 항염증 효과에 미치는 영향을 조사하고자 수행하였다. 담쟁이덩굴 열수 및 에탄올 추출물의 총 페놀함량은 각각 61.5 mg TAE/g과 122.1 mg TAE/g으로 에탄올 추출물에서 훨씬 높게 나타났다. 에탄올 추출물과 부탄올 분획물의 DPPH 라디칼 소거활성은 1 mg/ml에서 95%와 92%의 radical 소거능을 나타내어 positive control로 사용한 비타민 C와 유사한 항산화력을 보였다. SOD 활성은 에탄올 추출물과 부탄올 분획물 1 mg/ml 농도에서 91%, 97%로 높은 활성을 나타내었다. DPPH radical 소거능과 SOD 활성을 측정한 결과 항산화 활성은 모두 농도 의존적으로 증가하였으며 특히 열수추출물과 비교할 때 에탄올 추출물과 부탄올 분획물에서 높은 항산화 효과를 보였다. 담쟁이덩굴 추출물의 ${\alpha}$-glucosidase 활성억제 효과는 에탄올 추출물과 핵산 및 부탄올 분획층에서 높은 항당뇨 효과를 나타내었다. 이러한 결과는 지금까지 항당뇨 약물로 사용된 것보다도 높은 것으로 나타났다. LPS에 의하여 유도된 NO 합성은 1 mg/ml 농도의 담쟁이추출물을 처리함으로써 NO 합성이 약 40%로 감소하였다. 이러한 결과들은 담쟁이 추출물의 우수한 항산화, 항당뇨, 항염 효과를 시사하고 있으며 특히, 천연 항당뇨 기능성 소재로서 담쟁이 추출물의 활용 가능성이 높을 것으로 사료된다.

  • 목적: 상용화 된 superparamagnetic iron oxide (SPIO) nanoparticles과 transfection agent (TA)의 최적의 병용 용량을 알아보고자 하였다. 대상과 방법: Protamine sulfate (Pro), poly-L-lysin (PLL)과 ferumoxide, ferucarbotran을 다양한 농도에서 인체 중간엽 줄기세포에서 배양하여 세포 생존능을 알아보았다. 세포 철 섭취율은 정성적으로, 정량적으로 분석하였다. 결과: Ferumoxide 처리군의 생존능과 철 섭취율은 ferucarbotrn 처리군보다 통계적으로 의미있게 높았다 (p < 0.05). T2 이완시간은 ferumoxide 처리군에서 짧았다 (p < 0.05). 25 ${\mu}g$/ml ferumoxide와 3.0 ${\mu}g$/ml Pro 또는 PLL 병용군이 최적의 조건이었다. 결론: Ferumoxide 처리군의 세포 생존능과 철 섭취율은 ferucarbotrn 처리군보다 높았다. 25 ${\mu}g$/ml ferumoxide와 3.0 ${\mu}g$/ml TA는 줄기세포 표지에 적합하다.

In vitro antifilarial potential of the leaves and stem extract of Pluchea lanceolata on the cattle filarial parasite Setaria cervi

  • The effect of alcohol and aqueous extracts of the leaves and stem of Pluchea lanceolata on the spontaneous movements of both the whole worm and the nerve muscle preparation of Setaria cervi, and on the survival of microfilariae in vitro was studied. Alcohol and aqueous extracts of the leaves and stem of P. lanceolata caused the inhibition of spontaneous movements of the whole worm and the nerve muscle preparation of S. cervi, characterized by short lasting small increase in tone and amplitude of contractions followed by paralysis. The concentrations required to inhibit the movements of whole worm and nerve muscle preparations for alcohol extract were 200 and $25\;{\mu}g/ml$, and for aqueous extract were 250 and $100\;{\mu}g/ml$, respectively, suggesting a cuticular permeability barrier. Both the extracts (alcohol and aqueous) caused death of microfilariae in vitro, $LC_{50}$ and $LC_{90}$ being 12 and 18 ng/ml for alcohol extract and 25 and 40 ng/ml for aqueous extract, respectively.

