Effect of Osmotic and Solid Matrix Priming to Improve Germination and Early Growth of Carrot and Onion Seed (Osmotic Priming 및 Solid Matrix Priming 처리에 의한 당근과 양파종자의 발아증진과 초기생육에 미치는 영향)

  • The objective of this study was carried out to determine which method is effective for improving seed germination in carrot and onion among osmotic or solid matrix priming. Seeds were osmotic primed with -0.5 MPa polyethylene glycol 8000 (PEG) solution and solid matrix primed with Micro Cel E (seed, Micro Cel E, and water mixed with the ratio of 4.0:2.0:8.0 by weight for carrot, and the ratio of 4.0:1.2:5.0 for onion at $20^{\circ}C$ for 4 days). Seeds were rapidly absorbed water after osmotic priming and solid matrix priming treatment. The moisture content of solid matrix primed seeds was lower than that of osmotic primed seeds in carrot and onion. Osmotic priming and solid matrix priming did not influenced on increasing of percent germination, but $T_{50}$ of treated seeds was shorter than those of untreated seeds at four temperature regimes. The effects of osmotic or solid matrix priming in reducing the $T_{50}$ was greater when the seeds were germinated at $15^{\circ}C$ than at $25^{\circ}C$ and $30^{\circ}C$ temperature. Solid matrix primed seeds germinated faster than osmotic primed seeds at all temperature in both carrot and onion. After priming, surface-dried seeds germinated faster than dried-back seeds in carrot and onion. Emergence speed of carrot and onion seeds was faster than untreated seeds by osmotic priming or SMP treatment although the final emergence percentage was not significantly influenced. On the other hand, early growth was not significantly influenced by osmotic priming or SMP treatment of carrot and onion seeds.

  • Osmotic and solid matrix priming treatments enhanced germination performance. We compared osmotic with solid matrix priming to determine the more effective treatment for improving seed germination in pepper and tomato. Seed hydration was immediately observed after osmotic priming and solid matrix priming treatment. The moisture content of solid matrix primed seeds was lower than that of osmotic primed seeds in the two vegetable crops. Osmotic priming and solid matrix priming did not increased percent germination, but showed shorter number of days to 50% of the final germination percentage ($T_{50}$) compared with untreated seeds, regardless of germination temperature. $T_{50}$ value was reduced in osmotic or solid matrix primed pepper seeds about 6.0, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. While, that in tomato seeds was reduced about 3.3, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. The effectiveness of osmotic priming or solid matrix priming in reducing the $T_{50}$ was greater when the seeds were germinated at $15^{\circ}C$ than at temperature of higher than $20^{\circ}C$. Solid matrix primed seeds germinated faster than osmotic primed seeds at all temperature in pepper. However, there was no significant difference on the percentage germination between solid matrix and osmotic primed seeds in tomato. After priming, dried-bark seeds showed faster germination than surface-dried seeds in pepper. On the other hand, there was no significant difference in tomato. Emergence of pepper and tomato seeds was markedly enhanced by osmotic priming or SMP treatment although the final emergence percentage was not significantly influenced. On the other hand, early growth was not significantly influenced by osmotic priming or SMP treatment of pepper and tomato seeds.

SMP Treatment of Tobacco Seeds to Improved Stand Establishment (입묘율 향상을 위한 담배종자의 SMP 처리)

  • The purposes of this study were to examine effect by Solid Matrix Priming(SMP) ; method far enhanced rapid and uniform seedling emergence in tobacco seeds. The results were as follows. No enhancement of germination rate was by SMP treatment. In KF114 during 9 days treatment and in KB108 during 7 days treatment, germinative energy was highest, and T50 and mean germination time were the shortest. Cells in embryo and hypocotyl were divided and extended during SMP treatment. The contents of fatty acid were increased in SMP treatment. The changes of glucose content were little.

Effect of Prepriming on Improving Germinability of Pelleted Carrot Seeds (Priming에 의한 당근 Pelleting 종자의 발아성 향상)

  • Pelleting the seeds often reduces seed germinability. In an effort to alleviate this problem, the seeds of four carrot cultivars were, before pelleting, either solid-matrix primed (SMP) with Micro Cel-E or osmotically-primed with polyehtylene glycol 8000. Some batches were imbibed in 100-ppm $GA_3$ solution. In laboratory tests, pretreatment of the seeds increased the precentages of germination by as much as 50% of the controls, indicating the increase of the germination speed by 1.9 to 4.6 days as determined by the $T_{50}$ values. In field tests conducted in the year of 2000 and 2001, however, effects of SMP were not observed. There was no significant difference between the SMP seeds and the controls of the percentages and $T_{50}$ values. The emergence of the seedling in the field was affected greatly by soil water concentration.

Germination Promotion of Watermelon Seeds Using Solid Matrix Priming(SMP) Treatment (SMP 처리에 의한 수박종자의 발아촉진)

  • Early emergence of watermelon seeds was markedly enhanced by SMP treatment though the final emergence percentage was not significantly influenced. Mixing ratios of 10:1:5 and 10:1:6 (seed : Micro-Cel E : water by weight) were found to be most effective in promoting the early emergence of watermelon seeds primed for 72 hrs at $25^{\circ}C$. Even though the hypocotyl elongation was slightly promoted by SMP treatment, the hypocotyl diameter remained uninfluenced. Among the cultivars tested, 'House-Ilpum' and 'Surowang' showed the best response to the SMP treatment.

