Effect of Dietary Fat-soluble Vitamins on Growth Performance and Nutrient Digestibility in Growing Pigs

  • Two experiments were conducted to compare the effect of various vitamins on performance and digestibility in growing pigs. In experiment 1, a total of 54 pigs ($L{\times}Y{\times}D$, $42.73{\pm}2.40kg$) were assigned to three treatments in a randomized complete block design with three replicates (6 pigs/pen) for 40 days. The three dietary treatments were: 100% fat-soluble vitamins (FSV) and water-soluble vitamins (WSV); 150% FSV and 100% WSV of NRC (1998); and 100% FSV and 150% WSV of NRC (1998). In experiment 2, a total of 180 pigs ($L{\times}Y{\times}D$, $28.20{\pm}3.05 kg$) were assigned to four treatments in a completely randomized design with three replicates for four weeks (15 pigs/pen). The four dietary treatments were, 150% vitamin A and 100% other vitamins, 150% vitamin D and 100% other vitamins, 150% vitamin E and 100% other vitamins, and 150% vitamin K and 100% other vitamins. In experiment 1, there were significant differences in growth performance and digestibility of nutrients among the treatments. The ADG, ADFI and FCR of pigs fed 150% FSV diet were better (p<0.05) than those fed the control diet. However, there were no differences (p>0.05) in ADG, ADFI and FCR between pigs fed the control and 150% WSV diets. Digestibilities of dry matter, gross energy and calcium were improved in 150% FSV treatment group compared with control (p<0.05). However, the improvement was similar when compared with 150% WSV except for Ca. In experiment 2, there were no differences (p>0.05) in ADG, ADFI and FCR and nutrient digestibility between the fat-soluble vitamin treatments when fed at the 150% level. In conclusion, growing pigs were more responsive to additional fat-soluble vitamin supplements over the requirements suggested by NRC (1998) than to water-soluble vitamin supplements as measured by growth performance and digestibility of nutrients.

Role of Soluble Adiponectin Receptor 2 in Non-Alcoholic Fatty Liver Disease in Children

  • Purpose: The incidence of non-alcoholic fatty liver disease (NAFLD) in children is gradually increasing. The aim of this study was to investigate the use of serum adiponectin and soluble adiponectin receptor 2 (soluble Adipo R2) levels for the diagnosis of fatty liver disease in obese and overweight children. Methods: The study included 51 obese and overweight children between the ages of 6 and 18 years diagnosed with NAFLD using ultrasonography and 20 children without fatty liver disease. Patients whose alanine transaminase level was two times higher than normal (${\geq}80U/L$) were included in the non-alcoholic steatohepatitis (NASH) group. Results: NASH was observed in 11 (21.6%) of the patients with NAFLD. The incidence of obesity was higher in patients with NASH (80% and 45%, p=0.021). While the adiponectin levels were similar in patients with NAFLD and those without, they were below the normal level in the whole study group. Adiponectin and soluble Adipo R2 levels of patients with NASH were lower than those in patients without NASH; however, this difference was not statistically significant (p=0.064 and p=0.463). Soluble Adipo R2 levels in obese patients with NAFLD were higher than those in obese children without NAFLD (p<0.001). Conclusion: Soluble adiponectin receptor 2 level is a noninvasive marker that can be used for the diagnosis of NAFLD in obese children.

Effect of Environmental Factors on In Vivo Folding of Bacillus macerans Cyclodextrin Glycosyltransferase in Recombinant Escherichia coli

  • Effect of environmental factors on the expression of soluble forms of Bacillus macerans cyclodextrin glycosyltransferase in recombinant Escherichia coli BL21(DE3)pLysE:pTCGT1 were investigated. The amount of soluble CGTase produced in the cell was measured by determining its enzymatic activity. The soluble fractionof the enzyme was increased by lowering the culture temperature to $30{\circ}C$ and medium pH to 5.8 compared to the enzyme production in LB medium at $37^{\circ}C$ and pH7.0. Addition of 0.2 M NaCl enhanced enzyme expression levels at the expense of cell growth. Glycine betaine that was added after 3 h of induction protected not only the cell growth from hig osmotic pressue but also hepld in vivo folding of CGTase in recombinant E. coli. Addition of 1 mM $CaCl_2$ was also effective in the expression of soluble CGTase, resulting in 15 U/ml of the enzyme activity.

산란 노계육에서 추출한 염용성 단백질의 열변성에 관한 연구 (Heat-Induced Denaturation of Salt Soluble Protein Extracted from Spent Layer Meat)

  • Effects of protein concentration, ionic strength, pH, and temperature range on the heat-induced denaturation of salt soluble protein extracted from spent layer meat were investigated. Viscosity of salt soluble protein heated at 65$^{\circ}C$ for 30 min began to increase sharply above 7 mg/ml of breast protein concentration, and above 21 mg/ml of leg protein concentration, respectively. Both turbidity and viscosity showed the highest value in cooked protein solution with pH 6.0 and 1% NaCl. The turbidity of salt soluble protein started to increase continuously from 40$^{\circ}C$ to 80$^{\circ}C$. The viscosity increased rapidly from 45$^{\circ}C$ to 60$^{\circ}C$ in breast protein, and increased from 50$^{\circ}C$ to 55$^{\circ}C$ in leg protein, respectively, and then kept relatively constant. Breast protein had higher viscosity than leg protein during heat-induced gelation. Therefore, salt soluble protein from spent layer meat was associated with denatured protein (turbidity change) prior to gelation (viscosity change) during heating. Breast protein showed lower thermal transition temperature, and better gel formation than leg protein during heating.

