Effect of Triton X-100 on the Growth and Morphology of Trichoderma koningii (Triton X-100이 Trichoderma koningii의 성장 및 형태에 미치는 영향)

  • We investigated the effect of Triton X-100 on the growth and morphology of Trichoderma koningii by comparing various parameters representing the frowth of mold in the presence or absence of Triton X-100. The specific growth rate and doubling time of T. koningii were not affected by the addition of 0.05% Triton X-100 in batch culture. However, in the presence of Triton X-100, cultures reached its stationary phase earlier and showed reduced level in total yield of biomass. The addition of Triton X-100 into solid medium also resulted in decrease in the colony radial growth rate and this response was correlated with the formation of mycelia which showed increase in branching and septation in the presence of Triton X-100.

Development of Predictive Growth Model of Listeria monocytogenes Using Mathematical Quantitative Assessment Model (수학적 정량평가모델을 이용한 Listeria monocytogenes의 성장 예측모델의 개발)

  • Growth curves of Listeria monocytogenes in modified surimi-based imitation crab (MIC) broth were obtained by measuring cell concentration in MIC broth at different culture conditions [initial cell numbers, $1.0{\times}10^{2},\;1.0{\times}10^{3}\;and\;1.0{\times}10^{4}$, colony forming unit (CFU)/mL; temperature, 15, 20, 25, 37, and $40^{\circ}C$] and applied to Gompertz model to determine microbial growth indicators, maximum specific growth rate constant (k), lag time (LT), and generation time (GT). Maximum specific growth rate of L. monocytogenes increased rapidly with increasing temperature and reached maximum at $37^{\circ}C$, whereas LT and GT decreased with increasing temperature and reached minimum at $37^{\circ}C$. Initial cell number had no effect on k, LT, and GT (p > 0.05). Polynomial and square root models were developed to express combined effects of temperature and initial cell number using Gauss-Newton Algorism. Relative coefficients of experimental k and predicted k of polynomial and square root models were 0.92 and 0.95, respectively, based on response surface model. Results indicate L. monocytogenes growth was mainly affected by temperature and square root model was more effective than polynomial model for growth prediction.

Growth Performance and Digestive Characteristics of Starry Flounder Platichthys stellatus on the Moist and Extruded Pellets (습사료와 부상사료에 대한 강도다리(Platichthys stellatus)의 성장과 소화 특성)

  • A study was carried out to observe the effects of feed types on the growth, feed preference, and enteric feed transition rate of juvenile starry flounder, Platichthys stellatus for 45 days. Fifty fish (avg. 135 g) were stocked each in replication, and fed a commercial extruded pellet diet (EP, 45% protein) and a moist pellet diet (MP, 65% raw mackerel+35% feed powder in wet basis), respectively. The MP presented the higher performance than that of the EP on the feed efficiency ($68.3{\pm}0.9%$ for EP and $92.3{\pm}4.3%$ for MP) and the specific growth rate ($1.07{\pm}0.07$ for the EP and $1.20{\pm}0.05%$ for the MP). In contrast, the EP showed the higher feed preference in terms of the daily feed intake ($1.57{\pm}0.08%$ for the EP and $1.30{\pm}0.01$ for the MP) and the ad libitum feeding rate after a fast of 72 hours (1.73% for the EP and 1.35% for the MP). The feed transition rate through intestinal canals decreased exponentially in both the EP and the MP, showing the faster transition rate with the EP. In the result, starry flounder appeared to have the better feed preference to the EP, but have the higher feed efficiency and growth performance to the MP.

  • Under rate-controlled sintering, furnace power is controlled to maintain a specific specimen contraction rate. This thermal processing method guarantees continuous process with a minimum thermal energy applied over time and makes it possible to control the density of the sintered body precisely. In this study, the rate-controlled sintering is applied to the sintering of $B_4C$ in order to investigate how rate-controlled sintering variables can affect the sintering behavior and/or grain growth behavior of $B_4C$ and how the results can be interpreted using sintering theories to draw an optimal sintering condition of the rate-controlled sintering. Further, the applicability of the rate-controlled sintering into the study for sintering of unknown materials is also considered.

Hollow Fiber Dialysis Culture of E. coli (대장균의 실관투석배양)

  • A hollow fiber device was utilized to perform a dialysis culture for E. coli. Acetic acid inhibition on the growth of E. coli was relieved by dialyzing the acid from broth into a dialysate reservoir. The rate of acetic acid formation was very sensitive to the concentration of glucose and dissolved oxygen. Therefore it was found that the glucose permeation rate should be balanced with the oxygen supply rate. Specific growth rate of E. coli was determined by the glucose permeation rate through membrane. Under a low permeation rate, acetic acid formation was depressed in accordance with high dissolved oxygen concentration as well as low glucose concentration.

