Objective : Solar ultraviolet (UV) light can cause photoaging of human skin. Many researchers have focused on effective prevention to mitigate the aging process. This study was aimed to investigate anti-wrinkle effects of pomegranate concentration solution (PCS) using photoaged hairless mouse model. Method : To clarify the protective effect of PCS against UVB-induced photo-damage, water capacity was measured with transepidermal water loss (TEWL) in SKH-1 hairless mice. In addition, image of skin replicas was analyzed. Results : Our results showed that the PCS treatment protects skin against UVB-induced photoaging, based on the parameters including moisture capacity and TEWL. According to visual assessment of skin replica, application of 1 ml/kg PCS inhibited UV-induced wrinkle formation in mice skin as compared to the vehicle-applied control mice. Conclusion : Taken together, these results indicated that PCS could improve skin wrinkle formation induced by UVB irradiation in SHK-1 hairless mice. PCS could be applied as an anti-wrinkle agent.

Chronic exposure of solar ultraviolet (UV) light to human skin results in photoaging with wrinkle formation. This study was performed to investigate anti-wrinkle effects of Lindera obtusiloba extract (LO) on UVB-induced wrinkle formation. We first measured cell proliferation and type I pN collagen synthesis activities in normal human dermal fibroblasts. Cell proliferation and type I pN collagen synthesis were increased by 33.8 % and 91.8 %, respectively, compared with no treatment control. SKH-1 hairless mice were topically applied 5 % LO solution for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, application of 5 % LO solution inhibited UV-induced wrinkle formation in mouse skin as compared to the vehicle-applied control mice. These results indicated that LO could protect skin wrinkle formation caused by chronic photo-irradiation in hairless mice.
DNA samples extracted from Japanese cypress leaf tissues contain isopropyl alcohol-precipitable, high molecular weight compounds, which interfere ELISA for cyclobutane pyrimidine dimers (CPD). Removal of the compounds is achieved by DEAE ion-exchange column chromatography and improves the ELISA responses of the DNA. When extracting DNA repeatedly from the same leaf tissues, the DNA samples show CPD responses which increase with the order in sequential extraction, and hence for a reliable detennination of DNA lesion a thorough extraction of DNA is required. Clearing these two problems it was demonstrated that CPD level was slightly higher in the leaves of trees growing under full sunlight than in those growing under UBV -cut sunlight.
The dosal sites of 61 Koreans were exposed to increasing UVB doses in the 10mJ/$\textrm{cm}^2$ interval from 10mJ/$\textrm{cm}^2$ to 100mJ/$\textrm{cm}^2$ by solar-simulator, and at the 24hr after UV-B exposure we determined the first MED (Minimal Erythema Dose). For the precise measurements, we peformed the second exposures arround the first MED with 2.5mJ/$\textrm{cm}^2$ interval. The mean MED of all volunteers was 39.6 $\pm$ 15.7mJ/$\textrm{cm}^2$. As the distinction of sex, the means of male and female were 46.9 $\pm$ 18.1mJ/$\textrm{cm}^2$ and 31.5$\pm$8.5mJ/$\textrm{cm}^2$ respectively. According to the classification of skin type by Greiter. F.(1), the percent of skin type I to Ⅵ were 10.9%, 36.4%, 34.6%, 12.7%, 3.6%, and 1.8% respectively.
In recent years, titanium dioxide powders were produced as nanoparticles or nanowires or nanotubes in one-dimensional structure, and mesoporous spheres in 3-D to improve their crystallinities, which were further used as photo-electrode materials and for preventing photo-aging. In this study, a nano sol composed of titanium dioxide exhibiting a high crystallinity was synthesized using n-alcohol as a solvent. The crystallinity of the nano sol was confirmed by FE-SEM, and XRD, while the UV blocking rate confirmed by UV-Vis spectrometry results. Changes in the crystallinity were investigated by varying the types of solvents such as butanol, propanol and ethanol. The synthesized particle sizes were from 200 to 250 nm, and the optical transmittance showed a high blocking rate in the UVB and UVA range. It is expected that a high transmittance at 550 nm wavelength can increase the photoelectric conversion efficiency of solar cells and the UV blocking efficiency.
The skin exposure to solar irradiation and photoreactive xenobiotics may produce abnormal skin reaction, phototoxicity. Phototoxicity is an acute light-induced response, which occurs when photoreacive chemicals are activated by solar lights and transformed into products cytotoxic against the skin cells. Multifarious symptoms of phototoxicity are identified, skin irritation, erythema, pruritis, and edema that are similar to those of the exaggerated sunburn. Diverse organic chemicals, especially drugs, are known to induce phototoxicity, which is probably from the common possession of UV-absorbing benzene or heterocyclic rings in their molecular structures. Both UVB (290~320 nm) and UVA (320~400 nm) are responsible for the manifestation of phototoxicity. Absorption of photons and absorbed energy (hv) by photoactive chemicals results in molecular changes or generates reactive oxygen species and depending on the way how endogenous molecules are affected by phototoxicants, mechanisms of phototoxcity is categorized into two modes of action: Direct when unstable species from excited state directly react with the endogenous molecules, and indirect when endogeneous molecules react with secondary photoproducts. In order to identify phototoxic potential of a chemical, various test methods have been introduced. Focus is given to animal alternative test methods, i.e., in vitro, and in chemico assays as well as in vivo. 3T3 neutral red uptake assay, erythrocyte photohemolysis test, and phototoxicity test using human 3-dimensional (3D) epidermis model are examples of in vitro assays. In chemico methods evaluate the generation of reactive oxygen species or DNA strand break activity employing plasmid for chemicals, or drugs with phototoxic potential.

