Stain removal on ivory using cyclododecane as a hydrophobic sealing agent

  • Stain removal on ivory has been, for a long time, considered an undesirable treatment in conservation field because ivory is hygroscopic and anisotropic, having different physical properties in different directions. Cyclododecane, which sublimes at room temperature, has been investigated for its use in conservation field since 1995, as a reversible temporary consolidant, sealing agent or coating, water repellent, and barrier layer. This research aims to remove stains on ivory, temporarily protecting the none-stained area or painted area from methanol, acetone or the aqueous cleaning system using cyclododecane as a hydrophobic sealing agent. This research also aims to obtain information regarding whether cyclododecane can be safely and effectively used on archaeological wet ivory. Melted cyclododecane and saturated solutions of cyclododecane in mineral spirits, and hexanes were applied to ivory samples. Application methods, working properties of cyclododecane on ivory, and effect of cyclododecane coating on moisture content of wet ivory were evaluated. The sealing layer formed by molten cyclododecane or by saturated cyclododecane solution in hexane or saturated cyclododecane solution in mineral spirits did not form a secure contact with the surface of the highly polished ivory. The sealing formed with two different layers, in which saturated cyclododecane solution in hexane was applied initially and then molten cyclododecane was applied over the first layer, was found to securely protect the painted area. When the wet samples were kept in 100% RH environments for a month, active mold growths were observed except in the samples sealed with molten cyclododecane. In conclusion, cyclododecane was an efficient hydrophobic sealing agent to protect painting area while cleaning stains on ivory. It also prevented mold growing on wet ivory and wet bone. Evenness of cyclododecane film on ivory will be determined in UV light. Analytical techniques will include visual observation, polarized light microscopy, Scanning Electron Microscope, and Gas Chromatography.

$^{67}Ga$ Scan of Primary Hepatocellular Carcinoma; Correlation with Angiography (원발성 간암의 $^{67}Ga$ Scan소견 ; 혈관조영술 소견과의 비교)

  • The relationship between angiographic findings and those of $^{67}Ga$ scan was evaluated in 30 patients with primary hepatocellular carcinoma diagnosed by either pathological examination or laboratory, radiologic findings. Twenty-three cases revealed hot activities on $^{67}Ga$ scan and definite tumor stains on angiography. Main findings of $^{67}Ga$ scans of 7 cases were isoactivity in 5 and cold area in 2, 5 of which revealed faint or no tumor stain on angiography. Cold areas within the primary hepatocellular carcinoma were noted in 9 cases by $^{67}Ga$ scan. In 6 cases these were due to tumor necrosis. Remaining 3 cases had arterioportal shunt, portal vein thrombosis and one had necrosis as well. These results indicate that gallium uptake of primary hepatocellular carcinoma seems to be relatively correlated with tumor stains on angiography. It is well known that the necrotic portion of primary hepatocellular carcinoma does not uptake gallium and it's the main cause of cold areas on $^{67}Ga$ scan. And we suspect that the hemodynamic changes of primary hepatocellular carcinoma such as large arterioportal shunt, portal vein thromosis may cause the decreased activity on $^{67}Ga$ scan.

Effectiveness of Nanocrystalline Silver(Acticoat®) Dressing at Wound Infected by Multidrug Resistant Bacteria (다제내성(多劑耐性)을 지닌 균주에 감염된 상처에서 Nanocrystalline Silver(Acticoat®) 드레싱의 효용)

