Noncontact measurements of the morphological phenotypes of sorghum using 3D LiDAR point cloud

  • It is important to improve the efficiency of plant breeding and crop yield to fulfill increasing food demands. In plant phenotyping studies, the capability to correlate morphological traits such as plant height, stem diameter, leaf length, leaf width, leaf angle and size of panicle of the plants has an important role. However, manual phenotyping of plants is prone to human errors and is labor intensive and time-consuming. Hence, it is important to develop techniques that measure plant phenotypic traits accurately and rapidly. The aim of this study was to determine the feasibility of point cloud data based on a 3D light detection and ranging (LiDAR) system for plant phenotyping. The obtained results were then verified through manually acquired data from the sorghum samples. This study measured the plant height, plant crown diameter and the panicle height and diameter. The R2 of each trait was 0.83, 0.94, 0.90, and 0.90, and the root mean square error (RMSE) was 6.8 cm, 1.82 cm, 5.7 mm, and 7.8 mm, respectively. The results showed good correlation between the point cloud data and manually acquired data for plant phenotyping. The results indicate that the 3D LiDAR system has potential to measure the phenotypes of sorghum in a rapid and accurate way.

The Characteristics of Longitudinal Permeability and Hydraulic Resistance in Stem of Acer mono (고로쇠나무 줄기의 통수성(通水性)과 통수저항(通水抵抗)의 특성(特性))

  • This study was carried out to investigate the characteristics of longitudinal permeability and hydraulic resistance, and to compare the longitudinal permeability ($K_E$) calculated by the Hagen-Poiseuille's law (Siau, 1971) and the longitudinal permeability (K) measured in sapwood of Acer mono stem. The volume flow rate (Q) in a vessel was $0.80{\times}10^{-4}cm^3/sec$ and the hydraulic resistance ($R_S$) to viscous flow through a vessel was, on average, $1.37{\times}10^{10}dyn{\cdot}sec{\cdot}cm^{-3}{\cdot}cm^{-2}$. The average value of volume flow rate ($Q_N$) through the cross section of sapwood was $0.32cm^3{\cdot}sec^{-1}{\cdot}cm^{-2}$, and the average resistance ($R_{SN}$) was $3.59{\times}10^6dyn{\cdot}sec{\cdot}cm^{-3}{\cdot}cm^{-2}$. The values of K decreased as the diameter of stem increases, which was attributable to variations in the number of vessel per unit area rather than in vessel diameter, and to different resistances in the conducting tissues of each part of the stem. The average value of K measured at breast height was 31% of average value of $K_E$. The $K/K_E$ ratios were 100% in 4 to 6 year-old stems and more than 90% in 7 to 27 year-old stems. The $K/K_E$ ratio decreased as the age of stems increases, and was not more than 20% in near-ground parts of stem.

Studies on Growth and Biomass Production of Abies koreana Seedlings under Different Relative Light Intensity (상대광도 차이에 따른 구상나무 유묘의 생장과 물질생산에 관한 연구)

