Changes in CGRP-immunoreactive Nerve Fibers during Expansion of Midpalatal Suture of the Rat (백서 정중구개봉합 확대후의 CGRP 면역반응 신경섬유의 변화)

  • Midpalatal suture expansion is often used for patients haying narrow maxillary arch, cleft palate, respiratory handicap with narrow nasal cavity. CGRP has been known as a modulator of pain transmission in central nervous system and a local effector to peripheral tissue causing vasodilation, increase of blood flow, modulation of immune system, regulation of macrophagic function and stimulation of bone formation. To investigate changes of CGRP-immunoreactive nerve fibers in midpalatal suture during the expansion, immunohistochemical study was performed by using rats. Experimental rats (10 weeks, 250 gm) were divided into five groups (control, 1, 4, 7, 14 days group (each n=4) and applied orthodontic force (approximately 200gm) to upper anterior incisors. Frozen sections of midpalatal suture area were immunostained by using rabbit antisera. The results were as follows. ${\cdot}$ The CGRP-immunoreactive nerve fibers were hardly observed in control group. ${\cdot}$ In 1 day group, the CGRP-immunoreactive nerve fibers were more increased around the vessels than control group. ${\cdot}$ In 4 days group, the CGRP-immunoreactive nerve fibers were more increased than control group, but not more increased than 1 day group. Vascular diameter was more enlarged. ${\cdot}$ In 7 days group, especially, hematoxilin affinity of cells was remarkable and cells were arranged along the bone margin. The CGRP-immunoreactive nerve fibers were more reduced than 4 days group and vascular diameter was also reduced. ${\cdot}$ In 14 days group, the CGRP-immunoreactive nerve fibers were similar to those of 7 days group and the irregularity of bone margin was almost recoverd. In Conclusion, the CGRP-immunoreactive nerve fibers nay be related to initial neurogenic inflammatory reaction in expanding mid-palatal suture.

Biological Effect of Vaccinium uliginosum L. on STZ-induced Diabetes and Lipid Metabolism in Rats (들쭉이 약물에 의해 유도된 당뇨 및 지질대사에 미치는 생리활성 효과)

  • This study was conducted to investigate the effects of Vaccinium uliginosum L. (bilberry) on chemically induced diabetes and hypercholesterolemia. Sprague Dawley (SD) rats were divided into six groups, control (CON), bilberry added group (CBB), streptozotocin (STZ)-induced diabetic group (STZ), STZ and bilberry added group (SBB), high fat fed group (HFF) and high fat and bilberry added group (HFB). Diabetes was chemically induced by intravenous injection of 45 mg/kg body weight STZ in citrate buffer (pH 4.5). Serum triglycerides decreased significantly (p<0.05) in the STZ group that was fed bilberry. Additionally, the athrogenic index (AI) decreased significantly (p<0.05) when compared to the STZ group, while the liver triglycerides tended to decrease in the STZ group. HDL-cholesterol also increased significantly in response to bilberry. When compared to the STZ group, steady attenuation of the blood glucose level was observed upon fasting, 15 min, 30 min, 60 min and 120 min after oral glucose administration. The blood glucose level in the bilberry fed group decreased by 24% when compared to STZ group, while the superoxide dismutase (SOD) became significantly higher (p<0.05) in the STZ group when compared to the CON group. Overall, the results of this study suggest that bilberry stimulates lipid metabolism in both the serum and liver and has a positive effect on glucose metabolism in chemically induced diabetic rats.

Effect of Calcium Extracted from Salted Anchovy (Engraulis japonicus) on Calcium Metabolism of the Rat (멸치젓에서 추출한 칼슘이 흰쥐의 칼슘대사에 미치는 영향)

  • This study was conducted to examine the effect of calcium extracted from salted anchovy (Engraulis japonicus) on the calcium metabolism of rats. Sprague-Dawley male rats were fed low-calcium diets (0.15%) for 2 weeks after the adjustment period. Rats were divided into five groups and were fed experimental diet for four weeks. Experimental diets were low calcium (LC, 0.15% $CaCO_3$), 0.5% $CaCO_3$ (CC), seaweed calcium (SC), calcium lactate (LC), anchovy calcium (AC). The low-calcium diet group (LC) showed the lowest weight gain and had no differences among the groups with adequate calcium intake. Calcium retention was lowest in the LC group and higher in the CL, SC, AC groups than in SC groups. Serum alkaline phosphatase (ALP) level was highest in LC group, and significantly low in the CC and AC groups (p<0.05). Parathyroid hormone and osteocalcin levels showed no differences among experimental groups. The urine deoxypyridinoline (DPD) level was lower in AC and CC groups compared to the LC group (p<0.05). The dry weight of the femur showed no significant differences among normal calcium groups. The bone mineral density of the femur in AC and CC group were significantly higher than the LC group (p<0.05). From these results, calcium extracted from salted anchovy can be useful as a calcium supplement comparable with calcium carbonate.