Recently the harmfulness of W radiation is in creasing due to encironmental pollution. Environmental population may also play a role in global decrease of ozone layer, A major consequence of ozone depletion is increase in solar ultra violet radiation received at the earth's surface excessive exposure to W radiation cause a lot of problems in our skin. Plant extract that possess antioxidative activities has been reported to retard the oxidation process in product to which they have been added. Plant are alived under solar light. So it is expect the plants have so many protection mechanisms and UV absorbent ingredients against ultra violet radiation such as UVB, UVA. Plant extract which were flavonoids, alkaloids and others could be transformed into UV absorber by chemical modification. Therefore with the aim of finding alternative natural absorber that can safely be used in cosmetic, we have screened various extract for their UV absorbent effect. Thus, the cosmetic safety against human skin, antimicrobial effects and others could be improved by using the silicon.

Chronic exposure to solar radiation, particularly ultraviolet (UV) light, causes a variety of adverse reactions on human skin, such as sunburn, photoaging and photocarcinogenesis. Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental facts play critical roles in cellular damage. And, repeated-UV irradiation activated the expression of the matrix metalloproteinase (MMP) and induced skin irritation. Therefore, the development of effective and safe photoprotectants that can reduce and improve the skin damage has been required. The purpose of this study was to investigate the photo-protective effect of several chinese medical plants (Juniperus chinensis) on the UV -induced skin cell damages. We tested free radical and superoxide scavenging effect in vitro. Fluorometric assays of the proteolytic activities of MMP-1 (collagenase) were performed using fluorescent collagen substrates. UVA induced MMP-1 synthesis and activity were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in skin fibroblasts. We also examined anti-inflammatory effects by the determination test of proinflammatory cytokine, interleukin 6 in HaCaT keratinocytes. Expression of prostaglandin E$_2$ (PGE$_2$) after UVB irradiation was measured by competitive enzyme immunoassay(EIA) using PGE$_2$ monoclonal antibody. In the human skin we tested anti-irritation effect on the SLS-induced damage skin after appling the extract containing emulsion. We found that Juniperus chinensis extract had potent radical scavenging effect by 98% at 100$\mu\textrm{g}$/mL. The extract of Juniperus chinensis showed strong inhibitory effect on MMP-1 activities by 97% at 100 $\mu\textrm{g}$/mL and suppressed the UVA induced expression of MMP-1 by 79% at 25$\mu\textrm{g}$/mL. This extract also showed strong inhibition on MMP-2 activity in UVA irradiated fibroblast by zymography. In the test of proinflammatory cytokines of human keratinocytes Juniperus chinensis extract decreased expression of interleukin 6 about 30%. The amount of PGE$_2$ by HaCaT keratinocytes was significantly increased at the doses of above 10 mJ/$\textrm{cm}^2$ of UVB (p < 0.05). At the concentrations of 3.2-25$\mu\textrm{g}$/mL of this extract, the production of PGE$_2$ by HaCaT keratinocytes (24 h after 10mJ/$\textrm{cm}^2$ UVB irradiation) was significantly inhibited in culture supernatants (p < 0.05). In SLS-induced skin irritation model in vivo, we found to reduce skin erythema and improve barrier recovery after appling Juniperus chinensis extract containing emulsion when compared to irritated non-treated and placebo-treated skin. Our results suggest that Juniperus chinensis extract can be effectively used for the prevention of UV and SLS-induced adverse skin reactions and applied as anti-aging and anti-irritation cosmetics.

