Studies on the Determination of Synthetic Food Colors (합성식용색소의 정량분석법에 관한 검토)

  • Center for Food Standard Evaluation, Korea Food and Drug AdministrationThis study was performed to compare analytical methods of nine synthetic food colors and six food color lakes in Korea, Japan, Joint FAO/WHO Expert Committee of Food Additives (JECFA), and USA. The experimental protocol of this study consists of three parts: titration method with titanium chloride, gravimetric and spectrophotometric method. To measure the total contents of food colors, Korea and Japan used titration method with titanium chloride, USA used the average value of titration method with titanium chloride equipped with Kipp generator and spectrophotometric method. Also, JECFA used titration method with titanium chloride equipped with KiPP generator. However, All the low organizations used gravimetric method to measure the total content of coloring matter on Food Red No.3. Although all organizations use various methods for analysis of coloring matters, total contents of coloring matter on food colors tested fell into the standard showing $85.08-96.40\%$ in synthetic food colors and $10.00-36.86\%$ in food color lakes.

  • As chromium in urine remains as oxychromic acid ions, chromium was extracted by using ion pair formed by anion exchanger trioctylamine. then after it is noted whether this TOA-MIBK method is effective or not for the chromium analysis by using flame method of Atomic absorption Spectrophotometric Analysis. The result is as following. 1. Effects of various acids on the extraction of Cr with MIBK including TOA are good in order $NHO_3$, $H_2SO_4$, HCl in distilled water sample and its proper concentration of HCl is 0.2 N. 2. For the analysis of urine sample, the best result can be achieved by following condition. After finished pretreatment adjusted to pH 6.5-7.5 by NaOH and again controlled pH 0.5-0.6 by HCl. 3. Though TOA concentration slightly affects the analytic value, best result is noted in 1-3% concentration. 4. Recovery rates of urine samples made by $0.3mg/l{\cdot}urine$, $0.6mg/l{\cdot}urine$, $0.9mg/l{\cdot}urine$ are shown from 96.7% to 104.8%. 5. Recovery rates of urine samples made by $0.01mg/l{\cdot}urine$, $10.03mg/{\cdot}urine$, $0.05mg/l{\cdot}urine$ are shown from 89.3% to 98.6%.

  • Total and free tryptophan contents in the fore shank muscle of korean cow treated with proteolytic enzymes have been analyzed by the spectrophotometric method and the results are as follows: 1. By adding 0.01%, 0.05%, 0.1% of proteolytic enzymes, the total tryptophan and free tryptophan have incresed steadily 2. The rate of increase of the total tryptophan content was highest when 0.05% of the enzymes for the meat weight were added. 3. The change in free tryptophan incident to the addition of proteolytic enzymes was insignificant.

  • A spectrophotometric method for the determination of copper using N,N'-Oxalylbis(salicylaldehyde hydrazone) as a chromogenic reagent has been developed. Determination has been performed by measuring the absorbances of the copper complexes in solutions containing 60% dimethylformamide (pH 2) at 422 nm. The method allows the determination of 0.4${\sim}$1.8 ${\mu}$g/ml of copper and has been applied to its determination in synthetic mixtures and alloy samples.

  • We developed a spectrophotometric assay for the quantitative determination of chlorogenic acid based on the formation of green pigment at $50^{\circ}C$ under glycine and alkaline conditions in 96-well plates. The formation of green pigment was linear with a series of chlorogenic acid concentration (0-$300\;{\mu}M$). Using this method, the content of chlorogenic acid (12.42 mg/g dry weight) in the leaves of blueberry was quantified. This method is high-throughput, cost-effective, rapid, and easy to perform.

Highly Selective and Simple Zero and First Order Derivative Spectrophotometric Determination of Palladium by Using α-Benzilmonoxime in Triton X-100 Micellar Solution

  • The reaction of ${\alpha}$-Benzilmonoxime with palladium(II) produces a green complex in triton X-100 micellar media. Palladium has been determined using zero and first derivative spectrophotometric methods. The absorbances of Pd(II)- ${\alpha}$--benzilmonoxime complex at 441.8 and 677.0 nm in 0.10 M perchloric acid solution were monitored and linear working ranges of 0.3-12.0 and 0.7-20 ${\mu}$g mL$^{-1}$ with detection limits of 0.07 and 0.10 ${\mu}$g $mL^-1$ were obtained, respectively. Also, recoveries in the range of 92.8 to 100.1% and relative standard deviations in the range of 0.4 to 7.1% were obtained. First derivative spectrophotometry has also been applied for palladium determination under the optimum condition. The linear dynamic range of 0.2-24.0 ${\mu}$g $mL^{-1}$ palladium with relative standard deviations of 0.6-6.9% and recoveries in the range of 94.9-102.5% has been obtained by first derivative spectrophotometry. The method shows high selectivity because of the high concentration of acid used, which prevents formation of complexes of ${\alpha}$--benzilmonoxime with the other cations. The palladium complex formed was stable at least one day. The method was successfully applied to the determination of palladium in some synthetic palladium alloys and palladium-charcoal powder.

Analysis of Absorbance for Tinted Dye Absorbed into Tinted Lenses by Spectrophotometric Method (분광광도법에 의한 칼라 렌즈 내 염색제에 대한 흡광도 분석)

  • We studied absorbance of tinted dye absorbed into tinted lenses by Spectrophotometric method. Used lenses and tinted dye were CR-39 plastic optical lenses and Ons & $Lavas^{(R)}$ Blue, respectively. Absorbance values of tinted dye absorbed into tinted lenses were obtained from the differences of absorbance values between tinted lenses and CR-30 lenses. Average concentrations of tinted dye absorbed into tinted lenses were obtained from the mass differences between tinted lenses and CR-30 lenses. The relationship between absorbance and average concentration for tinted dye absorbed into tinted lenses was also confirmed to follow Beer’s law. The absorption coefficient calculated from the tangent of the calibration curve was determined to be a=0.983 $Lg^{-1}cm^{-1}$.

  • The interaction of cadmium (II) ion with protocatechuic acid ($H_3PA$) and ethylenediaminetetraacetic acid (EDTA) were investigated in aqueous solution at different pH. The protocatechuic acid and EDTA/cadmium stochiometries for cadmium (II) binding have been determined by UV-vis spectrophotometric method. The complexation of Cd (II) ion with protocatechuic acid was formed in solution. Among the two potential sites of chelation present in the protocatechuic acid structure, the carboxylic function presents higher complexation power toward Cd (II). 1 : 1 Cd (II)-complex had a maximum absorbance and showed the bathochromic shift of the long-wavelength band of the UV-vis spectra in the alkaline pH when interacted with protocatechuic acid in 0.2 M $NH_3$ - 0.2 M ${NH_4}Cl$ (pH 8.0) buffer. These results suggest that Cd $({H_2}PA)^+$ complex has the optimal condition of chelation in buffer solution at 64.22 ${\mu}M$ protocatechuic acid (A=1.01455).