Effects of Ethylacetate Fraction of Plantain (Plantago asiatica L.) on Experimentally-Induced Gastric Mucosal Damage and Gastric Ulcers in Rats (질경이가 실험적으로 유발된 흰쥐의 위염 및 위궤양에 미치는 영향)

  • Plantain has been used for antidiarrhea, antihemorrhage and the remedy of indigestion. Plantain was extracted with ethanol and fractionated systemically with n-hexane, chloroform, ethylacetate (EtOAC) and n-butanol. Antioxidant index (AI was expressed as induction period of oil containing various fractions/induction period of oil of 600 ppm) of EtOAC fraction was the highest among fractions in vitro. The protective effects of the EtOAC fraction of plantain (PE) administered 1 mL orally or intraduodenally on experimentally induced gastritis, gastric ulcer and gastric secretion were evaluated in rats. Sprague-Dawley rats weighing 250∼300 g were divided into 4 groups; negative control group (CON), PE 200 mg/kg treated group (PEL), PE 400 mg/kg treated group (PEH) and positive control group (cimetidine 100 mg/kg-CMT or omeprazol 100 mg/kg treated group-OMT), respectively, PE significantly suppressed HCl-ethanol induced gastric lesions and indomethacin-induced gastric ulcers (administered subcutaneouly) in rats. Specially PE 400 mg/kg showed significantly inhibitory effect, which was more potent than that of 100 mg/kg of commercial drug, cimetidine, and elevated an inhibitory effect to be close to the level in inhibitory ratio of omeprazol administered group in Shay's ucler. On gastric secretion in pylorus ligated rat, PE 200 mg/kg and 400 mg/kg decreased the gastric volume and acid output, but did not show an apparent effect on pepsin activity. In addition, PE 400 mg/kg depressed gastric ulcers induced by water immersion stress and duodenal ulcers induced by cysteamine administered subcutaneouly. These results suggest that the ethylacetate fraction of plantain can be used in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.

The Effect of Combined Estrogen/calcium Therapy on Bone Metabolism in Ovariectomized Rats I. A Study on Biochemical Parameters in Ovariectomized Rats (에스트로겐/칼슘 혼합요법이 난소절제 쥐의 골대사에 미치는 영향 I. 생화학적 변화에 관한 연구)

  • This study was implemented in order to investigate the effects of dietary calcium-salt, estrogentreatment, and estrogen/calcium treatment on bone metabolism. Ovariectomized rats were used as animal models. Female Sprague-Dawley rats with a body weight of 250~280g were underwent ovariectomy or sham-operation. The ovariectomized rats were divided into 9 different experimental groups including the saline-treated group, the estrogen-treated group, the high calcium salt-treated group, and the estrogen/calcium treated groups and fed for 6 weeks. Creatinine and hydroxyproline in urine were analyzed. Creatinine, calcitonin, osteocalcin, alkaline phosphatase and parathyroid hormone in plasma were also determined. The results of the experiment are as follows : The ovariectomy caused a significant increase in the level of food intake, food effciency ratio and body weight gain in comparison with sham-operation. The overall food intake, food effciency ratio and body weight gain were significantly decreased by estrogen. The ovariectomized animals developed obesity as a result of increased food intake. In addition, estradiol injections suppressed food intake with a concomitant loss in body weight. The level or urinary hydroxyproline, as an indicator or bone resorntion, was higher in the ovariectomized rats compared to sham-opertion, while these decresed in the estrogen/calcium treated group. Parathyroid hormone and calcitonin in the plasma, that were used as the indicator of calcium homeostasis, parathyroid hormone higher in the ovariectomized rats compared to sham-operation. It was lowered by estrogen and high calcium treated groups; thus, estrogen and estrogen/calcium treated groups were decreased by 32% compared to saline treated group. Osteocalcin and alkaline phosphatase which are indicators of bone formation, were significantly higher in ovariectomized group, while this showed to be decreased in the estrogen and the ostrogenicalcium treated groups. Estrogen and estrogen/calcium in ovariectomized rats resulted in lower bone loss. However, estrogen treated group its gradual reduction showed little effect on bone loss, while the gradual reduction of estrogen had a preventive effect on bone loss when the treatment was combined with calcium intensification.