Solid Matrix Priming with Hydrogels on Heteropappus arenarius Seeds

  • This study was initiated to evaluate hydrophilic polymers (hydrogels) as a new solid matrix medium for seed-priming of Heteropappus arenarius Kitam. Solid matrix priming (SMP)-media were prepared with the combination of Na- and K-based hydrogels and hydrogels with three different dry levels (DC; 70%, 80%, and 90%). Priming was performed in the dark at 15 or $20^{\circ}C$ for 24 hours, and all primed seeds were incubated at $20^{\circ}C$ in the dark for the germination test. Non-primed seeds and seeds primed with distilled water (DW) were also included. To reach the germination rate of 50% ($T_{50}$), it took 4.0 days for non-primed seeds, and 3.6 and 3.9 days for DW-primed seeds at 15 and $20^{\circ}C$, respectively. Na-based hydrogel-primed seeds with 70% DC (Na 70%) showed the fastest germination, which respectively took and 1.9 and 1.8 days at 15 and $20^{\circ}C$ to $T_{50}$. K-based hydrogel-primed seeds with 70% DC showed the fastest germination among K-based hydrogels with various DC levels, but it took 0.6 days more to $T_{50}$ compared to Na 70%. The hydration rate (HR) of DW-primed seeds was 37% lower than that of Na 70%-primed seeds at $15^{\circ}C$ priming temperature, which indicates that Na 70% priming is the best solid matrix priming condition for promoting the germination of H. arenarius seeds.

  • The purpose of this study is to select the appropriate herbicide, which is needed to control the bentgrass in kentucky bluegrass(Poa pratensis) fairway effectively. The best control of bentgrass was observed at /$0.3mL\m^2$ of dithiopyr applied on Kentucky bluegrass. A field experiment was conducted to determine the priming effects on seed germination of Kentucky bluegrass. Ground covering rates by turfgrass were investigated under field conditions for 70 treatments by thirty days after treatment. The solid matrix priming(SMP) treatments on Kentucky bluegrass reduced the number of days requited for emergence and improved final cover ratings. Primed seeds of Kentucky bluegrass (incubated for five days at $20^{\circ}C$) resulted in most rapid germination and covering rate.

Conditions for Solid Matrix Priming of Carrot Seeds and Physiological Changes in the Seed During the Treatment (당근 종자의 Solid Matrix Priming 적정 조건구명과 처리중 종자의 생리적 변화)

  • To improve germinability of carrot seeds of 'Inari', 'Mansan', 'Mussang', and 'Hongsim' cultivars, various levels of solid matrix priming (SMP) and the physiological changes of seeds associated with SMP were examined. As the ratio of water in a seed-Micro Cel E-water mix and priming duration increased, seed germinability enhanced. However, there was radicle protrusion during SMP in some cultivars, indicating the existence of cultivar different variations of the optimum conditions. The optimum ratio for a seed-Micro Cel E-water mix was 5 : 3 : 10.5. In this mix, the optimum durations for the treatment were 3 days for 'Mansan' and 5 days for other cultivars. SMP at $20^{\circ}C$ to $25^{\circ}C$ resulted in a higher germination percentage and a shorter $T_{50}$. Seeds absorbed water quickly within an hour and the percentage of moisture was stable at 43 to 49 after 24 hours of the treatment. Conductivity of the mix declined for the first 6 hours to $100{\mu}S{\cdot}cm^{-1}$ with little changes thereafter. On the other hand, the pH of the mixed increased slightly during the early phase, and then returned to the initial level as the treatment proceeded.

The Water Content and Treatment Period of SMP for Enhanced Seedling Emergence in Tobacco Seeds. (담배 종자의 입묘율 향상을 위한 SMP 수분함량과 처리기간)

  • The purposes of this study are to investigate water content and treatment period in Solid Matrix Priming(SMP; method for enhanced rapid and uniform seedling emergence in tobacco seeds). The higher water content made the faster imbibition speed of tobacco seed. But the radicle emergence shorten the period of water equilibrium. The range of water content in SMP treatment within the limits of no radicle emergence of tobacco seed was 28∼32% to dry weight of tobacco seed. The rate of radicle emergence and seedling emergence was higher in water content 30% and 40% treatment plot than that in 50% and 60% treatment plot. But it was not different between SMP treatment and non-SMP treatment. In water content 40% during 9 days treatment, germinative energy was highest, and T50 and mean germination time(MGT) were the shortest. The germination speed was higher in SMP treatment than that in non-SMP treatment. It was not different on the germination rate between non-SMP treatment and SMP treatment.

Changes of Fatty Acid during Germination by Seed Pretreatment, SMP, in Tobacco (SMP종자 전처리에 의한 담배 종자 발아과정의 지방산 변화)

  • We studied the effect of SMP (solid matrix priming) treatment, seed pretreatment for germination enhancement, to tobacco seeds by measurement of germination rate and contents of fatty acid, energy source for tobacco germination. The results were as follows. In result of germination test, germinative ability was the highest in KF109 at nine day treatment and in KB108 at seven day treatment. The composition of fatty acid in tobacco seed confirmed by gas chromatography were palmitic acid, stearic acid, oleic acid+elaidic acid, linoleic acid+linole­laidic acid, and $\alpha-linolenic$ acid. Palmitic acid, stearic acid, oleic acid and elaidic acid, and linoleic acid and linole-laidic acid were highest in KF109 at eight or nine day treatment and in KB108 at six or seven day treatment. Especially, content of oleic acid + elaidic acid, and linoleic acid+linolelaidic acid were changed largely by treatment, so these might be used for index to examine treatment effect.