Proliferation, Apoptosis, and Telomerase Activity in Human Cord Blood CD34+ Cells Cultured with Combinations of Various Cytokines

  • Umbilical cord blood (UCB), a rich source of hematopoietic stem/progenitor cells, has been proposed as an alternative to bone marrow and peripheral blood for transplantation treatment. Ex vivo expansion of cord blood stem cells could make the use of cord blood transplant feasible even for adult patients. However, the optimal cytokine cocktail for expansion of stem cells is yet to be established. This study compares proliferation, apoptosis, and telomerase activities in human cord blood stem cells cultured ex vivo with FLT3 ligand (FL)/thrombopoietin (TPO) or FL/TPO/stem cell factor (SCF), with a view to determine optimal combination of cytokines. CD34+ cells were cultured in DMEM containing either FL (50 ng/ml) and TPO (10 ng/ml) (FT group) or FL (50 ng/ml), TPO (10 ng/ml) and SCF (50 ng/ml) (FTS group). The cell proliferation rate was ten times higher in the FTS group. Although cells cultured with the two different combinations of cytokines were maintained for a long term (up to 8 weeks), a large number of cells underwent differentiation during this period. Cells cultured in FTS displayed lower levels of apoptosis compared to those of the FT group during the Initial 7 days of culture. The CD34+ fraction in both groups was markedly decreased to $21-30\%$ , and only $5-6\%$ was detected at 14 days of culture. Telomerase activity detected in human CD34+ cord blood at low levels was upregulated during the early phase of culture and decreased to baseline levels in the later phase. The telomerase activity of cord blood cultured in FT was lower than that of the FTS group. Our results suggest that, on adding stem cell factors to the FT cytokines, cultured CD34+ cord blood cells display a greater degree of cell proliferation and decreased apoptosis. However, during CD34+ cord blood cell culture, a Barge number of cells undergo differentiation, indicating that more potent novel cytokines or new culture conditioning methods should be developed to maintain their ability to engraft and sustain long-term hematopoiesis.

황칠나무 물 추출물의 항산화, 알코올 대사 효소 및 간 보호 활성 (Antioxidant, Alcohol Metabolizing Enzyme, and Hepatoprotective Activities of Dendropanax morbifera Water Extract)

  • 본 연구에서는 황칠나무 잎과 줄기 물 추출물의 항산화 활성과 알코올 대사 효소 활성 및 간 보호 효과를 알아보았다. 황칠나무 잎과 줄기 물 추출물의 총 폴리페놀 함량은 49.56 mg TAE/g으로 나타났다. DPPH radical 소거능은 황칠나무 잎과 줄기 물 추출물의 농도 의존적으로 소거능이 증가하였으며 1,000 ㎍/ml 농도에서 84.09%로 높게 나타났다. 알코올 분해 활성을 알아보기 위해 ADH 및 ALDH 활성을 측정한 결과, 두 가지 효소 모두 황칠나무 추출물의 농도 의존적으로 증가하였으며 1,000 ㎍/ml 농도에서의 ADH 및 ALDH 활성은 각각 37.68% 및 41.67%로 나타났다. 황칠나무 잎과 줄기 물 추출물은 tacrine으로 유도된 HepG2 세포주의 세포독성에 대한 보호 활성이 나타났다. 이상의 결과를 통해 황칠나무 잎과 줄기 물 추출물은 항산화 효과와 알코올 숙취해소 효과 및 간세포 보호 효과가 높은 것으로 나타나 기능성 식품 소재로서의 활용 가능이 높은 것으로 사료된다.

Effects of Photobiomodulation on Stem Cells Important for Regenerative Medicine

  • The use of stem cell therapy to treat various diseases has become a promising approach. The ability of stem cells to self-renew and differentiate can contribute significantly to the success of regenerative medical treatments. In line with these expectations, there is a great need for an efficient research methodology to differentiate stem cells into their specific targets. Photobiomodulation (PBM), formerly known as low-level laser therapy (LLLT), is a relatively non-invasive technique that has a therapeutic effect on damaged tissue or cells. Recent advances in adapting PBM to stem cell therapy showed that stem cells and progenitor cells respond favorably to light. PBM stimulates different types of stem cells to enhance their migration, proliferation, and differentiation in vitro and in vivo. This review summarizes the effects of PBM on targeted differentiation across multiple stem cell lineages. The analytical expertise gained can help better understand the current state and the latest findings in PBM and stem cell therapy.