Sunscreens have been advocated as an important means of preventing skin cancer. UV-induced immunosuppression is important for skin cancer development, yet the effectiveness of sunscreens in protecting the human immune system from UV radiation is unclear. The only currently accepted method of sunscreen rating is the Sun Protection Factor (SPF) based on prevention of erythema. We developed an in vivo non-invasive method for evaluating protection of the human immune system from UV radiation based on recall contact sensitivity to nickel, a common allergen. Using this system we showed that broad-spectrum sunscreens provide greater protection to the immune system than sunscreens which protect from UVB only. UVA was found to be immunosuppressive. We developed this technique to enable the study of solar simulated UV radiation dose responses and determined Immune Protection Factors (IPFs) for six commercially available sunscreens based on limits of protection from the dose response data. We found that the IPF did not correlate with the SPF and that protection from erythema therefore cannot be used to predict protection of the immune system. However, IPF was significantly correlated to the UVA protective capability of the sunscreens, indicating that sunscreen protection from UVA is important for prevention of immunosuppression. We recommend that sunscreens should be rated against their immune protective capability to provide a better indication of their ability to protect against skin cancer.

Skin is exposed to sunlight or artificial indoor light on a daily. The reached solar light on the earth surface consist of 50% visible light and 45% infrared (IR) except for ultra violet (UV). The negative effects of UV including UVB and UVA have been steadily investigated within the last decades. However, little is known about the effects of visible or IR light. In this study, we irradiated human dermal fibroblasts using light emitting diode (LED) to investigate the optimal parameter for enhancing cell growth and collagen synthesis. We found that red of 630 nm and green of 520 nm enhance the cell proliferation, but irradiation with purple and blue light exerts toxic effects. To examine the response of irradiation time and light intensity on the fibroblasts, cells were exposed to red or green light with intensities from 0.05 to $0.75mW/cm^2$. Procollagen secretion was increased of 1.4 fold by 10 min irradiation, while 30 min treatment decreased the collagen synthesis of dermal fibroblasts. Treatment with red of $0.3mW/cm^2$ and green of 0.15 and $0.3mW/cm^2$ resulted in enhancement of collagen mRNA. Lastly, we investigated the combinatorial effect of red and green light on dermal fibroblasts. The sequential irradiation of red and green light is an efficient way for the purpose of the increase in the number of fibroblasts than single light treatment. On the other hand, the exposure of red light alone was more effective method for enhancing of collagen secretion. Our study showed that specific light parameters accelerated cell proliferation, gene expression and collagen secretion on human dermal fibroblasts. In conclusion, we demonstrate that light exposure with specific parameter has beneficial effects on the function of dermal fibroblasts, and suggests the possibility of its cosmetically and clinical application.
