Age Variation in Extractive Nitrogenous Constituents of the Cultured Ascidian, Halocynthia roretzi Muscle (양식산 우렁쉥이, Halocynthia roretzi 함질소 엑스성분의 연령차)

  • The muscle extracts of the ascidian, Halocynthia roretzi cultured for two and three years old on the southern coast near Chungmu and the eastern coast near Pohang of Korea, were analyzed for extractive nitrogen (EN), free amino acids (FAA), combined amino acids (CAA), nucleotides and related compounds (NRC), quaternary ammonium bases and guanidino compounds using specimens collected in February 1989 and in April 1989, and compared for those contents with each other. As for the amount of EN, no remarkable difference was found between two- and three-year-old samples collected at St. 1 in the spring and winter seasons, while at St. 2 in the spring season the two-year-old sample was distinctly lower than the three-year-old one. Taurine, proline, glutamic acid, glycine and alanine were the major FAA in every sample. The amount of taurine, the most prominent FAA, was higher in three-year-old sample than in two-year-old one regardless of sampling station and season. Most of the other major FAA showed a similar tendency to EN at both sampling stations in both seasons. Adenosine 5'-triphosphate (ATP), adenosine 5'-diphosphate (ADP), adenosine 5'-monophosphate (AMP), inosine 5'-monophosphate (IMP), inosine (Ino) and hypoxanthine (Hyp) were detected in all the samples and ATP, ADP and AMP were the major ingredients. The amounts of total NRC were in parallel with those of EN and total FAA. As for the contents of betaines, two- and three-year-old samples collected in the winter season exhibited a great discrepancy each other, the former being clearly lower than the latter, but no remarkable difference was observed between two samples of two groups in the spring season. In proximate composition of the muscles, the two-year-old sample was considerably higher in moisture content and lower in protein and glycogen contents than the three-year-old one at St. 2 in the spring season. The large discrepancies observed between two- and three-year-old samples from St. 2 seems to be attributable to the difference in size of samples rather than to the difference in age.

Functional Characterization of the Gene Encoding UDP-glucose: Tetrahydrobiopterin $\alpha$-Glucosyltransferase in Synechococcus sp. PCC 7942

  • In this study, we attempted to characterize the Synechococcus sp. pee 7942 mutant resultant from a disruption in the gene encoding UDP-glucose: tetrahydrobiopterin a-glucosyltransferase (BGluT). 2D­PAGE followed by MALDI-TOF mass spectrometry revealed that phycocyanin rod linker protein 33K was one of the proteins expressed at lower level in the BGluT mutant. BGluT mutant cells were also determined to be more sensitive to high light stress. This is because photosynthetic O$_2$ exchange rates were significantly decreased, due to the reduced number of functional PSIs relative to the wild type cells. These results suggested that, in Synechococcus sp. pee 7942, BH4-glucoside might be involved in photosynthetic photoprotection.

Antifungical Activity of Autochthonous Bacillus subtilis Isolated from Prosopis juliflora against Phytopathogenic Fungi

  • The ability of Bacillus subtilis, strain ALICA to produce three mycolytic enzymes (chitinase, ${\beta}$-1,3-glucanase, and protease), was carried out by the chemical standard methods. Bacillus subtilis ALICA was screened based on their antifungal activity in dual plate assay and cell-free culture filtrate (25%) against five different phytopathogenic fungi Alternaria alternata, Macrophomina sp., Colletotrichum gloeosporioides, Botrytis cinerea, and Sclerotium rolfesii. The B. subtilis ALICA detected positive for chitinase, ${\beta}$-1,3-glucanase and protease enzymes. Fungal growth inhibition by both strain ALICA and its cell-free culture filtrate ranged from 51.36% to 86.3% and 38.43% to 68.6%, respectively. Moreover, hyphal morphological changes like damage, broken, swelling, distortions abnormal morphology were observed. Genes expression of protease, ${\beta}$-1,3-glucanase, and lipopeptides (subtilosin and subtilisin) were confirmed their presence in the supernatant of strain ALICA. Our findings indicated that strain ALICA provided a broad spectrum of antifungal activities against various phytopathogenic fungi and may be a potential effective alternative to chemical fungicides.

Novel Sources of Resistance to Phytophthora capsici on Pepper (Capsicum sp.) Landraces from Mexico

  • Phytophthora capsici Leonian is a major pathogen of pepper worldwide and few resistance sources to this pathogen have been identified so far. The goals of this study were to identify new sources of resistance against P. capsici in Capsicum landraces and analyze the relationship between the resistance indicator of plant symptoms and some plant phenotype parameters of plant height, stem width, leaf length and leaf width. Thirty-two landraces of pepper were collected from fourteen states in Mexico. From each population, 36 plants were inoculated with 10,000 zoospores of P. capsici under controlled conditions. This experiment was repeated twice. Out of the 32 landraces, six showed high level of resistance, four showed intermediate resistance and five showed low level of resistance when compared with the susceptible control 'Bravo' and the resistant control 'CM334', indicating that these landraces are promising novel sources of resistance to P. capsici. There was no correlation between the symptoms and plant phenotype parameters. However, these parameters were not affected in the group classified as highly resistant, indicating that P. capsici does not affect the growing of these resistant pepper landraces. The other resistant groups were significantly affected in a differently manner regarding their phenotype, indicating that this pathogen reduce their growth in different ways. This study reports novel resistance sources with great potential that could be used in breeding programs to develop new pepper cultivars with durable resistance to P. capsici.