A comparative study on the inhibitory effects of mast cell-mediated allergic reactions by artificially cultured and wild Acanthopanax senticosus

  • We compared the effect between CAS and WAS(root, stem) on mast cell-mediated allergic reaction. CAS, WAS-root and WAS-stem, significantly inhibited compound 48/80-induced systemic allergic reaction(1g/kg) and histamine release from RPMC(1mg/ml). CAS, WAS-root and WAS-stem also inhibited passive cutaneous anaphylactic reaction. In addition, IgE-induced $TNF-{\alpha}$ secretion from RBL-2H3 was inhibited by pretreatment of CAS, WAS-root or WAS-stem$(0.01{\mu}g/ml)$. Taken together, inhibitory effect on mast cell-mediated allergic reactions of WAS-root is greater than those of WAS-stem but less than those of CAS.

능소화 잎 및 줄기 추출물의 Human Immunodeficiency Virus Type I 억제활성 (Inhibitory Effects of Campsis grandiflora on HIV-1 reverse Transcriptase, HIV-1 Protease and α-glucosidase)

  • 능소화 추출물의 HIV-1에 대한 항바이러스 효과를 복제 관련 효소에 대한 억제실험과 바이러스 복제억제 실험을 통하여 살펴보았다. 역전사효소 억제활성을 ELOSA 방법으로 실험한 결과 능소화 줄기의 물 추출물이 100 ${\mu}g$/ml 농도에서 각각 37.9%의 활성을 나타내었고, 능소화 잎과 줄기의 메탄올 추출물에서 33.6% 및 31.5%의 HIV-1 protease 억제활성 나타 내었다. 그리고 HIV-1 복제 억제활성은 MT-4 세포에 대한 HIV-1 유도 세포변성억제를 광학현미경으로 관찰하여 살펴본 결과 줄기의 물 추출물이 100 ${\mu}g$/ml 농도에서 HIV-1 바이러스 증식을 완전히 억제하였다.

Optimization of Human Embryonic Stem Cells into Differentiation of Dopaminergic Neurons in Vitro: I. Additive Effect of Neurotrophic Factor on Human Embryonic Stem Cells

  • Embryonic stem cells are capable of differentiating into a variety of cell lineages. However, the ultimate results of differentiation in vitro greatly depend on the duration of treatment and kinds of differentiating inducers added. In order to investigate the efficiencies of various differentiation inducers and the methods of treatment, we examined differentiation patterns of human embryonic stem cell (hESC, MB03) according to several different protocols. Exp. I) Upon differentiation using retinoic acid and ascorbic acid (RA/AA), embryoid bodies (EB, for 4days) derived from hESC was exposed to Rh (10$^{-6}$ M) and AA (50 mM) for 4 days, and were allowed to differentiate in N2 medium for 7, 14, 21, or 28 days. Exp. II) When bFGF was used, neuronal precursor cells were selected for 8 days in N2 medium after EB formation. After selection, cells were expanded at the presence of bFGF (20 ng/ml) for another 6 days followed by a final differentiation in N2 medium for 7, 14, 21 or 28 days. Exp. III) In addition, to examine the effects of neurotrophic factors in the production of mature neurons, groups of cells were exposed to either BDNF (5 ng/ml) or TGF-$\alpha$(10 ng/ml) during the 28 days of final differentiation. Differentiation patterns of RA/AA or bFGF treated groups were very similar; approximately 82% and 83% of the cells, respectively, were positive for anti-NF200 antibody, while it was about 10% and 11%, respectively, for anti-NF160 antibody in 28 days in N2 medium. Alsor, cells expressing TH were as low as 5%, while the cells doubled when matured at the presence of either BDNF or TGF-$\alpha$. Cells immunoreactive to anti-GAD antibody were approximately 20%. These results suggest that a maturation step rather than differentiation induction step, which is formation of EB, effects more decisively to the ultimate differentiation pattern.