  • This study was carried out to investigate the growth and biomass production of Abies koreana seedlings under different relative light intensity. The 5-year-old potted seedlings were grown for 24 months under different light intensity of 100%, 50%, 30%, 10% and 2% of the natural full light intensity. The results were as follows: 1. Height growth was similar in the seedlings under different light intensities except in 2% of the full sun light intensity, having shown the lowest height growth. 2. Root collar diameter growth was reduced gradually by decreasing of the relative light intensity. 3. The seedlings which were grown under relatively low light intensity showed the later starting date of the stem growth in the next spring and showed the smaller number of new needles, and the smarter number and the shorter length of new shoot of the seedlings. 4. Total dry mass including the dry mass of leaves, shoot and root was reduced by decreasing of the relative light intensity. 5. In case of the seedling grown in one year shading condition, the ratio of the dry mass of current-year needles and stem to that of the above one-year-old needles and stem was ranged from 0.99$\pm$0.25 to 1.06$\pm$0.06 and was not significally different among the seedlings grown under 100%, 50% and 30% of the relative light intensity. And the radio was ranged from 0.60$\pm$0.04 to 0.52$\pm$0.03 in the seedlings grown under 10% and 2% of the relative light intensity, respectively. In case of the seedling grown in two years shading, the ratio of the dry mass of current-year needles and stem to that of the above one-year-old needles and stem was ranged from 0.79$\pm$0.04 to 1.00$\pm$0.03 and was not significally different among the seedlings grown under 100, 50, 30% and 10% of the relative light intensity. And the ratio was 0.29$\pm$0.03 in the seedlings grown under 2% of the relative light intensity, respectively. 6. T/R ratio of the seedlings grown in one year shading condition was ranged from 3.35$\pm$0.33 to 4.61$\pm$0.25 and was not significantly different among the relative light intensities of 100%, 50%, 30% and 10%. The seedlings grown under 2% of relative light intensity showed the highest T/R ratio of 7.36$\pm$0.15. In case of the two years shadings the T/R ratio was ranged from 1.76$\pm$0.11 to 3.12$\pm$0.19 and increased as relative light intensity decreased except 2.01$\pm$0.14 in 2% of relative light intensity.

Growth and Flowering of Cut Spray Chrysanthemum 'Charming Eye' and 'Pink Pride' by Daminozide (스프레이 절화국화 '챠밍아이'와 '핑크프라이드'의 daminozide에 의한 생육 및 개화)

  • This study was conducted to improve commercial quality of cut spray chrysanthemums 'Charming Eye' and 'Pink Pride' bred in Korea by foliar application of daminozide, suppressing excessive elongation of peduncle caused by high temperature in greenhouse. Applications were made at two floral-bud-developmental stages and concentrations used were 0, 500, 1000, and $2000mg{\cdot}L^{-1}$. As for 'Charming Eye', cut flower length, peduncle diameter, stem diameter, flower bud diameter, and the number of flower buds did not show any significant difference among all treatments including control. However, the suppressing effect of peduncle elongation, widening angle of flower cluster arrangement on apical part, and increasing parallel flower buds in stage I showed better than those in stage II regardless of daminozide concentration. As for 'Pink Pride', cut flower length, peduncle diameter, and stem diameter did not show any significant difference among all treatments including control but angle of flower cluster on apical part increased compared to control as daminozide was sprayed at stage I and II except $2,000mg{\cdot}L^{-1}$ daminozide-sprayed at stage II. The number of flower buds and flower bud diameter showed the greatest increment through $1,000mg{\cdot}L^{-1}$ daminozide-sprayed at stage I and did the least values as sprayed with $2,000mg{\cdot}L^{-1}$ daminozide at stage II. Daminozide also gradually reduced peduncle length in a concentration-dependant manner but elongation of peduncle foliar-sprayed at stage I showed the more suppressing effect than that at stage II. Increasing the number of parallel flower buds showed the best results when sprayed with $2,000mg{\cdot}L^{-1}$ at stage I. In conclusion, we recommended that foliar spraying with $500-1,000mg{\cdot}L^{-1}$ daminozide at stage I and foliar spraying with $1,000-2,000mg{\cdot}L^{-1}$ daminozide at stage I improved cut flower quality of 'Channing Eye' and 'Pink Pride', respectively.

Allometric Equations and Biomass Expansion Factors in an Age-sequence of Black Pine (Pinus thunbergii) Stands (곰솔임분의 임분연령별 상대생장식 및 현존량 확장계수)