Lithothamnion steneckii sp. nov. and Pneophyllum conicum: new coralline red algae (Corallinales, Rhodophyta) for coral reefs of Brazil

  • Nongeniculate coralline red algae are a common element of the Brazilian coastal zone, especially associated to coral reefs. During the course of ecological studies at Parque Municipal Marinho do Recife de Fora, two species of non-geniculate Corallinales were the major organisms covering the reef. Analyses of the vegetative and reproductive features of the species were analyzed; indicating that one new species of the genus Lithothamnion is proposed here based on the combination of several features associated with anatomy of the tetrasporangial conceptacles in relation to other species of the genus for which modern accounts are available. This new proposal along with other new species, new combinations and range extension of some species of the genus based in similar features clearly suggest that stability in species delimitation is possible. The second species found Pneophyllum conicum represents a range extension of more than 6,000 km from the Pacific in to the Atlantic Ocean suggesting that some nongeniculate species are widely distributed. The occurrence and abundance of these species supports and emphasizes the need for an extensive taxonomic reassessment of coralline red algae in the context of Brazilian coral reef biodiversity.

Par exemple, ce mouvement a $commenc{\acute{e}}$ avec les metteurs en $sc{\grave{e}}ne$ OH Tae-suk et GUI Gook-seo au $d{\acute{e}}but$ des $ann{\acute{e}}es$ 1990. Leur ont $succ{\acute{e}}d{\acute{e}}$ ensuite les metteurs en $sc{\grave{e}}ne$ CHOI Yong-hoon, de la troupe de $th{\acute{e}}{\hat{a}}tre$ Chak Eun Shin Hwa, et PARK Kun-hyung. Nitrite Removal Characteristics and Application of Bosea sp. Isolated from BFT System Culture Water (BFT 시스템 사육 수에서 분리한 Bosea sp.의 아질산 제거 특성과 활용)

  • This study was performed to isolate bacteria that could control the nitrite levels in a biofloc technology (BFT) culture tank. Nitrite-eliminating bacteria were isolated from a BFT culture tank rearing goldfish, and the isolated bacterium exhibiting the most potent nitrite eliminating ability was labeled as the "NOBSB1" strain. Sequencing the 16S rRNA revealed that NOBSB1 is a species in the genera Bosea. NOBSB1 had the following characteristics with regard to nitrite removal: (1) it removed nitrite by functioning heterotrophically in the presence of a carbon source (sugars); (2) it eliminated nitrite most effectively within a temperature range of $20-30^{\circ}C$, but its activity decreased at temperatures above $35^{\circ}C$ and below $20^{\circ}C$; (3) it had optimum nitrite removal ability within a pH range of 6.0-8.0; (4) it removed nitrite more effectively under hypoxic than aerobic conditions. NOBSB1 inoculation did not decrease ammonia or nitrate levels, but eliminated nitrite in a BFT culture tank rearing common carp (Cyprinus carpio). After inoculating the NOBSB1 strain in a BFT culture tank, NOBSB1 controlled and sufficiently reduced the nitrite concentration in the tank.

Preparation and Electrochemical Properties of LiFePO4-PSS Composite Cathode for Lithium-ion Batteries

  • In this study, we prepared $LiFePO_4$- poly (sodium 4-styrenesulfonate) (PSS) composite by the hydrothermal method and ball-milling process. Different wt% PSS were added to $LiFePO_4$. The cathode electrodes were made from mixtures of $LiFePO_4$-PSS: SP-270: PVDF in a weighting ratio of 70%: 25%: 5%. $LiFePO_4$-PSS powders were characterized by X-ray diffraction (XRD), and scanning electron microscopy (SEM). The electrochemical properties of $LiFePO_4$-PSS/Li batteries were analyzed by cyclic voltammetry, charge/discharge tests, and AC impedance spectroscopy. A Li/$LiFePO_4$-PSS battery with 4.75 wt% PSS shows the best electrochemical properties, with a discharge capacity of 128 mAh/g.

  • The Medical Subject Headings (MeSH) thesaurus is a controlled vocabulary for indexing biomedical documents that is used for document retrieval and other natural language processing purposes. However, although the oariginal English MeSH is freely available, its Japanese translation has a restricted license. We attempted to create an open alternative, and for this purpose we made a script for assigning MeSH UIDs to Japanese medical terms using Japanese-English glossaries. From the MeSpEn glossary and MEDUTX dictionary, we generated a 12,457-word Japanese-MeSH dictionary.

Screening of Salicylic acid Producing Rhizobacteria Isolated from Plant Roots and Rhizosphere (식물의 뿌리와 근권으로부터 Salicylic acid를 생성하는 근권세균의 검색)

  • Twenty two rhizobacteria were isolated from the roots and rhizosphere of radish, carnation, potato and tomato. There isolates produced a fluorescent pigment in King's B medium and identified as Pseudomonas spp. These isolates colonized roots and rhizosphere of the host plants. In the study of cultural characteristics of the bacteria, the pH of the culture broth was changed from neutral (7.0) to alkali (8.8∼9.41) and the numbers of cells were increased from 106 to 108 after 40 hr of incubation in basal standard succinate medium. The salicylic acid production identified by pink color reaction were observed in 7 bacteria. Out of these 7 salicylic acid producing bacteria, only 2 strains of bacteria such as Pseudomonas fluorescens RS006, and Pseudomonas sp. EN401 were confirmed as salicylic acid producers by optical density measurement. Therefore, for screening of salicylic acid producing bacteria from the roots and rhizosphere, color reaction of the culture medium should be done in the first step, and then optical density measurement of culture extract should be made for the confirmation of salicylic acid production.
