Effects of Legumes Consumption on the Association of Cholesterol and Bone Mineral Density in Ovariectomized Rats (난소 절제한 동물모델에서 콩의 섭취가 콜레스테롤과 BMD와의 상관관계에 미치는 효과)

  • Soy isoflavones have been suggested to improve bone loss and lipid profile in postmenopausal women and ovariectomized rats. In present study, we investigated the hypothesis that consumption of soybean, mung bean, cowpea and azuki bean has a beneficial effect on lipid profile which associates with bone mass in ovariectomized rats. Forty two female Sprague-Dawley rats were either sham-operated (Sham) or surgically ovariectomized (OVX). Sham and OVX groups were fed a regular AIN-93M diet, but ovariectomized rats with soybean (OS), mung bean (OM), cowpea (OC) or adzuki bean (OA) were fed AIN-93M diet replacing 35% of corn starch with powdered OS, OM, OC or OA for 10 weeks. Total- and LDL-cholesterol concentrations were significantly lower in Sham and OC groups than other OVX groups. There was significant negative association between total cholesterol concentration and bone mineral density (BMD) of tibia in only OC group. In conclusion, total-cholesterol concentration was significantly and negatively correlated with BMD in rats consumed cowpea, suggesting that the reduced cholesterol concentration may have a beneficial effect on bone mass.

Selection of rs2rs2titi Soybean Genotype with Yellow Seed Coat (rs2rs2titi 유전자형을 가진 노란 콩 계통 선발)

  • Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. But, antinutritional factors in the raw mature soybean are exist. Kunitz trypsin inhibitor (KTI) protein and stachyose are main antinutritional factors in soybean seed. The genetic removal of the antinutritional factors will improve the nutritional value of soybean seed. The objective of this research was to breed a new yellow soybean strains (rs2rs2titi genotype) with the traits of lacking of KTI protein and low content of stachyose. Breeding population was developed from the cross of "Jinyangkong" and 15G1 parents. Presence or absence of KTI protein was detected based on Western Blot technique. Content of stachyose in mature seed was detected by HPLC. Total four new strains (603-1, 603-2, 625, and 694) with KTI protein free and low content of stachyose were selected. Four strains (603-1, 603-2, 625, and 694) have yellow seed coat and hilum. Plant height of 603-1 strain was 65 cm and 100-seed weight was 29.2 g. Plant height of 603-2 strain was 66 cm and 100-seed weight was 26.2 g. Plant height of 625 strain was 64 cm and 100-seed weight was 27.1 g. Content of stachyose for four new strains was 3.0~3.50 g/kg. Four strains selected in this research will be used to improve new yellow soybean cultivar with KTI protein free, and low content of stachyose.

Identification of Quantitative Trait Loci Associated with Isoflavone Contents in Soybean Seed

  • Soybean seeds contain high amounts of isoflavones that display biological effects and isoflavone content of soybean seed can vary by year, environment, and genotype. Objective of this study was to identify quantitative trait loci that underlie isoflavone content in soybean seeds. The study involved 85 $F_2$ populations derived from Korean soybean cultivar 'Kwangkyo' and wild type soybean 'IT182305' for QTL analysis associated with isoflavone content. Isoflavone content of seeds was determined by HPLC. The genetic map of 33 linkage groups with 207 markers was constructed. The linkage map spanned 2,607.5 cM across all 33 linkage groups. The average linkage distance between pair of markers among all linkage groups was 12.6 cM in Kosambi map units. Isoflavone content in $F_2$ generations varied in a fashion that suggested a continuous, polygenic inheritance. Eleven markers (4 RAPD, 3 SSR, 4 AFLP) were significantly associated with isoflavone content. Only two markers, Satt419 and CTCGAG3 had F-tests that were significant at P<0.01 in $F_2$ generation for isoflavone content. Interval mapping using the $F_2$ data revealed only two putative QTLs for isoflavone content. The peak QTL region on linkage group 3, which was near OPAG03c, explained $14\%$ variation for isoflavone content. The peak QTL region on linkage group 5, which was located near OPN14 accounted for $35.3\%$ variation for isoflavone content. Using both Map-Maker-QTL $(LOD{\geq}2.0)$ and single-factor analysis $(P{\leq}0.05)$, one marker, CTCGAG3 in linkage group 3 was associated with QTLs for isoflavone content. This information would then be used in identification of QTLs for isoflavone content with precision

Comparison of agronomic characteristics of colored soybean landraces and selection of useful genetic resources