  • Purpose: The emergency of multi-drug resistant stains of bacteria represents a challenge in the field of plastic surgery. Especially, MRSA(methycillin-resistant Staphylococcus aureus) and Pseudomonas aeruginosa have strong pathogenicity as well as multi-drug resistance so that they have become a lot more problematic strains. This study has been planned to reduce the bacterial burden by applying $Acticoat^{(R)}$(Smith & Nephew Healthcare, Hull, England)dressing into the chronic wounds infected by multi-drug resistant strains and to facilitate their healing. Methods: Nanocrystalline silver dressings($Acticoat^{(R)}$) were applied to chronic wound infected by MRSA or Pseudomonas aeruginosa. Multi-drug resistant bacteria were smeared over a slide glass using sterilized cotton swabs and gram stains were performed directly before and after applying $Acticoat^{(R)}$ dressings at 1, 24, 48 and 72 hours. The gram-stained slides were observed using an optical microscope magnified 1000 times(${\times}1000$). The bacterial counts of the control group(0 hour) were compared to those of the experimental groups(1, 24, 48, and 72 hour). Paired T-test was used to assess a statistical significance. MRSA was cultured in two BAPs(blood agar plate) and two MacConkey plates with streak plate method. None were interventions on one culture plate, while on the other culture plate, $Acticoat^{(R)}$ was placed in a square shape and cultured for 72 hours at $37^{\circ}C$, then plates were examined. Pseudomonas aeruginosa was cultured in the same manner as MRSA. Results: There are the large amount of declination of bacterial counts with statistical significance after $Acticoat^{(R)}$ dressing. The bacteria grew in culture plate without specific intervention, but no bacteria grew in culture plate with applying of $Acticoat^{(R)}$ dressing. Conclusion: We believe that $Acticoat^{(R)}$ dressing could be used as an effective method of treating chronic wounds which are infected by multi-drug resistant organisms.

The Effect of Pretreatment of Veneer on the Improvement of Plywood Quality (단판(單板)의 약제처리(藥劑處理)가 합판성질개선(合板性質改善)에 미치는 영향(影響))

  • 1. In order to make the improvement of plywood quality, this study has been made. The pretreatments applied to the veneers are as follows. a) The soaking treatment of the veneer in 30 percent methanol solution for 24 hours was applied to remove some resinous materials which may cause many stain discolouring on the face of finished plywood. b) The preservative treatment of the veneer in 2 percent malenit solution for 2 to 3 minutes was adopted to make resistance against decay and insect damages. c) The fire retardant treatment of the veneer in 40 percent ammonium phosphate solution for 2 hours was applied to give retardation effect against fire burning. 2. The results summarized in this study are as follows. a) One percent resinous materials was extracted, after the soaking of the veneers in 30 percent diluted methanol. b) No marks of the dirty stains of resinous materials on the face of the treated plywood was shown, although many quite dirty stains on the face of untreated plywood have contrary seen. c) However, the strip shear test strength of the treated plywood was not decreased. It means that there is no difference in the strength between the treated plywood and the untreated plywood. The strength values were 25.08 kg/$cm^2$ and 24.98 kg/$cm^2$, respectively. d) The strip shear test strength of plywood made of the treated veneers in 2 percent malenit solution was not decreased. e) The slight decrease of the strip shear test strength of the treated plywood made of the treated veneer in 40 percent ammonuim phosphate solution was shown. However, the remarkable difference of the fire retardation activities between the treated specimens and the untreated specimens has seen as in Table 10, that is, the fire proofed specimens had taken about 28 seconds to start to burn, while the untreated specimens had taken 15 seconds to reach to burning. This means that the fire retardation effect of the fire proofed plywood was greater than that of the unproofed plywood.

Characterization of Bunker Oil-Related Compounds Degrading Bacteria Isolated from Pusan Coastal Waters (부산근해에서 분리한 Bunker Oil 관련화합물 분해세균의 특성)

  • Microorganisms utilizing petroleum as substrate were screened from the seawater in Pusan coastal area. Among them, fifty strains utilized bunker-A oil as a sole carbon and energy source. Five of these fifty strains were selected to experiment this study. According to the taxonomic characteristics of its morphological, cultural and biochemical properties, the selected stains were named Pseudomonas sp. EL-12, Flavobacterium sp. EL-15, Acinetobacter sp. EL-18, Enterobacter sp. EL-27 and Micrococcus sp. EL-43, respectively. The optimal medium compositions and cultural conditions for assimilation of bunker-A oil by the selected strains were 1.5-2% bunker-A oil, 0.1% $NH_4NO_3$, 1-1.5% $MgSO_4$.$7H_2O$, 0.05-0.15% KCl, 0.1-0.15% $CaCl_2$.$2H_2O$, 2.5-3.5% NaCl, initial pH 8-9, temperature 3$0^{\circ}C$ and aeration, respectively. The utilization and degradation characteristics on the various hydrocarbons by the selected stains were showed that bunker oil, n-alkane and branched alkane compounds were highly activity than cyclic alkane and aromatic hydrocarbon compounds.