Doping Effects of Water-Soluble Poly(3-thiopheneacetic acid) (P3TAA) and its Gel

  • The doping behaviors of water-soluble poly(3-thiopheneacetic acid) (P3TAA) and ie gel using I$_2$and concentrated HCIO$_4$aqueous solutions were investigated by UV-Visible absorption spectrometer. Electrical conductivity of tile doped polyhiophene gel was also studied. I$_2$-doping of water-soluble P3TAA gave rise to a new broad polaron peak at around 749 nm, which corresponds to localization of electron. It was found that doping ability of P3TAA gel was strikingly dependent on the concentration of HCIQ solution.

Genetic Analysis of Ginseng Germplasm by Lactate Polyacrylamide Gel Electrophoresis of Seed Protein

  • Systematic electrophoretic analysis of alcohol-soluble proteins and salt-soluble proteins of 247 Panax ginseng (P.g) and Panax quinquefolium (P.q) germplasms seed was carried out on an improved lactate-polyacrylamide gel electrophoresis, a method with high resolving power, good reproducibility and stability. The electrophoregrams of proteins, according to their migration rate, were classified into four groups such as ${\alpha}$, ${\beta}$, ${\gamma}$ and $\omega$ for the alcohol-soluble proteins and three such as I, II and III for the salt-soluble ones. Panax ginseng or Panax quinquefolium had their own unique band pattern distinguishable from each other, regarding as their specific "fingerprint". In this study, 3 of 168 (1.8%) P.g germplasms and 1 of 79 (1.3%) P.q germplasms had their own unique band pattern, showing that P.g and P.q germplasms have poor genetic diversity in species. The band patterns of dry seed and stratified seed (embryo rate=60%) were basically the same. The band number of the F, hybrid of p.gx p.q was exactly equivalent to the number of the common bands plus the specific bands of the two parents, indicating that the difference of band patterns was a genetic trait con- trolled by the nuclear genes. The electrophoregram of F1 of P.g x P.q could be predicted by that of the two parents and the band pattern of the F1 hybrids could be demnonstrated by that of the mixed seed extract from the two parents.

인산축적 시설 재배지 토양에서 작물재배에 따른 무기태 인의 형태별 함량 변화 (Changes in Inorganic Phosphorus Fractions in P Accumulated Plastic Film House Soils under Different Cropping Condition)

  • 인산이 과다하게 집적된 경작년수가 3, 8 및 16년 된 시설 재배지 토양을 대상으로 작물 연속재배에 따른 토양 무기태 인산의 형태별 함량 변화를 조사하고자 pot 재배실험을 수행하였다. 각 작물재배 전에 인산질 비료로서 용성인비를 사용하여 0, 100 또는 $200kg\;P\;ha^{-1}$(추천시비량), 200 또는 $400kg\;P\;ha^{-1}$(추천시비량의 2배)의 3수준으로 처리하여 옥수수-배추-배추-옥수수를 연속 재배하면서 토양 총 인산, 유효 인산 및 토양 무기태 인산의 형태별 함량을 정량하였다. 작물 재배 전 토양은 Ca-P와 Al-P 가 무기태 인산의 주성분으로서 총 인산에 대한 비율은 30% 이상이었다. 토양 중 이용성 인산과 Bray-1 P 간의 상관계수는 $0.839^{**}{\sim}0.952^{**}$, Lancaster P 와는 $0.895^{**}{\sim}0.967^{**}$, Olsen P 와는 $0.491^{**}{\sim}0.821^{**}$로서 토양 이용성 인산의 함량과 토양 유효인산 함량간에는 유의성 높은 정의 상관관계가 존재하였다. 작물재배에 따라 토양 이용성 인산의 감소량과 작물에 의해 흡수된 인산량 간에는 고도의 정의 상관관계가 ($r=0.644^{**}{\sim}0.822^{**}$) 존재하였다. 토양 이용성 인산 함량의 작물 재배회수에 따른 감소 경향은 1차 속도반응식으로 표현할 수 있었으며, 이 식을 이용하여 토양 이용성 인산의 함량이 $0.2mg\;P\;kg^{-1}$까지 감소하는데 소요되는 작물 재배 회수는 각 토양에 대하여 26~33회로 예측되었다. 토양 Al-P의 함량은 작물재배 기간동안 변화가 작았으며 작물의 인산 흡수량과는 상관관계가 없었다. 인산질 비료의 시용량과 관계없이 토양 총 인산에 대한 Fe-P의 비율은 작물 재배에 따라 증가하였다. 토양의 Ca-P 비율은 작물재배 전 토양에서도 30% 이상으로 매우 높은 수준에도 불구하고, 인산 무처리구에서도 작물 재배에 따라 지속적으로 증가하는 경향이었다. 작물 연속재배에 따라 토양 총 인산에 대한 reductant soluble-P의 비율은 인산 처리구에서는 변화가 거의 없었으나 인산 무처리구에서는 증가하는 경향이었으며, 토양의 residual-P의 비율은 인산질 비료의 시용량과 관계없이 작물 재배에 따라 감소하였다.