  • This study was conducted to evaluate age-specific and generalized allometric equations and biomass expansion factors (BEFs) for each tree component across three age-sequence stands (35-year-old, 51-year-old, 62-year-old) of black pine (Pinus thunbergii Parl.) in Jinju, located in the western part of Gyeongnam province, Korea. Biomass in each tree component, i.e. foliage, branch, and stem, was quantified by destructive tree harvesting. Allometric regression equations were significant (P<0.05) with diameter at breast height (DBH) or combination of DBH and height ($DBH^2H$) accounting for 55-98% of the variation (as indicated by coefficients of determination, $R^2$) in aboveground biomass except for foliage biomass of the 62-year-old stand. Generalized allometric equations can be used to estimate the biomass of black pine stands because the slopes of age-specific equations over 35-year-old stands were not significantly different by the age-sequence. The stem density and biomass expansion factor (BEFs) were not significantly different (P>0.05) from different stand ages and ranged from 0.45 to $0.51gcm^{-3}$, and from 1.32 to 1.38, respectively. The results indicate that allometric equations, stem density and aboveground BEFs in the matured black pine over 35-year-old are little influenced by different stand ages.

Efficiency of virus elimination in apple calli (cv. Hongro) derived from meristem culture of dormant buds (사과 품종 홍로의 휴면아 분열조직 배양을 통해 형성된 캘러스에서의 바이러스 제거효율)

  • Various sizes (0.2 ~ 1.2 mm) and developmental stages (referred to as Stage 1 ~ 3) of apical and lateral meristems were excised, together or separately, directly from dormant buds of apple 'Hongro'. They were mixed infected by Apple scar skin viroid (ASSVd), Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV) and Apple stem grooving virus (ASGV), which are major viruses attacking apples. A total of 31 callus lines (> 10 mm in diameter) were obtained by culturing the explants on Murashige and Skoog (MS) medium supplemented with 3% sucrose, 3.0 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-butyric acid (IBA), and they were subjected to RT-PCR analysis for virus detection. A high rate of virus elimination (expressed as the percentage of calli that did not amplify during RT-PCR, i.e., RT-PCR negative calli per total number of calli obtained) was achieved for ACLSV (100%), ASSVd (93.7%), and ASPV (93.7%), whereas it was only 25.8% for ASGV. ASPV was detected in the presence of 2 ~ 3 bracts. Simultaneous virus elimination of ASSVd, ASPV, ACLSV, and ASGV occurred during the meristem culture, in which the early stages of the dormant buds (Stage 1) were used, because ASGV was mostly eliminated during that stage. The results of the present study will be valuable for the production of virus-free apple trees.

Effect of Leaf and Stem Blight on Growth and Root Yield of Paeonia lactiflora Pallas (작약 지상부 고사가 뿌리의 생육과 수량에 미치는 영향)

  • The pathogenic fungi associated with blight of leaf and stem in peony were leaf spot (Alternaria sp.), powdery mildew (Erysiphe aquilegiae) and rust (Cronartium flaccidum). The infection of leaf spot and powdery mildew begins from late April to midMay and rust was infected in early June. Blight time of aerial part in peony started from late May and the ratio of blight on leaf and stem was more than 50% in late Aug. Yields of root by the incidence time of blight of leaf and stem were 69.1% in late June, 65.4% in late July and $87.6{\sim}92.7$% in August and September. The number of root of more than 10mm in root diameter blighted in late June and July was much lower than in August, but the paeoniflorin content in the former was much higher than the latter.

Stem Rot on Ligularia fischeri Caused by Sclerotium rolfsii in Korea (Sclerotium rolfsii에 의한 곰취 흰비단병)

  • In June 2012 and 2013, a destructive stem rot symptoms of Ligularia fischeri occurred sporadically in Hoengseong-gun and Pyeongchang-gun Gangwon-do, Korea. The typical symptom included water-soaking on the main stem, rotting, wilting and blighting, which eventually leads to death of the plant. White mycelial mats were spread over lesions and brown sclerotia were formed on stems and near soil surface. The sclerotia were white to brown, spherical or irregular, 1-3 mm in size on potato dextrose agar (PDA), The optimum temperature range of hyphal growth was $25-30^{\circ}C$ and the hyphal diameter was $4-10{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. The resulting sequence of 695 bp was deposited in GenBank. A BLAST search revealed that sequences of the this isolates showed >99% identity with those of Sclerotium rolfsii. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers ITS rDNA, the fungi were identified as S. rolfsii. A pathogenicity test was carried out to fulfill Koch's postulates. To our knowledge, this is the first report of S. rolfsii on Ligularia fischeri in Korea.