  • Adding chemical additives in bamboo paper making procedure in China became common in last decades to increase productivity. Supply of repair paper for paper based artefacts became more and more difficult due to this tendency. Furthermore, stains and spots on paper which happen to appear during dying procedure make it difficult to use modern bamboo paper for repair treatment. In this research, lime fermentation and sun bleaching were main elements which affect texture and color of paper. Impurities, however, add some effects on paper quality. Less thouroughly washed raw materials after fermentation also affects texture of paper substrate. One most significant impurity is lime. Minimum residue of lime can make stains and spots after dying. Reproduction of white bamboo paper would become useful resource in various conservation treatments as a repair paper, and also, for reproduction of paper woven painting. However, further research to improve quality at early stage of paper making procedure in China required.

Intergeneric Protoplast Fusion between Rhizopus oryzae and Aspergillus oryzae (Rhizopus oryzae와 Aspergillus oryzae의 속간 원형질체융합)

  • Conditions for the release and regeneration of protoplasts form Rhizopus oryzae and intergeneric protoplast fusion between Rhizopus oryzae and Aspergillus oryzae were studied. High yields of protoplast fusion between Rhizopus oryzae and Aspergillus oxyzae were studied. High yield of protoplasts from young germilings of R. oryzae were obtained by using lytic enzymes containing chitosanase (3 mg/ml), chitinase (3 mg/ml) and Novozym 234 (5 mg/ml). 0.5M glucose was used as the osmotic stabilizer and optimum pH of buffer was determined to be pH 7.5-8.0. Under these conditions, protoplasts were formed after about 3-4 hrs incubation. Approximately, 1.0%-4.9% of these protoplasts were formed after about 3-4 hrs incubation. Approximately, 1.0%-4.9% of these protoplasts regenerated on solid medium with a soft agar overlay. We have also carried out protoplasts fusion between R. oryzae and A. oryzae and have succeeded in obtaining three types of intergeneric fusants. In these experiments, 35% PEG-4000 and 10 mM CaCl$_{2}$ were used as fsogenic agents, and auxotrophic properties were used as a genetic marker to select fusants. Complementation frequency be protoplasts fusion of A. oxyzae and R. oryzae was 4.4% * 10$^{-5}$ . The fusant strains of the first type were prototrophs showing an Aspergillus type morphology with dark-yellow sporulation, those of the second type were also Apergillus type morphology but showed no sporulation. And the strains of the third type stopped growing when fusion products grown on regeneration minimal medium were transferred to fresh minimal medium. The formation of fusion products was observed by fluorescent vital stains for complementary labelling of protoplats from R. oryzae and A. oryzae. Rhodamine 6G and fluorescein diacetate wer useful complementary vital stains of Rhizopus and Aspergillus protoplasts for visualization of requency and type (dicell, multicell) of fusion.

The Effect of Tooth Whitener on the Color and Microhardness of Human Enamel in Vitro (미백제가 법랑질 명도 변화에 미세경도에 미치는 영향)