Effects of Human Milk Fortifier on Growth and Nutritional Status of Growing Rats Fed Infant Formula (모유강화제(HMF)가 흰쥐의 성장과 영양상태에 미치는 영향)

  • This study was conducted to evaluate the efficacy of human milk fortifier (HMF) on growth and nutritional status in growing rats fed infant formula supplemented with HMF. Three week-old male Sprague-Dawley rats were divided into three groups and fed regular formula (RF), premature formula (PF) and regular formula fortified with HMF (RF+HMF) diets for 3 weeks. There was no significant difference in weight gain among groups. However, a significant increase of food intake was observed in PF and RF+HMF groups compared with RF group. With increasing food intake, the intakes of carbohydrate and protein were significantly increased in PF and RF+HMF groups. The weight of perirenal fat was significantly increased in rats fed RF+HMF; however, the weights of liver, kidney and spleen were not significantly different among groups. Although total lipids, total cholesterol, HDL-cholesterol concentrations of serum were not significantly different among groups, triglyceride was significantly increased in PF group. The triglyceride and total-cholesterol of liver were significantly increased in rats fed regular formula fortified with HMF and PF compared with RF group. Glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT), creatinine (Cre) and blood urea nitrogen (BUN) in serum showed no significant difference among groups. The concentration of growth hormone was significantly increased in PF group compared with other groups. The concentration of hemoglobin was significantly increased in rats fed PF and RF+HMF. These results suggest that the supplementation of human milk fortifier in growing rats may promote growth as increasing food intake and lipid contents in tissues and prevent the anemia of infants.

CHANGES IN THE SHAPE AND ULTRASTRUCTURE OF THE ARTICULAR DISK OF RAT FOLLOWING POSTURAL HYPERPROPULSOR (백서 하악골의 기능적 전방위가 악관절 원판에 미치는 영향)

  • This study was conducted to examine the changes in the shape of the Sprague-Dawley rats' articular disk following postural hyperpropulsion by observing their articular specimens through light and electronic microscopes after following 2-week and 4-week postural hyperpropulsion from their four weeks of age. The findings of this study are summarized as follows. It was shown that as compared with the control group, the experimental group indicated a significant increase in thickness of the 2-week groups' anterior and postreior portion of the articular disc. The experimental group showed statistically more significant increase in thickness of the 4-week groups' anterior portion of the articular disc than the control group. Light micrograph showed that the experimental group had more fibroblast in the anterior portion of the 2-week and 4-week groups than the comparing group. The 2-week groups showed in the findings through the electronic microscope that there were found the well developed and dilated RER which seems to actively synthesize the extracellular matrix including collagen, the cells with the well developed RER without distention which seems to actively synthesize the intracellular microfilaments due to the well developed free ribosome, and the typical chondroid cells. In addition, there was more fibroblast cell with the distended and well developed RER in the anterior area of the experimental group than that of the control group. The 4 week experimental group's anterior area of the disk had more cells than that of the control group while fibroblast with the well developed RER and free ribosome was quite abundat. Based on the above result of this study, it was shown that the functional hyperpropulsion of the mandible causes the changes in the nature of the mechanical load to the certain portion of the articular disk. As a result, it seems that there may be occurred some changes in morphology of the disc by adaptation or confrontation with these changes at the cellular level.

Effects of Baked Garlic Powder on Lipid Metabolism in Rats Fed a High-Fat/High-Cholesterol Diet (구운 마늘 분말이 고지방-고콜레스테롤 식이를 급여한 흰쥐의 체내 지질대사에 미치는 효과)

  • This study examined the effects of baked garlic powder on the lipid metabolism in rats fed a high-fat/highcholesterol diet for 4 weeks to induce hyperlipidemia. Male Sprague-Dawley rats were assigned to four groups according to the dietary fat, cholesterol and baked garlic powder levels. The experimental groups were normal diet group (N), a high-fat/high-cholesterol diet group (C), a high-fat/high-cholesterol diet with 1.5% baked garlic powder group (GPL) and a high-fat/high-cholesterol diet with 3% baked garlic powder group (GPH). The body weight gain, food intake and food efficiency ratio were similar in the experimental groups. The epididymal adipose tissues weight of the C group was higher than that of the N group, whereas those of the groups fed baked garlic powder were decreased gradually. The ALT and ALP activities were similar in the C groups, but the serum AST and LDH activities elevated by a high-fat/high-cholesterol diet were decreased significantly by feeding a 3% baked garlic powder diet. The serum triglyceride, total cholesterol and LDL-cholesterol levels as well as the atherogenic index and cardiac risk factor tended to decrease in the groups fed baked garlic powder than the C group, whereas the serum HDL-cholesterol level was lower in the C group and remarkably in groups fed baked garlic powder than the control group. The total cholesterol level in the liver and mesenteric adipose tissue and the triglyceride level in epididymal tissue were lower in the groups fed baked garlic powder than the C group. These results suggest that baked garlic powder reduces the serum lipid components and improves the lipid metabolism in hyperlipidemic rats induced with a high-fat/high-cholesterol diet.