  • This study was conducted to compare the agronomic traits of well-known colored native soybean germplasms. Recently, we are increasingly interested in colored as various functional ingredient of soybeans have revealed. We used a total of 396 soybean genetic resources, consisting of ten "Seonbijabikong", 110 "Jyinunikong", 276 "Seoritaekong". We sowed on 10th June 2018 at the field of Nongsaensmyeongro in Jeonju city. The average number of days to flowering, days to maturing and days to growth of colored soybean were 53, 84 and 136 days, respectively. Days to flowering of "Seonbijabikong" were ranged from 41 to 50 days with an average 48 days, those of "Jyinunikong" were ranged from 39 to 72 days with an average 52 days. Days to flowering of "Seoritaekong" were ranged from 35 to 63 days with an average 54 days, which were earlier in "Seonbijabikong", and similar with "Seoritaekong" and "Jyinunikong". Days to growth of "Seonbijabikong" were ranged from 125 to 137 days with an average 132 days, those of "Jyinunikong" were ranged from 91 to 144 days with an average 130 days and those of "Seoritaekong" were ranged from 99 to 150 days with an average 139 days. they were shortest in "Jyinunikong" and longest in "Seoritaekong". The distribution of maturity period was from 6th September to 5th November. The maturity period was as early as September and yields were more than 100g per plant, which were all three accessions(IT161905, IT162602, IT269617), "Seoritaekong". They would be useful as breeding materials of colored soybean with early maturity. The 100-seed weight is important characteristics that distinguish the usage of soybeans. "Seoritaekong" and "Seonbijabikong" have large seed characteristics for cooking with rice, "Jyinunikong" has small seed it for medicine. The average 100 seed weight was 35.0g of "Seonbijabikong", 30.8g of "Seoritaekong" and 13.4g of "Jyinunikong", respectively. As for seed coat lust, the ratio of dull was as high as 100% of "Seonbijabikong" and 91% of "Seoritaekong", that of shiny was as high as 77% of "Jyinunikong". Cotyledon color of "Seonbijabikong" were all yellow, that of "Seoritaekong" were 94% of green. The other name of "Seoritaekong" is "Sokcheong", which means that cotyledon color is green. Therefore if cotyledon color of Seoritaekong is not green, it might be misidentified. In the future, we will increase the utilization through evaluation of functional component such as isoflavones and anthocyanins of colored soybean landraces.

Manipulating Isoflavone Levels in Plants

  • Metabolic engineering for production of isoflavones in nonlegume plants could distribute the health benefits of these phytoestrogens in more widely-consumed grains. Series of investigation to check the ability of the heterologous isoflavone synthase enzyme to interact with the endogenous phenylpropanoid pathway have been conducted. Overall, results provide possibility of production of isoflavonoids in several plant tissue systems including soybean and nonlegumes. In tissue that undergoes naturally enhanced synthesis of anthocyanins, genistein production was enhanced. In a monocot cell system, introduced expression of a transcription factor regulating genes of the anthocyanin pathway was effective in conferring the ability to produce genistein in the presence of the isoflavone synthase gene. However, in this case the intermediate accumulated to high levels indicating an inefficiency in its conversion. Introduction of a third gene, chalcone reductase, provided the ability to synthesize an additional substrate of isoflavone synthase resulting in production of the isoflavone daidzein. These research efforts provide insight into requirements for metabolic engineering for isoflavone production in nonlegume dicot and monocot tissues.

Effects of phytoestrogen on sexual development

  • Phytoestrogen is an estrogenic compound that occurs naturally in plants. The most common sources of phytoestrogen are soybean products, which contain high levels of isoflavones. This compound, which has structural similarity with estrogen, can act as an estrogen receptor agonist or antagonist. Animal studies provide evidence of the significant effects of phytoestrogen on sexual development, including altered pubertal timing, impaired estrous cycling and ovarian function, and altered hypothalamus and pituitary functions. Although human studies examining the effects of phytoestrogen on sexual development are extremely limited, the results of some studies agree with those of the animal studies. In this paper, we review the possible mechanism of phytoestrogen action and the evidence showing the effects of phytoestrogen on sexual development in animal and human studies.

Antioxidant activity and anti-inflammatory activity of ethanol extract and fractions of Doenjang in LPS-stimulated RAW 264.7 macrophages