  • Human teeth vary widely in color. Practitioner and patients are concerned with preventing and correcting discolored or dark teeth to achieve and maintain stain-free, white teeth. Tooth brushing cannot alter tooth color but it can remove adhering films and stains. The esthetics of natural dentition can be improved by bleaching and this process can be applied to intrinsically and extrinsically stained teeth. The need for a brighter, more attractive smile has made rapid growth in the market for tooth whiteners. There is no doubt these products work as whiteners, at least on mild to moderate stains, but the safety of these products are unclear. In this experiment, the effect of tooth whitener application on the color and microhardness of extracted human enamel was measured. RMS, RMT and NWT were used as tooth whiteners, and tooth paste(ETQ) and hydrogen peroxide solution(HPO) were used as controls. 35 caries-free extracted human molars were embedded and polished with the exposed enamel diameter of 4 mm. The tooth whiteners and control agents were applied according to the manufacturers' instructions or clinically simulated procedures for eight weeks, and measurements were repeated every two weeks. Value(L*) difference was measured using Differential Colorimeter(Model TC-6FX, Denshoku Co., Japan), and microhardness was measured using microhardness tester(Mitsuzawa Seiki Co., Japan). The results were as follows; 1. After application of agents for eight weeks, the Vickers hardness increased significantly in the ETQ, RMS and RMT application group(p〈0.01), and that decreased significantly in NWT application group(p〈0.01), but in HPO application group there was no significant change. The change in microhardness was greatest in NWT application group(p〈0.01). 2. After application of tooth whiteners and controls for eight weeks, the value change of toothpaste application group was significantly lower than those of other agents groups(p〈0.01), and there was no significant difference in value(L*) change among tooth whitener groups(p〉0.01). 3. The application of tooth paste and paste type tooth whitener made gradual value change, but hydrogen peroxide gel type tooth whitener and hydrogen peroxide solution made rapid value change during initial application period.

Effect of Washing Solution Characteristics on the Removal and Color of Cocoa Stains (세탁용수의 특성에 따른 코코아 오구의 세척성과 색상)

  • Cocoa is a popular drink for children and contains healthy polyphenols however; a deep brown stain is left when cocoa is spilled over clothes. The main pigments in cocoa are anthocyanins that change in washing solutions with different alkalinity and metals. The removal and color changes in a cocoa stain after washing with various pH solutions and water hardness were studied. Alkalinity and the water hardness of washing solutions were important factors for the removal of cocoa stains. The removal of cocoa things in washing solutions without detergent was low (and even became negative after removal and darker) in solutions with a pH 9 and above. The cocoa stain was not removed and only the fabric color faded, although the cocoa stained cloth was washed with Korea tap water that has a pH of 7. The cocoa stain removal in detergent solutions was conspicuously higher than for only water. Even in detergent solutions, the cocoa stain removal decreased as water hardness increased. Cocoa stain removal was more effective and the color dimmest when the stained cloth was washed in a solution without the metal cations, and the bleach added with the detergent at a temperature of $40^{\circ}C$ and for longer than 20 minutes. Effective and economical equipment for tap water softening for a washing machine should be developed and used to improve cocoa stain removal.

The Characteristics of Vacuum Drying Disks of Domestic Softwoods (I) (국산 침엽수재 원판(圓板)의 진공건조(眞空乾燥) 특성 (I))

  • This study was carried out to investigate the vacuum drying characteristics of 50mm- and 75mm- thick tree disks of some domestic softwoods for substituting the conventional hardwoods as the materials for wood crafts. Though the elapsed drying times from green to in-use moisture content(MC) were largely shortened by vacuum drying, the tree disks treated by EWS couldn't be dried to in-use MC, and so sapwood stains also were occurred severely. We suggest EWS treatment is undesirable for the species with very high sapwood portion or vacuum drying with hot water circulation heating system. Heart checks were slight, but sapwood checks, which have never been trouble in drying process of tree disks, were severe. For the reasonable explanation it is suggested MC differences between sapwood and heartwood were large and most of tree disks had already no barks before drying test. Ginkgo was vacuum-dried with very slight drying defects such as heart checks, sapwood checks. V-cracks and sapwood stains. In Korean red pine and pitch pine V-cracks were severely occurred. And it was found the special feature that most of these defected tree disks contained several V-cracks within one tree disk. It can be considered as the causes that the region of sapwood was defected by the several checks at the early drying stage because of the steep MC gradient along the radial direction, and then at the later drying stage the drying stresses due to differential shrinkage were concentrated on these brittle spots.