다양한 깔짚 첨가제 이용이 단 기간 깔짚내 pH와 수용성 인에 미치는 영향에 관한 연구 (A Study on pH and Soluble Reactive Phosphorus (SRP) from Litter Using Various Poultry Litter Amendments During Short-Term: A Laboratory Experiment)

  • The objective of this study was conducted to evaluate the effects of poultry litter amendments on pH and soluble reactive phosphorus (SRP) in poultry litter. Two laboratory studies were conducted for 42 d in Exp. 1 and for 10 d in Exp. 2, respectively. The poultry litter was treated with various amendments which included 4 g fly ash and 4 g $AlCl_3\;(AlCl_36H_2O)/100g$ litter in Exp. 1 and 4 g alum$(Al_2(SO_4){_3}\;14H_2O)$, 8 g alum, 8.66 g liquid alum, and 17.3 g liquid alum/100 g litter in Exp. 2; untreated litter served as controls. There were no differences in pH between control and T1(4 g fly ash) and SRP contents between T1(4 g fly ash) and T2(4 g $AlCl_3$) in Exp. 1. A significant difference in pH and SRP contents in Exp. 2 was observed among all treatments(P< 0.05). In experiment 1, T1(4 g fly ash) and T2(4 g $AlCl_3$) at 42 d decreased SRP in litter by 47.1% and 62.6% of that from litter alone, respectively. In experiment 2, T1(4 g alum), T2(8.66 g liquid alum), T3(8 g alum), and T4(17.3 g liquid alum) treatments at 10 days reduced SRP contents by up to 36.2%, 62.9%, 87.0%, and 83.9%, respectively, when compared with the controls. Decrease in SRP contents was chiefly associated with reduction in litter pH. These results indicate that use of various litter amendments to limit P solubility has potential and should be pursued as a means of reducing soluble reative phosphorus during short term.

소나무 및 편백나무 수용성 정유를 이용한 Trichoderma spp.의 생장억제 활성 (Antifungal Activity against Trichoderma spp. of Water Soluble Essential Oil extracted from Pinus densiflora and Chamaecyparis obtusa)

  • 본 연구는 수목으로부터 분리된 수용성 정유를 이용하여 버섯에 병해를 일으키는 푸른곰팡이병의 원인 미생물인 Trichoderma에 대한 항균활성을 확인하고자 시도되었다. 소나무(Pinus densiflora)와 편백나무(Chamaecyparis obtusa) 잎으로부터 GAP (Gas assisted process)을 이용하여 수용성 정유를 획득하였다. 버섯의 푸른 곰팡이병을 발생시키는 Trichoderma spp. 곰팡이 5종은 $25^{\circ}C$에서 균사생장이 가장 높았으며, pH 5.0의 배지조건에서 가장 양호한 균사 생장을 나타내었다. 소나무와 편백나무 잎의 수용성 정유는 3.9% 및 3.7%의 수율을 나타내었다. 소나무와 편백나무 잎 정유의 화학적 조성은 동일 화합물이 많았으며, 동일 화합물로는 $\alpha$-Terpineol acetate, Terpinen-4-ol 및 $\alpha$-Terpineol이 확인되었다. 소나무 잎의 수용성 정유는 5000 ppm 농도에서 Trichoderma harzianum에 대하여 가장 높은 항균활성을 나타냈으며, 편백나무 잎의 수용성 정유 또한 5000 ppm 농도에서 Trichoderma atroviride에 대하여 가장 높은 항균활성을 나타내었다.

Camembert Cheese 숙성 중 Casein의 변화에 관한 연구 (Changes in Caseins during the Ripening of Camembert Cheese)

  • P. caseicolum V.B.와 복합유산균을 사용하여 제조한 Camembert cheese를 45일간 숙성시키면서 카제인 변화와 그에 따른 기호성을 조사하였다. 숙성과정 중 수용성 질소, 비카제인태 질소, 비단백태 질소량은 증가하였다. pH 4.6-insoluble casein의 전기영동 결과 숙성 10일에는 4개의 band에서 45일에 12개의 band로 분리되었는데 ${\alpha}_{s1}-casein$은 숙성 17일에 완전히 분해되었으며, ${\beta}-casein$도 숙성 45일 동안 거의 대부분이 분해되었다. 또한 pH 4.6-soluble casein fragments의 gel filtration에서 숙성 10일에는 3개의 fraction, 24일까지는 4개의 fraction이 나타났고 31일부터는 5개의 fraction으로 분별되었다. 관능검사 결과 숙성 31일된 Camembert cheese의 기호성이 제일 높았으며, 숙성이 진행됨에 따라 hardness는 감소하였고 관능적 기호성과는 0.1%의 유의수준으로 높은 상관관계를 나타냈다.