  • BACKGROUND/OBJECTIVES: Fermentation can increase functional compounds in fermented soybean products, thereby improving antioxidant and/or anti-inflammatory activities. We investigated the changes in the contents of phenolics and isoflavones, antioxidant activity and anti-inflammatory activity of Doenjang during fermentation and aging. MATERIALS/METHODS: Doenjang was made by inoculating Aspergillus oryzae and Bacillus licheniformis in soybeans, fermenting and aging for 1, 3, 6, 8, and 12 months (D1, D3, D6, D8, and D12). Doenjang was extracted using ethanol, and sequentially fractioned by hexane, dichloromethane (DM), ethylacetate (EA), n-butanol, and water. The contents of total phenolics, flavonoids and isoflavones, 2,2-diphenyl-1 picryl hydrazyl (DPPH) radical scavenging activity, and ferric reducing antioxidant power (FRAP) were measured. Anti-inflammatory effects in terms of nitric oxide (NO), prostaglandin (PG) E2 and pro-inflammatory cytokine production and inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expressions were also measured using LPS-treated RAW 264.7 macrophages. RESULTS: Total phenolic and flavonoid contents showed a gradual increase during fermentation and 6 months of aging and were sustained thereafter. DPPH radical scavenging activity and FRAP were increased by fermentation. FRAP was further increased by aging, but DPPH radical scavenging activity was not. Total isoflavone and glycoside contents decreased during fermentation and the aging process, while aglycone content and its proportion increased up to 3 or 6 months of aging and then showed a slow decrease. DM and EA fractions of Doenjang showed much higher total phenolic and flavonoid contents, and DPPH radical scavenging activity than the others. At $100{\mu}g/mL$, DM and EA fractions of D12 showed strongly suppressed NO production to 55.6% and 52.5% of control, respectively, and PGE2 production to 25.0% and 28.3% of control with inhibition of iNOS or COX-2 protein expression in macrophages. CONCLUSIONS: Twelve month-aged Doenjang has potent antioxidant and anti-inflammatory activities with high levels of phenolics and isoflavone aglycones, and can be used as a beneficial food for human health.

Effects of Soybean Sprouts Extract Isoflavone In the Osteoporosis of Rats Caused by Ovariectomy (콩나물 추출 이소플라본이 난소적출로 유발된 골다공증 백서의 골대사에 미치는 영향)

  • The overall purpose of this study was to investigate the effects of level of isoflavones supplementation on bone metabolism in osteoporosis rats. The effects of level of isoflavones supplementation on calcium and osteocalcin blood level, femur/body weight, bone mineral density(BMD) and bone strength were inspected in this study. This study classified 28 of 12 weeks-old male Sprague Dawley rats which have osteoporosis caused by ovariectomy into four groups of 7 rats and made the subjects medicated them isoflavone. Group I was non-treatment after osteoporosis(control); Group II was low-dose isoflavone(20 mg/kg) feeding after osteoporosis; Group III was middle-dose isoflavone(40 mg/kg) feeding after osteoporosis; Group IV was high-dose isoflavone(80 mg/kg) feeding after osteoporosis; In the calcium and osteocalcin level as one of bone formation indexes, there was a statistically significant difference between the group II, III, IV compared to group I. In respect to the femur/body weight, there was a statistically significant difference between the group II, III, IV compared to group I. In the bone mineral density and bone strength test, there was a statistically significant difference between the group II, III, IV compared to group I. The above results suggests that isoflavone medicated is effective to prevention and treatment of osteoporosis.

Quality properties on soy bean pastes made with microorganisms isolated from traditional soy bean pastes (전통된장으로부터 분리한 균종으로 제조된 된장의 품질특성)

  • Quality properties of soy bean pastes made with Aspergillus oryzae and 5 Bacillus strains isolated from traditional soy bean pastes were examined. The pH decreased gradually and contents of amino-type nitrogen increased during fermentation. There were small differences in moisture and crude-protein contents, whereas big difference was observed in reducing sugar, isoflavones and organic acid contents. Isoflavones in the samples made with Bacillus licheniformis F2358 and Bacillus subtilis F2362 were high. Samples made with Bacillus licheniformis F2382 had high contents of organic acid and good score for taste and overall acceptability in sensory evaluation.

Physicochemical Properties of Soybean Leaf by Cultivar and Development of Soybean Curd Prepared with Soybean Leaf Powder (콩잎 품종에 따른 이화학적 특성 비교 및 콩잎 분말을 첨가한 두부 개발)

  • The objectives of this study were to investigate the physicochemical characteristics of soybean leaves in different cultivars and to develop a soybean curd prepared with soybean leave powder as a functional food. Four cultivars (Daewonkong leaf, Daepungkong leaf, Hwangguemkong leaf, and Seoritae leaf) were selected for this experiment. A significant difference was observed in the proximate composition of soybeans leaves (p<0.05). Soybean leaves had the highest content of carbohydrates. The mineral composition and isoflavone content in soybean leaves were significantly different among the cultivars (p<0.05). In particular, Daepongkong leaf had the highest content of genistein, daidzein, and total isoflavones. This study was also conducted to determine the quality characteristics of a soybean curd developed from daepongkong leaf with various concentrations (0, 0.1, 0.2, and 0.3%) of soybean leaf powder (SLP). The soybean curd yield rate increased according to the level of SLP added, whereas the L and a color values decreased. In the sensory evaluation, intensity scores for color, after taste, leafy taste, and chewiness were highest for the 0.3% soybean curd. The soybean curd with 0.2% SLP soybean milk attained the highest overall acceptability score. These results showed that soybean leaf was preferred over soybean curd containing 0.2% soybean leaf powder.
