Bacillus subtilis JW-1 was isolated from rhizosphere soil as a potential biocontrol agent of bacterial wilt caused by Ralstonia solanacearum. Seed treatment followed by a soil drench application with this strain resulted in >80% reduction in bacterial wilt disease compared with that in the untreated control under greenhouse conditions. The antibacterial compound produced by strain JW-1 was purified by bioactivity-guided fractionation. Based on mass spectroscopy and nuclear magnetic resonance spectral data ($^1H$, $^{13}C$, $^1H-^1H$ correlation spectroscopies, rotating frame nuclear Overhauser effect spectroscopy, and heteronuclear multiple-bond correlation spectroscopy), the structure of this compound was elucidated as a cyclic lipopeptide composed of a heptapeptide (Gln-Leu-Leu-Val-Asp-Leu-Leu) bonded to a ${\beta}$-hydroxy-iso-hexadecanoic acid arranged in a lactone ring system.
Actinomycetes CS0707 has been isolated in soil sample from location in the Jeju province. Korea, and produces thermostable extracellular proteases. Actinomycetes CS0703 showed the highest protease activity at late exponential phase when grown in OSYM medium (oatmeal 2.0%, soybean meal 1 %, dried yeast 1 %, mannitol 1 %) at $48^{\circ}C$. Three forms of protease(Ta-1, TA-2 and Ta-3) were fractionated by Ultrogel AcA 54 column chromatography, and further purified through ammonium sulfate fractionation, ultramembrane filtration, and DEAE-sepharose CL-6B column chromatography. Organic farming (OF) is regarded as alternative farming types against general farming system for sustainable agriculture, recently. However, there is little information on effects of OF on soil properties and watershed condition. In order to determine the effects of OF on soil properties, 36, 10 and 8 sites of organic farming (OF) fields of plastic film houses, paddy and orchard were selected in the national scale, respectively, to evaluate their chemical properties and phosphorus distributing characteristics. The average organic matter (OM) contents in organic farming fields were with $44g\;kg^{-1}$ in plastic film houses, $26g\;kg^{-1}$ in paddies and $39g\;kg^{-1}$ in orchard soils higher than the average OM contents in conventional farming (CF) soils. Available phosphates were accumulated to 986 in plastic film house soils and $754mg\;kg^{-1}$ in orchard soils, respectively, over the optimum range. Furthermore, total P (T-P) reached to $2.973mg\;kg^{-1}$ in plastic film houses and $2303mg\;kg^{-1}$ in orchards in OF soils. It could be attained by applying repeatedly low N/P ratio of manure-based compost. In two types of soils inorganic P was dominant with the ratio of 62~80% of T-P, and then residual and organic Ps followed. However. residual-P was dominant in paddy soils with the rate of 50% of T-P. Fractionation of soil extractable P showed that Ca-P was dominant with about $1,330mg\;kg^{-1}$ in upland soils in OF fields, which is affected by high soil pH of over 6.0. However. Fe-P of extractable P was dominant in paddy soils. Water-soluble P was very high with 65 and $26mg\;kg^{-1}$ in plastic film house and orchard soils in OF. From this results. OF regarded as an environment-friendly farming system may cause serious soil deterioration by accumulated phosphorus and may also cause water pollution.
Beryllium (Be), cobalt (Co), thallium (Tl) and vanadium (V) are candidates of 21 priority soil pollutants in Korea. The distribution of their concentration in soils from three contamination sources including industrial, roadside and mining areas was investigated. Concentrations of the metals were evaluated quantitatively using pollution indices and the fractionation of metals was conducted using modified SM&T (Standards Measurements and Testing programme) sequential extraction. Concentrations of the metals for all samples from industrial and roadside soils were within the range of natural background levels, while some of Be in soils from abandoned mines exceeded that the range. Enrichment Factor (EF) and Nemerow Integrated Pollution Index (NIPI) for Be, Co, Tl and V showed that there are effects or possibilities of anthropogenic activities. Pollution Load Index (PLI) analyses indicated all investigated sites needed further monitoring. The results of sequential extractions indicated mobile fractions (F1+F2) of Be, Tl and V were below 30% except some of Co in soil, which implies their low mobility to neighboring environment media. Variable tools like sequential extraction, comparison with background/actual concentration and pollution indices, as well as aqua regia extraction should be considered when evaluating Be, Co, Tl, V in soil.