Effect of Mixture of Lactobacillus plantarum CECT 7527, 7528, and 7529 on Obesity and Lipid Metabolism in Rats Fed a High-fat Diet (Lactobacillus plantarum CECT 7527, 7528, 7529 혼합물이 고지방식이를 급여한 흰쥐의 비만 및 지질 대사에 미치는 영향)

  • In the present study, we examined the effects of a mixture of three strains, Lactobacillus plantarum CECT 7527, 7528, and 7529 (L. plantarum mixture), on body weight and lipid metabolism in Sprague-Dawley rats fed a high-fat diet. Rats were fed a high-fat diet and subjected to oral gavage with vehicle or the L. plantarum mixture ($0.6{\times}10^9$, $1.2{\times}10^9$, $2.4{\times}10^9$ colony-forming units (CFU)/day/rat, respectively) for 8 weeks. In rats fed a high-fat diet, oral administration of $2.4{\times}10^9CFU/day$ of the L. plantarum mixture significantly reduced body weight gain as well as weights of liver and epididymal fat. Leptin levels in sera were significantly reduced by oral administration of $2.4{\times}10^9CFU/day$ of the L. plantarum mixture. The L. plantarum mixture ($1.2{\times}10^9$ or $2.4{\times}10^9CFU/day$) also reduced the concentrations of total cholesterol and LDL-cholesterol in sera when it administered orally. Further, the L. plantarum mixture significantly reduced the atherogenic index and cardiac risk factor. In addition, oral administration of $2.4{\times}10^9CFU/day$ of the L. plantarum mixture markedly reduced levels of total lipids, triglycerides, and total cholesterol in the liver. The results of this study indicate that the L. plantarum mixture may exhibit anti-obesity and cholesterol-lowering effects, which suggest that the L. plantarum mixture has the potential to be a probiotic in the management of obesity and hypercholesterolemia.

Effects of the Artemisia capillaris Extract on the Hepatotoxicity in Ethanol-induced Rats (에탄올 투여 랫트에서 인진쑥 열수 추출물이 간 독성 저하에 미치는 영향)

  • This study was conducted to find out the effects of aqueous extract from the leaves of Artemisia capillaris (AA) on the reduction of hepatotoxicity induced by ethanol in rats. In this experiment, Sprague Dawley rats were used in the experimental groups, which were divided into 5 groups; normal group, ethanol+UDCA (ursodeoxycholic acid)-treated group (positive control), ethanol-treated group (control), ethanol+Artemisia capillaris aqueous extract-treated group (200 mg/kg of BW) and ethanol+Artemisia capillaris aqueous extract-treated group (400 mg/kg of BW). AST (aspartate aminotransferase), ALT (alanine aminotransferase), GGT (gamma(${\gamma}$)-glutamyl transferase) and LDH (lactate dehydrogenase) activities of the ethanol+Artemisia capillaris aqueous extract-treated group (400 mg/kg of BW) were significantly decreased compared to that of the ethanol-treated group (P<0.05). The triglyceride level of the ethanol-treated group was significantly increased and the HDL-cholesterol level was significantly decreased compared to the normal group (P<0.05). On the other hand, the triglyceride level was significantly decreased (P<0.05) and the HDL-cholesterol level was significantly increased (P<0.05) in the ethanol+Artemisia capillaris aqueous extract-treated groups. Superoxide dismutase (SOD) activity was enhanced significantly (P<0.05) in the ethanol+Artemisia capillaris aqueous extract-treated groups. Also, malondialdehyde contents were decreased in this group (P<0.05). Histologically, in the control group, there was a mild degenerative change around central venule. The AA treated group showed well preserved lobular architectures with no evidence of steatosis or liver damage in aqueous extract from the leaves of Artemisia capillaris treated group (H&E, ${\times}20$). As the results of this study, it is thought that Artemisia capillaris aqueous extract may have effects on the improvement of hepatic damage by ethanol.