Effects of sodium bicarbonate as an inorganic carbon source on the growth of scenedesmus dimorphus (무기탄소원으로서의 NaHCO3가 미세조류 Scenedesmus dimorphus의 성장에 미치는 영향 평가)

  • This study investigates the effect of sodium bicarbonate ($NaHCO_3$) on growth of S.dimorphus. $NaHCO_3$ concentration was varied from 0 to 2 g-C/L. As a result, the increase in concentration of $NaHCO_3$ up to 1.5 g-C/L increased dry weight of algae. The highest specific growth rate of S. dimorphus was $0.36day^{-1}$ which was obtained at concentration of 0.5 g-C/L $NaHCO_3$. pH showed a large variation range at the concentrations lower than 0.5 g-C/L $NaHCO_3$ whereas inorganic carbon, nitrate and phosphorus removal rates were almost same at the concentrations higher than 0.5 g-C/L $NaHCO_3$ (0.75, 1, 1.25, 1.5, 2 g-C/L $NaHCO_3$). Their average inorganic carbon, nitrate and phosphorus removal rate were 70 mg-C/L/d, 11.3 mg-N/L/d, and 1.6 mg-P/L/d, respectively. Thus, $NaHCO_3$ didn't effect on inorganic carbon, nitrate and phosphorus removal rate of S. dimorphus.

Optimal Temperature and Light Intensity for Improved Mixotrophic Metabolism of Chlorella sorokiniana Treating Livestock Wastewater

  • Mixotrophic microalgal growth gives a great premise for wastewater treatment based on photoautotrophic nutrient utilization and heterotrophic organic removal while producing renewable biomass. There remains a need for a control strategy to enrich them in a photobioreactor. This study performed a series of batch experiments using a mixotroph, Chlorella sorokiniana, to characterize optimal guidelines of mixotrophic growth based on a statistical design of the experiment. Using a central composite design, this study evaluated how temperature and light irradiance are associated with $CO_2$ capture and organic carbon respiration through biomass production and ammonia removal kinetics. By conducting regressions on the experimental data, response surfaces were created to suggest proper ranges of temperature and light irradiance that mixotrophs can beneficially use as two types of energy sources. The results identified that efficient mixotrophic metabolism of Chlorella sorokiniana for organics and inorganics occurs at the temperature of $30-40^{\circ}C$ and diurnal light condition of $150-200{\mu}mol\;E{\cdot}m^{-2}{\cdot}s^{-1}$. The optimal specific growth rate and ammonia removal rate were recorded as 0.51/d and 0.56/h on average, respectively, and the confirmation test verified that the organic removal rate was $105mg\;COD{\cdot}l^{-1}{\cdot}d^{-1}$. These results support the development of a viable option for sustainable treatment and effluent quality management of problematic livestock wastewater.

Growth Characteristics of Juvenile Abalone, Haliotis discus hannai by Reared Methods in order to High Density Intermediate Culture in Land-based Tank (육성수조 내 북방전복, Haliotis discus hannai 치패 고밀도 중간양성 사육방식별 성장특성)

  • The effect of different intermediated rearing method by expanding the attached floor space in order to highly density culture on the growth characteristics and survival rate of the juvenile abalone, Haliotis discus hannai, were investigated in land-based tanks. The intermediated culture methods was determined thru the shelter counts and layer for 10 month with two replicates : the single layer shelter (SLS), the double layer shelter (DLS), the triple layer shelter (TLS) and the single layer shelter under net cage (SLSNC). In addition, the culture on shallow race way tank had to set up as culture of the ditch raceway tank (CDRT) and the floor race way tank (CFRT). In the growth performance of reared abalone (initial mean shell length $54.18{\pm}7.39mm$ and weight $1.93{\pm}0.14g$) at experimental tanks, that the absolute growth rate (ARG), daily growth rate (DGR) and specific growth rate (SGR) to the shell length and shell breadth was not significant at each experimental tanks except SLSNC. As well as too, weight gain (WG), daily weight gain (DWG) and specific weight gain (SWG) to weight was not significant at each experimental tanks except SLSNC, too. Survival rates of CDRT and CFRT was lower than those of different experimental tanks (P < 0.05). Therefore, these results is showed that high density different intermediated rearing method by expanding the attached floor space for juvenile H. discus hannai was not have difference as growth performance and survival rate both one layer shelter and multi layer shelter. Also, it is considered that shallow race way tank was not useful rearing for the juvenile intermediate culture of H. discus hannai in land based.

Kinetics of L-Phenylalanine Production by Corynebacterium glutamicum (Corynebacterium glutamicum에 의한 L-Phenylalanine 생산의 동역학적 특성)

  • Microbial production of L-phenylalanine using Corynebacterium glutamicum ATCC 21674, a tyrosine auxotroph resistant to aromatic amino acid analogues, has been studied and kinetic analysis was performed. Even though the strain was reported as a tyrosine auxotroph, it produced tyrosine and was able to grow on the minimal medium where no tyrosine was present. The average specific growth rate at the exponential growth phase was 0.087 hr-1. There was a dissociation of growth from the formation of the product. Linear correlation between biomass production and total CO2 production was obtained. The relationship between CO2 evolution rate and sugar consumption rate was also found to be linear.