The molecular size distribution and fluorescence properties of soil fulvic acids (FA) were characterized by using gel filtration chromatography (GFC) and luminescence spectroscopy. The objectives of this work were to fractionate the FA extracted from a forest soil into different nominal molecular size using GFC system and to characterize the fluorescence properties (excitation, emission and synchronous) of these fractions using luminescence spectrometer. The GFC column was calibrated with polyethylene glycols, acetone and dextrane Blue. The total permeation volume of the GFC system was 404 mL and the void volume 130 mL. The GFC molecular weight of the soil FA was in the range of 190~8,900 Dalton and the molecular weight at the peak on the chromatogram was 930 Dalton. The fluorescence intensity ratio ($I_{498nm}/I_{390nm}$) was found to be increased with an increasing molecular weight. This results may suggest that the fulvic acid fractions with high molecular weight have large amount of the condensed aromatic compound.
In order to compare phosphorus (P) behavior of livestock manure compost (LMC) and chemical fertilizer (CF) applied to the sea-reclaimed land soils, incubation experiments were conducted for five weeks. Four soils differing textural classes, sandy loam and clay loam, and electric conductivity (EC) value, high and low, were applied with CF and LMC. LMC was applied at the level of 0, 1, 2, and 3% on the soil weight basis and CF was applied at the same levels of P as LMC. The results showed that increase of P application rate of CF and LMC led to linearly increase available $P_2O_5$ and $0.01M\;CaCl_2$ extractable P contents regardless of soil texture and EC. However, 0.01M $CaCl_2$ extractable P from soil applied with CF was significantly higher than with LMC. Correlation analysis between $0.01M-CaCl_2$ extractable P and fractionated P by different extraction methods showed that $0.01M-CaCl_2$ extractable P positively correlated with KCl-P (soluble and exchangeable P) and HCl-P (Ca and Mg bound P). However, NaOH-P (Fe and Al bound P and organic P) and residual P was adverse. The amount of NaOH-P significantly influenced to the amount of 0.01M $CaCl_2$ extractable P of CF and LMC in the soils. The application of LMC at sandy loam soil could be carried out in the consideration of nutrient leaching and crop uptake.
Soil organic carbon (SOC) in the Arctic is vulnerable to climate change. However, research on SOC stored in the high Arctic regions is currently very limited. Thus, this study was aimed at understanding the distribution and characteristics of SOC with respect to geomorphology and vegetation in Svalbard. In August 2011, soil samples were collected near the Vestre Lov$\acute{e}$nbreen moraine. Sampling sites were chosen according to altitude (High, Mid, and Low) and differences in levels of vegetation establishment. Vegetation coverage, aboveground biomass, and SOC contents were measured, and density-size fractionation of SOC was conducted. The SOC content was the highest in the Mid site ($126.9mg\;g^{-1}$) and the lowest in the High site ($32.1mg\;g^{-1}$), although aboveground biomass and vegetation coverage were not different between these two sites. The low SOC content measured at the High site could be related to a slower soil development following glacial retreat. On the other hand, the Low site contained a high amount of SOC despite having low vegetative cover and a high ratio of sand particles. These incompatible relationships between SOC and vegetation in the Low site might be associated with past site disturbances such as runoff from snow/glacier melting. This study showed that geomorphological features combined with glacier retreat or melting snow/glacier effects could have affected the SOC distribution and vegetation establishment in the high Arctic.

Micellar enhanced ultrafiltration (MEUF) is a surfactant-based separation process and it can remove heavy metal ions from aqueous stream effectively. However, it is necessary to recover and reuse surfactants for economic feasibility because surfactant is expensive. Foam fractionation was investigated for both anionic and cationic surfactant recovery. Chelating agent such as ethylenediaminetetraacetic acid (EDTA) was studied for the separation of heavy metals from surfactant solution. Anionic surfactants bound with heavy metals can be recovered by lowering pH (acidification). In this study, citric acid and imminodiacetic acid (IDA) were applied to release copper from sodium dodecyl sulfate (SDS) micellar solution and compared with EDTA. Precipitation of copper by ferricynide and sodium sulfide were also investigated. As a result, ca. 100 % of copper was released from SDS micellar solution by 5 mM of EDTA and citric acid. And 3.3 mM of ferricyanide formed precipitate with 82.7 % of copper. 5 mM of IDA and sodium sulfide released or formed precipitate 82.5 % and 58.9 % of copper, respectively. Citric acid is harmless to environments and ferricyanide precipitates with Cu easily. Therefore, it is considered that citric acid and ferricyanide have competiveness over a famous chelating agent, EDTA, for the separation of Cu from SDS solution.