Effects of Varieties and Seeding Dates on the Yield Components, Protein and Oil Content in Soybean (품종 및 파종기이동이 대두의 수량형질과 단백질 및 유지함량에 미치는 영향)

  • This experiment was conducted to study the effexts of seeding time and varieties on yield components, protein and oil content of soybean. Four varieties of soybean(Chung Buk Baek, Iksan, Keum doo, Chang dan Baek mok)wrere applied in this experiment and were seeded at May 15th, June 5th, and June 25th, the results obtained are as follows. 1.When the soybean was seeded at May 15th, stem height, stem diameter and number of nodes on main stem came to an end about 70days after seeding. In case that the seeding time was delayed 20 days and 40 days from May15th, respectively, the time to be almost cimpleted the growth of the characteristics was shortened 10days and 20days during ther debelopment. Among varieties, Chung Buk Beak grew faster than any other varieries. 2.When seeded at May 15th, the plants produced many pods, and had high proportion of branch pods. When seeded late, the proportion of branch pods were low. Branches had large proportion of empty pods and one grain pods, but on main stem, the proportion of two grain pods and three grain pods were more than that of branch. Among varieties, Chung Buk Baek had high proportion of two and three grain pods, Keum doo and Chang dan Baek mok had high proportion of one and two grain pods, and Iksan had high proportion of one and three grain pods. 3.Tields were less by the late seeding.The proportion if decreased yields to the yields when seeded at May 15th, that of Chung Buk Baek was decreased 6% when seeded at June 5th and 14% when seeded at June 25th, that of Ik san was decreased 9% and 17%, that of Keum doo decreased 15% and 26% and that of Chang dan Baek mik decreased 18% and 27%. 4.Protein content was highest in the plots seeded on May 15th, but oil content was highest in the plots seeded in June 25th. Among varietiesm Xhang dan Baek mok had the highest proportion of protein content and Iksan had the highest of oil content.

Growth and Yield of Sedum sarmentosum as Affected by Planting Density in Cultivation System Using a Rice Nursery Tray (벼 육묘상자를 이용한 돌나물 재배에서 삽식밀도에 따른 생육 및 수량)

  • To establish a cultivation system of Sedum sarmentosum using a rice nursery tray ($30{\times}60{\times}3\;cm$) in non-heating plastic film house, cuttings of two local strains (Wanju and Keumsan) were prepared and planted at different planting densities (40, 60 and 80 cuttings per tray) on September 7th, and subsequent growth and yield at pre- and post-winter season were investigated. When pre-wintering growth were measured at 60 days after planting, high planting density (80 cuttings/tray) decreased leaf length and lateral shoot number per plant compared to 40 cuttings/tray. When post-wintering growth were measured on April 20th, no significant growth differences as affected by planting density were observed in Keumsan strain, while decreased stem diameter, node number and leaf number were observed in Wanju strain planted at 80 cuttings/tray compared to 40 cuttings/tray. Fresh and dry weights were not significantly different as affected by planting density, but those were significantly increased in Wanju strain compared to Keumsan strain. When regrowth characteristics followed by the first harvest were measured on June 5th, significantly increased stem number was observed at 80 cuttings/tray compared to 40 cuttings/ tray. Fresh and dry weight were significantly increased in Keumsan strain planted at 80 cuttings/tray compared to 40 cuttings/tray, and dry weight were increased in Wanju strain compared to Keumsan strain. Wanju strain showed higher shoot yield, thicker stem, shorter node and larger leaf, and non-succulent stem under high planting density compared to Keumsan strain. Accordingly, the optimum density was 40 cuttings per tray, and Wanju strain was adequate for the cultivation system using a rice nursery tray.
